ABSTRACT:   The genetic diversity of wild and hatchery-released Pacific herring Clupea pallasii collected from three brackish lakes and two bays in Honshu and Hokkaido, Japan was examined with five microsatellite loci. All loci showed high genetic variability with expected heterozygosities ranging from 0.815 to 0.945. Significant differences in genotypic and allelic distributions were detected among all locations except for between the two bays in Honshu Island. Pairwise population analysis based on the F ST values showed close genetic relationships among the locations in Hokkaido Island, and the hierarchical analyses of molecular variance showed significant genetic difference between the two islands. Those results suggest the existence of subpopulations due to natal homing. In addition, stocked fish showed as much genetic diversity as the wild fish. The pairwise population analyses also showed close relationships between the hatchery fish and the wild fish in respective stocking areas, showing that no effects of stocking programs on genetic diversity of wild populations were detected.  相似文献

ABSTRACT:   Barfin flounder and spotted halibut have been selected as target species for stock enhancement in Japan. Understanding the genetic condition of the wild stock is a principal requirement in any stock enhancement program. The genetic variability of barfin flounder and spotted halibut, and the population structure of spotted halibut were evaluated using microsatellite DNA markers (msDNA) and the control region of the mitocondrial DNA (mtDNA). Barfin flounder and spotted halibut showed high genetic variability at the msDNA level. Barfin flounder A was 16.7 and H e was 0.860; spotted halibut A n ranged from 7.7 to 10.2 and H e ranged from 0.710 to 0.774. At the mtDNA level, high haplotype ( h  = 0.922) and low nucleotide (π = 0.002) diversities were observed for barfin flounder; however, low haplotype and nucleotide diversities ( h  = 0.603–0.620 and π = 0.001–0.002), and very low haplotype and nucleotide diversities ( h  = 0.193 and π = 0.0003) were observed for spotted halibut in the north and south locations, respectively. Slight genetic differentiation among spotted halibut sampling locations was observed from the msDNA. MtDNA analyses showed genetic differentiation between north and south locations, but not within them. The designation of north-specific and south-specific management units in the future stock enhancement activities of spotted halibut is recommended.  相似文献

ABSTRACT:   Stocking of exogenous, hatchery-reared white-spotted charr Salvelinus leucomaenis has been conducted throughout much of their range in Honshu Island, Japan, to increase angling opportunities. Although the native charr populations are thought to have declined because of hybridization with introduced fish, their distribution and genetic status have been uncertain. Fine population structures of charr in the upper Tone River drainage were examined using mitochondrial DNA and microsatellite analyses so as to clarify the presence of native populations. One common mtDNA haplotype was detected in all populations in the Ohashi River and Watarase River, and four and one tributary populations were monomorphic for such haplotypes, respectively. However, several haplotypes, considered to have originated from stocked hatchery fish, were observed in the stocked and the remaining populations. Judging from the genetic integrity over a fine geographic scale, the former were considered as indicative of native populations and the latter as admixtures with hatchery fish. Comparisons of genetic diversity, deviations from the Hardy–Weinberg equilibrium, principal component analysis, and relatedness estimations based on microsatellite DNA can also provide evidence for distinguishing native populations from those influenced by hatchery fish.  相似文献

Twelve populations of Girella punctata, from widespread locations of the species’ range in Japan and Korea, were screened for sequence variability within the mitochondrial DNA (mtDNA) control region (n = 128) and at five polymorphic microsatellite loci (n = 547) to determine the genetic structure maintaining population integrity. mtDNA variability of 132 variable sites within a 334-bp region reveals shallow genetic differentiation across populations. The weak differentiation of G. punctata was partly supported by the screening of five polymorphic microsatellite loci. However, hierarchical analysis of molecular variance and principal component analysis on the basis of allele frequencies in microsatellite loci extracted a subtle substructure in a subsidiary population and in near-subsidiary populations in the semi-enclosed Seto Inland Sea.  相似文献

