Taste effects of oligopeptides in a Vietnamese fish sauce   总被引:4,自引:0,他引:4  
ABSTRACT: A Vietnamese fish sauce Nuoc mam used in the present study was rich in oligopeptides of which nitrogen content accounted for 20% of total nitrogen. The high-molecular-weight peptide fractions fractionated by ion-exchange chromatographies and ultrafiltration enhanced sweetness and umami as well as sourness and bitterness of the fish sauce, and increased several flavor characteristics including continuity, first taste and aftertaste. Thus, it is apparent that large amounts of peptides produced during the long-term fermentation of fish sauce are responsible for the complicated taste of the sauce. From each fraction, 17 peptides in total were isolated and determined for their amino acid sequences. These di-, tri- and tetra-peptides synthesized by solid- or liquid-phase method gave any one of bitter, sour, umami taste, or practically no taste in the absence of salt. In the presence of 0.3% NaCl, however, almost all peptides showed sweet and umami tastes. Other than these 17 peptides, there existed many other kinds of peptides in the fish sauce. Thus, these peptides are thought to contribute to the overall taste of the fish sauce.  相似文献

研究了不同酶解的鱼可溶性产物苦味及呈味氨基酸组成的差异 ,以及其水解时间、水解率对苦味的影响。结果表明 ,随着酶解时间延长和水解率的提高 ,几种水解液的苦味值都有不同程度增强。胃蛋白酶和胰蛋白酶的复合水解液表现出随水解时间的延长 ,水解率提高明显 ,苦味强度上升平缓。当水解率从4 2 %提高到 78%时 ,苦味值只提高了 2 ,水解液总的苦味分值为 3.5 ;枯草杆菌蛋白酶水解 2h后苦味分值急剧提高 ,水解 4h后苦味分值达到 6,水解液味道很苦 ,水解率从 32 %提高到 62 %时 ,苦味分值提高到了 7,水解液总的苦味值为 8.5。两种方法水解获得的可溶性产物氨基酸及必需氨基酸组成含量接近 ,但胃蛋白酶、胰蛋白酶复合水解的可溶性产物具有更高的氨基酸分值 ,主要鲜味氨基酸含量略高于枯草杆菌蛋白酶水解产物 ,苦味氨基酸则相反。  相似文献

Solid wastes from threadfin bream (Nemipterus spp.) surimi production composed of head and frame were hydrolyzed by various commercial proteases (Alcalase, Flavourzyme, Neutrase, Protamex, papain, and pepsin) to produce protein hydrolysates with bioactive properties. An Alcalase-hydrolyzed sample at 24.4% degree of hydrolysis (DH) displayed the highest antioxidant activity based on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, ferric reducing antioxidant power (FRAP) assay, and potassium ferricyanide method. In addition, it showed an inhibitory activity toward angiotensin-converting enzyme (ACE) of 25.5%. Antioxidant activity of threadfin bream by-product hydrolysates increased with hydrolysis time and reached the highest DPPH activity after 6 h, while that hydrolyzed for 3 h showed the highest reducing power based on FRAP and potassium ferricyanide assays. In addition, ACE inhibitory activity was found to be at an optimum after 3 h of hydrolysis. The hydrolysates (1 mg/mL) also retarded oxidation of a linoleic acid emulsion system to a similar extent as 0.1 mg/mL 3-tert-butyl-4-hydroxy anisole (BHA), indicating a potential use in the food system. Protein hydrolysates from threadfin bream surimi by-products could be tailor-made to possess both antioxidant and ACE inhibitory activity through controlling DH of Alcalase-catalyzed reactions.  相似文献

