采用埋植和离体方法进行克氏原螯虾的大颚器(MO)对卵巢发育作用的研究,埋植大颚器7次能显著提高成熟系数和促进卵径增大,处于不同卵巢发育时期的克氏原螯虾大颚器提取物(MOE)的离体研究发现,卵黄发生期的MOE对初级和次级卵黄发生期卵径增大均有极显著的作用,而对卵黄发生前期的卵径增大无作用,处于卵巢发育早期的MOE对卵黄发4生期卵巢小块总RNA含量升高无显著作用,处于卵黄发生前期,卵黄发生期和恢复期卵  相似文献

The effects of fulvic acid (FA) on survival and immune‐related gene expression were investigated in Litopenaeus vannamei challenged with Vibrio parahaemolyticus by immersion. Shrimp were fed with different dietary FA concentrations (1, 2, 4 and 6 g/kg feed) for 20 days (first bioassay) or 8 days (second bioassay, 2 g/kg feed of FA added every 2 days) and then challenged with V. parahaemolyticus. In a third bioassay, the expression of three immune‐related genes (translationally controlled tumour protein [TCTP], superoxide dismutase [SOD] and heat‐shock protein 70 [HSP70]) in haemocytes or hepatopancreas of experimental shrimp was measured by real‐time quantitative PCR at 0, 6, 12, 24, 48, 72 and 96 hr after FA (2 g/kg feed) administration. Fulvic acid increased survival at a concentration of 2 g/kg feed supplied every two days. Interestingly, TCTP gene expression was upregulated, whereas gene expression of SOD and HSP70 was downregulated. In conclusion, dietary fulvic acid improves survival in white shrimp challenged with V. parahaemolyticus and modulates the immune response. Therefore, FA merits further evaluation as prophylactic treatment in commercial shrimp farms.  相似文献

采用组织学方法系统研究了人工养殖条件下半滑舌鳎亲鱼卵巢的组织发育周年变化特征.结果表明,卵母细胞发育可分为6个时相,卵巢发育分为6期.卵巢不同发育时期都由不同类型的卵母细胞组成,半滑舌鳎为非同步分批多次产卵类型.周年发育过程中,6月龄前的鱼卵巢发育处于第Ⅰ期;9~12月龄卵巢可发育至第Ⅱ期;12月龄半滑舌鳎性腺可发育达到Ⅲ期,并一直保持至24月龄.3龄雌鱼达到性成熟,卵巢可发育至第Ⅳ期;随着水温和光周期的调控,卵巢发育进入成熟期(Ⅴ期),发生水合现象是卵母细胞成熟并准备排卵的信号;产卵结束后卵巢退化至第Ⅵ期,排出的卵母细胞退化吸收,以Ⅲ期卵巢越冬后,卵巢退化至Ⅱ期并持续至下一个繁殖周期.人工养殖条件下,卵巢内存在卵母细胞闭锁现象.性腺周年发育过程中,性腺指数(GSI)在性腺达到成熟期时达到峰值,产卵结束后性腺快速退化,GSI值各月份差异显著(P<0.05),亲鱼性腺重量在卵巢发育成熟过程中一直处于生长状态.亲鱼肝脏指数(HSI)在产卵前7~8月份(性腺达到成熟期前30 d左右)达到最大,表明7~8月份是性腺进行卵子发育、卵黄能量储备的重要时期,亲鱼在产卵前通过积极摄食储备体内能量,肝脏在卵母细胞卵黄积累方面具有重要的生理作用.肥满度(CF)值在繁殖盛期10~11月份达到最大,表现出与GSI类似的变化趋势.  相似文献

Eyestalk ablation is often used to induce ovarian development in female shrimp, but it possibly alters the animal’s hormonal balance, and affects its physiology and immune system. Therefore, this study was conducted to investigate alterations to the immune response during induced ovarian development by unilateral and bilateral ablation in Litopenaeus vannamei. Sub-adult female shrimp could be induced to undergo ovarian development over a 20-day period by both unilateral and bilateral eyestalk ablation; however, ovaries did not reach the final mature stage in which cortical rods are observed. In terms of immune response, total hemocyte count decreased significantly at 4 h after unilateral ablation, and at 20 days after bilateral ablation. After 20 days, expression in the hemocytes of the immune-related genes prophenoloxidase and peroxinectin in bilaterally ablated shrimp increased significantly, whereas that for penaeidin, for lipopolysaccharide- and β-glucan-binding protein, and for transglutaminase mRNA expression did not change significantly. The results of this study suggest that, while eyestalk ablation induces ovarian development, it may also bring about a hormonal imbalance that leads to immune-related genes being activated.  相似文献

