This experiment was conducted to study the effects of Bacillus preparations on immunity and antioxidant activities in grass carp. A total of 315 grass carp, with similar initial weight (average weight of fish 45?g), were randomly divided into three groups with three replicates. The control group was fed the basal diet without Bacillus; treatment group 1 was added Bacillus preparation no. 1 with 1?×?108?cfu/m3 per 7?days in culture water and also fed the basal diet; treatment group 2 was fed the basal diet mixed with 0.5?% Bacillus preparation no. 2, and the culture water was added 1?×?108?cfu/m3 Bacillus preparation no. 1 per 7?days. After 4?weeks of culture, 12 grass carp from each replicate were taken randomly for the determination of immune response and oxidization resistance indices. The objective of the study was to evaluate the efficacy of 2-phenoxyethanol (2-PE) as an anesthetic in great sturgeon under two experiments. First, fish were exposed to 0.1, 0.3, 0.5, 0.7 and 0.9?mL/L 2-PE, and time to induction (deep anesthesia) and recovery from anesthesia were measured. At concentration of 0.1?mL/L, 2-PE failed to induce deep anesthesia in fish, whereas at concentrations of 0.7 and 0.9?mL/L, all the fish were anaesthetized within 3?min of exposure. For assessing the impact of effective concentrations of 2-PE on physiological responses of great sturgeon, hematological indices, plasma metabolites, electrolytes, enzymes and cortisol levels were measured. The use of 2-PE induces a significant increase in RBC values at 0.3?mL/L concentration and a parallel increase in hemoglobin and hematocrit values. 2-PE anesthesia had no effect on WBC, MCV, MCH and MCHC levels when compared to control group. Serum glucose, cholesterol and cortisol levels were significantly high in 0.3 and 0.5?mL/L 2-PE. Moreover, AST levels were increased in fish exposed to the 0.3?mL/L 2-PE comparing with the control group (P?<?0.05). There were no significant differences in serum levels of total protein, triglycerides, ALP, ALT, Cl?, Na+, K+, Ca2+ and Mg2+. In this study, alteration in hematological and serum biochemical indices was time-dependent. This study demonstrates that rapid induction of deep anesthesia with a relatively high concentration of 2-PE (0.7 and 0.9?mL/L) was associated with the lowest effects on the hematological and serum biochemical indices in great sturgeon and therefore would be recommended as eligible doses for hematological studies in this species.  相似文献

The aim of this study was to evaluate the effects of Bacillus subtilis and Bacillus licheniformis on growth, gut microbiota, and digestive enzyme activities of Artemia urmiana. Three diets containing 102 (T1), 104 (T2), 106 (T3) CFU of probiotics/g feed, and a control diet (C) without probiotic were used through a completely randomized design (treatments with triplicates). Twelve plastic tanks with the capacity of 60- l and density of 20 nauplii/ml were used and the trial continued for 15?days. Results showed that probiotics significantly increased the total length of A. urmiana (P?<?0.05). Although the total aerobic gastrointestinal bacteria count showed no significant differences among the treatments, the total Bacillus count significantly increased in experiments (P?<?0.05). The ratio of TCBS to total aerobic bacteria count was significantly lower in T1 (0.31?±?0.05), T2 (0.27?±?0.15), and T3 (0.25?±?0.05) compared to the control (0.76?±?0.34) (P?<?0.05). The objective of this study was to assess the effects of stocking density on growth performance, serum biochemical parameters, and muscle texture properties of genetically improved farmed tilapia (Oreochromis niloticus, GIFT). Juvenile GIFT with an average initial weight of 12.54?±?0.45 g (mean?±?SD) were randomly stocked in 16 tanks (80 L) in a recirculation aquaculture system at four densities of 10 (D1), 20 (D2), 30 (D3), and 40 (D4) fish per tank for 56 days, with quadruplicate for each density. Nitrite (NO2?) contamination of water can severely impact the health of aquatic life and is a major concern for commercial aquaculture. In order to study the effect of nitrite on Aristichthys nobilis, we investigated the oxygen-carrying capacity, NF-κB/HIF-1α pathway, and the gill tissue structure under nitrite stress. In the current study, bighead carp (initial weight 180.05?±?0.092 g) were exposed to nitrite (48.634 mg/L) for 96 h and then for 96 h recovery test. After nitrite exposure for 6 h, hypoxia-inducible factor-1α (HIF-1α) and nuclear factor-κB (NF-κB) mRNA expression increased significantly in the gill of bighead carp (P?<?0.05). The present study was carried out to evaluate the effect of dietary taurine (Tau) on performance, digestive enzymes, antioxidant activity, and resistance of common carp, Cyprinus carpio L., fry to salinity stress. A 5-week feeding trial was conducted to evaluate the effects of biofloc in situ and commercial probiotic supplementation on white shrimp (1.87?