Prevalence of Lyme borreliosis in canine sentinels has been shown to correlate with infection in humans. One thousand canine sera (917 dogs, 83 coyotes) obtained from animal control authorities and area veterinarians were screened by ELISA for antibodies to Borrelia burgdorferi. Results were validated by Western blot and indirect fluorescent antibody (IFA) tests at referee laboratories. Criterion for a positive Western blot was presence of 5 of 10 of the most common antigen IgG bands; for IFA, >1:128 or the equivalent when correcting for interlaboratory variability. Twenty-two of 1,000 canines were confirmed serologically positive (21 dogs and 1 coyote; seroprevalence 2.3% and 1.2%, respectively). Lifestyle, breed size, gender, and age were not statistically predictive of seropositive status. No regional clustering of seropositive animals was detected. The low prevalence of seropositivity in sentinel canines suggests the Lyme borreliosis hazard in San Diego County is minimal.  相似文献

The prevalence of serum antibodies against canine parvovirus-2 (CPV-2), as measured by a standard hemagglutination-inhibition test, was determined in serum samples collected from 1,184 coyotes (Canis latrans) in Texas, Utah, and Idaho from 1972 to 1983. No evidence of parvoviral infection was found before 1979, after which seroprevalence rapidly increased to greater than 70% at all sites by 1982. There were minor differences in prevalence between age groups and among sites, but no consistent differences between sexes. The onset of CPV-2 seroprevalence in free-ranging coyotes coincided remarkably with the recognition of the clinical disease and seroprevalence in domestic dogs in the United States.  相似文献

The objectives of this study were (1) to evaluate the performance and agreement of serological assays (ELISA, IFAT, Neospora caninum agglutination test and immunoblot) using reference sera and field sera from foxes and coyotes and (2) to estimate the N. caninum seroprevalence in foxes and coyotes on Prince Edward Island, Canada. With fox and coyote reference sera the test performance of the ELISA, IFAT and IB was excellent (100% sensitivity and specificity). NAT showed a low sensitivity (50%). Serum was collected from 201 coyotes and 271 foxes. The seroprevalence observed in the different assays ranged from 0.5 to 14.0% in coyotes and 1.1 to 34.8% in foxes. The seroprevalence, when taking more than one test positive as cut-off value was 3.3 and 1.1% for coyotes and foxes, respectively. From the N. caninum-positive group, all coyotes were older than 3 years. Agreement among assays (measured as prevalence-adjusted bias-adjusted kappa) using the field sera ranged from 0.17 to 0.97. Best agreement was observed between ELISA and IFAT, poor agreement was observed between NAT and the other assays. Positive agreement was moderate to poor among all assays utilized in this study. Although the seroprevalence observed was low, N. caninum antibodies are present in foxes and coyotes on Prince Edward Island (PEI) and their role in the N. caninum epidemiology needs further study.  相似文献

OBJECTIVE: To estimate the prevalence of paratuberculosis in purebred beef cattle in Texas and identify risk factors for seropositivity. DESIGN: Epidemiologic survey. ANIMALS: 4,579 purebred cattle from 115 beef ranches in Texas. PROCEDURE: Blood was collected, and serum was analyzed for antibodies with a commercial ELISA. Fecal samples were collected and frozen at -80 degrees C until results of the ELISA were obtained, and feces from seropositive cattle were submitted for mycobacterial culture. Herd owners completed a survey form on management factors. RESULTS: Results of the ELISA were positive for 137 of the 4,579 (3.0%) cattle, and 50 of the 115 (43.8%) herds had at least 1 seropositive animal. Results of mycobacterial culture were positive for 10 of the 137 (7.3%) seropositive cattle, and 9 of the 50 (18%) seropositive herds had at least 1 animal for which results of mycobacterial culture were positive. Risk factors for seropositivity included water source, use of dairy-type nurse cows, previous clinical signs of paratuberculosis, species of cattle (Bos taurus vs Bos indicus), and location. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that seroprevalence of paratuberculosis among purebred beef cattle in Texas may be greater than seroprevalence among beef cattle in the United States as a whole; however, this difference could be attributable to breed or regional differences in infection rates or interference by cross-reacting organisms. Veterinarians should be aware of risk factors for paratuberculosis as well as the possibility that unexpected serologic results may be found in some herds.  相似文献