vasa和PL10同属于DEAD-box基因家族,具有依赖ATP的RNA解螺旋酶活性。本研究中,克隆了日本沼虾DEAD-box家族的两个成员,分别为vasa(Mnv-asa)和PL10(MnP-L10)。其中Mn-vasa全长2 408bp,包含1 803 bp的开放阅读框,编码601个氨基酸;Mn-PL10全长2 607 bp,包含2 127 bp的开放阅读框,编码709个氨基酸。成体组织的组织检测表明,Mnv-asa仅在性腺中表达,而Mn-PL10在性腺和其他器官中均有表达。Mn-vasa和MnP-L10在卵巢的原位杂交结果显示:Mn-PL10和Mnv-asa都在卵黄发生前期,卵黄发生期的早期和中期表达,而在卵黄积累末期不表达,但Mn-PL10在卵黄发生中期定位于卵母细胞的两端,而Mn-vasa则是均匀的分布在卵母细胞细胞质中。  相似文献

研究了银鲳(Papus argenteus)肝及卵巢组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性、总抗氧化能力(T-AOC)及丙二醛(MDA)含量在整个卵黄发生过程中的变化情况,并分析了饲料n-3 LC-PUFA对卵黄发生期间组织中抗氧化水平的影响。分别以100%鱼油(FO组)、70%鱼油和30%大豆油(FSO组)、30%鱼油和70%大豆油(SFO组)、100%大豆油(SO组)为脂肪源,配制了4组等氮、等能及等脂的试验饲料。以1年龄雌性银鲳为试验对象,每组饲料设3重复,试验周期185 d。研究结果表明,肝与卵巢组织SOD、CAT活性、T-AOC水平及MDA含量在卵黄发生过程中均呈现逐渐升高的趋势,且卵黄发生后期各指标水平均显著高于卵黄发生前期(P0.05)。肝SOD(除SO饲料组外)、CAT活性、T-AOC水平及MDA含量在卵黄发生中期与前期之间未表现出显著性差异(P0.05),而卵黄发生中期FO与FSO饲料组卵巢SOD、CAT活性及T-AOC水平则均显著高于卵黄发生前期(P0.05)。FSO饲料组肝与卵巢SOD、CAT活性在卵黄发生过程中均为最高值,且在卵黄发生中、后期均显著高于SO饲料组(P0.05),但与FO饲料组无显著性差异(P0.05)。各饲料组间肝与卵巢T-AOC水平在卵黄发生前期均未表现出显著性差异(P0.05),而在卵黄发生后期,FO与FSO饲料组肝与卵巢T-AOC水平均显著高于SO饲料组(P0.05),但FO与FSO饲料组间无显著性差异(P0.05)。肝与卵巢中MDA含量随着饲料n-3 LC-PUFA含量的升高而呈现出升高趋势,且这种升高趋势在肝组织中表现更为明显,卵巢组织MDA含量在卵黄发生中、后期仅FO饲料组表现出显著性升高趋势(P0.05),其他各饲料组在卵黄发生各期均未表现出显著性差异(P0.05)。统计分析表明,银鲳卵黄发生过程中组织中抗氧化水平逐渐升高,适宜的饲料n-3 LC-PUFA含量(4.01%,FSO饲料组)可明显改善银鲳卵黄发生中期与后期组织中的抗氧化水平。双因素方差分析结果表明,试验饲料与卵黄发生时期对银鲳组织抗氧化水平均具有极显著性影响(P0.01),且两者对肝T-AOC水平与MDA含量存在显著性的交互作用(P0.05)。  相似文献

