Bovine mastitis caused by Escherichia coli can range from being a subclinical infection of the mammary gland to a severe systemic disease. Cow‐dependent factors such as lactation stage and age affect the severity of coliform mastitis. Evidence for the efficacy of antimicrobial treatment for E. coli mastitis is very limited. Antimicrobial resistance is generally not a limiting factor for treatment, but it should be monitored to detect changes in resistance profiles. The only antimicrobials for which there is some scientific evidence of beneficial effects in the treatment for E. coli mastitis are fluoroquinolones and cephalosporins. Both are critically important drugs, the use of which in animals destined for food should be limited to specific indications and should be based on bacteriological diagnosis. The suggested routine protocol in dairy herds could target the primary antimicrobial treatment for mastitis, specifically infections caused by gram‐positive bacteria. In E. coli mastitis with mild to moderate clinical signs, a non‐antimicrobial approach (anti‐inflammatory treatment, frequent milking and fluid therapy) should be the first option. In cases of severe E. coli mastitis, parenteral administration of fluoroquinolones, or third‐ or fourth‐generation cephalosporins, is recommended due to the risk of unlimited growth of bacteria in the mammary gland and ensuing bacteremia. Evidence for the efficacy of intramammary‐administered antimicrobial treatment for E. coli mastitis is so limited that it cannot be recommended. Nonsteroidal anti‐inflammatory drugs have documented the efficacy in the treatment for E. coli mastitis and are recommended for supportive treatment for clinical mastitis.  相似文献

The accurate identification of mastitis‐causing bacteria assists in effective management by both dairy farmers and veterinarians and can be used to implement the selective use of antimicrobials for treatment. The purpose of this study was to evaluate the ability of our developed anti–ribosomal protein-L7/L12 antibody–coated immunochromatographic strip (ICS) test to detect coliforms in milk by comparing the results with the bacteriological culture method. We investigated the performance of the ICS test as compared with the bacteriological culture method using 308 milk samples from clinical bovine mastitis. First, to determine the optimal ICS test cutoff point for detecting coliform mastitis, we developed a receiver-operating characteristic curve. The result showed that the cutoff point was at 0.5 of our index. The sensitivity, specificity, positive predictive value (PPV), and negative predictive value of the ICS test were 81.3%, 84.8%, 69.2%, and 91.54%, respectively. As the clinical signs increased in severity, the F-measure, a weighted harmonic mean of the sensitivity and overall PPV performance, increased. Because it is especially important to treat clinical mastitis appropriately in the early stages of detection, the ICS test, which can be used by both dairy farmers and veterinarians on dairy farms, is considered to be a useful tool for detecting coliform mastitis, which often presents with severe signs.  相似文献

为评价复方阿莫西林乳房注入剂治疗临床型奶牛乳房炎的疗效,在北京某牛场选择25头(48个乳区)临床型乳房炎患牛进行疗效观察。将临床型乳房炎患牛随机分为受试药物组(25个乳区)和对照药物组(23个乳区)。受试药物组患病乳区灌注复方阿莫西林乳房注入剂,对照药物组则灌注速诺LC。采用临床学与细菌学治愈率相结合的方法进行药效学评价。复方阿莫西林乳房注入剂治疗临床型乳房炎的临床学治愈率为68.0%,细菌学治愈率为83.3%,而速诺LC治疗组分别为69.6%、81.8%,两者治疗效果无显著性差异(P>0.05)。结果表明,复方阿莫西林乳房注入剂对临床型乳房炎具有较好的治疗效果。  相似文献

Mastitis remains one the most important diseases in dairy cattle despite the progress made in improving general udder health in recent years. Epidemiological studies have revealed that following treatment with antimicrobials, bacteriological cure rates vary between 0% and 80% but with no evidence of a significant loss of activity of the major classes of antibiotics licensed for the treatment of bovine mastitis. Recurrent infections are often attributable to biofilm growth of bacteria and this review provides an overview of those mechanisms related to bacterial biofilm growth in mastitis. Biofilm formation is accompanied by significant genetic and subsequent physiological changes in the microorganisms resulting, inter alia, in a loss of sensitivity to virtually all classes of antibiotics.  相似文献

