2002年采用PCR-核酸探针斑点杂交法检测了乳山对虾养殖场1000余份样品携带白斑综合征病毒(WSSV)的情况。结果显示,639例对虾样品中阳性检出率为26·6%;77例蟹类样品中阳性检出率为18·2%;266例浮游动物样品中阳性检出率为38·3%,3~9月份浮游动物阳性率呈下降趋势,消毒后水体中浮游动物的阳性率仍很高;30例贝类样品检测均为阴性;204例底泥样品中,阳性检出率为17·6%,22例抽滤海水样品检测均为阴性。结果表明,虾、蟹类在传播WSSV中起着重要作用,贝类、海水传播WSSV的可能性很小,浮游动物、底泥在传播WSSV中的作用和机制应引起高度重视。  相似文献

摘要:免疫活性物质可以调动或激活虾类自身的免疫系统,提高动物的免疫机能,增强动物的抗病毒能力。有关卵黄抗体对对虾体内酶活力及抗病毒能力的影响,国内外尚未见报道。本实验通过连续投喂的方法,用3个水平(1%、0.5%、0.1%)的Ig-Guard(shrimp)制成的试验饲料,同时以基础饲料为空白对照饲喂凡纳滨对虾(Litopenaeus vannamei) 20 d,分别测定了第5,10,15,20 d血淋巴的酚氧化酶(PO)、溶菌酶(UL)、酸性磷酸酶(ACP)及肌肉匀浆液的超氧化物歧化酶(SOD)等非特异性免疫因子活性,并对血清及肌肉匀浆液中蛋白进行定量。结果表明,免疫组的PO、UL、ACP、SOD活力均显著高于对照组(P<0.05)。免疫20 d后,用白斑综合征病毒(WSSV)投喂感染。攻毒后第7 d各免疫组的相对免疫保护率分别为17.95%、23.08%、35.90%。实验结果说明,Ig-Guard(shrimp)能有效提高对虾免疫因子的活性,对于提高抗WSSV感染能力也有一定作用。将对虾免疫因子活性和累计死亡率协同分析,摄食低浓度Ig-Guard(shrimp)组较之高浓度组的酶活力高,其累计死亡率低,故笔者建议适当地投喂低浓度Ig-Guard(shrimp)更为合理。  相似文献

Dendronereis spp. (Peters) (Nereididae) is a common polychaete in shrimp ponds built on intertidal land and is natural food for shrimp in traditionally managed ponds in Indonesia. White spot syndrome virus (WSSV), an important viral pathogen of the shrimp, can replicate in this polychaete (Desrina et al. 2013); therefore, it is a potential propagative vector for virus transmission. The major aim of this study was to determine whether WSSV can be transmitted from naturally infected Dendronereis spp. to specific pathogen‐free (SPF) Pacific white shrimp Litopenaeus vannamei (Boone) through feeding. WSSV was detected in naturally infected Dendronereis spp. and Penaeus monodon Fabricius from a traditional shrimp pond, and the positive animals were used in the current experiment. WSSV‐infected Dendronereis spp. and P. monodon in a pond had a point prevalence of 90% and 80%, respectively, as measured by PCR. WSSV was detected in the head, gills, blood and mid‐body of Dendronereis spp. WSSV from naturally infected Dendronereis spp was transmitted to SPF L. vannamei and subsequently from this shrimp to new naïve‐SPF L. vannamei to cause transient infection. Our findings support the contention that Dendronereis spp, upon feeding, can be a source of WSSV infection of shrimp in ponds.  相似文献

