The fluorescence of bovine tissues was measured post mortem by microscopy of frozen sections and by using optical fibres to excite fluorescence and to measure fluorescence emission spectra. Mechanical disruption of the tissue (by comminution or sectioning) did not appreciably change tissue fluorescence spectra. Ligamentum nuchae had the strongest fluorescence and lung tissue had the weakest. In samples measured with a minimum prior exposure to ultraviolet light, the peak fluorescence emission was at 410 or 420 nm (with excitation at 365 nm). Exposure to ultraviolet light for about 1 minute shifted the fluorescence peak to 450 to 470 nm. Further exposure (about 30 minutes) caused a loss of the 450 to 470 nm fluorescence peak, while emissions above 530 nm were maintained or strengthened. Microscopy showed that the fluorescence that was measured by fibre optics from intact connective tissues originated mostly from collagen and elastin fibres.  相似文献

White light from a xenon arc was focused onto the upper surface of 25-mm-thick transverse sections of pork longissimus muscle. A servo motor moved an optical fiber across the lower surface of the muscle to collect transmitted light, which then was passed through a grating monochromator and onto a photomultiplier for spatial measurements of scattering (SMS) and transmittance spectra. The SMS were calculated as the slope of the logarithm of transmittance relative to path length through the sample (which was calculated trigonometrically). Pale, soft, exudative (PSE) pork was measured with an index that included a subjective evaluation of meat color and objective measurements of reflectance, drip loss, and centrifugation fluid loss. The strongest correlation of SMS with PSE was at 610 nm (r = .86, P less than .005) and the strongest correlation of transmittance with PSE was at 650 nm (r = -.95, P less than .005). This supports the use of a red laser at 633 nm for the detection of PSE pork.  相似文献

We examined animal species differences in the postprandial thermic effect of meat and investigated the postprandial thermic effect of mutton in rats. After intake of experimental diets containing each meat, body temperatures of rats fed mutton or venison were significantly higher than that of rats fed rabbit meat. After intake of experimental diets containing fractionized mutton, the body temperatures of rats fed diets containing lean mutton protein were higher than those of rats fed diets without lean mutton protein. In a two‐dimensional fluorescence difference gel electrophoresis study of brown adipose tissue, it was shown that the intake of mutton up‐regulated the expression of many signaling molecules that are involved in energy metabolism. The postprandial thermic effect of mutton seems to be due not to catecholamine and adrenocorticotropic hormone but to thyroid hormones. The results suggest that intake of lean mutton protein stimulates the secretion of thyroid hormones and facilitates energy metabolism in rats.  相似文献

Six Belgian Blue bulls (double-muscled type) and six Friesian bulls were offered a fattening diet for 34 weeks. Plasma samples were obtained once a week and also every 20 min over a 24 h period, 7 weeks before slaughter.No differences were observed between the breeds in plasma glucose, urea and free amino nitrogen concentrations, while creatinine was significantly higher in the Belgian Blue bulls. Tri-iodothyronin, tetra-iodothyronin, insulin-like growth factor 1, insulin and testosterone concentrations were higher in the Holstein group. In contrast, the Belgian Blue bulls appeared to produce more growth hormone. The slaughter weight, carcass weight, dressing percentage and proportion of lean meat were significantly higher in the Belgian Blue group. The characteristics of muscle mass (carcass weight, dressing percentage and proportion of lean meat) were positively correlated with creatinine and with the total peak area or peak amplitude of growth hormone. The insulin concentration was positively correlated with the proportion of adipose tissue in the carcass and negatively correlated with the proportion of muscle. There were no correlations between the carcass characteristics and insulin-like growth factor 1 or testosterone. No further information was provided when the ratios of the hormones were correlated with carcass characteristics.  相似文献

