旨在研究褪黑素对妊娠牦牛子宫的调控作用,采用real-time PCR法和免疫组织化学S-P法,对未妊娠和妊娠不同时期牦牛子宫中褪黑素受体(MT1)mRNA和褪黑素受体(MT1)蛋白的表达进行研究。结果表明:牦牛未妊娠和妊娠的不同时期,牦牛子宫基质细胞(SC)和肌层平滑肌细胞(MM)中均有MT1蛋白表达(P0.05);血管内皮细胞(EBV)和血管平滑肌细胞(VSM)中的MT1蛋白表达,在妊娠28~35d差异不显著,随妊娠时间的增加,MT1蛋白表达增加(P0.05);腺上皮细胞(GE)中的MT1蛋白,在妊娠初期(妊娠28~35d)差异不显著,妊娠50~60d时,差异显著(P0.05),妊娠90~100d时,差异极显著(P0.01)。而在腔上皮细胞(SE)中,MT1蛋白表达量在妊娠28~35d比未妊娠时低(P0.05),而这种趋势随妊娠时间的增加继续降低(P0.01)。未妊娠牦牛子宫中MT1mRNA表达量显著高于妊娠牦牛(P0.01)。综上表明,在妊娠期,牦牛子宫对褪黑素的需求量降低,褪黑素主要作用于牦牛GE,调节子宫腺功能,使牦牛子宫产生适应妊娠的生理反应。  相似文献

为了研究日粮中添加N-氨甲酰谷氨酸(NCG)对妊娠前期鄂尔多斯细毛羊子宫内膜层及胎儿发育的影响,试验选择受孕鄂尔多斯细毛羊母羊40只,随机分为3组:NCG1组饲喂基础日粮+0.30 g NCG/d(n=13),NCG2组饲喂基础日粮+0.40 g NCG/d(n=13),对照组饲喂基础日粮(n=14),开展妊娠期0 d至90 d的饲喂试验。结果表明:妊娠期第90 d,NCG1组胎儿肺脏重显著高于对照组,NCG2组胎儿肝脏重显著高于对照组(P0.05);妊娠期第17天,NCG1组母羊子宫内膜OPN基因表达量显著高于NCG2组(P0.05)和对照组(P0.01),NCG2组母羊子宫内膜Integrinαvβ3基因表达量显著高于对照组(P0.05);妊娠期第90 d,NCG2组和NCG1组胎儿肾脂肪BMP7(P0.05)和UCP-1基因表达量显著高于对照组(P0.01),而NCG1组(P0.01)胎儿肾脂肪PGC-1α基因表达量显著低于对照组,NCG2组(P0.01)胎儿肾脂肪PRDM16基因表达量显著低于对照组。在本试验条件下,妊娠前期日粮中添加NCG有效促进了鄂尔多斯细毛羊妊娠识别、早期胚胎定植和子宫内膜层发育,保障了胎儿早期程序化健康生长。  相似文献

胎盘在母体-胎儿间物质交换时发挥着重要作用,而其物质交换动力来自胎盘线粒体通过氧化磷酸化产生ATP。为探讨随着动物妊娠阶段的持续,线粒体相关基因的表达量变化,本研究采用RT-PCR技术检测大足黑山羊妊娠20,25,30,60,90和120d胎盘线粒体COXIV、SDHA、PGC-1α、NRF-1和TFAM基因的相对表达量。结果显示,随着大足黑山羊妊娠持续,胎盘子叶和子宫阜线粒体被检测的5项基因相对表达量均呈现先升高后降低、再缓慢变化的趋势。尤其在妊娠25d时相对表达量均达到高峰,但妊娠30d时相对表达量急剧下降。在胎盘子叶组织中COXIV、SDHA和NRF-1的相对表达量分别由0.84,1.53和1.06,降至0.09,0.32和0.22,差异显著(P0.05);在子宫阜中COXIV、SDHA和PGC-1α的相对表达量分别由0.50,4.36和1.36降至0.11,0.84和0.48,差异极显著(P0.01)。妊娠60d后,在胎盘子叶组织中,这些基因相对表达量不断升高;在子宫阜,相对表达量呈递减趋势,但差异均不显著(P0.05)。相关性分析结果显示,子宫阜组织中线粒体相关基因的表达与胎儿及胎盘生长发育指标如胎儿重、胎儿长、胎盘重、胎盘效率、子叶个数和子叶直径呈显著负相关(P0.05);而子叶组织中线粒体相关基因的表达与胎儿长和子叶直径显著性相关(P0.05),但与胎儿重、胎盘质量、胎盘效率和子叶个数无显著相关性(P0.05)。  相似文献

