通过考察乙酰甲喹对小新月菱形藻(Nitzschia closterium f.minutissima)、等鞭金藻(Isochrysis galbana Parke3011)的生长抑制和对叶绿素a、总超氧岐化酶(T-SOD)活性以及丙二醛(MDA)浓度的影响,研究了乙酰甲喹对2种微藻的毒性效应,为评价兽药乙酰甲喹的使用安全性和保护养殖环境提供基础数据。结果表明,乙酰甲喹对2种微藻的生长抑制作用随质量浓度的增加而增大,乙酰甲喹对小新月菱形藻的24 h-EC50(半数有效浓度)为4.73 mg·L~(-1)、对等鞭金藻3011的24 h-EC50为2.14 mg·L~(-1),乙酰甲喹对2种微藻均属于高毒物质,等鞭金藻3011更敏感。暴露24 h后,随着乙酰甲喹质量浓度的升高,小新月菱形藻叶绿素a浓度下降而等鞭金藻3011叶绿素a浓度基本不受影响;2种微藻的T-SOD活性随着乙酰甲喹质量浓度的升高均显著增加,各实验组MDA浓度也高于对照组,等鞭金藻3011更敏感,说明乙酰甲喹对2种微藻均能造成氧化胁迫,对细胞造成氧化损伤。  相似文献

球等鞭金藻作为一种优良的单细胞生物饵料具有选育价值。目前我国水产养殖上使用的大多数球等鞭金藻品系都来源于同一株藻株Isochrysis galbana OA-3011,经过多年的地理隔离形成了不同的品系,为了揭示地理隔离对球等鞭金藻表型性状的影响,从而为球等鞭金藻的选育工作提供依据,我们对5个品系的球等鞭金藻(I.galbana FACHB-861,OA3011-QD,OA3011-LK,OA3011-AY和OA3011-LZ)的种属关系、生长和脂肪酸组成进行了比较分析。实验在相同条件下对5个品系球等鞭金藻培养6 d,提取基因组DNA并PCR扩增测定18S rDNA基因序列,每天定时记录各品系的OD_(660),绘制生长曲线,在进入稳定期后收获并进行生物量的测定及脂肪酸的气相色谱分析。18S rDNA基因序列的进化分析表明,OA3011-LK、OA3011-AY和OA3011-LZ具有一致的序列,而OA3011-QD因1个碱基的差异与其他OA3011出现了分离,FACHB-861与OA3011品系亲缘关系较远;对生长和脂肪酸进行对比分析后,结果显示,5个品系的球等鞭金藻在不同的时间达到稳定期,其中FACHB-861表现出了与其他品系明显不同的生长模式。FACHB-861和OA-3011品系的生物量具有显著差异,OA3011-LK、OA3011-AY和OA3011-LZ之间也有显著差异(P0.05),但特异生长率各品系间无显著差异(P0.05)。FACHB-861和OA3011-LZ的总脂肪酸(TFA)含量显著低于OA3011-LK、OA3011-QD和OA3011-AY(P0.05)。FACHB-861的脂肪酸各成分与OA-3011各品系明显不同。然而,在OA-3011各品系间脂肪酸组成也有明显差异,这表明球等鞭金藻存在广泛和频繁的数量性状表型差异。总之,地理隔离造成的这些差异为球等鞭金藻的选育提供了可行性。根据生长速度或DHA、EPA、多不饱和脂肪酸含量等筛选标准初步分析显示,可对OA3011-LK,OA3011-QD进行进一步的选育。  相似文献

为探究球等鞭金藻(Isochrysis galbana)的最优培养条件,采用实验生态学方法和响应面法,以球等鞭金藻的比生长速率为指标,设置不同温度、盐度、接种量单因素实验及Box-Benhnken(BBD)模型多因素交叉实验。结果表明:1)在一定温度、盐度及接种量范围内,球等鞭金藻的比生长速率随着各因素的增大,呈先升高后下降的趋势;2)通过响应面BBD模型优化出球等鞭金藻的最佳培养条件为:温度26.38℃、盐度26.61、接种量1.29×10~6个·mL~(-1),此时的球等鞭金藻的比生长速率最大,为0.44 d~(-1)。  相似文献