We determined the prevalence and mean intensity of Trichinella sp. infection in coyotes from six counties in Oklahoma and one in northern Texas. Tongues from 77 coyotes were examined using histology and artificial tissue digestion. Histological examination showed a prevalence of 3.9% (3 of 77) whereas the prevalence was 6.5% (5 of 77) based on artificial digestion of 5.0 g of muscle from coyote tongues. One sample was positive for Trichinella sp. on histology but negative by artificial digestion. Combining data from both diagnostic techniques showed that six of 77 (7.8%) coyotes were infected with Trichinella spp. The mean intensity of Trichinella sp. larvae ranged from 0.2 to 66.2 with an average of 16.0 larvae per gram (LPG) of tongue. Genotyping results demonstrated that the coyotes were infected with Trichinella murrelli. This is the first report of T. murrelli infection in coyotes in Oklahoma. T. murrelli had previously been isolated from coyotes in Texas.  相似文献

Serum antibody titers to canine parvovirus (CPV), canine adenovirus-1 (CAV-1), and canine distemper virus (CDV) were measured in dogs with known immunization status. The dogs represented 3 groups: nonvaccinated dogs less than 12 months old; vaccinated dogs less than 12 months old; and adult dogs greater than 12 months old. For practical reasons, the population from which the specimens were obtained could be considered as free from natural infection with CAV-1 and CDV. In nonvaccinated dogs less than 12 months old, antibodies against all 3 viruses were measured at the time the dogs were given their first vaccination. Altogether, 50.7% of the dogs had titer greater than or equal to 1:10 to CPV, and 26.1 and 46.2% had titer greater than or equal to 1:8 to CAV-1 and CDV, respectively. The concentration of maternal antibody seemed to be of major importance for failure of immunization with use of inactivated CPV vaccine, but not with CAV-1 and CDV vaccination. In dogs less than 12 months old and vaccinated against CPV infection with inactivated virus, only 11.5% had titer greater than or equal to 1:80. In dogs vaccinated against infectious canine hepatitis and canine distemper, 63.2 and 78.3%, respectively, had titer greater than or equal to 1:16. In adult dogs greater than 2 months old and vaccinated against CPV infection, less than 50% had titer greater than or equal to 1:80, regardless of time after vaccination. The seroprevalence and seasonal trend of antibody titers against equine monocytic ehrlichiosis (Potomac horse fever) were determined in apparently healthy horses in selected areas of Illinois in 1986. Sera from 1,367 horses (6 months to 29 years old) were evaluated for the presence of antibodies against Ehrlichia risticii with indirect immunofluorescence. The majority (88%) of the horses were Thoroughbred or Standardbred racehorses. The number of horses with antibodies against E risticii was 229/1,367 (16.75%). The titers in these horses ranged from 1:10 to 1:640. As the year progressed, the number of seropositive horses (titers greater than or equal to 1:10) and the magnitude of the titers increased significantly, both reaching a maximum in July and August, respectively (P less than 0.05). A relationship between seropositivity and gender was not detected. In the year prior to sampling, 56.8% of the seropositive horses had not been ill, whereas 0.8% had diarrhea, an episode of acute abdominal pain, or laminitis. It was concluded that a large number of horses in Illinois are exposed to E risticii, that maximal exposure occurs in July, and that the most common form of the disease in Illinois is not associated with clinical signs.  相似文献

Goats are important domestic animals in the south of Thailand due to the minimal cost of rearing and maintaining them, and their production of both meat and milk. Toxoplasmosis is one of the most threatening parasitic zoonoses and the causative agent Toxoplasma gondii uses a wide range of warm-blooded intermediate hosts including the goat. The objective of this study was to assess the seroprevalence of antibodies to T. gondii in goats of Satun Province in Thailand. A total of 631 goat sera were examined for antibodies against toxoplasmosis with commercial latex agglutination test kits (Toxocheck-MT 'Eiken'). Of these, 176 (27.9%) were found to be positive to T. gondii; antibody titers ranged from 1:64 to 1:4096 (1:64 cut-off). Female goats were 1.73 times more likely than male to be seropositive (odds ratio [OR]=1.73; 95% confidential interval [CI]=1.11, 2.73). Dairy goats were more likely to be seropositive than meat goats (OR=1.36; 95% CI=0.84, 2.20). Goats were infected with T. gondii with acquisition of age because older goats were more likely to be seropositive than young goats under 1-year-old (for 1-2 years, OR=19.6; 95% CI=0.92, 4.15, for >2 years, OR=2.70; 95% CI=1.26, 5.80). The high seroprevalence of T. gondii antibodies found in the present study suggested widespread exposure of goats in Satun Province to T. gondii.  相似文献