采用组织学和形态计量学的方法对发情周期牦牛子宫的组织结构及形态变化进行了观察。结果表明,牦牛子宫壁具有3层结构,分别为黏膜(子宫内膜)、肌层(子宫肌层)和浆膜(子宫外膜)。子宫内膜表面上皮和腺上皮在卵泡期为单层柱状上皮,在黄体期上皮高度明显增加,为假复层柱状上皮。表面上皮主要由分泌细胞及少量淋巴细胞组成。分泌细胞可分为亮细胞和暗细胞2种。亮细胞呈柱状,细胞体积较大,胞质弱嗜酸性;暗细胞呈两侧压扁的细条状,胞质强嗜酸性。腺上皮的细胞组成与表面上皮相似。PAS反应可见黄体期子宫内膜表面上皮、腺上皮和基质中阳性分泌颗粒较卵泡期的多。卵泡期固有层内偶见嵌套腺管、扩张腺管和弥散淋巴组织,但黄体期固有层内仅偶见嵌套腺管。此外,与卵泡期相比,黄体期子宫内膜和子宫肌层的厚度显著增加(P0.01)。黄体期子宫内膜腺管的平均直径、单个腺管的面积和周长、每平方毫米内腺管的总面积和总周长都较卵泡期的大,差异极显著(P0.01),而每平方毫米内腺管的数量差异并不显著(P0.05)。可见,牦牛子宫的组织结构在发情周期发生着有规律的变化。  相似文献

为研究亚丁牦牛肉的营养及品质特性,对6头成年亚丁牦牛(5.5岁,公母各半)的肌肉组织进行常规营养成分含量、脂肪酸含量、矿物质元素含量、氨基酸含量、肉质嫩度、熟肉率测定,并与昌台牦牛肉质进行联合分析。结果表明:常规营养成分中,亚丁牦牛肉的蛋白质显著高于昌台牦牛(P0.05);矿物质元素中,亚丁牦牛肉中钠的含量极显著低于昌台牦牛(P0.01),而铜的含量极显著高于昌台牦牛(P0.01);氨基酸测定中,亚丁牦牛肉的必需氨基酸占总氨基酸的比例和必需氨基酸与非必需氨基酸的比例均极显著低于昌台牦牛(P0.01);在冈上肌中,亚丁牦牛的肉色亮度值L~*极显著高于昌台牦牛(P0.01),而红度值a~*和黄度值b~*均极显著低于昌台牦牛(P0.01);在背最长肌中,亚丁牦牛的红度值a~*及黄度值b~*均极显著低于昌台牦牛(P0.01);在半腱肌中,亚丁牦牛的肉色亮度值L~*极显著高于昌台牦牛(P0.01),而红度值a~*及黄度值b~*亚丁牦牛均极显著低于昌台牦牛(P0.01);亚丁牦牛肉的pH值冈上肌为6.40,背最长肌为6.62,半腱肌为6.34;在药物检测中,亚丁牦牛肉中均未检测到药物残留。表明亚丁牦牛肉是具有蛋白质含量高、脂肪含量低,氨基酸含量丰富、肌肉嫩度高等特点的安全健康、无污染食品。  相似文献