Three probes were evaluated for their effectiveness in measuring connective tissue fluorescence within carcasses or primal cuts: 1) a quartz-glass rod, 2) a light guide formed from bundles of optical fibers, and 3) a single optical fiber. The shape of the fluorescence emission spectrum of tendon was altered by the method of measurement, probably because of differences in the intensity of excitation. The single optical fiber design provided the best solution to the problem caused by the irregular distribution of connective tissue in meat, and a modified fat-depth probe was tested as a prototype. Beef shank had more fluorescence peaks per millimeter (P less than .01), a greater area above minimum fluorescence (P less than .01), and a greater mean peak intensity (P less than .005) than did psoas major.  相似文献

The belly region composition of a representative commercial pig sample of 130 castrates was examined by in vivo spiral computed tomographic (CT) scanning. The lean meat percentage, the muscle/fat tissue ratio within the total body and separately within the bacon part were estimated by image analysis. The lean meat content of the total body was determined by the EU reference method. A correlation of r = 0.97 was found between lean meat content of the total body determined by CT estimation and the slaughtering results. The pigs were divided into seven groups on the basis of their lean meat content. The muscle percentage of the bacon part was 26% and 67%, respectively, in the two groups representing the two extremes of lean meat percentage (36% and 67%, respectively). The total volume of the bacon part was relatively constant (8.6 +/- 1 dm3) in the groups representing different lean meat categories. At the same time, the fat tissue volume of the bacon part was considerably higher while its muscle tissue volume was markedly lower in the group of the lowest than in that of the highest lean meat content (6.6 vs. 2.5 dm3 and 2.4 vs. 5.5 dm3, respectively). To describe changes in the tissue composition of the bacon part during fattening, 10 castrates were scanned repeatedly at 70, 90 and 110 kg of liveweight. A linear regression model was developed to characterise the continuous decrease observed in the muscle/fat ratio within the bacon part due to increasing liveweight.  相似文献

Moderately PSE (pale, soft, exudative) and moderately DFD (dark, firm, dry) pork was examined by x-ray diffraction for interfilament separation, by differential interference contrast microscopy for interfiber area, and was centrifuged to measure water holding capacity (WHC). Internal reflectance spectra were measured by fiber optics. For PSE to DFD pork, filament separation ranged from 39 to 48 nm, interfiber area from 42 to 3%, and WHC from 49 to 64%, respectively. The correlation of reflectance with interfilament separation varied considerably with wavelength (reaching r = -.83 at 680 nm, P less than .005). The correlation of reflectance with interfiber area was more uniform across the spectrum (reaching r = .90 at 450 nm, P less than .005), as was the correlation of reflectance with WHC (reaching r = -.80 at 400 nm, P less than .005). At 24 h postmortem, fiber-optic spectrophotometry may be used as a rapid, nondestructive method to predict WHC and potential fluid losses from commercial pork with a moderate range from PSE to DFD. Interfiber area was correlated negatively with filament lattice area and WHC, but no significant correlation was found between filament lattice area and WHC. Filament separation was decreased only slightly by centrifugation. These results indicate that at 24 h postmortem the extra fluid released from PSE pork already has been lost from the myofilament lattice and is awaiting release from compartments downstream such as interfiber and interfascicular spaces.  相似文献

1. Three trials were carried out to compare the effects of different degrees of fatness in broilers and of dietary a‐tocopherol (AT) concentrations on the stability of meat and adipose tissue at 5 to 9 weeks of age.

2. In spite of higher concentrations of AT in carcass fat, the meat and abdominal fat of lean broilers were less stable than those of obese birds, apparently due to an increase in the unsaturation of their carcass fat.

3. Dietary AT improved significantly the stability of meat and adipose tissue, irrespective of the degree of fatness. However, the difference in the stability of these tissues between obese and lean broilers remained apparent even with 60 mg AT/kg of the diet.

