Downy mildew, incited by Peronospora parasitica (Pers.: Fr.) Fr., is a destructive disease of broccoli (Brassica oleraceaL., Italica Group). Resistant cultivars represent a desirable control method to provide a practical, environmentally benign, and long-term means of limiting damage from this disease. Doubled-haploid (DH) lines developed by us exhibit a high level of downy mildew resistance at the cotyledon stage. To determine the mode of inheritance for this resistance, a resistant DH line was crossed to a susceptible DH line to make an F1, from which F2 and backcross (BC) populations were developed. All populations were evaluated for response to artificial inoculation with P. parasitica at the cotyledon stage. All F1 plants (including reciprocals) were as resistant as the resistant parent, indicating no maternal effect for this trait. F2 populations segregated approximately 3resistant to 1 susceptible, BC populations using the resistant parent as the recurrent parent contained all resistant plants, and the BC to the susceptible parent segregated 1 resistant to 1 susceptible. These results indicate that resistance is controlled by a single dominant gene. This gene should be easily incorporated into F1 hybrids and used commercially to prevent downy mildew at the cotyledon stage. Summary An analysis of adult plant resistance of powdery mildew in 15 F1, F2 and F3 populations of pea derived from crossing 15 diverse and susceptible lines with one resistant line revealed that resistance to powdery mildew is controlled by duplicate recessive genes. Cucurbita pepo (pumpkin, squash,gourd) is an economically important species that is susceptible to the cucurbit powdery mildew fungus, Podosphaera xanthii(syn. Sphaerotheca fuliginea). ‘True French’, an open-pollinated cultivar of the Zucchini Group of C. pepo, was crossed with an unnamed powdery-mildew resistant straight neck-type accession, the resistance of which was apparently derived from an interspecific cross with a resistant wild species of Cucurbita,and resistant plants were selected in the F2 generation. This was followed by six cycles of backcross-pedigree selection for resistance, and resulted in the development of an accession true-breeding for resistance to powdery mildew and nearly isogenic to ‘True French’. The resistant and susceptible near-isogenics were crossed and seeds of the filial and backcross generations were produced. Plants of the parental accessions and their progenies were grown together in a controlled-environment chamber, exposed to the pathogenic fungus, and scored as resistant, partially resistant, or susceptible 27–33 days after sowing. The results indicated that resistance is conferred by a single incompletely dominant gene, designated Pm-0. Summary In studies of the inheritance of resistance, pea seedlings of seven lines in which stems and leaves were both resistant to Mycosphaerella pinodes were crossed with a line in which they were both susceptible. With seven of the crosses resistance was dominant to susceptibility. When F2 progenies of five crosses were inoculated on either stems or leaves independently, phenotypes segregated in a ratio of 3 resistant: 1 susceptible indicating that a single dominant gene controlled resistance. F2 progenies of one other cross gave ratios with a better fit to 9 resistant: 7 susceptible indicating that two co-dominant genes controlled resistance. The F2 progeny of another cross segregated in complex ratios indicating multigene resistance.When resistant lines JI 97 and JI 1089 were crossed with a susceptible line and leaves and stems of each F2 plant were inoculated, resistance phenotypes segregated independently demonstrating that leaf and stem resistance were controlled by different genes. In two experiments where the F2 progeny of the cross JI 97×JI 1089 were tested for stem and leaf resistance separately, both characters segregated in a ratio of 15 resistant:1 susceptible indicating that these two resistant lines contain two non-allelic genes for stem resistance (designated Rmp1 and Rmp2) and two for leaf resistance (designated Rmp3 and Rmp4). Evidence that the gene for leaf resistance in JI 1089 is located in linkage group 4 of Pisum sativum is presented.  相似文献