Plasma chemistry, lipid metabolism and vitellogenin gene expression of captive Sterlet sturgeon Acipenser ruthenus were studied in different maturity stages. A total of 32 fish were sampled, and maturity stages were identified on the basis of histological criteria and direct observation. Females were classified to four groups: previtellogenic, vitellogenic, post-vitellogenic, and atresia. Blood, gonad and liver tissue samples were taken through non-lethal biopsy. Our results showed that plasma levels of glucose, cholesterol, triacylglycerol, high-density lipoprotein, low-density lipoprotein, very low-density lipoprotein, calcium, phosphorus, alkaline phosphatase activity, albumin and total protein increased during ovarian development and were highest at post-vitellogenic stage. The lowest amounts in atresia stage demonstrate that lipid and energy imbalance was related to reabsorption and digestion of the yolk. These results suggested that the VLDL was the main plasma lipoprotein component of Sterlet. We determined that lipoprotein lipase and hepatic lipase activity increased during vitellogenesis process which suggested the role of lipase enzymes in regulating blood lipid metabolism. RT-PCR analysis indicates that Vitellogenin (VTG) mRNA could be detected both in livers and ovaries of female Sterlet. Throughout the study, the expression level of VTG gene showed an increase both in ovaries and in livers reaching its peak at late vitellogenesis stage. This strongly indicated a relation between VTG mRNA and ovarian development.  相似文献

The activity of the ovary-inhibiting hormone in the eyestalks of female prawns, Parapenaeopsis hardwickii, was determined during its ovarian developmental cycle. In the inactive (stage I) and post-spawned phases of ovarian development, the ovary-inhibiting hormone activity was found to be very high. During the phase of full vitellogenesis (stage IV), the inhibiting activity is almost negligible. After spawning (stage V) it is present, but at a lower level than during inactive and post-spawned phases. Hence it is inferred that ovary-inhibiting hormone appears to be responsible for the phase of ovarian quiescence and to control the rate of vitellogenesis.  相似文献

Heat shock proteins (HSPs) are a group of highly conserved molecular chaperones. Shrimp HSPs have recently been a topic of increasing interest because of their roles in shrimp immunity and homeostasis. In penaeid shrimp, HSP70s and the cognate forms, heat shock cognate (HSC) 70s, have been reported, but their responses towards various stimulations are different. We found a novel type HSP70 (MjHSP70-2) from the hyperexpansion of the large segmental duplication that is present in kuruma shrimp Marsupenaeus japonicus, which shows about 60 % identity with reported shrimp HSP70s. In a phylogenetic tree, MjHSP70-2 formed a sister clade with eukaryote HSP70 family while MjHSP70 was located close to the shrimp HSP70 and HSC70 group. MjHSP70-2 gene expression was not significantly increased by heat shock or pathogen challenge by Vibrio penaeicida, but it was significantly increased by infection with white spot syndrome virus. In contrast, MjHSP70 gene expression was increased by heat shock but decreased by infection with V. penaeicida. The kuruma shrimp genome was found to have 400-fold more copies of the MjHSP70-2 gene than the putative single-copy gene transglutaminase. In conclusion, our results reveal the presence of a novel type HSP70 gene, HSP70-2, from kuruma shrimp. There are multiple forms of HSP70 in crustaceans, and these HSP70s behave differently under various stressors.  相似文献

In this study, four species of penaeid shrimp from Pakistan's inshore waters were studied. These were Penaeus penicillatus (Alcock), Penaeus merguiensis (de Man), Metapenaeus affinis (Milne Edwards) and Parapenaeopsis stylifera (Milne Edwards). During maturation, shrimp ovaries pass through a series of colour changes. The various colours observed in the ovaries of the four species of penaeid shrimps were translucent, white, cream, yellow, green‐yellow, green‐white, light green and dark green. The different type of oocytes observed in histological sections of the penaeid ovaries are as follows: chromatin nucleolar oocytes, perinucleolar oocytes, yolkless oocytes, yolky oocytes and oocytes with cortical bodies. The ovarian developmental stages recognized in these shrimps are: undeveloped, developing, nearly ripe, fully ripe, resorbing and resorbing/ developing. The ovarian developmental stages were similar in all the four species with the exception of the fully ripe stage. In the fully ripe stage of P. penicillatus and P. merguiensis, the cortical bodies were present at the periphery in two shapes; spherical as well as rod‐like. On the other hand, in the fully ripe stage of M. affinis and P. stylifera, the cortical bodies were present only at the periphery and were only spherical. A relationship between colour of ovaries and histological stages of ovarian development was established. A female shrimp with a dark‐green ovary is an indication of the ready‐ to‐spawn condition. In P. merguiensis and Penaeus penicillatus, respectively, 100% and 88.1% females with dark‐green ovaries had oocytes with cortical bodies, whereas in M. affinis and P. stylifera, respectively, 50% and 28.6% females with dark‐green ovaries had oocytes with cortical bodies.  相似文献