在水温(22±1)℃和连续光照强度为50μmol·m~(-1)·s~(-2)下,取主要由乳酸菌、硝化菌、芽孢杆菌、光合菌,及酵母菌等组成的培藻肥水型EM菌0.5m L、2.5m L或5.0m L,分别加入到200m L密度相同的小新月菱形藻Nitzschia closterium f.、球等鞭金藻3011 Isochrysis galbana parke和小球藻Chlorella sp.中培养15d,以f培养基为对照,研究不同浓度培藻肥水型EM菌对这3种藻类生长的影响。结果表明:EM菌对金藻、硅藻、绿藻的生长都有一定的促进作用,其中对金藻及硅藻生长的促进作用更加显著,生长率提高20%~50%,最终细胞密度有所提高,添加5m L促生长作用更持久。  相似文献

通过分析喹烯酮对小新月菱形藻(Nitzschia closterium f.minutissima)和等鞭金藻(Isochrysis galbana Parke 3011)的生长抑制、叶绿素a含量、总超氧岐化酶(T-SOD)活性以及丙二醛(MDA)含量的影响,研究了喹烯酮对两种微藻的毒性效应。实验结果如下:1)喹烯酮对微藻的生长抑制作用随浓度的增大而增大。喹烯酮对小新月菱形藻的24 h-EC_(50)为1.85 mg/L。喹烯酮对等鞭金藻3011的24 h-EC_(50)为0.41 mg/L,表明喹烯酮对等鞭金藻3011的生长抑制作用较小新月菱形藻更敏感。2)暴露24 h后,随着喹烯酮浓度的增大,小新月菱形藻叶绿素a含量下降;而等鞭金藻3011叶绿素a的含量基本不受影响。3)暴露24 h后,两种海洋微藻的总超氧化物歧化酶(T-SOD)活性随着喹烯酮浓度的升高均显著增加(P0.05),各实验组丙二醛(MDA)含量也高于对照组,其中等鞭金藻3011对喹烯酮更敏感,说明喹烯酮对两种微藻均能造成氧化胁迫,对细胞造成氧化损伤。研究表明,喹烯酮对两种微藻均具有显著急性毒性效应,对两种微藻而言,喹烯酮属于高毒物质,等鞭金藻3011对喹烯酮更敏感。本研究可为正确评价兽药喹烯酮的使用安全性提供基础数据。  相似文献

高温胁迫对球等鞭金藻3011和8701叶绿素荧光特性的影响   总被引:5,自引:3,他引:2  
摘要:本文以球等鞭金藻(Isochrysis galbana)3011和8701为实验材料,研究了高温胁迫(35-50℃)对其叶绿素荧光特性的影响。结果表明,与对照组相比,高温胁迫下2株金藻的叶绿素荧光参数Fv/Fm(PSⅡ的最大光能转化效率)、Fv/Fo(PSⅡ的潜在活性)、ΦPSⅡ(PSⅡ的实际光能转化效率)、rETR(相对光合电子传递效率)和qP(光化学淬灭)均明显降低,并且随着胁迫温度的升高,胁迫时间的延长,下降幅度也逐步增大。NPQ(非光化学淬灭)的变化趋势则与其他荧光参数不同,3011的NPQ随着胁迫温度的升高和胁迫时间的延长先上升后下降;8701的NPQ变化则与胁迫温度有关,35℃和40℃处理下NPQ随着胁迫时间的延长而逐渐上升,45℃处理下NPQ则先上升后下降。3011与8701的耐热性存在一定差异。3011在40℃下处理10min后,经过一定的时间后各荧光参数基本恢复,而在45℃和50℃下处理10min后,各参数均不能恢复。8701在35℃和40℃处理10min后,经过一定时间后,荧光参数Fv/Fm和Fv/Fo基本恢复到对照水平,而在45℃下处理10min后,各参数均不能恢复。本文还对高温胁迫下2株金藻的响应机制以及叶绿素荧光技术在筛选耐高温微藻品系中的应用进行了初步探讨。  相似文献

在室内采用正交试验的方法,对湛江等鞭金藻(Isochrysis zhanjiangensis)氮、磷、铁、碳、维生素B1、维生素B12等主要营养成分进行了优化,获得了以天然海水为基础的优化培养基:NaNO3 60mg/L,KH2PO4 4mg/L,FeC6H5O7 0.5mg/L,NaHCO3 1.0g/L,维生素B1 150μg/L,维生素B12 200 ng/L.试验结果表明,使用优化培养基培养湛江等鞭金藻具有生长速度快、产量高、成本低的优势.应用微生物流加技术培养湛江等鞭金藻可取得较好效果.  相似文献