Lathyrus odoratus L. × Lathyrus belinensis L. hybrids were produced using L. belinensis as the pollen parent, with fertile seed produced by the L. odoratus parent. The F1 hybrid plants were completely self-sterile, but produced viable seeds when backcrossed to L. odoratus. The plants produced by backcrossing resembled L. odoratus, the flower colour being purple/magenta, and were self-fertile. Both hybrid plants and those produced by back crossing to L. odoratus were resistant to Erysiphe pisi DC that causes powdery mildew in sweet peas. Continued backcrossing resulted in hybrid plants, that closely resembled the L. odoratus parent, but segregated for complete resistance/susceptibility to E. pisi,with a ratio of 2.46:1 resistant to sensitive plants. This suggests the presence of a single dominant gene that confers resistance. When resistant and sensitive plants were inoculated with E. pisi and their leaf surfaces examined,using a Scanning Electron Microscope, it was found that although spores germinated on the leaves of both resistant and sensitive plants, spores present on resistant plants collapsed shortly after germination. Summary The oat line Pc54 was found to be resistant to powdery mildew under both field and glasshouse conditions. The ratio of resistant to susceptible F2 and F2 progeny of a cross between a selection from the Pc54 line (Cc7422) and a susceptible cultivar (Selma) showed that, in addition to carrying the crown rust resistance gene Pc54 and the pg15 gene for stem rust resistance, the mildew resistance of the Pc54 line was conditioned by a single incompletely dominant gene along with additional factors which modified the expression of resistance. Previous results, that there was no linkage between genes Pc54 and Pg15, were confirmed. In addition, there was no evidence of linkage between the mildew resistance gene and gene Pc54. Evaluation of selections from within the Pc54 line showed that the expression of both stem rust and mildew resistance was modified by, or linked to, plant height. The effectiveness of genes Pc54 and Pg15, as measured by virulence frequencies, in central and eastern Europe is described.  相似文献

Summary The current powdery mildew (Sphaerotheca fuligninea) resistant cucumber varieties suffer from leaf chlorosis during autumn, winter and early spring cultivation in the Netherlands. Therefore screening was carried out for novel powdery mildew resistance genes. From 177 accessions, derived from different sources, 108 accessions proved to be partially resistant to S. fuliginea. Crosses were made with 53 resistant accessions to distinguish the presence of novel genes. It is likely that the accessions C. sativus 2145, C. sativus LV 41, PI 188807, Vladivostokij, White and Yellow 1 have one or more recessive powdery mildew resistance genes, different from powdery mildew resistance genes of the line NPI, which was used for variety breeding. The inheritance of the Mlk powdery mildew resistance originating from ‘Heine 2174.50’ was analyzed by crossing the Mlk resistant cultivar ‘Ralle’× cv. ‘Amor’ (highly susceptible) and vice versa and by observing the reactions of F1- and F2-plants after inoculation with two different Mlk avirulent powdery mildew isolates. In all cases, a 3 (resistant): I (susceptible) segregation was found in F2. The reactions of the F2plants against the two powdery mildew isolates were identical in each case. Therefore, it is supposed that one dominant resistant gene is responsible for the resistant reactions against these two isolates. These results support the earlier assumption of Heun and Fischbeck (1987b) that the whole Mlk resistance pattern is controlled by a single gene.  相似文献

Summary The genetics of resistance to angular leaf spot caused by Pseudomonas syringae pv. tabaci in Nicotiana tabacum cultivars Burley 21 and Kentucky 14 was investigated by studying disease reactions to three isolates of parental, F1, F2 and backcross generations derived from crosses between the resistant cultivars and the susceptible cultivar Judy's Pride. Studies were conducted in the greenhouse and in field plant beds. Chi-square values were computed to determine whether the observed ratios for disease reactions deviated from expected Mendelian ratios for a single, dominant gene controlling resistance to angular leaf spot in tobacco. Based on the resistance of the F1 and the backcross generation to the resistant parent (BC-R), a 3 resistant: 1 susceptible segregation ratio in the F2, and a 1 resistant: 1 susceptible segregation ratio in the backcross generation to the susceptible parent (BC-S), it was concluded that resistance to three isolates of Pseudomonas syringae pv. tabaci is governed by a single, dominant gene.  相似文献

Summary F2-plant progenies, derived from seven crosses between susceptible erectoides breeding lines and conventional, nutans breeding lines with partial resistance to leaf rust incited by Puccinia hordei Otth., derived from Vada x Cebada Capa, were examined in a field and a glasshouse experiment. In the field experiment, the nutans plant progenies generally were more resistant to leaf rust and powdery mildew than the erectoides progenies. In the glasshouse experiment, the components of partial resistance to leaf rust of six erectoides and six nutans progenies derived from one cross were studied. The nutans progenies generally had lower infection frequencies than erectoides progenies with a similar infection level in the field experiment. The variation for leaf rust and powdery mildew infection in the field was similar for the nutans and erectoides progenies. This suggests that erectoides lines with a fairly high level of partial resistance to leaf rust and powdery mildew can be selected from these populations.  相似文献

Summary Using the cultivar Arina as the recurrent parent, six backcrosses were made with two donor lines carrying the leaf rust resistance genes Lr1 and Lr9, respectively. Selection for leaf rust resistance occurred at the seedling stage in the greenhouse; the first plants transferred to the field were BC6F4s. Frequency distribution of the 332 Lr1/7 × Arina and the 335 Lr9/7 × Arina lines showed continuous variation for yellow rust resistance and heading date in these leaf rust near-isogenic lines (NILs). Similar results were also obtained for plant height, for resistance to powdery mildew and glume blotch, as well as for baking quality characters in another set of more advanced NILs. The available information on the behaviour of one of the parents of cultivar Arina led to the conclusion that the expressed yellow rust resistance is quantitative and might possibly be durable.  相似文献