4. A linear relationship was observed between dietary AT and the stability of adipose tissue, whereas the relationship between AT intake and meat stability was logarithmic.  相似文献

以定远猪为母本,分别与瘦肉型猪种杂交所产F1代仔猪进行肌肉生长性状及肌肉蛋白质含量、氨基酸组分的分析与比较。结果发现:①眼肌重、眼肌面积与蛋白质含量三者间达到较高的正相关(r=0.74~0.85),蛋白质含量高者,眼肌重和眼肌面积均明显增加;②杂交后,F3代猪肌肉蛋白质含量均有提高,并以长白×定远杂交组合改良效果最好,达到22.6%;⑧杂交后F1代猪肌肉中赖氨酸、亮氨酸、异亮氮酸、缬氨酸含量均较对照组有不同程度提高,并且赖氨酸、亮氨酸、异亮氮酸与骨骼肌生长间有较强的正相关,其中赖氨酸与肌肉增长相关性为r=0.51~0.96,亮氨酸、异壳氨酸与肌肉增长相关性为r=0.40~0.80;④杂交后可有效提高猪瘦肉产出量,明显提高瘦肉率,并适应改进猪后躯的发育。  相似文献

In vitro lipolytic rate was determined in adipose tissue from genetically obese and lean pigs. There were about 20 pigs/genetic strain at 25 and 80 kg and 10 pigs/strain at 50 kg body weight. When expressed on a cellular basis, the in vitro adipose tissue basal (no exogenous hormone) lipolytic rate was similar in obese and lean pigs at 25 and 50 kg body weight. At 80 kg body weight the basal rate was greater in obese than in lean pigs. The in vitro adipose tissue epinephrine-stimulated lipolytic rate expressed on a cell basis was greater at 25 kg, was similar at 50 kg body weight and tended (P less than or equal to .1) to be greater at 80 kg in obese compared with lean pigs. The in vitro sensitivity of lipolysis to epinephrine was slightly greater in lean compared with obese pigs. The data obtained in vitro indicate that obese pigs do not have low adipose tissue lipolytic rates compared with lean pigs. Consequently, adipose tissue lipolysis does not appear to be a major metabolic factor leading to the excessive fat accretion in these obese pigs.  相似文献

试验旨在研究不同pH对淀粉样纤维形成的影响。以蛋清溶菌酶(hen egg-white lysozyme,HEWL)为模型蛋白,在不同pH的甘氨酸(50 mmol/L)溶液中(57.0±0.1)℃孵育0~8 d,使其形成不同聚集程度的淀粉样纤维,pH设为2.0、6.5、7.5和8.0;用透射电镜定性观察淀粉样纤维生成状况,采用硫磺素T (ThT)荧光法和刚果红染色法测定淀粉样纤维生长状况,8-苯胺基-1-萘磺酸(ANS)荧光法测定淀粉样纤维疏水性变化,圆二色谱法测定二级结构转化,BeStSel软件预测β-折叠含量。结果显示,透射电镜下观察第8天纤维生长状况发现,pH为2.0时HEWL形成了大量的短杆状淀粉样纤维,同时ThT荧光法结果表明淀粉样纤维生长状况最好(P<0.01),且生长趋势呈时间依赖性;刚果红染色法显示,在490~510 nm出现红移、540 nm出现特征性肩峰,疏水性显著增强,并且二级结构转化明显,β-折叠含量从天然HEWL蛋白的6.1%增加到37.6%;pH 6.5组仅在透射电镜下观察到极少量的淀粉样纤维,pH 7.5组在视野中未观察到明显的淀粉样纤维,仅观察到大量的球状或杆状蛋白聚集体;pH 8.0组未产生淀粉样纤维。高温条件下,HEWL在强酸性(pH 2.0)条件较易形成淀粉样纤维,且此时疏水性和二级结构转化程度最高,这为解释朊病毒病及其他淀粉样变性疾病的淀粉样纤维形成机制提供了理论基础。  相似文献