The seroprevalence of Toxoplasma gondii, Dirofilaria immitis (heartworm), feline immunodeficiency virus (FIV) and feline leukemia virus (FeLV) infections was examined using serum or plasma samples from 746 pet cats collected between May and July 2009 from clinics and hospitals located in and around Bangkok, Thailand. The samples were tested for heartworm, FIV, and FeLV using a commercial ELISA. Of the 746 samples, 4.6% (34/746) were positive for heartworm antigen, 24.5% (183/746) had circulating FeLV antigen, and 20.1% (150/746) had antibodies against FIV. In addition, the first 348 submitted samples were tested for T. gondii antibodies using a modified agglutination test (MAT, cut off 1:25); 10.1% (35/348) were seropositive. Of the 348 cats sampled for all four pathogens, 11, 10, and 1 were positive for T. gondii antibodies and FIV antibodies, FeLV antigen, or D. immitis antigen, respectively. Of the 35 T. gondii-seropositive cats, 42.9% (15/35) were co-infected with at least one of the other three pathogens. The presence of antibodies to FIV was significantly associated with both age and gender, while FeLV antigen presence was only associated with age. In the case of FIV, males were twice as likely to be infected as females, and cats over 10 years of age were 13.5 times more likely to be infected than cats less than 1 year of age. FeLV antigen was more common in younger cats, with cats over 10 years of age being 10 times less likely to be FeLV positive than cats under 1 year of age. This is the first survey for these four pathogens affecting feline health in Thailand.  相似文献

OBJECTIVE: To examine sera obtained from dairy and beef cattle to detect antibodies against vesivirus and compare seroprevalence among cattle within the sample population. SAMPLE POPULATION: Cattle sera from 8 western states and Maryland submitted to the Washington Animal Disease Diagnostic Laboratory during 1999 and 2000. PROCEDURE: Sera were analyzed for vesivirus-specific antibodies by use of a recombinant vesivirus-San Miguel sea lion virus serotype 5-capsid peptide antigen in an indirect ELISA. RESULTS: Overall, 693 sera were tested and 105 (15.2%) had positive results. Seropositive cattle were from 7 states (all cattle from Montana and Maryland 10 and 4, respectively were seronegative). Overall seroprevalence for antivesivirus antibody in herds ranged between 0% and 80% (median, 14%). Higher antibody prevalence was significantly associated with older age, dairy rather than beef cattle, and reasons for submission. Logistic regression of factors (abortion, respiratory tract disease, and all other reasons for sample submission) revealed that older age and other reasons were independently associated with higher seroprevalence. Higher seropositive optical density values for the ELISA were observed among older cattle and cattle that aborted, compared with values for cattle with respiratory tract disease or other reasons for submission. CONCLUSIONS AND CLINICAL RELEVANCE: This laboratory-based surveillance sample provided a point estimate of seroprevalence against vesivirus among cattle in 9 US states. This suggests that vesivirus infection is widespread with high prevalence in some herds. Risk factors associated with vesivirus seroprevalence in beef and dairy cattle should be confirmed in population-based studies.  相似文献