Abstract –  Conservation of life-history diversity found in Oncorhynchus mykiss requires knowledge of the underlying population structure and genetic basis of this variability. We analysed variation at 10 microsatellite loci from seven rivers across Kamchatka to identify population structure and to test for divergence between life-history forms. We found lower heterozygosity in Kamchatkan populations compared with North American populations, but population structure was substantial (region-wide F ST = 0.11) and followed an isolation-by-distance pattern similar to that reported for older North American populations. We found no evidence for genetic divergence between resident and anadromous individuals in the Sopochnaya River or between typically anadromous individuals and 'half-pounders' in the Utkholok River. A review of other studies of reproductive isolation, in combination with our results, suggests: (1) that pristine populations of steelhead should be expected to exhibit partial anadromy; and (2) that managing anadromous and resident individuals separately without demonstrating reproductive isolation is biologically unsound.  相似文献

Salmo trutta abanticus is a non‐anadromous trout species native to Lake Abant and Seven Lakes in Turkey. A restocking programme by captive breeding was initiated in 1999 to support S. trutta abanticus population. Reared 2‐year‐old juveniles from randomly caught wild parental individuals in Maçka breeding farm were introduced into Lake Abant. We aimed to compare genetic and morphological divergences between wild‐ and captive‐bred populations using seven microsatellite loci and geometric morphometric measurements. A significant genetic and morphological divergences were detected between all population in Fst and canonical variate analysis based on geometric morphometric with 10 homolog landmark. Eighty‐six microsatellites alleles were recorded across loci. Number of private alleles, observed alleles and observed heterozygosity are statistically significant higher in Maçka captive‐bred population than Lake Abant and Seven Lakes populations. Of 42 tests, three departures from Hardy–Weinberg equilibrium were detected in all populations after Bonferroni correction. Two pairs of loci (Ssa85 – Str73 and Str73‐Str543) in Maçka, one pairs of loci (Ssa85‐Str73) in Abant and two pairs of loci (Ssa85‐Str60 and Str73‐Str543) in Seven Lakes populations show linkage disequilibrium. Population structure analysed with Structure software showed three genetic groups (?K = 3) in our studied populations. Relatedness estimates show higher mean relatedness values (r = 0.220 ± 0.230) for Maçka captive‐breed population than wild populations of Abant Lake and Seven Lakes (r = 0.140 ± 0.210 and r = 0.170 ± 0.200 respectively).  相似文献

ABSTRACT:   In order to assess a daily change of genetic variability during spawning season, hatched larvae of red sea bream sampled on different dates were assayed by polymorphic markers such as microsatellite DNA (msDNA) and mitochondria DNA (mtDNA) control region. Based on the microsatellite loci, the average number of alleles per locus ranged between 13.7 and 18.3. The expected heterozygosities ranged between 0.843 and 0.919. A total of 23 mtDNA haplotypes were detected via digestion of mtDNA D-loop sequences with five endonucleases: Taq  I, Alu  I, Mbo  I, Rsa  I and Hinf  I. Significant fluctuation of genetic variability during spawning season was detected by both types of DNA markers. It was suggested that the genetic variability was maintained by pooling the seed fish collected on different spawning dates in a hatchery.  相似文献

利用同工酶技术结合聚丙烯酰胺电泳对江苏海州湾、浙江象山港、辽宁辽东湾三个海区毛蚶群体的同工酶遗传多样性进行了研究。在三个群体的9种同工酶中,共检测到了18个基因位点,海州湾、象山港和辽东湾三个群体的多态位点比率(P.95)分别为61.11%、55.56%、64.71%,平均杂合度观测值(Ho)变化范围在0.1689~0.1770间,位点有效等位基因数分别为1.4417、1.3917、1.4219,遗传多样性较丰富。各群体普遍存在着杂合子缺失现象,近交系数(Fis)分别为0.2140、0.1232、0.1797。比较了三个群体间的遗传分化指数(Fst)和群体每代迁移数(Nm),结果表明,海州湾群体和象山港群体间遗传分化较小,Fst=0.0448;而辽东湾群体与其它两个群体间的遗传分化较大,分别为Fst=0.0755,Fst=0.0812。聚类分析结果同样表明,海州湾群体首先与象山港群体相聚,后与辽东湾群体聚类。讨论了毛蚶群体分化机制,近交系数与繁殖生物学的相关性。  相似文献