Exacting demands have to be met by milk testing routine laboratories where mycoplasma species are to be cultured from milk samples. Cleanliness of sampling, immediate cold storage, and no-delay transport of the milk samples to the testing centre are of greatest importance to the informative value of all mycoplasma tests. In the summer season, it is recommended to freeze the milk samples immediately after sampling. Mycoplasma broth should not be inoculated to animals of the herd from which the samples had been taken. A modified mycoplasma culturing medium is described in this paper. It will enable wider introduction of mycoplasma diagnosis by more Regional Institutes of Veterinary Medicine. Also reported in this paper are diagnosis and successful control of enzootic mastitis caused by Acholeplasma laidlawii and Acheloplasma axanthum. Repeated bacteriological testing of milk and secretion of all cows in the herd helped in picking out all bacteriologically positive animals and isolating them from the negative individuals. All animals that had produced positive responses to bacteriological testing were killed, notwithstanding clinical udder and general milk findings. Definite success of any control action undertaken against mycoplasmic mastitis will depend strongly on no-delay bacteriological testing of milk and secretion samples from all cows of the given herd, as early as in the acute phase of mastitis caused by mycoplasma.  相似文献

Numerous culture-based diagnostics are available on the Australian and international markets for on-farm detection of bacterial pathogens in milk. Use of such diagnostics may provide an opportunity to improve the prudent use of antimicrobials in udder health management. Farms are low-resource settings in terms of diagnostic microbiology capacity. The World Health Organisation has identified criteria for the evaluation of diagnostic tests in low resource settings based on Accuracy, Sensitivity, Specificity, User-friendliness, being Rapid or Robust, Equipment-free and being Deliverable (ASSURED). Here, we review how those criteria can be interpreted in the context of microbiological diagnosis of mastitis pathogens, and how on-farm diagnostics that are currently available in Australia perform relative to ASSURED criteria. This evaluation identifies multiple trade-offs, both with regard to scientific criteria and with regards to convenience criteria. More importantly, the purpose of testing may differ between farms, and test performance should be evaluated relative to its intended use. The ability of on-farm mastitis diagnostics to inform mastitis treatment decision-making in a timely and cost-effective manner depends not just on test characteristics but also on farm-specific pathogen prevalence, and on the farm enterprise's priorities and the farm manager's potential courses of action. With most assay evaluations to date conducted in professional laboratories, there is a surprising dearth of information on how well any of the diagnostic tests perform on-farm and, indeed, of the on-farm decision-making processes that they aim to inform.  相似文献

Bovine mastitis remains as the disease causing the biggest economic losses to the dairy industry, despite the intensive research and prevention measures at herd level carried out for decades. Antibiotics are widely used to combat mastitis, but focus should be shifted from treatment to more economical and efficient prevention. The bacteriological aetiology of mastitis has changed from contagious to environmental pathogens, which has reduced the efficacy of the traditional mastitis control strategies. Considerable progress in the understanding of epidemiology, immunology, diagnostics and pathogenesis of mastitis has been made. The modern molecular biological methods offer good possibilities for the research of the epidemiological and virulence aspects of bacteria, which may help in building‐up specific mastitis control strategies for dairy herds. Studies on the host response and relationship between somatic cell count and susceptibility to mastitis offer tools for genetic improvement of dairy cows. Biotechnological approaches for mastitis prevention are in the developmental stage, but many problems are associated, e.g. with vaccination of dairy cows against mastitis. Different methods of immunomodulation for the prevention of mastitis have shown promise in experimental trials, but the evidence is not yet enough to support commercial applications. Improving nutrition, housing and environment of dairy cattle are still crucial in the prevention of mastitis, especially during the most susceptible period after parturition. New milking techniques including robotic milking may provide better possibility for proper milking and improved udder health. Mastitis control should be part of the herd health programme in the dairy herds. In this paper, results from recent research and proposals for new prevention strategies in the field of mastitis are reviewed.  相似文献