One of the most important diseases of barley (Hordeum vulgare) is powdery mildew, caused by Blumeria graminis f. sp. hordei. Spring barley line 173-1-2 was selected from a Moroccan landrace and revealed broad-spectrum resistance to powdery mildew. The objective of this study was to map and characterize the gene for seedling powdery mildew resistance in this line. After crossing with the susceptible cultivar ‘Manchuria’, genetic analysis of F2 and F3 families at the seedling stage revealed powdery mildew resistance in line 173-1-2 conditioned by a single recessive gene. Molecular analysis of non-segregating homozygous resistant and homozygous susceptible F2 plants conducted on the DArTseq platform (Diversity Arrays Technology Pty Ltd) identified significant markers which were converted to allele-specific PCR markers and tested among 94 F2 individuals. The new resistance gene was mapped on the long arm of chromosome 6H. No other powdery mildew recessive resistance gene has been located on 6H so far. Therefore, we concluded that the 173-1-2 barley line carries a novel recessive resistance gene designated as mlmr.  相似文献

The objective of this study was to determine the mode of inheritance of field resistance to downy mildew (Peronospora parasitica (Pers. ex Fr.) Fr.) in broccoli (Brassica oleracea var.italica) at the adult plant stage. The F1, F2 and F3 progeny of resistant and susceptible plants of broccoli were tested in the field under natural infection, in central Portugal, from August to December in two successive years. The plants were evaluated for resistance to downy mildew at maturity using a five-class scale of increasing susceptibility to the disease, which took into account the number of infected leaves and the size of the sporulating lesions. The F1 was completely resistant, the F2 segregated a clear 3 resistant: 1susceptible and the F3 confirmed the F2 segregation, which suggests a dominant character controlled by a single locus. This resistance has good potencial for direct use in commercial broccoli breeding or for transfer to other Brassica vegetables. Screening for resistance to powdery mildew of Solanum melongena and wild related species was made in the field under natural infection conditions. A total of 172 accessions originating from several geographical parts of the world were tested. Single plant selection for resistance was carried out and open-pollination was used. Most S. melongena accessions were susceptible or highly susceptible to the disease. By S0 to S3 selection, an increase in the overall powdery mildew resistance level of S. melongena population was obtained and four S. melongena lines possessing a high level of resistance were obtained. Among the wild Solanum species, S. laciniatum and S. nigrum showed to be non-host plants of L. taurica. S. quinquangolare showed no symptoms of powdery mildew, S. linnaeanum, S. aculeatissimum, S. aviculare, S. pseudocapsicum were highly resistant, S. spinosissimum was resistant, S. gilo, S. capsicoides were susceptible or highly susceptible, and plants of S. sisymbriifolium showed a widely variable disease reaction. Four S. melongena resistant lines were obtained: PAVEG 10187 S3, PAVEG 10196 S3, P.I. 230279 S3 and P.I. 419198 S3. These S. melongena lines together with the resistant wild species could be used for genetic studies, classical breeding programs and biotechnological applications.  相似文献

Summary Aegilops umbellulata acc. Y39 and Triticum carthlicum acc. PS5, immune to many powdery mildew isolates, were crossed to make an amphidiploid line Am9. The powdery mildew resistance of Am9 was transferred to common wheat cultivar Laizhou953 by crossing and backcrossing. In this study, the origin of powdery mildew resistance in a BC3F4:5 population derived from a cross of Am9 and Laizhou953 was identified. Microsatellite markers analysis showed that markers Xgwm257, Xgwm296, and Xgwm319, co-segregated with the powdery mildew resistance, whereas markers Xgwm210, Xgwm388/140, Xgwm388/170 and Xgwm526 were related to susceptibility and linked to resistance in repulsion. Of three markers related to resistance, Xgwm257 and Xgwm319 were codominant, whereas Xgwm296 was dominant. All three markers were Ae. umbellulata-specific indicating that resistance in the test population originated from Ae. umbellulata acc. Y39. The chromosome location and mapping of these linked microsatellite markers, the chromosome numbers of derived BC3F4:6 families, and chromosome pairing in F1 plants from a cross of a homozygous resistant BC3F4:5 plant and Laizhou953, showed that wheat chromosome 2B was substituted by Ae. umbellulata chromosome 2U. This is the first gene conferring powdery mildew resistance transferred to wheat from Ae. umbellulata, and it should be a novel resistance gene to powdery mildew. Summary Hordeum chilense is a wild barley with high crossability with Triticum, Hordeum and Secale. Its amphiploid with wheat, tritordeum, has potential as a new crop. H. chilense is highly resistant to the powdery mildew diseases of both wheat and barley. Whereas tritordeum is resistant to barley powdery mildew, its reaction to wheat powdery mildew is similar to that of its wheat parent. However H. chilense contributes to a reduced density of mildew colonies. This quantitative resistance of tritordeum is diluted at higher ploidy levels.  相似文献