ABSTRACT:   Allozyme variation of the littleneck clam Ruditapes philippinarum was evaluated in four samples from Nameishi and Matsuo in the Ariake Sea, Ryugatake and Ushibuka in the Shiranui Sea off Kyushu Island, Japan, and in one sample from Jinzhou, China, in the Bohai Sea. A Ruditapes bruguieri sample imported from the Korean Bay off Nampo, North Korea was also studied. Among the R. philippinarum samples, heterozygosity varied from 0.265 to 0.301 and F IS estimates indicated significant homozygosity excess in 15 of 40 loci analyzed. Deviations from Hardy–Weinberg equilibrium were significant in all samples ( P  < 0.05). Pairwise F ST estimates indicate that genetic differences between the Chinese and Japanese samples were very low, but significantly different from zero. Mixture proportions with 95% confidence intervals of Chinese R. philippinarum in Nameishi and Matsuo were estimated at 0.4098 [0.2512, 0.5705] and 0.4899 [0.3262, 0.6540], respectively. However, genetic invasion of stocked Chinese R. philippinarum into wild populations in the Ariake Sea remains uncertain due to the low precision of the estimates caused by the high similarity of allele frequencies between Jinzhou and the Ariake Sea.  相似文献

ABSTRACT:   In relation to the stock enhancement program for barfin flounder, hatchery juveniles produced in 2001 were genotyped using microsatellite DNA markers (msDNA) and then released to natural waters. Subsequently, recaptured individuals, designated as 'tentative recaptured', were examined using msDNA. In order to evaluate the effectiveness of the stock enhancement program, pairwise F ST and genic differentiation tests were used to estimate the genetic divergence between the wild samples, the hatchery broodstock, and the tentative recaptured samples. Analysis showed significant differentiation among these three groups. Pedigree determination by msDNA was used to establish the origin of the tentative recaptured individuals, in order to elucidate whether they were hatchery produced, 'real recaptured', or wild specimens. Wild individuals were not found. The effective population size of the real recaptured stock was very low ( N e = 16.6). Equal family survivability was observed between the released and real recaptured stocks, indicating that the genetic variability of the released stock was maintained in the natural environment. Future broodstock management, breeding designs, and family contribution equalization of the offspring to be released will be required in this rare species to avoid unintended genetic differentiation between the wild population and the hatchery broodstock.  相似文献

ABSTRACT:   Elongate ilisha Ilisha elongata is a commercially important fishery resource in both Japan and China. Age and growth of the species were investigated by scale analysis from June 1996 to July 1997 in Ariake Sound, Japan. Scale annuli were formed once a year mainly between June and July. The age was 1–6 years for both male and female individuals. There was no significant difference in the regression of fork length (FL) on scale radius ( R ) between sexes by a covariance analysis. The combined sex von Bertalanffy growth equations were expressed as L t  = 495.4 × (1 − e−0.3176( t  + 0.4108)), where L t is FL in mm at age t . Comparison of the age and growth among different populations from coastal waters in the western North Pacific Ocean suggested that the FL of the Ariake population is the smallest at each age. The spatio–temporal variations in growth are possibly relevant to environmental conditions of the growth grounds of subadults; however, there was no gradient change in the growth of I. elongata with latitude. Water temperature may not be the main factor affecting the growth of this species . The growth of the tropical population from Sarawak differs greatly from those of temperate and subtemperate populations, which implies genetic variation or phenotypic plasticity in different climate zones.  相似文献

Various hypotheses have been put forward to explain the mechanisms leading to recruitment variability in fish populations. These have been based on either physical (wind stress, upwelling) or biological (food and predation) processes. In the Bay of Biscay, the hypothesis of a physical influence on anchovy recruitment has been confirmed. Oceanographic conditions in the Bay of Biscay in the spring and summer, influenced by north-easterly winds of medium and low intensity, seem to induce good levels of recruitment to the anchovy ( Engraulis encrasicolus ) population. An index of upwelling was significantly correlated with annual recruitment of anchovy ( P  < 0.0001) for the period 1967–1996. This physical parameter explains about 59% of the variability in the recruitment of the Bay of Biscay anchovy. Two factors affecting productivity in the Bay of Biscay may be linked to north-easterly winds, namely weak upwelling and an extension of the area influenced by river outflows. Both of these factors, together with low turbulence and stability, may act to enhance survival of the early life-history stages of anchovy by increasing food availability. The potential use of this upwelling index to forecast the recruitment of the Bay of Biscay anchovy offers possibilities for improving the management of this population.  相似文献