±?0.03 g) inorganic nitrogen control, growth, and immunophysiological response. Effect of ambient copper on the immune, antioxidant and lipid peroxidation parameters of Fenneropenaeus indicus and its susceptibility to white spot syndrome virus under heavy metal (Cu2+) exposure has been studied. Adult shrimps were acclimated to 25‰ salinity for a period of 7?days, and after 12?h of starvation, shrimps were dosed with 0.075, 0.150, 0.225 and 0.30?ppm Cu2+ by adding appropriate quantities of copper sulphate solution. After 14?days of metal exposure, the shrimps were challenged with white spot syndrome virus through oral administration and the immune and antioxidant parameters were analysed. Analysis of variance showed that there were significant differences (P?<?0.05) in the immune and antioxidant parameters and lipid peroxidation product in different treatment groups of F. indicus compared to untreated group. The immune parameters and antioxidant enzymes have been significantly lowered (P?<?0.05) in the haemolymph of shrimps dosed with higher concentrations of copper sulphate leading to lipid peroxidation and accumulation of malondialdehyde. However, there was less oxidative stress in shrimps exposed to 0.075?ppm Cu2+. The present study showed that high levels of Cu2+ enhanced the mortality of F. indicus with concomitant reduction in immune and antioxidant parameters. A concentration of 0.075?ppm Cu2+ in the rearing water was found to have beneficial effect in shrimps in terms of immunostimulation and higher survival against WSSV infection. In this study, the effects of oral administration of different levels of Dunaliella salina (a natural β-carotene source) on growth parameters, immunological and hematological indices, as well as skin carotenoids, of Heros severus were investigated. One hundred and eighty H. severus weighing 27 ± 0.5 g were divided randomly into four groups in triplicate (15 fish in each replicate). Groups 1–4 received food supplemented with 0, 50, 100 and 200 mg kg?1 D. salina powder, respectively. After 6 weeks, the growth parameters were compared among the groups. Blood samples were taken from each group, and hematological parameters including red blood cell count (RBC), white blood cell count (WBC), hematocrit (PCV), hemoglobin (Hb) and immunological indices (serum and mucus lysozyme and bactericidal activity, resistance against Aeromonas hydrophila infection) as well as carotenoid content of skin were evaluated. Results showed that some growth indices increased significantly in fish fed with 100 and 200 mg kg?1 D. salina-supplemented food (P < 0.05). Although serum lysozyme activity was increased in fish fed with food supplemented with 100 and 200 mg kg?1 D. salina (P < 0.05), no significant change was observed in serum and mucus bactericidal activity and mucus lysozyme activity among the groups (P > 0.05). Most of the hematological parameters such as WBC, RBC, PCV and Hb significantly increased in D. salina-treated fish compared with controls (P < 0.05). Mortality induced after challenge with A. hydrophila in 200 mg kg?1 D. salina-treated fish was 36.67 %, which significantly decreased compared with control (P < 0.05). Skin carotenoid content in all D. salina treatments was statistically higher than that of control (P < 0.05). The effects of replacing fish meal (FM) with corn gluten meal (CGM) on growth and physiological performance were evaluated in common carp (Cyprinus carpio). Six experimental diets were formulated by substituting 0 (control), 20, 40, 60, 80, and 100% CGM protein for FM protein. The concentrations of dietary protein in the experimental diets were 27.8–29% and the P:E ratios were 14.7–15.46 mg/kJ. Calpain is known to be the most important endogenous protease responsible for degrading myofibril protein, thus affecting meat quality. This study aimed to investigate the activation of calpain and its potential influence in fish after slaughter. Calpain activity, expressions of the calpain inhibitor and activator (calpastatin and UK114, respectively), and effects of influencing factors on calpain activity and on the necessary Ca2+ activation concentration was investigated. Calpain activity increased significantly during the first 5 h of storage followed by rapid decline (P?<?0.05). Three calpastatin expression bands were detected, and the 120-kDa calpastatin was significantly down-regulated with storage time (P?<?0.05), whereas the level of UK114 in tilapia muscle was significantly up-regulated during postmortem storage (P?<?0.05). Calpastatin and UK114 showed opposite effects on calpain activity; a significant decrease and increase in calpain activity were observed when crude calpain was incubated with calpastatin and UK114, respectively (P?<?0.05). Furthermore, a significant increase was observed in calpain activity following co-treatment with calpastatin and UK114 (P?