“对虾白斑病的围栏封闭预防技术”是一项针对沿海低位池塘、以切断传染病源为主旨、以设置防蟹围栏为特色的对虾白斑病综合预防技术。其主要措施为:首先在贮水(兼水处理)池和养殖池周围设置防蟹围栏,将该等池塘封闭起来,使杂蟹不会从陆地侵入池内;然后通过60目滤网进水,并用敌百虫或敌杀死处理上述两种池塘,杀灭以蟹类为主的一切甲壳动物;解毒后,放养经检测不带白斑症病毒的虾苗;采取封闭式养殖,养殖期中只向养殖池加水以补充渗漏和蒸发的水损失;加水时,先将海水在贮水池中用药物处理,杀灭其中的传染该病的媒介生物(主要是桡足类),药物解毒后再注入养殖池。此外,作为辅助措施,在养殖期中从多方面尽量保证虾池中有一个良好的生态环境。该技术已经过预备研究、实验研究和扩大试验等阶段,均证明效果良好。已扩大试验了近33.33hm^2(其中实际养虾池约17.33hm^2),成功率达96%左右。  相似文献

对使用同种饲料喂养的海水和淡水养殖凡纳滨对虾(Litopenaeusvannamei)进行了感官评价、一般成分和呈味物质分析。海水养殖对虾感官评价总体得分高于淡水养殖对虾,海水养殖对虾在鲜味、甜味上优于淡水养殖对虾,而淡水养殖对虾带有明显的土腥味(P<0.01)。海水养殖对虾的灰分高于淡水养殖对虾,粗蛋白、水分及有机酸含量两类对虾差异不大。通过分析氨基酸、核苷酸、甜菜碱和有机酸等呈味物质,发现主要是谷氨酸、AMP、IMP、甜菜碱这几种物质对两类对虾的口味差异起着重要作用,其他成分作用不明显。  相似文献

An experiment was conducted, in a dark room with controlled temperature (27.3–28.4 °C), to determine the acute toxicity of chlorine concentration to black tiger shrimp (Penaeus monodon fabicus) of sizes 0.02 g, 2.75 g, 8.47 g and 23.65 g. Toxicity tests on each of these shrimp sizes were run in triplicate in glass jars under static conditions without media renewal. The concentration of active chlorine that killed 50% of the shrimp of each size after 24‐h exposure (LC50‐24 h) was used as an indicator of acute toxicity. Chlorine concentrations applied in the shrimp toxicity test ranged from 2.0 to 14.5 mg L?1 in shrimp pond water. As the test water contained total suspended solids of 22.0–85.0 mg L?1 and total ammonia nitrogen of 0.18–0.40 mg L?1, the resultant concentrations of combined residual chlorine ranged from 0.6 to 3.5 mg L?1, which were the effective doses causing shrimp mortality. The test results showed that 24‐h LC50 for average shrimp size at 0.02, 2.75, 8.47 and 23.65 g occurred in water containing combined residual chlorine at a concentration of 0.91, 1.39, 1.74 and 1.98 mg L?1, for which the original application doses were 6.96, 2.05 11.50 and 13.34 mg L?1 respectively.  相似文献

The effect of dietary protein level and natural food management on the production parameters of blue and white shrimp, as well as on water quality, was evaluated in a microcosms system (plastic pools simulating aquaculture ponds). Two experimental trials were carried out in the facilities of DICTUS, University of Sonora, Northwest México. Treatment with low protein diet (LP) consisted of a low protein input (diet with 250 g kg?1 crude protein) through the culture period; treatment with high protein diet (HP) consisted of a high protein input (diet with 400 g kg?1 crude protein) through the trial, and finally treatment VP consisted of an adjustment of protein input (diets with 250, 350 or 400 g kg?1 crude protein), depending on the abundance of biota (zooplankton and benthos) in the system. Each species responded differently to the treatments. For blue shrimp, low protein input resulted in the lowest final body weight (12.9 ± 0.6 g) and biomass (696.0 g pool?1). Survival and feed conversion ratio were similar in the three treatments. For white shrimp, the best growth, biomass and food conversion ratio were obtained in the low protein input treatment. Water quality parameters such as nitrate, ammonia and organic matter during the two trials, were better for LP and VP treatments. White shrimp seems to have lower protein requirements than blue shrimp. For the blue shrimp culture, adjusting protein input according to natural food abundance (zooplankton and benthos) in the system, seems to be advantageous because of the possibility of getting a production similar to that obtained with a high protein input through the farming period, but at lower feed cost, and with a lower environmental impact. It is concluded that a high protein input through the whole farming period is not the best feeding strategy for any of the two species.  相似文献