There is often a need to reduce sample size in clinical trials due to practical limitations and ethical considerations. Better comparability between treatment groups by use of stratification in the design, and use of continuous outcome variables in the evaluation of treatment results, are two methods that can be used in order to achieve this. In this paper the choice of stratification factors in trials of clinical mastitis in dairy cows is investigated, and two score-scales for evaluation of clinical mastitis are introduced. The outcome in 57 dairy cows suffering from clinical mastitis and included in a clinical trial comparing homeopathic treatment, placebo and a standard antibiotic treatment is investigated. The strata of various stratification factors are compared across treatments to determine which other factors influence outcome. The two score scales, measuring acute and chronic mastitis symptoms, respectively, are evaluated on their ability to differentiate between patients classified from clinical criteria as responders or non-responders to treatment. Differences were found between the strata of the factors severity of mastitis, lactation number, previous mastitis this lactation and bacteriological findings. These factors influence outcome of treatment and appear relevant as stratification factors in mastitis trials. Both score scales differentiated between responders and non-responders to treatment and were found useful for evaluation of mastitis and mastitis treatment.  相似文献

OBJECTIVE: To determine factors associated with implementation and use of an on-farm system for bacteriologic culture of milk from cows with lowgrade mastitis, including information on how producers used the on-farm bacteriologic culture system to guide antimicrobial selection practices and the resulting impact on patterns of antimicrobial use. DESIGN: Retrospective cohort study. SAMPLE POPULATION: Producers of 81 dairy farms. PROCEDURE: Farms that used an on-farm system for bacteriologic culture of milk from January 2001 to July 2003 were surveyed. RESULTS: Over half of those producers continuing to use the on-farm culture delayed antimicrobial treatment pending results of bacteriologic culture. Most other producers initiated empirical antimicrobial treatment while bacteriologic culture results were pending. Several barriers to the use of an on-farm system were identified. Significant reductions in rates of antimicrobial use were detected when comparing antimicrobial use rates before and during use of the on-farm system. Most producers chose to treat cows with mastitis caused by gram-positive pathogens with antimicrobials, whereas treatment choices for cows with mastitis caused by gram-negative bacteria and in cases in which no growth was detected varied. CONCLUSIONS AND CLINICAL RELEVANCE: Readily available results permit antimicrobial selections to be made on the basis of the causative agent of mastitis. Adoption of an on-farm system for bacteriologic culture of milk may result in significant reductions in the percentage of cows treated with antimicrobials. Decreasing antimicrobial use may have several benefits including preventing unnecessary discarding of milk, decreasing the potential for drug residues in milk, and improving treatment outcomes as a result of targeted treatments.  相似文献

Mastitis affects the quality of milk and is a potential health risk for the other cows. In a well managed dairy herd, in addition to clinical mastitis, subclinical mastitis should be efficiently detected. Bacteriological sampling is not feasible as a routine test to identify subclinical mastitis, and indirect tests of mastitis are more suitable for selecting cows with intramammary infections for subsequent bacteriological sampling. Mastitis affects the composition of milk, and the degree of changes depends on the infecting agent and the inflammatory response. Indicators of inflammation in the milk which can be determined using rapid, reliable and easy routine techniques, can be used for the early detection of mastitis. The measuring of the somatic cell count in milk is the standard method, but the analysis technique is problematic for routine use in herds. The most promising parameters for monitoring subclinical mastitis are milk N-acetyl-beta-D-glucosaminidase activity, lactose, and electrical conductivity along with some other indicators such as optical and milk flow measurements, preferably with an inter-quarter evaluation included in the test. Acute phase proteins, haptoglobin and serum amyloid A, are also potential candidates for mastitis monitoring. New mastitis detection systems which can be adapted into on-line use are urgently needed, since dairy units are growing bigger and automatic milking systems are being taken into use.  相似文献

The most important characteristics of Mycoplasma mastitis on dairy farms are described, based on two case studies. Clinical symptoms, diagnostics, epidemiology, and a plan of action are presented. In the herds investigated, Mycoplasma mastitis was characterized by multiple affected quarters unresponsive to treatment with antibiotic and/or anti-inflammatory agents. Most striking were a sandy sediment, brown colouring, and rice-like structure of the milk of affected animals. Clinical symptoms differed in the two affected herds. Diagnosis was based on bacteriological investigation of samples of milk and synovial fluid taken from infected cows. Affected animals were culled immediately, and the herds were monitored by repeated testing of bulk milk samples. It was concluded that a consequence of the increasing size of cattle herds in the Netherlands is that subclinical/clinical Mycoplasma mastitis may be diagnosed more frequently than in the past. In the case of Mycoplasma mastitis, farmers and veterinary practitioners are advised to draw up a plan of action together, incorporating aspects such as diagnostics at cow level, direct culling of affected animals, hygiene during milking, including post-milking teat disinfection, and routine monitoring of bulk milk. AIM: To test the non-inferiority of a novel combination intramammary product containing penicillin and cloxacillin to a reference intramammary product containing oxytetracycline, oleandomycin, neomycin and prednisolone with regard to bacteriological cure and clinical cure.