OBJECTIVE: To estimate seroprevalence of antibodies against the serogroup of epizootic hemorrhagic disease viruses (EHDVs) and describe spatial distribution of antibodies against EHDV among cattle herds in Illinois and western Indiana. SAMPLE POPULATION: 9,414 serum samples collected from cattle in 60 herds over 3 transmission seasons. PROCEDURES: Serum samples were tested for antibodies against EHDV by use of an ELISA. Seroprevalence for 4 zones covering the length of Illinois and parts of Indiana were estimated. A multivariable mixed-effects logistic regression model with a random effect for herd was used to estimate seropositive risk for zone (1 through 4), age (yearling, adult), herd type (beef, dairy), transmission season (2000 to 2002), and zone by year interaction. Isopleth maps of seroprevalence at the herd level were produced. RESULTS: Antibodies against EHDV were detected in 1,110 (11.8%) samples. Estimated seroprevalence in 2000, 2001, and 2002 was 15.3%, 13.4%, and 5.2%, respectively. Seroprevalence was highest in the southernmost zone and lowest in the northernmost zone, but risk of seropositivity for EHDV among and within zones varied by year. Clusters of high seroprevalence in the south, low seroprevalence in the north, and outliers of high and low seroprevalence were detected. Risk mapping revealed areas of higher seroprevalence extending northward along the western and eastern ends of the study region. CONCLUSIONS: Seroprevalence of antibodies against EHDV in cattle was higher in the south than north; however, local complexities existed that were not observed in a serosurvey of antibodies against bluetongue virus from the same cattle population.  相似文献

The main aim of the present study was to establish the prevalence of antibodies against Neospora caninum dogs from the Czech Republic and to examine the dynamics of antibody titers during a long-term period. For this purpose, sera of 858 dogs were examined for the presence of anti-N. caninum antibodies using an indirect immunofluorescent antibody test (IFAT). Four groups of dogs of various origins were included in the survey: the first group (A, n=470) comprised dogs purchased by the Czech Army from the civilian sector throughout the Czech Republic, with 22 (4.7%) N. caninum-positive dogs, second group (B, n=115) represented police dogs with no seropositive animal, third group (C, n=195) were pet dog sera collected for veterinary clinic with 5 (2.6%) anti-N. caninum sera and the fourth group (D, n=78) of canine shelter dogs with the seroprevalence of 19.2%. The differences in seroprevalence were significant (P< or =0.01) between groups B and A, and between D and A. None of the serologically positive animals had clinical signs of neurological disorders. Coprological examination did not reveal any dog shedding N. caninum oocysts. The seropositivity rates for N. caninum were analyzed in relation to other data, such as age, breed and gender. Increased prevalence rates of anti-N. caninum antibodies were found in the older age strata of the dog population sample tested in the present study. We found significantly higher (P=0.02) prevalence in 3-3.5-year-old dogs (11.1% of 36), as compared to 1-1.5-years-old dogs (2% of 98). A longitudinal study of antibody dynamics was carried out in 19 initially seropositive dogs over a period of 4 years. The second and third examinations revealed that antibody titers decreased in majority of positive dogs (10, 52.6%), of which in seven cases (36.8%) the titers fell to levels that are currently considered as being seronegative (titer <1:50), or even became undetectable (titer <1:25).  相似文献

Captive coyotes (Canis latrans) were inoculated intragastrically (1 coyote), and intratracheally (6 coyotes) with Histoplasma capsulatum to simulate natural routes of exposure. In addition, the infectious organism was inoculated IV into another coyote and into a dog (C familiaris) to simulate hematogenous dissemination of H capsulatum. Isolation of the pathogen from various tissue specimens, using direct plating and mouse inoculation methods, provided mycologic evidence of infection. The presence of anti-Histoplasma serum antibodies also was monitored. Of the 6 coyotes challenged intratracheally, one developed a benign pulmonary form of histoplasmosis. This coyote developed a transient cough, and intracellular yeasts typical of H capsulatum were observed microscopically in pulmonary tissue specimens. The pathogen was isolated from specimens of pulmonary tissue and associated lymph nodes. The coyote that was fed Histoplasma-infected mice (intragastric challenge) did not develop clinical disease. The coyote and the dog inoculated IV developed a multifocal, nonsuppurative infection, without clinical evidence of histoplasmosis. Pathologic findings in the coyote and dog were similar; both animals had splenitis, lymphadenitis, and petechiation of the lungs. The liver and spleen were considered the primary extrapulmonary target organs on the basis of mycologic and histologic findings.  相似文献