金藻3011和8701与球等鞭金藻的18S rRNA基因研究   总被引:1,自引:0,他引:1

对金藻3011和8701的18S rRNA基因序列进行测定,分别获得1695 bp和1684 bp的DNA序列。应用DNAMAN,DNAstar和RnaViz 2.0生物软件,将获得的DNA序列与球等鞭金藻的18S rRNA基因序列进行DNA序列和RNA二级结构对比分析。DNA序列分析显示,三者的18S rRNA基因序列非常保守,相似性在99.5%以上,三者应同为球等鞭金藻;RNA二级结构分析显示,三者的RNA二级结构既存在球等鞭金藻种的特异性茎环结构,又存在明显的差异,并且从相近水域(山东)分离出来的3011和8701相对于采自挪威水域的球等鞭金藻具有更大的结构相似性;相对于3011,8701可能与球等鞭金藻具有更高的同源性。由此可见,单纯的18S rRNA基因序列分析可能不适用于球等鞭金藻种下水平的研究,但是其RNA二级结构分析对球等鞭金藻的物种鉴定,甚至是种下地理株的研究都具有非常重要的意义。  相似文献

运用单体培育法和群体培育法研究了绿色巴夫藻(Pavlova viridis)等4种藻类对褶皱臂尾轮虫(Brachionus plicatilis)的饵料价值并测定了它们对褶皱臂尾轮虫被甲长、被甲宽和侧棘距的影响。结果表明,绿色巴夫藻和亚心形扁藻(Platymonas subcordiformis)的饵料效果分别明显地优于大溪地等鞭金藻(Tahitian Isochrysis aff.galbana)和小球藻(Chlorela sp.)(P<0.01),而绿色巴夫藻与亚心形扁藻之间及大溪地等鞭金藻与小球藻之间无显著差异(P>0.01)。不同藻类对褶皱臂轮虫的被甲长、被甲宽和侧棘距有极显著的影响(P<0.01)。  相似文献

Fatty acids of three cultured unicellular algae wldely used in aquaculture ( Isochrysis galbana , clone T-iso, Chaetoceros calcitrans , and Chaetoceros gracilis ) were analyzed in detail by gas-liquid chromatography and mass spectrometry. Fatty acid compositions of the individual species, harvested at monthly intervals during a four month period, showed a very consistent pattern. Shorter chain (C16) fatty acids accounted for more than 70% of total fatty acids in the diatoms but less than 20% in I. galbana . Medium chain (C18) and the long chain 22:6w3 unsaturated fatty acids represented more than 50% of the total fatty acids in I. galbana but less than 10% of the total In the Chaetoceros spp. However, in the latter, 20:5w3 replaced 22:6w3 at an equivalent level. Fatty acid profiles of c. calcitrans and c. gracilis were similar, except that the latter contained lower levels of 16:2w4, 16:3w4 and 16:4w1, and higher levels of 18:2w6 and 18:3w6. In particular C. gracilis contained a higher proportion of the biochemically important fatty acid 20:4w6 (arachldonic acid). One of the salient features of I. galbana lipids was the presence of the fatty acid 18:5w3. This fatty acid is not generally reported in algae or invertebrate lipid analyses, but is a typical component of I. galbana and of other marine photosynthetic flagellates. Its potential for utilization by aquatic organisms is unknown.  相似文献

This research evaluated ozone treatment of three unicellular algal species commonly used in the culture of marine larvae. Isochrvsis galbana and Nannochloropsis oculata were found to be resistant to total residual oxidants (TRO); whereas, Chaetoceros gracilis was relatively sensitive to TRO. Isochrvsis galbana remained in log-phase growth after treatment with 0.05 mg/L to 0.50 mg/L TRO, and N. oculata exhibited strong growth after being exposed to TRO levels up to 0.67 mg/L. Only TRO levels of 0.92 mg/L and 0.90 tng/L reduced cell counts of N. oculata and I. galbana , respectively. Furthermore, algal cultures having higher concentrations at the time of ozone treatment may respond more favorably to the treatment. Initially, C. gracilis was reduced in cell count at TRO levels above 0.06 mg/L, but log-phase growth was resumed 2 d after exposure to TRO levels as high as 0.31 mg/L. It is concluded that I. galbana and N. oculata can be successfully treated with ozone in the mass production stages without hampering the timely production of concentrated cultures. Although C. gracilis was found to be relatively sensitive, ozone treatment of starter cultures can be performed routinely with this species. We have demonstrated that ozone treatment of algal cultures is a feasible technique, but studies need to be conducted on ozone treatment of other species of unicellular algae and on ozone treatment of varying concentrations of algae in order to enhance the applications of this technique. Also, in order to determine the extent of disinfection of algal cultures, bacterial and viral composition of the cultures should be evaluated in future studies.  相似文献