对虾WSSV病是亚洲对虾养殖业中的一个棘手问题。本研究采用Kimura引物 ,用PCR技术对不同生长期的中国对虾 (Penaeuschinensis)进行了WSSV的检测 ,同时也检测了对虾发病时养殖池中多见的野生厚蟹 (Helicesp .)和矛尾刺虎鱼 (Acanthogobiushasta)。检测结果表明 :分别在检测的 5尾亲虾中的 1尾 ,6尾仔虾中的 1尾 ,5尾稚虾中的 3尾及所检测的 5尾病虾和 2只厚蟹中获得到 982bp的PCR扩增产物 ,说明为WSSV感染阳性。在检测的 2尾矛尾刺虎鱼中均未获得PCR扩增产物 ,说明为WSSV感染阴性。在亲虾、虾苗以及虾池内的野生厚蟹中检测到WSSV感染的阳性结果表明 :WSSV感染的亲虾有可能是病毒的储主 ,WSSV感染的野生厚蟹有可能是病毒中间宿主或病毒的携带者 ,它们在对虾WSSV病的感染、传播中起了重要的作用  相似文献

应用克氏原螯虾作为动物模型研究对虾白斑综合征病毒(WSSV)的感染增殖特性,涉及感染温度,感染途径,继发感染,半数致死量(LD50),免疫保护及保存期等,结果显示,22-25℃时,接种WSSV的螯虾一般于2-7d内死亡,温度升高对病毒增殖影响不显著,30-32℃时,平均死亡时间2-6d,温度降低对病毒增殖影响较显著,15-19℃时,接种螯虾平均2-10d内死亡,8-10℃时,平均死亡时间3-6d,感染途径分别用腹节肌肉,腹节皮下洲射及口服,均能使螯虾感染发病,而浸泡方式不能使螯虾发病,细菌分离和细菌定量结果表明,寄考于螯虾心脏,肝胰腺内的阴沟肠杆菌在感染后期大量增殖,菌量分别是正常螯虾的25和30倍,形成继发感染,用螯虾心脏,肝胰腺内的阴肠杆菌在感染后期大量增殖,菌量分别是政党螯虾的25和30倍,形成继发梁。用螯虾测定WSSV的LD50为10^-6.5.mL^-1种毒液,将病毒56℃,30min灭活后免疫螯虾,不能使螯虾形成免疫保护,WSSV匀浆液-30℃冻存1年后失活,而-30℃冻存于螯虾体内WSSV保存1年后仍有活力。  相似文献

Advances in the domestication and selective breeding of Australian Black Tiger shrimp, Penaeus monodon, opens the opportunity for world producers to reconsider the benefits of farming this species. Just over a decade ago this species was the world's most farmed shrimp species, however, difficulty in its domestication, in part, led to the widespread establishment of Penaeus vannamei (Pacific White shrimp) as the most farmed shrimp species in the world. This study empirically evaluates the productivity benefits of commercially domesticating P. monodon against production from wild broodstock of the same species. The evaluation compared the relative production from commercial ponds stocked with the progeny of wild P. monodon broodstock and ponds stocked with the progeny of domesticated stocks. The production data were from 164 ponds of domesticated stocks and 30 ponds of wild stocks, collected over 4 years (2009–2013) from two separate farm sites of the same Australian shrimp farming company. The wild stocks were sourced from the east coast of Australia. The results suggested that the productivity of the selectively bred stocks was 39% greater compared with production from wild stocks given equivalent amounts of feed and other inputs. Furthermore, productivity was additionally enhanced depending on the choice of feeds and whether stocking took place in September rather than later in the year (i.e. in early spring rather than late spring/early summer in the Southern hemisphere). This suggests that there is significant potential to further enhance the productivity of P. monodon farms via integrating advances in domestication, feeds and management practices.  相似文献