<?0.05), but lower than that of treatment with UK114. The calpain activity in the UK114-treated group was higher than that of the control and calpastatin with the same Ca2+ concentration. Calpastatin inhibited calpain activation, whereas UK114 promoted the activation of calpain by reducing the concentration of Ca2+ required. This experiment was carried out to investigate the potential probiotic properties of Bacillus subtilis and suitable methods of administration to the commercial feed in juvenile Macrobrachium rosenbergii (de Man) from 25 August to 25 November 2008. Putative Bacillus subtilis bacterium isolated from juvenile M. rosenbergii intestine was added to commercial prawn feed as a probiotic. Five types of diets were prepared by mixing B. subtilis at level of 108 cells g?1 with commercial feed using different methods consisting mixing, soaking, spraying and bathing. After 60?days, the prawns fed diets at soaking method treated group, showed a higher mean weight gain (2.09) or 328.84% increase in growth over control. There were significant differences (P?<?0.05) in weight gain, feed intake and FCR among soaking method and other treated and control groups. There was significant difference (P?<?0.05) in survival among treated and control groups, but no significant difference (P?>?0.05) in water quality and biochemical composition among treated and control groups. Clearly, B. subtilis-treated diets appeared to enhance growth and survival of juveniles M. rosenbergii. The effect of four iso-nitrogenous and iso-lipidic diets containing soy lecithin and lysolecithin with fish oil (sardine) and palm oil on growth, digestibility, and fatty acid composition of tail muscle and non-muscle portions of tiger shrimp, Penaeus monodon, was evaluated. Shrimp fed with lysolecithin diets had significantly (P?<?0.05) higher daily growth coefficient values (1.40–1.45% day?1) than those fed with soylecithin containing diets (1.32–1.37% day?1). Arachidonic, eicosapentaenoic, and docosahexaenoic acid contents of muscle and non-muscle portions of shrimp were significantly (P?<?0.05) higher in lysolecithin-supplemented diet compared to soylecithin-supplemented diet. The aim of this study was to explore the artificial hibernation of crucian carp for?waterless preservation and to characterize the quality and biochemical properties during and after the hibernation. Anesthetized crucian carp using eugenol were stored at 8?°C with 90?% oxygen and 95–100?% relative humidity for 38?h and then transferred to fresh water to recover. Liquid loss and cooking loss had no significant changes (p?>?0.05). The total volatile basic nitrogen content and 2-thiobarbituric acid value in hibernated fish were significantly higher (p?<?0.05) than fresh and recovered groups. Serum cortisol, glutamic-oxaloacetic transaminase (GOT), alkaline phosphatase (AKP), and acid phosphatase (ACP) activities significantly increased (p?<?0.05) during hibernation, while glutamic-pyruvic transaminase (GPT) had no significant change (p?>?0.05). Both ACP and AKP activities decreased upon the fish recovered, but only the ACP activity returned to normal. However, there were increased serum glucose concentration, GOT and GPT activities in recovered fish. On the basis of these findings, it can be concluded that the artificially hibernated life of crucian carp was 38?h by the combination of anaesthetizing and low temperature. The increase of water temperature due to global warming is a great concern of aquaculturists and fishery biologists. In the present study, we examined the effects of high temperature on hematological parameters and blood glucose levels in striped catfish, Pangasianodon hypophthalmus exposed to three temperature conditions (28, 32, and 36 °C) for 7 days. Fish were sacrificed at days 1, 3, and 7. Erythroblasts (Ebs), erythrocytic cellular abnormalities (ECA), and erythrocytic nuclear abnormalities (ENA) were assayed using peripheral erythrocytes of the sampled fishes. Hemoglobin (Hb) and red blood cell (RBC) significantly (P?<?0.05) decreased at 36 °C after 3 and 7 days of exposure, whereas white blood cell (WBC) showed opposite scenario. Blood glucose levels significantly (P?<?0.05) increased at 36 °C on day 3. Frequencies of Ebs, ECA, and ENA were found to be elevated at increased temperature. Differential leucocytes count showed significant increases in neutrophil and decreases in lymphocytes in the highest temperature (36 °C). Dissolved oxygen decreased and free CO2 increased significantly (P?<?0.05) with increasing temperature, while the pH and total alkalinity of the water were almost unchanged throughout the study period. Eleven isoproteic (ca. 47%) and isoenergetic (ca. 20.5 MJ kg?1) diets were formulated including a control diet, in which 60% of dietary nitrogen were provided by intact protein (fish meal, gelatin, and wheat meal) and 40% by crystalline AA. The other 10 diets were formulated by 40% reduction in each EAA from the control diet. At the end of the experiment, fish fed with threonine-deficient diet showed the lowest survival rate (P?<?0.05), whereas growth performance decreased in fish fed all EAA-deficient diets, although the reduction in body growth varied depending on the EAA considered. Pancreatic enzymes (trypsin, lipase, α-amylase, and carboxypeptidase A) activities significantly decreased in fish fed the EAA-deficient diets in comparison with fish fed the control diet (P?