试验旨在研究紫花苜蓿鲜草对长大母猪生产性能、血液理化指标、初乳IgG及经济效益的影响,为优化利用紫花苜蓿饲料资源提供依据。试验选择健康和胎次、年龄、历史产仔哺育性能接近的长大二元母猪60头为试验对象,采用完全随机化设计,试猪分为4组,每组3个重复,每个重复5头。对照组饲喂基础饲粮,3个试验组在妊娠、泌乳期母猪饲粮中分别添加20%、30%和40%紫花苜蓿鲜草。结果表明:1)在妊娠母猪、泌乳母猪饲粮中添加不同比例的紫花苜蓿鲜草,不同程度地提高了母猪的总产仔数、产活仔数、泌乳量及仔猪的初生窝重、初生个体重、21日龄窝重、28日龄断奶成活率,其中30%紫花苜蓿鲜草组显著提高了仔猪21日龄窝重及母猪泌乳量(P0.05);与对照组产后7 d母猪体重相比,各试验组产后21 d、28 d母猪体重减重率较对照组增大(P0.05)。2)妊娠母猪阶段,各组间干物质、粗蛋白质、粗纤维、粗脂肪消化率差异不显著(P0.05);泌乳母猪阶段,各组间的干物质、粗蛋白质、粗纤维消化率差异不显著(P0.05),而各试验组的粗脂肪消化率显著或极显著高于对照组。3)在妊娠期、泌乳期母猪饲粮中添加20%、30%、40%的紫花苜蓿鲜草,3个试验组的血清尿素氮均低于对照组,其中30%紫花苜蓿鲜草组显著或极显著低于对照组;3个试验组的血清总蛋白均高于对照组,30%、40%紫花苜蓿鲜草组显著或极显著高于对照组;3个试验组的血糖低于对照组,其中泌乳母猪30%、40%的紫花苜蓿鲜草组显著低于对照组(P0.05);3个试验组血清中的甘油三酯有所降低,其中30%紫花苜蓿鲜草组极显著低于对照组(P0.01);3个试验组的血清IgG均高于对照组,30%、40%紫花苜蓿鲜草组极显著高于对照组(P0.01);妊娠期、泌乳期相比,3个试验组的血清尿素氮、总蛋白、血糖、甘油三酯差异不显著(P0.05),而泌乳期血清IgG均极显著高于妊娠期IgG(P0.01)。4)各试验组初乳中的IgG均有所提高,30%、40%紫花苜蓿鲜草组的IgG极显著高于对照组(P0.01)。结果显示,在妊娠母猪、泌乳母猪饲粮中添加不同比例的紫花苜蓿鲜草,提高了母猪的生产性能,改善了血液和初乳理化指标,节本增效明显。在该试验条件下,推荐紫花苜蓿鲜草替代妊娠、泌乳母猪饲粮的适宜比例为30%~40%。  相似文献

为探讨脑源性神经营养因子(BDNF)及其受体酪氨酸激酶受体B(TRKB)在妊娠期山羊子宫和胎盘中的表达情况,本实验分别于妊娠15、25、65 d采集子宫和胎盘样品,免疫组织化学方法分析BDNF和TRKB蛋白的分布情况。结果表明:妊娠15 d时,BDNF和TRKB蛋白在子宫内膜上皮和腺上皮细胞中强表达,BDNF在肌层弱表达,而TRKB强表达;妊娠25 d和65 d时,2种蛋白在子宫内膜上皮和腺上皮细胞中维持强表达,同时在胎儿绒毛膜滋养层细胞中表达强烈;与妊娠15 d相比,妊娠25 d和65 d时2种蛋白均在浅层子宫腺强烈表达,深层子宫腺弱表达。综上可知,随着妊娠的进行,BDNF和TRKB蛋白的表达情况也发生变化,提示BDNF对山羊妊娠活动有一定的调节作用,与妊娠期山羊子宫和胎盘的组织结构变化相关联,可能通过与其受体TRKB的特异性结合来实现其功能。  相似文献

本研究旨在探讨镰刀菌毒素对断奶仔猪子宫的组织结构、Hsp70的分布和mRNA表达量的影响。选择35日龄体重为(8.45±0.94)kg的健康三元杂交(杜洛克×长白×大白)雌性断奶仔猪20头,仔猪随机分为2组,每组10头。对照组饲喂基础饲粮,镰刀菌毒素组饲喂含有镰刀菌毒素(玉米赤霉烯酮0.90mg·kg~(-1),呕吐毒素1.43mg·kg~(-1),烟曲霉毒素5.85mg·kg~(-1))的试验饲粮,预试期7d,正试期35d,试验结束进行屠宰取出子宫。结果表明:与对照组相比,镰刀菌毒素极显著降低了仔猪的平均日增重和平均日采食量(P0.01),引起子宫器官指数极显著增加(对照组:0.91±0.03,镰刀菌毒素组:1.90±0.10,P0.01),子宫腺腺泡数量增多、密度增大,子宫内膜厚度极显著增加(对照组:1 002.55±101.22μm,镰刀菌毒素组:1 343.09±104.57μm,P0.01)。肌层增厚一倍以上(对照组:355.58±28.26μm,镰刀菌毒素组:779.56±40.38μm),与对照组相比均差异极显著(P0.01)。子宫中的Hsp70阳性细胞主要分布在子宫内膜上皮和子宫腺柱状上皮内,为胞质着色。与对照组相比,镰刀菌毒素组子宫Hsp70的免疫阳性反应明显增强(P0.01),且内环肌层也可见有Hsp70阳性细胞存在,子宫腺上皮细胞Hsp70IOD较对照组增加2倍多。镰刀菌毒素组子宫Hsp70mRNA相对表达量显著高于对照组(P0.05)。综上表明,本试验条件下,镰刀菌毒素可引起断奶仔猪子宫氧化应激反应,子宫内膜上皮和子宫腺上皮通过增加自分泌Hsp70能力,一定限度内能够对抗镰刀菌毒素对子宫正常生理机能的影响。  相似文献