This study examines puberty in female yellowtail flounder, correlating macroscopic, histological and endocrine parameters at the gonadal level. Short-term ovarian tissue incubations were used to study estradiol-17β production in relation to oogenic stage. Examining 2 year classes of young flounder demonstrated that cultured females retain phenotypic plasticity in reproductive age with pubertal onset occurring in one-year, two-year and three-year-olds. Immature ovaries were steroidogenic and capable of responding to gonadotropic stimulation. Endocrine puberty in females was detected by a peak in estradiol-17β production during the cortical alveolar oocyte stage prior to any ovarian evidence of vitellogenesis. Puberty, once initiated, proceeded to ovulation within 8 to 12 months; vitellogenic oocyte development followed the group synchronous pattern. Estradiol-17β was clearly the dominant of the two measured plasma hormones during pubertal onset and throughout vitellogenesis. Plasma testosterone was consistently detectable at low levels by mid-to late-vitellogenesis. Ovaries showed the highest sensitivity to gonadotropic stimulation in vitro during late-vitellogenesis. Variable plasma levels in both estradiol-17β and testosterone occurred in preovulatory and ovulating females during the captive spawning period. Together the results show that yellowtail flounder can mature at a young age and small size when culture conditions permit. In addition, the early sensitivity to gonadotropin by the immature ovaries may be an important physiological determinant for the timing of puberty in this species.  相似文献

十足目甲壳类卵巢中卵黄的来源,在过去的几十年研究中一直存在争议,内源性合成和外源性合成都有报道。本文以雌性中国对虾为实验材料,根据组织学观察确定其发育阶段,试验虾可以分为:卵巢未发育期;卵黄发生前期;初级卵黄发生期和次级卵黄发生期4个时期。从处于不同卵巢发育期对虾的卵巢和肝胰腺中提取总RNA,用RT-PCR的方法初步探讨不同发育期的中国对虾卵巢和肝胰腺中卵黄蛋白原mRNA的表达,确定是否存在卵黄蛋白原的合成功能。结果发现,从卵巢未发育期到次级卵黄发生期的卵巢和肝胰腺中都有卵黄蛋白原mRNA的表达,说明二者为卵黄蛋白的合成部位。  相似文献

Molecular cloning, characterization, and functional analysis of follicle-stimulating hormone receptor (FSHR) in female turbot (Scophthalmus maximus) were evaluated. Results showed that the full-length FSHR cDNA was 3824 bp long and contained a 2202 bp open reading frame that encoded a mature protein of 733 amino acids (aa) and a signal peptide of 18 aa. Multiple sequence analyses showed that turbot FSHR has high homology with the corresponding genes of other teleosts and significant homology with that of Hippoglossus hippoglossus. Turbot FSHR has the typical structural architecture of glycoprotein hormone receptors consisting of a large N-terminal extracellular domain, seven transmembrane domains and short C-terminal intracellular domain. FSHR mRNA was found to be abundant in the ovaries, but deficient in eyes, intestine, brain, muscle, gills, spleen, stomach, heart and kidney. Furthermore, FSHR mRNA was found to increase gradually from pre-vitellogenesis to migratory nucleus stages, with the highest values observed during the late vitellogenesis stage of the reproductive cycle. However, FSHR mRNA was found to decrease dramatically during the atresia stage. Meanwhile, functional analysis with HEK293T cells continual expressing FSHR demonstrated that FSHR was specifically stimulated by ovine FSH, but not ovine LH. These results indicate that turbot FSHR is mainly involved in the stimulation of vitellogenesis, regulation of oocyte maturation as well as promotion of ovarian development via specific ligand binding. These findings open doors to further investigation of physiological functions of FSHR, which will be valuable for fish reproduction and broodstock management.  相似文献

Heat shock proteins (HSPs) are proteins that are expressed more strongly when the cells are exposed to physiological and stressful conditions. In this study, the full-length cDNAs of heat shock proteins 40 (MjHSP40), 70 (MjHSP70) and 90 (MjHSP90) were cloned from kuruma shrimp Marsupenaeus japonicus. The open reading frames (ORFs) of the cDNA clones have lengths of 1,191, 1,959 and 2,172 bp and encode 396, 652 and 723 amino acid residues, respectively. The predicted MjHSP40 amino acid sequence contains a J domain, a glycine/phenylalanine-rich region, and a central domain containing four repeats of a CxxCxGxG motif, indicating that it is a type I HSP40 homolog. The signature sequences of the HSP70 and HSP90 gene families are conserved in the MjHSP70 and MjHSP90 amino acid sequences. The deduced amino acid sequences of MjHSP70 and MjHSP90 share high identity with previously reported shrimp HSP70s and HSP90s, respectively. The expression of MjHSP90 mRNA increased at 32°C. Additionally, the expressions of MjHSP40, MjHSP70 and MjHSP90 mRNAs increased in defense-related tissues (i.e., hemocytes and lymphoid organ) when the shrimp were challenged with white spot syndrome virus.  相似文献