Threadfin shad, Dorosoma petenense, or fathead minnows, Pimephales promelas, were co‐cultured with channel catfish, Ictalurus punctatus, in earthen ponds to determine the effects of planktivory on plankton community dynamics and catfish fillet quality. Fathead minnows had no effect on the plankton community structure or catfish fillet flavor, color, and fatty acid composition. Fillet color was also unaffected by the presence of threadfin shad. Small differences were found in fillet fatty acid composition for catfish from ponds with shad, but these differences probably have no biological significance. Threadfin shad did, however, have important impacts on the plankton community structure and catfish flavor. Size‐selective filter‐feeding by shad reduced cyanobacterial abundance relative to ponds with catfish‐only and fathead minnows. Relative abundance of smaller phytoplankton in the groups Chlorophyta, Cryptophyta, Bacillariophyceae, and Euglenophyta increased in ponds with shad. Relative abundance of small zooplankton (rotifers) also increased in shad ponds. Reduced abundance of large, colonial cyanobacteria that are known to produce odorous metabolites caused a corresponding reduction in off‐flavor prevalence and intensity in catfish from ponds with threadfin shad when sampled in September. Although threadfin shad dramatically reduced catfish off‐flavor prevalence during the warm season, they apparently caused a high prevalence of “fishy” off‐flavors in the February sample. This undesirable flavor appeared to be caused by catfish foraging on shad killed during a preceding period of exceptionally cold water temperatures. Use of threadfin shad for phytoplankton biomanipulation therefore presents a dilemma: catfish–shad polyculture reduces prevalence of cyanobacteria‐related off‐flavors in warm months but may cause undesirable forage‐related off‐flavors in the colder months. Catfish farmers must consider these benefits and risks when deciding to use threadfin shad as a management tool.  相似文献

ABSTRACT:   By conducing organoleptic analysis, the authors investigated the effect of oil droplet size on human preference to the taste of oil-in-water (o/w) emulsions to which several tastants were added. Two singly dispersed o/w emulsions, different in oil droplet sizes, consisting of 10% triacilglycerol purified from tuna, sardine or soybean oil, 0.5% emulsifier and water were prepared by ceramic membrane filtration. Organoleptic analysis showed that the impressions of sweetness, bitterness and umami, which were developed by addition of certain taste substances, were different between o/w emulsions with different oil droplet sizes. The results of two-bottle choice tests, which were carried out using olfactory-blocked mice, showed that o/w emulsions with 1.00-µm droplets, with added sweet or bitter substances, were preferred to emulsion with 5.50-µm droplets. These results suggest that the droplet size of o/w emulsion remarkably influences certain taste impressions created by added taste substances.  相似文献

ABSTRACT: Transglutaminase seems to be related to the setting phenomenon of fish meat paste that occurs at temperatures below 40°C. In many reports on the relationship between transglutaminase and setting phenomenon, the enzyme activity has been measured at 25°C. However, it is known that the setting phenomenon is complicated and the effect of calcium and the 3-D structure of myofibrils, which are sensitive to temperature, play important roles in the reaction. In the present study, total activities of transglutaminase of threadfin bream, white croaker, red sea bream and carp meats were measured at various temperatures. Total transglutaminase activities at 25°C of tested fish meats are arranged in order as follows; white croaker, red sea bream, carp, threadfin bream. In contrast, optimal temperature of carp meat is 30°C, red sea bream 40°C, threadfin bream 50°C and white croaker 50–55°C. In carp meat, the enzyme activity at optimal temperature (30°C) became 8.5-fold higher than at 25°C. The data of the total activity at various temperatures are useful in order to comprehend the details of the reaction.  相似文献