为了快速、方便地检测出这2种病毒,研究将PCR核酸扩增的高灵敏度和胶体金免疫层析技术快速、简便、直观的优点结合起来,建立了同时检测2种对虾病毒(WSSV和IHHNV)的同步PCR-胶体金免疫层析检测方法。采用了生物素标记和地高辛(Digoxigenin)标记引物。PCR扩增产物利用胶体金免疫层析技术进行检测。胶体金免疫检测试剂条的检测线(T线)处的层析膜处点上地高辛的单抗,经地高辛标记的PCR扩增产物可在T线上被检测到,在10 min内即可得到检测结果。同时,在PCR过程中使用UNG(尿嘧啶-DNA糖基酶),极大地控制了遗留污染。通过此方法检测对虾WSSV和IHHNV病毒的灵敏度均可达到10~100个拷贝,高于国家标准PCR检测法10~100倍。此检测方法让工作人员避免了接触有毒和诱导突变的试剂,免除基层检测实验室购买电泳设备和荧光检测系统的投资。  相似文献

The bunyavirus Mourilyan virus (MoV) occurs commonly in Black tiger (Penaeus monodon) and kuruma shrimp (Penaeus japonicus) farmed in eastern Australia. There is circumstantial evidence of MoV causing mortalities among P. japonicus moved from farm ponds to tanks for rearing as broodstock. To directly assess its pathogenic potential, independent cohorts of pond- (n = 24) or tank-reared juvenile (n = 21) P. japonicus were challenged intramuscularly with a cephalothorax tissue homogenate of P. monodon containing high loads of MoV (1.48 ± 0.28 × 108 MoV RNA copies/µg total RNA). In each trial, mortalities accumulated gradually among the saline-injected controls. Mortality onset occurred 12–14 days earlier in the pond-reared shrimp, possibly due to them possessing low-level pre-existing MoV infections. Despite the time to onset of mortality differing, Kaplan–Meier survival analyses confirmed mortality rates to be significantly higher in both the pond- (p = .017) and tank-reared shrimp (p = .031) challenged with MoV. RT-qPCR data on shrimp sampled progressively over each trial showed high loads of MoV to establish following challenge and discounted GAV and other endemic viruses from contributing to mortality. Together, the data show that acute MoV infection can adversely compromise the survival of juvenile P. japonicus.  相似文献

An experimental study was conducted during 20 weeks in Bahia Kino, Sonora, Mexico, in order to evaluate the feasibility of promoting biota in low‐water exchange ponds farming blue shrimp, Litopenaeus stylirostris. The effect of that promotion on the production parameters of cultured shrimp as well as on the water quality parameters was evaluated. Treatments consisted of: (i) ponds fed formulated food (FF), and (ii) ponds fed formulated plus promoted natural food (NFF). Phytoplankton, zooplankton and benthos were effectively promoted during some weeks of the culture period. Growth and feed conversion ratio (15.16 g and 1.79 respectively) were significantly better in treatment NFF than in treatment FF (13.89 g and 2.02 respectively). Differences in some of the water quality parameters were observed among treatments. Phosphates (0.15 mg/L versus 0.53 mg/L), and total ammonia‐N (0.09 mg/L versus 0.12 mg/L) presented greater concentrations in treatment FF than in the NFF.  相似文献

The requirements for essential amino acids by kuruma shrimp, Marsupenaeus japonicus (Bate) have not yet been determined using the dose–response method due to poor utilization of crystalline amino acids (CAA). In a previous study, we produced coated CAA that could be used to retard leaching losses. Therefore, to determine the arginine requirement of juvenile kuruma shrimp, a growth study was conducted using test diets containing six levels of coated arginine (from 1.20% to 3.19% of diet) for 42 days. Crystalline amino acids was supplemented to correspond to the amino acid pattern in the whole body protein of the kuruma shrimp except for arginine. To reduce leaching losses, CAA were coated with carboxymethylcellulose (CMC) and diets were further bound with CMC and k‐carrageenan. Test diets were fed to triplicate groups of juveniles (0.25±0.02 g) twice a day. Proximate composition and the amino acid composition of shrimp whole bodies were analysed at the end of the feeding trial. Percent body weight gain (WG), specific growth rate, feed intake, feed efficiency ratio and protein retention efficiency were significantly affected by the dietary arginine concentrations. Estimating the arginine requirement of kuruma shrimp using broken‐line analysis of percent WG resulted in a value of 2.66% of diet or 5.32% of protein. After correcting for leaching of arginine from the diets, the optimum dietary value of arginine for kuruma shrimp was a little lower than the estimated value.  相似文献