利用cDNA末端快速扩增技术克隆出凡纳滨对虾(Litopenaeus vannamei)核自身抗原精子蛋白(NASP)基因,NASP基因c DNA全长2258 bp,其中包括92 bp 5′端非编码区,147 bp 3′端非编码区及2019 bp开放阅读区,编码673个氨基酸,预测分子量为74.18 k D。该序列已提交至Gen Bank,序列号为KT274811。在NCBI上进行序列比对后发现,其与斑节对虾(Penaeus monodon)NASP序列相似性最高,为90%。采用荧光定量PCR方法测定了不同发育时期卵巢与肝胰腺中NASP m RNA相对表达水平,结果表明,NASP在卵巢中的表达量高于肝胰腺中的表达量,且在Ⅱ期表达量最高,Ⅲ期表达量最低。此外,通过构建系统进化树比较了凡纳滨对虾NASP与其他物种间的遗传距离。实验结果为进一步研究该基因在凡纳滨对虾卵巢发育中的作用提供了依据。  相似文献

Changes in serum concentrations of estradiol-17β, testosterone, 17α,20βdihydroxy-4-pregnen-3-one, and vitellogenin were investigated during ovarian development induced by injection of a salmon pituitary extract in cultured European eel Anguilla anguilla . Vitellogenesis was induced with a weekly dose of 50 mg pituitary extract/kg body weight. In eels receiving that dose, gonadsomatic indices ranged from 20–43% after the 10th–11th weekly injection. Body weights were relatively stable during vitellogenesis, but increased dramatically during final maturation. Serum estradiol-17β levels increased slightly during vitellogenesis and peaked at an average of 6.95 ng/mL at final maturation. The profile of serum vitellogenin followed that of estradiol-17β which increased markedly from an average of 0.36 to 20.72 mg/mL. Control levels of vitellogenin were undetectable throughout the study. Average serum levels of testosterone increased to a peak of 17.74 ng/mL in the early stage of vitellogenesis, followed by a sharp drop to initial levels (3.86 ng/mL) in the late stage of vitellogenesis, and then increased again to an average of 8.84 ng/mL at the final maturation stage. Serum 17α,20β-dihydroxy-4-pregnen-3-one was not detected (<50 pg/mL) throughout the experiment. Profiles of serum estradiol-17β, testosterone, and vitellogenin observed during ovarian development appear different from those found in salmonids and other teleosts. This study indicates, however, that cultured European eels are a useful model for study of gonadal maturation in the eel.  相似文献

We studied the effects of dietary n‐3 LC‐PUFAs on the activities and mRNA expression levels of tissue lipoprotein lipase (LPL) and fatty acid synthase (FAS) during vitellogenesis and ovarian fatty acid composition in female silver pomfret broodstock. Broodstock were fed one of four experimental diets for 185 days: FO (100% fish oil), FSO (70% fish oil + 30% soybean oil), SFO (30% fish oil + 70% soybean oil) or SO (100% soybean oil). The results revealed that hepatic LPL and FAS and ovarian FAS activities and mRNA expression levels significantly increased at vitellogenesis and postvitellogenesis relative to previtellogenesis, with no significant differences between these two stages, except for hepatic LPL mRNA expression. Dietary n‐3 LC‐PUFAs decreased tissue FAS and increased LPL activities and mRNA expression levels. The ovarian concentrations of 20:4n‐6 (ARA), 20:5n‐3 (EPA), 22:6n‐3 (DHA) and n‐3 LC‐PUFAs were directly influenced by n‐3 LC‐PUFA levels. Total n‐3 LC‐PUFA concentrations in SO were 57% lower than those in FO, while 18:2n‐6 concentrations in SO were 4.7 ×  higher than those in FO. These results revealed that high dietary n‐3 LC‐PUFAs levels significantly affected tissue lipid metabolism in female silver pomfret broodstock during vitellogenesis by upregulating LPL and downregulating FAS.  相似文献