生物酶解是提高水产品蛋白质利用率的有效手段,且酶解过程往往伴随肽类、氨基酸、小分子挥发性成分等的生成或反应,进而引起酶解液风味变化。仿刺参(Apostichopus japonicus)性腺富含蛋白质和多种功效成分,具有良好的开发利用前景。为探究仿刺参性腺酶解过程中蛋白质和风味的变化规律,采用中性蛋白酶对其进行酶解,对酶解过程中可溶性蛋白质、氨基酸态氮、游离氨基酸组成、挥发性成分的变化情况进行了检测分析。结果显示,仿刺参性腺匀浆液中可溶性蛋白质和氨基酸态氮的初始含量分别为1.14和0.15 g/100 g,可溶性蛋白质在酶解前30 min内迅速增加,之后基本保持不变,氨基酸态氮含量在前90 min内随时间延长而逐渐增加,90 min后略有下降,90 min时水解度最大,达43.66%。随着酶解时间的延长,酶解液的鲜味有所增强,腥味减弱,甜味和苦味也略有增强。酶解后游离氨基酸含量显著增加(P<0.05),其中,呈鲜味的谷氨酸含量最高,其次为呈甜味的甘氨酸和丙氨酸。仿刺参性腺酶解过程气味发生明显变化,烃类种类和相对含量均明显增加,二甲基硫醚含量显著降低,这可能是酶解液腥味减弱的主要原因  相似文献

The effect of addition of trypsin and chymotrypsin (0.3%, w/w) at various proportions (100:0, 50:50, and -0:100) was investigated to accelerate fish sauce fermentation using herring as raw material. Results showed that supplementations with trypsin and chymotrypsin significantly increased protein hydrolysis (P<0.05). Fish sauce prepared form herring with enzyme supplementation contained significantly more total nitrogen, soluble protein, free amino acid content and total amino acid content compared to fish sauce with no added enzyme (P<0.05). Supplementation with tryspin and/or chrymotrypsin mixture *ratios 50:50 and 0:100) enzyme produced the most favorable results in term of protein hydrolysis, while there was no difference among enzyme treated sauce in term of free amino acid composition. With the exception of the amino acid profile, and microbiological composition compared to Nampla, a first grade commercially produced Thai fish sauce. Sensory analysis of products for color, flavor and aroma indicated that panelist preferred the lighter colored fish sauce prepared with enzyme supplementation compared to the darker colored commercially produced Nampla. However, There was no significant preference (p<0.05) for aroma and flavor of the enzyme supplemented sauce or Nampla. The results of this stuffy indicate that an acceptable fish sauce product can be produced form herring and the addition of enzymes has the potential to reduce the fermentation time to --2 months without compromising product quality.  相似文献

An “abnormal” female Scylla paramamosain, named “Butter crab” (B_♀) is more popular than wild‐type mud crab (CK) with people. Histologically, “butter crab” ovaries are partially degraded, with full of yellow oily substances in body. Composition of the essential nutritional components is quite similar in “butter crab” and CK. In a comparison of fatty acids, B_♀ is richer. A comparison of flavour amino acids and nucleotide in muscle showed that sweet amino acids and bitter amino acids in B_♀ is lower than CK_♀, but umami amino acids are significantly higher in B_♀. However, the umami amino acid content in the gonads is significantly less in B_♀ than CK_♀. Moreover, umami nucleotides in B_♀ muscle is significantly higher. From the perspective of EUC evaluation, the umami taste of CK_ ♀ gonads is strongest, followed by B_ ♀ gonads. In the hepatopancreas, the umami taste ranks CK_♀> B_♀ >CK_♂, and there are no significant differences among muscles in the three groups. In conclusion, the characteristics of “butter crab” are more fragrant and nutritious, with sweet and umami taste. The article systematically scientifically analysed the nutritional composition and flavour characteristics of “butter crab”, providing a novel insight for development of S. paramamosain industry.  相似文献

ABSTRACT

A protein hydrolysate was prepared from proteins of tuna dark muscle by-product. The hydrolysis conditions (time, temperature, pH, and enzyme concentration) using Alcalase was optimized by response surface methodology (RSM). The regression coefficient close to 1.0, observed during experimental and validation runs, indicated the validity of the model. The hydrolysate produced under the optimum conditions determined by RSM has a low rate of peptide fraction of molecular weight of 4–1 kDa. Meanwhile, the results obtained by hydrolysis under optimal conditions determined by a complementary study (temperature 55°C, time 60 min, 1% enzyme concentration, and pH 8.5) show that the hydrolysate produced has a height rate of the peptide fraction of molecular weight of 4–1 kDa. The amino acid composition of the protein hydrolysate prepared proved to have the potential for application as an ingredient in balanced fish diets and as a source of nitrogen in microbial growth media.  相似文献