为了研究紫花苜蓿对猪生长及肉质性能的作用,选用16头长×大外二元育肥猪,在基础饲料中添加4%苜蓿,应用免疫组织化学、酶联免疫吸附试验、实时荧光定量PCR研究脂肪代谢相关因子在mRNA水平和蛋白质水平表达的差异。结果显示:饲料中添加苜蓿后,杂交猪骨骼肌细胞中瘦素(leptin)基因(P<0.05)和蛋白(P>0.05)的表达量下降,与日增重呈轻度负相关(r=-0.314);转录因子C/EBPβ(CCAAT/en-hancer binding proteins,CEBPβ)基因和蛋白的表达量变化不明显;过氧化氢酶体激活增殖受体(peroxisome proliterator actiated receptors,PPARr)基因和蛋白水平高于对照组(P>0.05),与日增重(r=-0.837)、瘦肉率(r=-1.000)、pH1(r=-1.000)呈高度负相关,与脂率(r=1.000)、熟肉率(r=1.000)高度正相关;脂联素(adiponectin)基因(P>0.05)和蛋白(P<0.05)表达量高于对照组,与脂率(r=-1.000)、熟肉率(r=-1.000)、胴体重(r=-0.500)、屠宰率(r=-0.500)、肉色评分(r=-0.500)、pH24(r=-0.500),肌内脂肪含量(r=-0.500)负相关,与日增重(r=0.837)、瘦肉率(r=1.000)、pH1(r=1.000)、眼肌面积(r=0.500)、失水率(r=0.500)正相关;脂肪分化相关蛋白(adiposedifferentiation-related protein,ADFP)基因(P>0.05)和蛋白(P<0.05)表达量上调,但与肉质、体增重等指标相关性不大;脂蛋白脂酶(lipo-protein lipase,LPL)基因和蛋白变化不明显,与各项屠宰指标不相关。这些结果表明一定量的紫花苜蓿下调Leptin、通过CEBPβ上调PPARr、ADFP和Adiponectin 3个基因和蛋白的表达,增加猪的日增重、肌内脂肪,影响肉质性状,对LPL的作用较小。这些研究为紫花苜蓿在养猪业中的开发利用奠定了理论基础。  相似文献

Carcass, muscle and meat characteristics of lean and obese pigs   总被引:1,自引:0,他引:1  
Six pigs obtained from a lean selected strain and six pigs obtained from an obese selected strain were slaughtered at about 110 kg live-animal weight. Carcasses were evaluated; hams were dissected into bone, skin, fat and lean, and loin samples were obtained for fiber type characteristics, percentage of fat and moisture, collagen analysis, sensory characteristics, textural properties and objective color analysis. Carcasses from lean pigs were longer, had less backfat and larger longissimus muscle cross-sectional areas than carcasses obtained from obese pigs. Hams from lean pigs had less fat, more bone and more lean than hams from carcasses of obese pigs. The percentages and cross-sectional areas of red and white muscle fibers of the longissimus muscle from lean and obese pigs were not different. However, lean pigs had intermediate fibers that were only 79% as large (P less than .10) as intermediate muscle fibers from obese pigs. Intermediate fibers represented only 7 and 10% of total fiber area, whereas white fibers represented 84 and 79% of total fiber area in longissimus muscle of lean and obese pigs, respectively. Overall, lean pigs tended to possess fewer fibers (-16%) per unit of area than obese pigs, indicating that total muscle fiber hypertrophy was partially responsible for the increased longissimus muscle area of the lean strain. Sensory properties of longissimus meat samples from lean and obese strains were not different. However, the shear force requirement of the longissimus samples from the lean strain were slightly, but significantly (P less than .10), higher than those from the obese strain. The effects of four protein sources (soybean meal, sunflower meal, pea, and fish meal as the main protein source) and three apparent ileally digestible Lys:DE ratios (0.50, 0.43, 0.36 and 0.42, 0.36, 0.30 g Lys/MJ DE for 30 to 60 kg BW and 60 to 105 kg BW, respectively) in pig diets on growing-finishing performance, and carcass and meat quality traits were investigated. Eight individually housed animals per treatment received the diets from 30 to 105 kg BW at a level of 3.0 times maintenance requirements of energy. The ileal digestibility of protein sources was determined in a previous digestibility experiment. Protein sources showed no differences in growth performance from 30 to 105 kg BW. From 30 to 60 kg BW soybean treatment had lowest performance. The protein sources had no effect on lean meat percentage, liver weight, or meat quality (intramuscular fat content, pH at 45 min and 24 h after slaughter, drip loss, and meat color measured 24 h and 4 d after slaughter). The experimental diets formulated on the basis of similar apparent ileal digestible lysine content resulted in similar body composition regardless of the protein source used (P > 0.05). Reducing the Lys:DE ratio from 0.50/0.43 to 0.36/0.30 (by about 28%) reduced BW gain by 119 g/d from 30 to 60 kg and by 151 g/d from 60 to 105 kg BW. The gain:feed ratio increased by 82 g/kg in the first phase and by 47 g/kg in the second phase for the highest Lys:DE treatment compared with the lowest. Reducing Lys:DE ratio did not modify meat quality traits. A high Lys:DE ratio was associated with a high lean meat percentage. Differences between the medium- and low-Lys:DE groups were not significant. Lowering the Lys:DE ratio increased (P < 0.05) crude fat and fatty tissue content and decreased (P < 0.05) protein and muscle content in the body. Ash content and bone volume were not affected by Lys:DE ratio (P > 0.05). The chemical composition of the carcass can be predicted with moderate accuracy (R2 = 0.39 to 0.58) using volumetric composition data of previously frozen carcasses. In conclusion, similar growth performance, carcass and meat quality, and body composition can be expected if diet formulation is based on the apparent ileally digestible amino acid contents of feedstuffs, independent of dietary protein sources. Diminishing Lys:DE ratios reduce growth performance but do not modify meat quality traits. The chemical composition of the carcass can be predicted with moderate accuracy using the volumetric composition of thawed carcasses.  相似文献