METHODS: Clinical cases of mastitis were sourced from 30 spring-calving dairy farms in the Southland region of New Zealand. Affected quarters were infused three times at 24 hourly intervals with either the novel combination product containing 1?g penicillin and 200?mg cloxacillin, or a reference product containing 200?mg oxytetracycline, 100?mg oleandomycin, 100?mg neomycin and 5?mg prednisolone. Cows were enrolled when a farmer detected a case of clinical mastitis. Milk samples were collected for microbiological culture immediately before treatment (Day 0) and on Days 9, 16 and 23. Bacteriological cure was compared for 187 and 178 quarters treated with the reference and novel product, respectively, and clinical cure was compared for 235 and 223 quarters, respectively. Non-inferiority was assessed by calculating the difference in cure rates between the two products and constructing a 95% CI around the difference, using the variance inflation factor to account for herd level clustering. The non-inferiority margin was 20% for both bacteriological and clinical cure. Generalising estimating equation models were used to determine predictor variables.

RESULTS: The bacteriological cure percentage, adjusted to account for herd-level clustering, was 8.5 (95% CI=?1.7–21.8)% higher for quarters treated with the novel than the reference product. The adjusted clinical cure percentage was 0.3 (95% CI=?11.2–12.0)% higher for clinical quarters treated with the novel than the reference product. Bacterial species was the only covariate for bacteriological cure (p=0.003), and quarter score at enrolment (indicating udder inflammation) was the only covariate for clinical cure (p=0.032) in the multivariable models.

CONCLUSION: The novel combination product was demonstrated to be non-inferior to the reference product with regards to both bacteriological cure and clinical cure.

CLINICAL RELEVANCE: Clinicians treating mastitis now have access to this novel combination intramammary product, and demonstration of its non-inferiority compared to the existing reference product will provide options for treatment approaches. The novel product contains fewer antimicrobials; which are of a narrower spectrum of activity.  相似文献

乳酸链球菌素(Nisin)是由乳酸链球菌产生的合有34个氨基酸的抗菌肽,对人体无害。本研究将患临床型乳房炎奶牛92头(107个乳区)和隐性乳房炎奶牛90头(90个乳区)随机分成试验组和对照组。试验组的治疗方法是在患病乳区灌注Nisin;临床型乳房炎以庆大霉素乳房内灌注作对照,隐性乳房炎的对照组不作任何处理。临床型乳房炎的Nisin治疗组细菌学治愈率和临床治愈率分别为60.8%和90.2%,而庆大霉素治疗组则分别为44.6%和91.2%,两者无统计学差异。隐性乳房炎经Nisin治疗后,高体细胞数奶牛的数量和牛奶中NAGase的活性均显著下降,细菌转阴率提高。乳酸菌发酵试验表明,经Nisin乳房内灌注治疗临床型乳房炎后,牛奶在第36小时恢复正常发酵,而庆大霉素治疗则需要72h。  相似文献

Antibiotics are widely used in treatment and prophylaxis of mastitis, but their antibacterial and therapeutic efficacy continues to decrease. New methods of prophylaxis of mastitis with the use of vaccines and biological response modifiers are reviewed. Prophylactic activity of staphylococcal vaccines should be investigated in the future because of difficulties in successful treatment of infections caused by these organisms. Vaccination with J5 bacterin (0111:B4) reduced duration of IMI and local signs of clinical coliform-mastitis. Some immunomodulators proved to be effective in treatment of mastitis during lactating period. Promising results were obtained with the use of giseng saponin, herbal gel, herbal extracts, propolis, lysosubtilin, antibacterial proteins, and lysozyme dimer especially.  相似文献