Pacific threadfin Polydactylus sexfilis (Bloch & Schneider 1801) and bluefin trevally Caranx melampygus (Cuvier 1833) are warmwater marine finfish currently under development for aquaculture in the Pacific. Differences in specific activities of digestive enzymes extracted from the stomach and mid-gut were compared to gain insight into their feeding habits in the wild and to understand their nutritional needs. Adult fish were maintained in captivity and fed a commercial pelleted feed. Serine protease measured in all tissues was at least 20 times higher in threadfin than in trevally. Aspartic proteases were the major digestive enzymes found in trevally. There was a 34-fold increase in collagenase activity in the intestine of threadfin from the prefed to the fed state. Chitinase activity was found in the stomach, pylorus and intestine of both species. However, specific activity in pylorus and intestine of threadfin increased 2.75 and 4 times, respectively, but showed little change in trevally. Amlyases were found only in trevally. Increase in lipase specific activity in the gut of trevally was higher than that for threadfin. The results indicated that the two species have diverse digestive capabilities. This appears to be consistent with their feeding habits in nature. Threadfin are more adapted to a wider range of food protein sources than trevally, but appear to be less well adapted than trevally to using complex carbohydrates. These observations may provide a basis for practical diet formulations for these two species.  相似文献

Rest raw materials as viscera, heads, and frames from farmed Atlantic salmon (Salmo salar) were hydrolyzed with the use of endogenous enzymes and the commercial enzymes Protamex and a mixture of Papain and Bromelain. Composition of the prehydrolysis mixture clearly influenced the quality of final hydrolysate and process kinetics. An increased proportion of viscera increased the amount of endogenous enzymes, which influenced hydrolysis kinetics, the extent of hydrolysis, and increased bitterness. Commercial enzymes, in addition to endogenous enzymes, are not always more yield efficient or economically beneficial but can be used to improve the taste and to ease the separation of oil from the hydrolysate fraction. Hydrolysis of denatured proteins in rest raw materials was hardly detectable. Optimum temperature should therefore be selected to avoid protein denaturation and simultaneously maintain high enzymatic activity. Hydrolysates preferably contain a low concentration of oil, and this work shows that high-quality oil can be separated before hydrolysis by mild thermal treatment. The previous separation of oil did not influence hydrolysate yield and decreased the concentration of lipids in the final hydrolysate. The initial separation of oil also increased productivity of the hydrolysis reactor, due to the reduction of hydrolysis volume.  相似文献

Extractive components were determined for the mantle muscle and liver of three species of Loliginidae squid, Sepioteuthis lessoniana, Loligo bleekeri, and L. edulis, and one species of Ommastrephidae, Todarodes pacificus, as a control. Total free amino acid levels and the major free amino acids in muscle, taurine, proline, glycine, alanine, and arginine, were significantly higher in Loliginidae squids than in T. pacificus. The main nucleotide was adenosine 5′-monophosphate, which did not differ significantly between species. Malate was the organic acid found in muscle in highest concentrations. The muscle of these species contained a large amount of trimethylamine oxide. A large amount of glycine betaine was also detected in the muscle, but showed no large species difference. From these results, the muscle of Loliginidae species is considered to have a much sweeter taste than that of T. pacificus. Compared with muscle, squid liver was characteristic with high contents of taurine, glutamate, bitter amino acids, succinate, propionate, trimethylamine, and glycine betaine, and with low contents of sweet amino acids, arginine, nucleotides, malate, and trimethylamine oxide. These results suggest that squid liver is characterized by a complicated taste containing umami, bitterness, sourness, fishy flavor, and less sweetness.  相似文献