球等鞭金藻生长抑制物对自身藻细胞的抑制效应   总被引:2,自引:0,他引:2  
研究了生长抑制物1-羟基,丙二酸二乙酯-十二烯酸异丙酯(HDMA)对自身细胞生长、叶绿素和丙二醛(MDA)含量、胞外可溶性蛋白质和多糖含量、硝酸还原酶(NR)、超氧化物歧化酶(SOD)和过氧化物酶(POD)活性等生理指标的影响,分析HDMA对球等鞭金藻的抑制效应。结果表明,HDMA显著降低了细胞密度、叶绿素含量以及NR、SOD和POD活性,显著增大胞外可溶性蛋白质和多糖的比值,并且还明显提高MDA含量。因此,我们认为HDMA能够影响微藻细胞的某些生理生化过程。其中包括降低叶绿素含量,影响光合作用;改变胞外可溶性蛋白质和多糖的比值,增强细胞的疏水性,促使细胞絮凝;降低微藻硝酸还原酶和抗氧化体系酶的活性,影响细胞内营养平衡,并加速细胞体内活性氧的积累,导致细胞发生过氧化反应,从而影响微藻生长。  相似文献

The nutritive value of Isochrysis galbana (Tahitian), Chaetoceros calcitrans, Chaetoceros muel-leri . and their bi- and trialgal combinations for Pacific scallop Argopecten ventricosus spat culture was studied. Ingestion rate, growth, survival, and biochemical composition were the variables evaluated. Growth results showed C. calcitrans and I. galbana + C. calcitrans diets have the best nutritive values. Poorest results were obtained with I. galbana diet. The acceptability and a high lipid content of C. calcitrans could explain the good results of C. calcitrans and I. galbana + C. calcitrans diets along with a better balanced diet effect in the bialgal mixture. The low growth of the scallops using the C. calcitrans + C. muelleri diet, despite the high ingestion rate, is explained by a low energy content and an unbalanced nutrient supply. The gross biochemical composition of the diets did not show a clear relationship to spat gross biochemical composition.  相似文献

研究了等鞭金藻、三角褐指藻、扁藻对对虾育苗中致病菌鳗弧菌、假单胞菌的抑菌效果。结果表明,等鞭金藻、三角褐指藻、扁藻的水溶性物质及脂溶性物质对鳗弧菌有抑制作用;等鞭金藻、三角褐指藻的水溶性物质及等鞭金藻、三角褐指藻、扁藻的脂溶性物质对假单胞菌有抑制作用。在抑菌试验中,等鞭金藻、三角褐指藻、扁藻密度达10×104/ml时,对对虾育苗池水的抑菌作用较好,特别是对弧菌的抑制效果明显。  相似文献

To investigate the nutritional value of the marine micro‐alga Isochrysis galbana Tahitian Isochrysis strain (T.iso) as an alternative feed for aquaculture during culture age, its biochemical composition was studied under autotrophic and controlled culture conditions at different growth stages: exponential phase, early and late stationary phases and decay phase. Analysis showed that C14:0, C16:0, C16:1, C18:4 (n‐3) and C22:6 (n‐3) were the most abundant fatty acids in this alga at different growth stages. The highest values of monounsaturated fatty acids were recorded at the late stationary and the decay phases. However, the highest levels of polyunsaturated fatty acids were observed at the early stationary phase. At all growth stages, I. galbana (T.iso) contained arabinose, xylose, mannose, galactose and glucose. Glucose represented the main sugar, and its content per dry alga biomass weight increased with increasing age of the culture and reached about fourfold in the decay phase. The maximum protein content was also observed during this last phase.  相似文献