本试验旨在探讨澳大利亚肉类标准(MSA)体系在我国川西北牦牛肉质量评价和等级分级应用中的可行性。选取舍饲育肥180 d的麦洼青年公牦牛、成年公牦牛、成年母牦牛各3头以及3头放牧麦洼成年公牦牛,取其背最长肌,在分析背最长肌理化指标基础上,采用4×4拉丁方设计,参考MSA体系以涮牛肉的形式对牦牛背最长肌的感官食用品质进行评价。结果表明:3个舍饲育肥组牦牛背最长肌的亮度(L*)显著高于放牧成年公牦牛(P0.05),红度(a*)显著低于放牧成年公牦牛(P0.05),4组牦牛肉的黄度(b*)差异不显著(P0.05);放牧成年公牦牛肉的p H45min、p H24h、滴水损失和蒸煮损失都最高,并显著高于3个舍饲育肥组牦牛肉(P0.05);4组牦牛肉的剪切力具有显著性差异(P0.05),由大到小依次是放牧成年公牦牛、舍饲成年公牦牛、舍饲青年公牦牛和舍饲成年母牦牛;3个舍饲育肥组牦牛肉的嫩度、多汁性、风味、整体喜欢程度、MQ4和质量等级等感官评价指标评分都显著高于放牧成年公牦牛肉(P0.05),但3个舍饲育肥组间无显著性差异(P0.05);嫩度、多汁性、风味和整体喜欢程度4项基础感官评价指标之间具有极显著的正相关性(P0.01)。本试验结果说明,MSA体系可以被借鉴用于川西北牦牛肉感官食用品质的评价,其中MQ4评分有被用于预测牦牛肉质量和分级的潜力。  相似文献

Entire reproductive tracts were removed from seven normal healthy heifers and qualitative unbiased estimates made of endometrial gland volume density and glandular surface density. After examining approximately 55 microscopic fields of endometrium from each tract, a mean glandular surface density value of 10.2 mm(2)/mm(3) (CE 3.1%) was obtained. The stereological method was then employed in optimising the design of the main study. The endometrial height was measured for 17 healthy cycling heifers and 19 similar cows. Subsequently, unbiased estimates were made of intercaruncular endometrial gland volume per unit surface for all cattle were investigated; differences between heifers and cows generally, and the possible effect of the follicular and luteal phases of the oestrous cycle were compared. The mean surface area of glands per unit area of endometrium at the intercaruncular site in heifers and cows was approximately 18 mm(2)/mm(2) in the follicular phase and 26 mm(2)/mm(2) in the luteal phase, figures similar to the gland area found in women. The intercaruncular gland volume increased significantly, by about 30% during the luteal phase of the bovine oestrous cycle in heifers, from 0.01 to 0.13 per mm(3). The differences in endometrial anatomy between site of sampling and either follicular or luteal phases of the oestrous cycle were always more significant in heifers than cows. The endometrial thickness in cows was always greater than for heifers, irrespective of the site of sampling. It was concluded that the intercaruncular endometrium of cattle was far more active physiologically than recognised previously.  相似文献