<?0.05). Fish fed with the arginine-deficient diet had the highest plasma and liver alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase levels (P?<?0.05). Plasma and liver lactate dehydrogenase and superoxide dismutase showed the highest and lowest values, respectively, in fish fed the arginine and lysine-deficient diets (P?<?0.05). Plasma metabolites were significantly affected by dietary EAA deficiencies (P?<?0.05). The results of this study suggesting dietary EAA deficiencies led to reduction in growth performance as well as pancreatic and liver malfunction. This study was conducted to evaluate the effects of Euphorbia hirta leaf extract on the growth performance, hematological and organosomatic indices of hybrid catfish, Clarias macrocephalus?×?C. gariepinus. The fish were treated with 0 (control), 300, 500 and 800 mg/kg (ppm) for 90 days. The weight gain, average daily growth rate, and specific growth rate were at significantly higher levels in fish from all the treatment groups on days 75 and 90, while the feed efficiency and protein efficiency ratio were consistently higher in fish from all the treatment groups from day 60 up until day 90. The feed conversion rate significantly decreased from day 60 until day 90 in all treatment groups when compared with the control group, and the survival rate was significantly different from day 30 until day 90; a consistently higher rate was observed in fish fed 800 mg/kg. The highest viscerosomatic index and intraperitoneal fat were observed in the group fed 500 mg/kg (p?<?0.05). The hepatosomatic index was significantly increased alongside increasing levels of E. hirta extract. The total white blood cell count in the control group was significantly higher on day 30, but on day 90 all the treatment groups showed higher levels. Hematocrit percentage was significantly different on days 30 and 90. Lymphocyte, eosinophil and thrombocyte levels were shown to be significantly different (p?<?0.05) when different groups of fish were compared. In the current study, laboratory evaluations were made to assess the immunomodulatory effect of cypermethrin on fingerlings of common carp (Cyprinus carpio L.). Results showed that 96-h LC50 of cypermethrin in common carp was estimated at 0.85 μg/L. Fish were exposed for 21 days to cypermethrin at three sub-lethal concentrations of 0.042, 0.085, and 0.17 μg/L that represented 5, 10, and 20%, respectively, of the 96-h LC50 of the pesticide for this fish species. Blood samples were taken after 7, 14, and 21 days of exposure. Immunological indices and resistance against bacterial infection were determined. Compared to the control group, the fish exposed to cypermethrin showed a significant increase in neutrophil ratio but exhibited a significant decrease in leukocyte number and lymphocyte ratio in treatments exposed to 0.17 and/or 0.085 μg/L after 21 days of exposure (p < 0.05). Serum protein level was significantly decreased in group exposed to 0.17 μg/L on day 14 and also in groups exposed to 0.085 and 0.17 μg/L on day 21 (p < 0.05). Immunoglobulin value was significantly reduced in groups exposed to 0.085 and 0.17 μg/L after 21 days of exposure (p < 0.05). Serum lysozyme activity and phagocytic activity were significantly decreased following exposure to 0.17 μg/L determined on days 14 and 21, post-exposure (p < 0.05). Mortality rate following the challenge with Aeromonas hydrophila significantly increased in fish exposed to 0.17 μg/L of cypermethrin. Overall, the present results indicate severe immunotoxicological effects of cypermethrin in common carp. Therefore, the use of cypermethrin in the proximities of common carp farms should be carefully considered.  相似文献

Replacement of fish meal with squilla silage in practical diet for the juvenile giant freshwater prawn, Macrobrachium rosenbergii, was evaluated. Five iso-nitrogenous and iso-caloric diets were prepared in which protein requirements were met with 100% fish meal (control, diet 1) and squilla silage replacing fish meal at 25, 50, 75 and 100% (diets 2, 3, 4 and 5, respectively). Water stability and water absorption of five feeds revealed significant differences (P?<?0.05). The weight gain varied between a maximum of 248.50?±?34.07% in diet 4 to lowest 132.45?±?14.34% in diet 1. The average daily gain in weight in grams varied from 0.091?±?0.01 (diet 5) to 0.154?±?0.04 (diet 3), whilst the specific growth rate and gross growth coefficient values were recorded highest in diet 4. Highest survival rates (86.66?±?5.77%) were recorded in both experimental diets 4 and 5. Low feed conversion ratio of 1.19?±?0.21 was recorded from diet 4, whilst the highest value of 2.16?±?0.09% was estimated from the diet 5. Proximate carcass composition showed highest crude protein content in post-larvae fed with diet 5 (47.24%) in contrast to lowest diet 1 (36.12%). Lipid values fluctuated significantly between 2.45 and 7.18% (P?<?0.05) between treatments. The ash content significantly increased from diet 1 (16.34%) to diet 5 (20.69%) (P?<?0.05). Highest feed efficiency (gain/feed) value was recorded in diet 4 (1.52?±?0.22), whilst the diet 5 showed lowest value (0.99?±?0.06).