A luminol-dependent chemiluminescence (LDCL) assay was used to assess the response of polymorphonuclear leukocyte (PMN) preparations from 4 species of ruminants (ie, cattle, sheep, goats, and antelopes) and 6 species of nonruminants (ie, swine, dogs, cats, rabbits, horses, and persons) to both opsonized and nonopsonized preparations of living and heat-killed Pasteurella haemolytica and Staphylococcus aureus and to opsonized and nonopsonized heat-killed strains of each bacterium in the presence of sterile culture supernatant (leukotoxin) from P haemolytica. The LDCL responses of PMN preparations from each of the species studied were greater for living than for heat-killed S aureus. The most efficient LDCL emission was observed with reaction mixtures containing opsonized living S aureus. Regardless whether they contained killed or living bacteria, the opsonized S aureus preparations elicited LDCL emissions more efficiently than did the corresponding nonopsonized preparations. Living P haemolytica cells and their sterile culture supernatant inhibited the LDCL emissions of phagocytically stimulated PMN preparations from ruminants, but not those from nonruminants. The LDCL response of ruminant PMN to nonopsonized living P haemolytica was characterized by the development of a peak response at 10 minutes of incubation followed by a precipitous decrease and a subsequent complete cessation of chemiluminescence. The peak LDCL response was higher for opsonized living P haemolytica than for nonopsonized living bacteria, and the increased response lasted longer. However, opsonization of living P haemolytica with the serum samples tested only temporarily spared the ruminant PMN preparations from the detrimental effects of leukotoxin.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Genetically obese and lean pigs were fed isonitrogenous-isoenergetic (digestible energy) amounts of a high or low fat diet from 25 kg body weight. Obese pigs gained less and required more feed per unit gain than lean pigs. Lean pigs were more muscular with less fat than obese pigs. Obese pigs utilized more dietary amino acids for energy (greater plasma urea N) than did lean pigs. Weight gain was similar at all intermediate periods in obese pigs fed the two diets. However, gain tended (P less than or equal to .10) to be greater and the ratio of dietary energy intake to gain tended (P less than or equal to .10) to be less in obese pigs fed high compared with low fat diets. Similar results were observed in lean pigs fed the two diets. The high fat diet produced more carcass adipose tissue deposition in both strains after 20 wk of feeding (detectable by ultrasound at 14, but not at 7 wk). Adipose tissue lipogenic rate (glucose incorporation) was similarly depressed by fat feeding in both obese and lean pigs. Obese and lean pigs both utilized dietary carbohydrate and fat differentially but there was no indication of genetic divergence regarding this utilization. In both strains of pigs, energy from the fat-enriched diet was preferentially partitioned into carcass adipose tissue.  相似文献