To determine effects of age and administration and withdrawal of a synthetic progestin (P) on endometrial development and DNA synthesis, ewe lambs were ovariectomized on d 0 (birth) and assigned to one of four groups (n = three/group) that provided (by means of hemihysterectomy) the following uterine tissue types: 1) d 0 control, 2) d 13 control, 3) d 26 control, 4) d 13 after 13 d exposure to P (13P) and 5) d 26 after P exposure from d 0 to 13 (26P). Uterine tissues were processed for histology or explanted with [methyl-3 H] thymidine for autoradiography. Labeling index (LI) was determined for stroma and epithelium in caruncular and intercaruncular endometrial areas and for lumenal and glandular epithelium in uteri with glands. Endometrial glands were absent on d 0, evident at d 13 and well developed by d 26. Day 0 LI was greater (P less than .05) in caruncular than in intercaruncular areas, and greater in stromal than in epithelial tissues. Relationships were reversed in d 13 endometrium (day X endometrial area, P less than .07). Caruncular stromal LI was greater on d 0 than later (P less than .02), whereas intercaruncular epithelial LI was greater after d 0 (P less than .05), but decreased from d 13 to 26 (P less than .05). Glandular epithelial LI was higher on d 13 than on d 26 (P less than .03). Administration of P inhibited endometrial gland development and suppressed d 13P intercaruncular LI (P less than .05). Withdrawal of P was followed by endometrial gland development and increased (P less than .01) intercaruncular epithelial LI in d 26P uteri. Ovary-independent initiation of endometrial gland development involves age- and region-specific alterations in DNA synthesis and could involve negative control.  相似文献

为揭示牦牛皱胃组织结构和黏膜免疫相关细胞的分布与数量变化的规律,采用组织化学法、图像分析法及透射电镜技术,对牦牛皱胃组织结构及上皮内淋巴细胞、浆细胞和肥大细胞变化进行了研究.结果表明:牦牛皱胃胃壁由黏膜层、黏膜下层、肌层和浆膜构成.皱胃幽门腺区胃小凹深度最深、腺体最长、肌层最厚;3个腺区肌层厚度、腺体长度之间差异极显著(P<0.01);幽门腺区与胃底腺区、贲门腺区之间胃小凹差异极显著(P<0.01),胃底腺区与贲门腺区之间差异不显著(P>0.05).牦牛皱胃黏膜上皮内淋巴细胞数量和浆细胞数量,3个腺区之间差异不显著(P>0.05);肥大细胞数量以胃底腺区最多,幽门腺区最少,两者之间差异极显著(P<0.01),贲门腺区与胃底腺区和幽门腺区之间差异不显著(P>0.05).皱胃各腺区固有层中均有大量的弥散淋巴细胞和孤立淋巴小结.电镜观察表明,胃小凹柱状上皮细胞排列紧密.幽门腺区固有层中有大量的黏液细胞,黏液细胞呈高柱状或锥体状,核位于基底部,在细胞顶端常聚集有较多的电子密度较高的颗粒.胃底腺区和贲门腺区有大量的壁细胞和主细胞.牦牛皱胃的组织结构和其他反刍动物基本相似,但各层有其明显特点.牦牛皱胃各腺区固有层中均有大量弥散淋巴细胞和孤立淋巴小结,使牦牛皱胃具有比其他反刍动物更强的黏膜免疫功能.  相似文献

An experiment was performed to evaluate the types and quantity of proteins secreted by intercaruncular endometrium at days 20, 60, 100 and 140 of gestation and caruncular endometrium from days 100 and 140 of gestation. Tissues were obtained from ewes made unilaterally pregnant. Based on SDS polyacrylamide gel electrophoresis (SDS-PAGE), the major proteins present in uterine fluid at days 60–140 of gestation were the uterine milk proteins (UTM-proteins), a pair of structurally-related, progesterone-induced polypeptides with molecular weights of 55,000 and 57,000. These proteins were also present in uterine fluid at day 20, but the major protein at this time migrated coincident with albumin. Cultured explants of endometrium at all days of pregnancy produced UTM-proteins as their major radiolabelled product for both caruncular and intercaruncular endometrium. The amount of protein secretion in vitro was greater (P<.04) for intercaruncular endometrium than for caruncular endometrium but was not significantly affected by stage of gestation or local presence of the conceptus. Immunohistochemical analysis revealed that UTM-proteins were present in glandular and luminal epithelium of intercaruncular endometrium and in both epithelial and stromal elements of caruncular endometrium. It was concluded that the UTM-proteins are produced earlier than previously described (i.e., day 20). In addition, caruncles contribute to the uterine secretory protein milieu through the secretion of proteins that are similar to that produced by the glandular intercaruncular epithelium.  相似文献