To investigate the extent of genetic differentiation among wild populations of the Pacific cod Gadus macrocephalus, we have examined genetic polymorphism at five locations within Korean waters [Boryeong in the West Sea (WC-BR); Jinhae Bay in the South Sea (SC-JH); Jumunjin (EC-JM), Jukbyeon (EC-JB), and Bangeojin (EC-BJ) off the eastern coast of Korea] using mitochondrial DNA (mtDNA) control region and microsatellite DNA (msDNA) markers. Nucleotide sequence analysis of 584 bp in the variable portion of the 5′ end of the mtDNA control region revealed 27 variable nucleotide sites among 184 individuals, which defined eight, three, and 11 haplotypes in the western, southern, and eastern coast populations, respectively. The mtDNA analysis revealed a low variability but significant local differentiation among populations from these three areas within Korean waters. msDNA analysis also revealed moderate polymorphism in the wild populations, with a mean of 13.8–22.6 alleles per locus for the five msDNA markers and observed (and expected) heterozygosities of 0.755 (0.825) for the WC-BR, 0.793 (0.810) for the SC-JH, 0.920 (0.905) for the EC-BJ, 0.783 (0.865) for the EC-JB, and 0.804 (0.812) for the EC-JM populations. Analysis of msDNA loci indicated that Pacific cod sampled at the WC-BR, SC-JH, and EC-JB sites belong to genetically distinct populations. However, no significant difference was found between the Pacific cod population from SC-JH and that from EC-BJ. Consequently, three genetically distinct populations, namely, WC-BR, SC-JH and EC-BJ, and EC-JB, were identified using msDNA analysis. These results indicate that genetically distinct populations of Pacific cod are present in Korean coastal waters where spawning aggregations occur.  相似文献

ABSTRACT:   The genetic diversity and population structure of Cyclina sinensis were assessed using amplified fragment length polymorphism (AFLP). Three hundred and fifty-four AFLP loci were analyzed in 160 individuals collected from Lvshun, Lianyungang, Yueqing and Dongxing of coastal China. High levels of genetic diversity were detected, where the percentage of polymorphic loci and average expected heterozygosity ranged 88.4–98.9% and 0.304–0.365, respectively. Analysis of molecular variance showed that variation among populations (24.4%) was highly significant ( P  < 0.001). Accordingly, the global fixation index ( θ B) averaged over loci was high (0.205). The large genetic differences among populations indicate restricted gene flow, congruent with limited dispersal capability of C. sinensis . The unweighted pair group method with arithmetic mean (UPGMA) tree topology constructed on the basis of Nei's genetic distances between populations showed a clear separation of the northern two populations from the southern two populations, suggesting that gene flow between the northern and southern regions is extremely low. This finding is additionally supported by the separate clustering of the four populations in the results of principal coordinate analysis. The useful information obtained in this study will offer insights to fine-tune conservation and fishery management measures in the future.  相似文献

Abstract –  This study examined the degree and pattern of variability in trophic morphology in Arctic charr ( Salvelinus alpinus L. ) at three spatial scales: across 22 populations from Scotland and between and within two adjacent catchments (Laxford and Shin) in northern Scotland. In addition, the variability at six microsatellite loci between and within the Laxford and Shin systems was determined. Habitat use by charr differed significantly between populations. The pattern of variability in trophic morphology, known to influence foraging ability in charr, showed a very high degree of between-population variation with at least 52% of population pairs showing significant differences in head shape. Trophic morphology and genetic variation was also high over small geographical scales; variation being as high between charr from lakes within the same catchment, as between adjacent catchments. The pattern of both phenotypic and genotypic variation suggests a mosaic of variation across populations with geographically close populations often as distinct from each other as populations with much greater separation. Very low levels of effective migrants between populations, even within the same catchment, suggest that this variation is being maintained by very low straying rates between phenotypically and genetically distinct populations, even when there is no apparent barrier to movement. We conclude that the genetic and phenotypic integrity of charr populations across Scotland is high and that this adaptive radiation constitutes a 'hidden' element of diversity in northern freshwater systems. Two consequences of this are that the population (rather than the species) makes a more rational unit for the consideration of conservation strategies and that the habitat requirements and therefore management needs may differ significantly between populations.  相似文献