In the present study, spray-dried pink perch meat protein hydrolysate (PPH) and hydrolysate with maltodextrin and gum arabic (PPHMG) were characterized in relation to their physical, rheological, functional, antioxidant, thermal, and sensory properties. The Herschel–Bulkley model was suitable to explain the flow behavior of PPH and PPHMG emulsions. Both the samples exhibited functional and antioxidant properties. Antioxidant activities were significantly higher for PPH compared with PPHMG (p<0.05). PPH and PPHMG alone did not exhibit antimicrobial activity but enhanced the activity of chitosan against pathogens. Sensory acceptability of the samples in soup revealed that PPHMG of up to 4% was highly scored without any traces of bitterness, whereas for PPH, a concentration of up to 3% was acceptable.  相似文献

以南极磷虾虾(Euphausia superba)肉糜为原料,通过测定南极磷虾虾肉糜在反复冷冻和解冻过程中ATP及其关联产物、游离氨基酸等的变化,分析冻融循环对虾肉糜滋味成分的影响。结果显示,随着冻融循环次数的增加,南极磷虾虾肉糜的ATP及其关联产物降解加剧,呈鲜味的核苷酸含量逐渐减少,呈苦味的核苷酸含量逐渐增加。1次冻融对应的鲜味苦味比为3.32;2次及以上的冻融循环会造成鲜苦比的显著降低(P<0.05),同时K值迅速增大,表明鲜度发生大幅下降;3次冻融循环后,K值显著增大(P<0.05);4次冻融对应的K值为(86.88±3.58)%,南极磷虾虾肉糜已不新鲜。冻融循环次数的增加导致游离氨基酸总含量增加,鲜味氨基酸和甜味氨基酸比例逐渐减小,苦味氨基酸的比例逐渐增大,导致虾肉糜的滋味进一步劣化。研究表明,冻融循环次数控制在2次以内,可较好地保持南极磷虾虾肉糜的滋味。  相似文献

To make use of underutilized fish species and produce high-quality condiments substitutable for those of mammalian origin, fish sauces were produced on a small industrial scale from three fish species, the flyingfish Cypselurus agoo agoo, the small dolphinfish Coryphaena hippurus, and the deepsea smelt Glossanodon semifasciatus, using salt and koji mold. During 180 days of fermentation, the pH decreased to approximately 4.5 and total nitrogen and free amino acids increased to approximately 2 g/100 mL and 6000 mg/100 mL, respectively. Halophilic and extreme halophilic bacterial counts dramatically increased from 106 to 108 colony forming units/mL between days 14 and 30 and decreased the reafter. In the final products, the pH and salt concentration decreased to between soy sauce and a Vietnamese fish sauce, Nuoc mam. The total amino acid and organic acid contents were almost the same as those of Nuoc mam and soy sauce. Sensory evaluation found these products to have lower smell, saltiness, and bitterness, and higher sweetness and umami taste than Nuoc mam. During fermentation, the histamine content increased in one or two of three fermentation tanks for each fish species. As a result, the histamine content of the final products was higher than that in Nuoc mam.  相似文献

ABSTRACT

The effect of Alcalase hydrolysis conditions on antioxidant activities of shark skin gelatin hydrolysate (SSGH) byproduct was studied. Optimal hydrolysis condition for SSGH at 60°C, pH 7.5, and 2.70 AU kg?1 gelatin protein was used for varied hydrolysis times (10–90 min). The IC50 against DPPH radicals of SSGH hydrolyzed for 90 min (SSGH-90) was 27.39 mg ml?1, which is greater than ascorbic acid. SSGH-90 was predominantly composed of 15–20 kDa protein fragments that regulated increases in peroxide value and thiobarbituric acid reactive substances values and suppressed decrease in DHA of a fish oil-in-water emulsion during storage at 30°C for 7 days.  相似文献