This study was undertaken to determine acceptable dietary concentrations of high-fibre canola meal (CMHF) and low-fibre canola meal (CMLF) for juvenile shrimp, Penaeus vannamei. Four groups of 0.78 g shrimp held in running, 24.0–27.8°C sea water on a 12 h light: 12 h dark cycle were each fed one of seven isonitrogenous (340 g kg?1 protein) and isoenergetic (18.5 MJ of gross energy kg?1) diets to satiation four times daily for 56 days. Each of the test canola protein products comprised either 150, 300 or 450 g kg?1 of the protein in a basal (practical) diet by replacement of one-third, two-thirds or all of the menhaden meal protein. Shrimp that ingested diets in which CMHF and CMLF comprised 450 and 300 g kg?1 of the protein, respectively, exhibited significant reductions in growth and feed intake relative to those fed the basal diet. Feed and protein utilization were not significantly depressed unless menhaden meal in the basal diet was completely replaced by CMHF or CMLF. In general, percentage survival and final whole-body levels of protein, minerals, and thyroid hormones were not significantly affected by dietary treatment. Terminal whole-body levels of moisture were raised significantly in shrimp fed diets containing the highest levels of CMHF and CMLF. Potassium levels were significantly higher in shrimp fed the diet containing the lowest level of CMLF relative to those fed the basal diet and the diet with the highest level of CMLF. Water stability of the diet pellets was negatively correlated with their levels of CMHF and CMLF. It is concluded that commercial high-fibre canola meal can constitute 300 g kg?1 of the dietary protein of juvenile shrimp (Penaeus vannamei) without compromising growth, feed intake and feed and protein utilization. However, because of a trend towards reduced shrimp survival at this dietary concentration of canola meal, it is recommended that this protein source not exceed 150 g kg?1 of the protein in practical juvenile shrimp diets. Fibre-reduced canola meal did not have improved nutritive value for shrimp. However, we postulate that one or more fibre-reduced, and solvent-extracted canola protein products may be cost-effective substitutes for fish meal protein.  相似文献

Both MI and MII triploids were successfully produced by heat shock in Chinese shrimp Fenneropenaeus chinensis. The inducing conditions for MI and MII triploids were optimized. The highest inducing rate obtained for MI triploids reached more than 90%, and that for MII triploids reached nearly 100% at the nauplius stage as evaluated using flow cytometry. Comparisons of survival rates at larval stages between triploids and diploids or diploids experiencing treatment and diploids without treatment were performed. At larval stage from nauplii to postlarvae, heat shocks lowered survival at larval stages even if the ploidy was not changed. Ploidy did not affect shrimp larvae survival, and no significant difference was found in the survival of shrimp larvae between MI and MII triploids. Highly significant differences were observed in the morphology of triploids and diploids, and no apparent difference was found in the morphology of MI and MII triploids at the grow‐out stages. Discriminating formulae for triploid and diploid shrimp at grow‐out stage were developed and could be used to distinguish triploids from diploids based on morphological parameters. MI and MII triploids of shrimp have the potential to be used in aquaculture.  相似文献

The relationship between fishing line height (FLH), shrimp catch and bycatch in a semi-pelagic ocean shrimp (Pandalus jordani) trawl was investigated using a newly developed recording inclinometer. The inclinometer was effective at measuring FLH and indicating trawl performance deficits. FLH was determined to be stable during a haul and also between hauls within a given footrope and groundline configuration. FLH was readily adjusted with simple modifications to the footrope “dropper” chains. Inclinometer data showed that FLH can be unequal between double-rigged nets of identical configuration. Shrimp catch and the bycatch of flatfish and juvenile rockfish varied inversely with FLH, suggesting FLH can be adjusted to equalize the catch of shrimp, flatfish and juvenile rockfish between two double-rigged shrimp nets.  相似文献