X‐ray microanalysis was used to determine the occurrence of transudation as a source of intrauterine fluid in mares during oestrus. The content of some selected elements was studied in secretory vesicles of the epithelium of endometrial glands and in intraluminal fluid of healthy mares of varying age and parity. An in situ cotton tampon was used to absorb intrauterine fluid from 23 gynaecologically healthy mares during oestrus. Immediately after tampon withdrawal, endometrial tissue for biopsy was removed from the uterine body/horn junction while videoendoscopy was performed. The tissue specimens were frozen ultra‐rapidly against a copper surface chilled with liquid N2. X‐ray microanalysis of frozen fully hydrated endometrial samples and of microdroplets of intraluminal fluid (n = 20) was performed, using a SEM microscope equipped with a cryo‐stage and an X‐ray microanalysis system. The concentrations of sodium (Na), potassium (K), chlorine (Cl), sulphur (S), phosphorus (P) and calcium (Ca) in the epithelium of endometrial glands and in intraluminal fluid were compared with those in serum. There was a significant difference in the concentrations of all analysed elements between the secretory gland epithelium and the intraluminal fluid apart from S. The major elements in glandular epithelium were K and P. Although there was a significant difference in the concentrations of Na, Cl, S and Ca between the uterine fluid and serum, there was still a conspicuous resemblance in the relationship between the different elements in uterine fluid and serum, with Na and Cl being the dominant elements. Sulphur and especially Ca were present only in small amounts throughout. Neither age nor parity affected the elemental concentrations of Na, K, Cl, S, P and Ca in gland epithelium, uterine fluid or serum. It was concluded that serum transudation contributes to a great extent to the composition of uterine fluid, having a diluting effect upon the specific secretion of the uterine glands.  相似文献

应用HE染色方法和免疫荧光组织化学技术对小鼠生后不同发育阶段子宫组织结构的发育以及极性调控蛋白Crb1的定位表达进行了研究,结果显示,随个体发育,子宫管腔及子宫腺腔不断扩大,子宫内膜上皮持续增厚、固有层内子宫腺逐渐发达;4周龄时子宫腔面出现纤毛,随发育进程越来越发达;在各发育阶段的小鼠子宫组织中均有Crb1的表达,随个体发育表达呈现先增强后减弱的趋势,在8周龄时达到最强;Crb1主要定位于子宫内膜上皮细胞和子宫腺上皮细胞的胞膜和胞质,提示Crb1可能与小鼠子宫内膜上皮细胞和腺上皮细胞的极性建立和维持有关。  相似文献

为明确牦牛不同妊娠阶段孕酮的来源,分别对53头不同妊娠阶段牦牛的黄体组织、肾上腺组织和胎盘组织进行体外细胞培养,同时对44头牦牛的上述组织进行匀浆处理,采用放射免疫分析法(RIA)测定培养液及组织匀浆上清液中孕酮的含量。结果显示,黄体细胞在体外培养时,妊娠前2个月内孕酮分泌量显著升高(P〈0.05):妊娠2-5个月,培养液中孕酮含量变化不明显(P〉0.05),但在妊娠5个月以后,其浓度又开始显著增加(P〈0.01)。肾上腺组织体外培养时,分泌孕酮的能力随不同个体差别较大,孕酮浓度在整个妊娠期变化不明显(P〉0.05)。胎盘组织体外培养时仅分泌少量孕酮,且在整个妊娠期其浓度变化不明显。黄体组织匀浆上清液中孕酮浓度在妊娠前2个月内呈上升趋势(P〈0.05),之后其浓度开始下降,至妊娠第4个月时下降到最低水平,以后又逐渐上升(P〈0.05),妊娠6-7个月时达到峰值,临近分娩时,又下降至较低水平。肾上腺组织匀浆中孕酮含量的变化差异较大,且无明显的变化趋势,胎盘组织匀浆中孕酮的含量也很低,甚至检测不到。结果表明,黄体是牦牛整个妊娠期孕酮的主要来源器官。  相似文献