ABSTRACT:   This study was carried out to investigate the genetic variability and the population genetics of Sparus auratus. In fact, despite its importance in Mediterranean fisheries and aquaculture, very little it is known concerning its population structure. Samples of wild gilthead sea bream were collected in seven different localities along the Mediterranean and Atlantic coasts and were genetically characterized by means of microsatellite markers. Genotyping at four microsatellite loci revealed high polymorphism (7–38 alleles/locus) and expected heterozygosities, which ranged from 0.80 to 0.85. A slight but significant population structure was found ( F ST = 0.010). In fact, at least three populations of gilthead sea bream within the Western Mediterranean Sea were identified (Sardinian Sea, Sardinian Channel and Central Tyrrhenian Sea), which are also genetically differentiated from those of the Atlantic Ocean and the Adriatic Sea.  相似文献

Abstract –  Wild salmonid populations with only a few breeding adults may not exhibit a significant reduction in genetic variability compared with larger populations. Such an observation suggests that effective population sizes are larger than population size estimates based on direct adult counts and/or the mating strategy maximises outbreeding, contributing to increased heterozygosity. In the case of wild Atlantic salmon Salmo salar populations, stratification by age classes and sexes on the spawning grounds avoids inbreeding and increases genetic variability. We studied the breeding composition of four Spanish salmon populations. Over a 7-year period we concluded that the probability of within-cohort mating is very low: females generally reproduce after two sea-winters whereas males reproduce mostly as one sea-winter ( grilse ) and/or mature parr. Considering different levels of contribution of mature parr to spawning derived from field surveys, we developed a simple model for estimating effective population sizes and found that they doubled with 65% parr contribution expected for rivers at this latitude (43°N), and ranged from 100–800 individuals. The effect of between-cohort mating was modelled considering different ranges of differences in allele frequencies between cohorts and resulted in 28–50% increases in heterozygosity when considering a 65% parr contribution. The complex mating strategy of Atlantic salmon contributes to explain the high levels of genetic variability found for small populations of this species. This model can probably be extended to other animal species with mating strategies involving different cohorts.  相似文献

ABSTRACT:   The Japanese mitten crab Eriocheir japonica is a common grapsid species found throughout freshwater and estuarine regions in Japan. In order to obtain information on the genetic variation and population structure of this species, a polymerase chain reaction (PCR) restriction fragment length polymorphism (RFLP) analysis was conducted on the cytochrome oxidase subunit I (COI) of mitochondrial DNA, on 666 individuals from 19 sample sites covering the three main geographic regions of Japan (Main Islands, Okinawa, and Ogasawara). Genetic analysis using seven restriction enzymes produced an array of 61 composite haplotypes. Three regional groups corresponding to the three geographic regions were clearly identified by cluster and molecular variance model ( amova ) analyses. Each of the three groups showed dominant haplotypes that were almost completely absent in populations from the other geographic areas. Comparison with published information for other species indicates that the degree of genetic divergence between these three main groups is equivalent to the genetic distance between congeneric species. Thus, the population structure of the Japanese mitten crab, as inferred from mtDNA analysis, is formed by genetically distinct groups that closely reflect their geographic distribution in the Japanese archipelago as well as restricted gene flow.  相似文献

河南境内四水系宽鳍鱲野生群体的遗传多样性   总被引:2,自引:1,他引:1  
为准确掌握河南野生宽鳍鱲(Zacco platypus)群体遗传结构,本研究利用线粒体CO I基因检测了海河、黄河、淮河和长江水系宽鳍鱲群体的遗传多样性和种群分化情况。110个样品共检测出40个单倍型,平均单倍型多样性为0.92077,平均核苷酸多样性为0.02439。AMOVA分析显示,大多数遗传变异存在于宽鳍鱲群体间(60.59%),群体内的遗传变异为37.67%。系统发育树结果表明,长江水系宽鳍鱲群体遗传分化较大,一部分群体单独聚为一支,另外一部分群体与海河、黄河、淮河水系宽鳍鱲聚在一起,没有显示出明显的南北分化格局。种群历史动态结果推测宽鳍鱲群体经历过瓶颈效应,且该过程可能与第四纪中更新世气候波动有关。建议对河南境内的宽鳍鱲种群,特别是遗传多样性低的种群(海河水系)进行保护。  相似文献