This review presents a framework for assessing the efficacy of antimicrobials used to control necrotic enteritis (NE) caused byClostridium perfringens (CP) in the context of susceptibility testing and clinical efficacy, and their potential interactions with the intestinal microbiota of poultry. Practitioners have traditionally based their choice of antimicrobial agent on antimicrobial susceptibility testing, but there appears to be a lack of correlation with clinical efficacy for in-feed antimicrobials (particularly bacitracin and virginiamycin). Resistance patterns of CP and antimicrobials have been monitored using epidemiological cutoffs for minimal inhibitory concentration (MIC), which are not intended to guide therapy. Also, most data have been determined using CP isolates from healthy birds (i.e., potentially from commensal strains not known to be clinically relevant). It is believed that NE is caused by specific virulent CP strains (and potentially other bacteria) that proliferate and displace these commensals. The presence of resistant commensals is not necessarily detrimental (and may even be beneficial) if they inhibit the single CP strain dominance effect observed in acute NE. The choice of antimicrobial therapy in a clinical setting should thus be based on a variety of factors, including an accurate diagnosis, results of efficacy studies, prior experience at the premises in question, and interpretation of MIC data, recognizing that it is not necessarily well correlated with clinical efficacy.  相似文献

Information regarding antimicrobial use in sheep is scarce. In 2001, a scrapie surveillance program was initiated in Alberta that also provided a mechanism for collecting other sheep health data including antimicrobial use information between April 2001 and April 2002. A major objective of this study was to describe antimicrobial use in the Alberta sheep industry. This was done by obtaining qualitative antimicrobial use information from all flocks (n = 212) providing cull ewes to the program using a brief, primarily flock-level, questionnaire. The respondents' flocks represented 13.6% of the total provincial flock in Alberta in 2001. By a substantial amount, the most frequent method of administering antimicrobials was through injection followed by in-feed, oral (liquids, pills, boluses), and in-water routes, respectively. Drug-specific use data were collected for injectable antimicrobials only, with the most commonly used antimicrobial classes being penicillins followed by tetracyclines. Producers rarely treated some or all of their flock with injectable antimicrobials after discovering an individual sick animal. Adult sheep were the most common age group treated with injectable antimicrobials and the most frequent reason for injectable antimicrobial use was mastitis followed by respiratory problems. This study provides some initial insight regarding antimicrobial use in Alberta sheep flocks. However, collection of more drug-specific data (drug type, dose/concentration, duration of treatment) for noninjectable routes of administration should be conducted in future studies. Assessing antimicrobial use in other sectors of the Alberta sheep industry (feedlots) and other provinces across Canada would also be beneficial.  相似文献

The coagulase-negative staphylococcal species causing mastitis in lactating cattle were identified and possible differences in the clinical characteristics or persistence of mastitis caused by different CNS were evaluated. The effect of antimicrobial treatment was also assessed. In addition, AFLP-typing of CNS was compared with the phenotypic identification. A total of 133 clinical or subclinical quarter cases of intramammary infection caused by CNS from the practice area of the Ambulatory Clinic of the University of Helsinki were studied. Bacteriological diagnosis was based on biochemical (API) testing. Staphylococcus simulans (43.6%) followed by S. chromogenes (23.3%) were the most common CNS species isolated from the milk samples. Ninety-nine isolates were genotyped using AFLP-analysis. Only 75.0% of S. chromogenes and S. simulans isolates identified with API test were clustered with the type strains of these species. Approximately half of the mastitis cases were clinical, and in the majority clinical signs were mild. The severity and persistence of intramammary infection were unaffected by CNS species. Fifty-nine percent of the quarter cases were treated with antimicrobials, and the rest were left without treatment. Mastitis due to beta-lactamase-negative CNS was treated with penicillin G and that due to beta-lactamase-positive CNS with cloxacillin. Nineteen percent of the isolates were beta-lactamase-positive. The bacterial cure rate for quarters treated with antimicrobials was high, 85.9%, as opposed to only 45.5% for untreated quarters. Bacterial cure rates for the most common CNS species or AFLP clusters were not statistically different. Further studies on identification of CNS species are needed.  相似文献