在结扎子宫颈的情况下,在同一子宫的两侧子宫角分别注入生理盐水(对照组)和精液(试验组)以研究精液对子宫免疫和孕向发育的影响。试验共用母兔8只。子宫注入后48 h处死试验用兔,取子宫进行测量,并制片染色,用图像采集分析系统进行研究。结果显示:1)与对照组相比,兔子宫注入精液后可以显著(P〈0.05)增加子宫的长度、直径和子宫角指数;可明显增加子宫壁(P〈0.05)、黏膜上皮层(P〈0.01)和黏膜层(P〈0.05)的厚度;可显著增加黏膜中腺管(P〈0.05)和血管(P〈0.05)数量,可明显增加腺上皮的厚度(P〈0.05)。2)子宫注入精液后,子宫冲洗液中的细胞数(53.3±4.03×109/L)较对照组极显著增加(P〈0.01);单位面积子宫黏膜中肥大细胞的数量(2.85±0.05个/mm2)较对照组(3.72±0.04个/mm2)显著减少(P〈0.05)。子宫冲洗液涂片染色检查发现,子宫冲洗液中的细胞主要为嗜中性粒细胞。本研究表明,兔精液确实能够促进母兔子宫的孕向发育,能够改变子宫的免疫状态。  相似文献

以卵泡期小尾寒羊和萨福克羊为研究对象,采用免疫组织化学技术,对促凋亡基因(Bad)在生殖器官卵巢、输卵管和子宫中的表达与定位进行初步研究。结果表明,在卵巢组织中Bad基因在原始卵泡、初级卵泡和次级卵泡中呈中度着染,在三级卵泡中呈轻度着染,阳性细胞在卵泡上皮细胞和颗粒细胞中分散分布,呈轻度着染;输卵管上皮分泌细胞和部分纤毛细胞胞质中见Bad基因中等强度表达,输卵管壶腹部上皮中,Bad基因多表达于分泌细胞,纤毛细胞中有部分呈阳性且强度弱,宫管结合部、峡部和壶腹部Bad基因阳性细胞染色强度弱,均呈中度着染;子宫内膜子叶和基质中的阳性细胞数量极少,阳性细胞呈轻度着色。利用计算机图像分析技术测量小尾寒羊和萨福克羊生殖器官各组织细胞中Bad基因表达的阳性细胞率和平均光密度,结果Bad基因在这2品种绵羊组织中的表达规律大体相似,但也存在一些明显的差异:萨福克羊三级卵泡颗粒细胞Bad基因阳性细胞率显著高于小尾寒羊(P0.01);萨福克羊三级卵泡和成熟卵泡膜细胞Bad基因阳性细胞率和平均光密度均显著高于小尾寒羊(P0.01);萨福克羊子宫内膜腺体卵泡期Bad基因阳性细胞平均光密度显著高于小尾寒羊(P0.01);由此表明,小尾寒羊和萨福克羊卵泡期生殖器官中细胞凋亡的差异与Bad基因表达的差异,可能是造成两者繁殖力高低差异的基础。  相似文献

The secretions of the equine endometrial glands are essential for the survival, growth, and development of the conceptus in early pregnancy, and endometrial gland density is directly related to successful pregnancy outcome. Endometrial biopsy is routinely used to assess the reproductive potential of broodmares. Some previous studies have shown that equine endometrial glands are uniformly distributed throughout the uterus; however, other work has shown variation of the endometrial architecture between biopsy sites, suggesting that a single biopsy is not representative of the entire endometrium. The aims of this study were to assess and compare the endometrial gland density and thickness at four sampling sites in the uterus (the central segment of each uterine horn, the uterine horn-body junction, and the caudal portion of the uterine body). Endometrial samples from five nulliparous Thoroughbred mares in diestrus were obtained at necropsy and used for subsequent histomorphometric analysis. The caudal uterine body had a significantly lower endometrial gland density and endometrial thickness than the other sites. This may result in nutrient deprivation and reduced survival of embryos or fetuses in this region of the uterus. The endometrial gland density and endometrial thickness did not significantly differ between the other regions sampled, indicating that they are similarly suitable for embryonic implantation and fetal development. Our results suggest that the endometrial structure of the caudal uterine body of the mare is not representative of the endometrial morphology at other sites. Thus, the caudal uterine body is not a suitable site for routine endometrial biopsy.  相似文献