Summary Partial resistance to powdery mildew in spring barley was evaluated in three plot types: large isolation plots, in 1.4 m2 plots in chessboard design with guard plots of spring wheat and in single rows. Percentage leaf area covered by powdery mildew was scored four to six times during the season and partial resistance was characterized by the area under the disease progress curve. Varietal differences were revealed in al three plot designs, differences between the most resistant and susceptible genotypes being of a factor five. Differences between varieties decreased with decreasing plot size. The relationship between single scores of amount of powdery mildew on the upper four leaves and the area under the disease progress curve was high in all plot designs during the first two to three weeks after heading, allowing selection for the trait by one or two scorings. Differential ranking of varieties between different plot designs was observed, and is assumed to be due to increasing plot interference with reduced plot size and reduced distance between plots. A reliable selection for partial resistance could be made in large isolation plots and in 1.4 m2 plots, but hardly in single rows.  相似文献

Small variant wheat populations created by induced mutagenesis (n = 69) or adventitious regeneration (n = 66) were intensively screened for an altered response (compared to the parent variety ‘Guardian’) to the causal pathogen of powdery mildew in wheat, Blumeria graminis f. sp. tritici. Intensive field screening following natural infection of replicated plots of wheat lines over two years revealed a total of 13 mutants exhibiting significantly greater resistance than ‘Guardian’: eight from induced mutagenesis (11.6%) of the M2 population and five from adventitious regeneration (7.6%). Complete resistance was identified in two lines, (one (M66) developed following induced mutagenesis, and the other (SC240) by adventitious regeneration). The complete resistance in the induced mutant was stable over two generations and was associated with a high frequency of leaf flecking, and consequently a low grain yield. Resistance in SC240 proved to be unstable; SC240 exhibited complete resistance to powdery mildew in the SC2 and SC3 generations, but only 20% of the SC4 plants were completely resistant, while the remainder were indistinguishable in mildew response to ‘Guardian’. The mildew response of all the SC5 generation of SC240 was not significantly different from ‘Guardian’. Yield analysis of the thirteen mutants with increased resistance in the presence of powdery mildew indicated that eleven exhibitedgrain yields at least as high as that of ‘Guardian’, while the mutant M19 exhibited a yield significantly higher than that of ‘Guardian’. V97‐3000 is a maturity group (MG) V soybean breeding line derived from SS 516 × V90‐2592 (Vance × V81‐1325) with high stachyose, small seed and powdery mildew resistance. A total of 53 F2:3 families were derived from a cross between V97‐3000 and a powdery mildew susceptible line V99‐5089. The 53 F2:3 families, each with 30 plants, were grown in the greenhouse for powdery mildew evaluation, and the corresponding 53 F2 plants were genotyped using simple sequence repeat (SSR) markers. Results showed that the 53 F2:3 families segregated in ratio of one resistant : two segregating : one susceptible (13 : 26 : 14) and the 26 segregating F2:3 families each exhibited a good fit to three resistant : one susceptible, indicating that resistance to powdery mildew is conditioned by a single dominant gene. The gene for powdery mildew resistance in V97‐3000 was mapped on chromosome 16 [linkage group (LG) J] flanked by Satt547 and Sat_396 on one side and Sat_393 on the other side with 3.8 cM and 3.9 cM distance, respectively. This study provides a new source of powdery mildew resistance and information of genetic location of the resistance gene and linked markers, which is useful for breeders selecting powdery mildew resistance through marker‐assisted selection (MAS) in soybean breeding programmes.  相似文献

Summary Inheritance studies were conducted to determine the genetic basis of adult-plant resistance in pepper (Capsicum annuum L.) to Phytophthora capsici. F1, backcrosses and F2 populations were developed using the resistant parent Criollo de Morellos 334 and susceptible parents Agronômico 10-G and Yolo Wonder. Pepper plants, at 36 days post-emergence, were inoculated near the base of the stem with an inoculum suspension of 5×104 zoospores/ml. Segregation ratios in the F2 generation of 13 resistant to 3 susceptible plants fit a 2-gene model for resistance with dominant and recessive epistasis.  相似文献