Leptin is the hormone product of the obese gene that is synthesized and predominantly expressed by adipocytes. This study estimated the genetic variation in serum leptin concentration and evaluated the genetic and phenotypic relationships of serum leptin concentration with performance, efficiency of gain, and carcass merit. There were 464 steers with records for serum leptin concentration, performance, and efficiency of gain and 381 steers with records for carcass traits. The analyses included a total of 813 steers, including those without phenotypic records. Phenotypic and genetic parameter estimates were obtained using SAS and ASREML, respectively. Serum leptin concentration was moderately heritable (h2 = 0.34 +/- 0.13) and averaged 13.91 (SD = 5.74) ng/mL. Sire breed differences in serum leptin concentration correlated well with breed differences in body composition. Specifically, the serum leptin concentration was 20% greater in Angus-sired steers compared with Charolais-sired steers (P < 0.001). Consequently, ultrasound backfat (27%), carcass 12th-rib fat (31%), ultrasound marbling (14%), and carcass marbling (15%) were less in Charolais- than Angus-sired steers (P < 0.001). Conversely, carcass LM area (P = 0.05) and carcass lean meat yield (P < 0.001) were greater in Charolais- compared with Angus-sired steers. Steers with greater serum leptin concentration also had greater DMI (P < 0.001), greater residual feed intake (P = 0.04), and partial efficiency of growth (P = 0.01), but did not differ in feed conversion ratio (P > 0.10). Serum leptin concentration was correlated phenotypically with ultrasound backfat (r = 0.41; P < 0.001), carcass 12th-rib fat (r = 0.42; P < 0.001), ultrasound marbling (r = 0.25; P < 0.01), carcass marbling (r = 0.28; P < 0.01), ultrasound LM area (r = -0.19; P < 0.01), carcass LM area (r = -0.17; P < 0.05), lean meat yield (r = -0.38; P < 0.001), and yield grade (r = 0.32; P < 0.001). The corresponding genetic correlations were generally greater than the phenotypic correlations and included ultrasound backfat (r = 0.76 +/- 0.19), carcass 12th-rib fat (r = 0.54 +/- 0.23), ultrasound marbling (r = 0.27 +/- 0.22), carcass marbling (r = 0.76 +/- 0.21), ultrasound LM area (r = -0.71 +/- 0.19), carcass LM area (r = -0.75 +/- 0.20), lean meat yield (r = -0.59 +/- 0.22), and yield grade (r = 0.39 +/- 0.26). Serum leptin concentration can be a valuable tool that can be incorporated into appropriate selection programs to favorably improve the carcass merit of cattle.  相似文献

Clenbuterol(CLB,盐酸克伦特罗),俗称瘦肉精。由于其在动物体内广泛残留,可能导致食用此类产品的人中毒而被禁止使用,但已有研究表明该制剂可显著减少猪脂肪沉积,因此,对其分子机制进行深入研究将具有重要的理论与实践价值。本研究通过对家猪饲喂高剂量clenbuterol(≥1.5mg/kg体重),提高瘦肉率10%左右。通过实时荧光定量PCR技术对脂肪组织中与脂类代谢密切相关的4个基因研究发现,饲喂clenbuterol的猪脂肪组织内载酯蛋白D(apolipoprotein D,apoD)mRNA丰度增加超过2倍,而脂蛋白酯酶(lipoprotein lipase,lpl)、硬酯酰辅酶A去饱和酶(stearoyl-CoA desaturase,scd)和激素敏感脂酶(hormone sensitive lipase,hsl)mRNA丰度变化不明显。因此,推断clenbuterol通过改变部分脂代谢关键基因的表达来实现减少脂肪积累。载酯蛋白D可能是CLB应答基因之一。  相似文献