OBJECTIVE: To estimate seroprevalence of bluetongue virus (BTV) and the geographic distribution of seropositive cattle herds in Illinois and western Indiana. SAMPLE POPULATION: 10,585 serum samples obtained from cattle in 60 herds during 3 transmission seasons (2000 through 2002). PROCEDURES: In a longitudinal study, serum samples were tested for BTV antibodies by use of a competitive ELISA. Four geographic zones were created by use of mean minimum January temperature. A multivariable mixed-effects logistic regression model with a random effect for herd was used to estimate seropositive risk for zone, age of cattle, herd type, and transmission season. RESULTS: Overall, BTV antibodies were detected in 156 (1.5%) samples. Estimated seroprevalence in 2000, 2001, and 2002 was 1.49%, 0.97%, and 2.18%, respectively. Risk of being seropositive for BTV was associated with geographic zone and age. Seroprevalence increased progressively from northern to southern zones, with no evidence of BTV infection in the northernmost zone. In the southernmost zone, annual seroprevalence ranged from 8.65% to 11.00%. Adult cattle were 2.35 times as likely as juvenile cattle to be seropositive. CONCLUSIONS AND CLINICAL RELEVANCE: Overall seroprevalence was lower than has been reported for Illinois cattle. Bluetongue virus antibodies were distributed heterogeneously in this region. Only in the southernmost zone was seroprevalence consistently > 2%. Regionalization of BTV risk based on state borders does not account for such variability. Serologic data could be combined with landscape, climate, and vector data to develop predictive models of BTV risk within transitional regions of the United States.  相似文献

Sera were collected using a systematic random sampling from 348 cattle herds in Ontario, in proportion to the cattle population in different areas. One cow in five from 296 dairy herds and one in three from 52 beef herds were sampled. The sera were analyzed for prevalence of antibodies to Leptospira interrogans serovar grippotyphosa, hardjo, icterohaemorhagiae and pomona using the microscopic agglutination test. Herd seroprevalence (one or more animals with titer greater than or equal to 80) in beef and dairy herds combined was grippotyphosa 2%, hardjo 13.8%, icterohaemorrhagiae 10.1% and pomona 25.8%; 39% of all herds showed evidence of leptospiral infection with one or more serovars; 44.2% of 52 beef herds had serological evidence of infection with serovar hardjo compared to 8.4% of 296 dairy herds (P less than 0.0001). Seroprevalence of other serovars was not significantly different between beef and dairy herds. The proportion of beef animals seropositive for hardjo and for pomona increased with age, particularly for hardjo; 26.5% of beef animals aged nine years or over were seropositive for hardjo. Dairy animals showed a significant rise of hardjo but not pomona titers with age. The seroprevalence of pomona infection was significantly higher in dairy cattle in eastern Ontario than in other regions. Thirty-four (6.1%) of 553 aborted bovine fetuses had leptospires detected by immunofluorescence techniques. Sixty-five percent of these fetuses were from submissions made between November and January. Leptospires were identified as serovar hardjo by specific immunofluorescence. There appeared, however, to be a paradoxical serological response in that eight aborting cows had antibody titers to pomona rather than hardjo.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Serological tests offer a potentially powerful tool for monitoring parasites in wildlife populations. However, such tests must be validated before using them with target wildlife populations. We evaluated in coyotes (Canis latrans) the performance of a commercially available serological test used to detect canine heartworm (Dirofilaria immitis) in domestic dogs. We obtained 265 coyote carcasses and serological specimens from 54 additional coyotes from several regions of California, USA. We necropsied coyotes to determine the adult heartworm infection status. Blood was collected at necropsy on filter paper strips and allowed to dry; it was later eluted in a buffer solution, and the supernatant was tested for heartworm. Receiver operating characteristic (ROC) analysis was used to assess discriminatory power of the test and indicated a 93% probability that a randomly selected infected coyote would exhibit a higher enzyme-linked immunosorbent assay (ELISA) value than a randomly selected uninfected coyote. We estimated specificity at 96% (95% CI: 92-98%) for 165 uninfected coyotes and sensitivity at 85% (77-91%) for 100 infected coyotes, results similar to published values for the commercial serological test used with dog serum or plasma. Test performance was similar for filter paper specimens and supernatant of frozen whole blood collected in EDTA tubes (i.e. hemolyzed plasma). We found no difference in test performance among geographic or demographic coyote groups. Our findings support application of the test to filter paper or standard serological specimens for detection of heartworm in coyote populations.  相似文献

Canine distemper virus (CDV) can cause mortality in domestic dogs, which is easily prevented by the consistent application of vaccination protocols. The aim of this study was to determine if the dog populations of three strategically located islands in the Torres Strait of Australia, adjacent to Papua New Guinea, are infected by CDV. Eighty-four serum samples were collected from 70 dogs resident on Saibai, Dauan and Boigu Islands during 2017–2018. Sera were tested for CDV antibodies by a virus neutralization test (VNT). Overall, 7 (8.3%) sera from 6 (8.6%) dogs resident on all three islands were test positive. VNT titres ranged from 20 to >1280. Male adult dogs were more commonly seropositive than female and juvenile dogs. Considering the origin and age of test positive dogs, and veterinary visits to these islands, it was concluded that there is evidence of exposure to a field strain of CDV – rather than previous vaccination – in 4 of the 70 dogs (5.7%) tested in this study. Given the strategic location of these islands in a zone of high biosecurity risk, ongoing surveillance of pathogens such as CDV could inform on potential disease spread pathways in this region. In addition, the presence of high serological titres in the apparent absence of clinical disease requires further investigation.  相似文献

Serum samples from 1,000 dairy goats from northwest United States (1982 to 1984) were examined for Toxoplasma gondii antibodies by a modified agglutination test. Toxoplasma gondii antibody titers were less than 1.40 for 779 goats, 1.40 for 153 goats, and greater than or equal to 1:400 for 68 goats. Seroprevalence increased with age of goats; 3.7% of 54 six-month-old goats were seropositive (greater than or equal to 1:40) vs 17.8% of 218 one-year-old goats.  相似文献

OBJECTIVE: To determine regional seroprevalence estimates of Toxoplasma gondi-specific IgM and IgG in clinically ill cats throughout the United States. Sample Population-Sera from 12,628 clinically ill, client-owned cats. PROCEDURE: Toxoplasma gondii-specific IgM and IgG antibodies were detected by use of ELISAs. Sera from clinically ill cats previously submitted for T. gondii antibody testing were sequentially selected from our serum bank and the sample submission paperwork reviewed. The country was divided into 12 geographic regions. Overall prevalence as well as prevalence for each region, age group, season, sex (male vs female), and breed (domestic shorthair vs other) was calculated. Data were analyzed by logistic regression analysis. RESULTS: Overall, 31.6% of the cats were seropositive for T. gondii-specific IgM, IgG, or both. Percentage of cats seropositive for T. gondii antibodies ranged from 16.1% (southwestern United States) to 43.5% (northeastern United States). As age increased, odds of positive T. gondii antibody assay results (IgM alone, IgG alone, and any combination of IgM or IgG) increased. Males were more likely than females to be seropositive for T. gondii antibodies (IgG alone and any combination of IgM or IgG). Domestic shorthair cats were more likely than other breeds to be seropositive for T. gondii antibodies (IgM alone, IgG alone, and any combination of IgM or IgG). CONCLUSIONS AND CLINICAL RELEVANCE: Toxoplasma gondii-specific antibodies are common in serum samples of clinically ill cats from all regions of the United States. Seroprevalence increases as cats age and is higher in male and domestic shorthair cats, compared with females and other breeds.  相似文献

为初步调查北京地区犬猫弓形虫感染的流病学特征,用酶联免疫吸附试验(ELISA)方法检测2010年5月至2011年4月采集的家养犬猫、流浪猫血清样本.其中,家养犬血清样本1876份,家养猫血清样本561份,检测发现,家养犬动物弓形虫IgG抗体阳性率24.9%;家养猫弓形虫IgG阳性率21.2%;同时检测流浪猫样本201份,阳性率30.3%.家养犬弓形虫血清抗体阳性率不同季节间差异显著(P<0.05),夏季最高,为30.0%,家养猫弓形虫血清抗体阳性率不同季节间无显著差异(P>0.05).不同性别犬猫弓形虫血清抗体阳性率无显著差异(P>0.05).随着年龄增长,犬猫弓形虫抗体阳性率均有明显增长.对25例弓形虫抗体阳性家养犬病例和37例弓形虫抗体阳性家养猫病例进行了回访调查,结果发现,该62例动物主人的弓形虫检测结果均为阴性.  相似文献
