用禽网状内皮组织增生病病毒(REV)感染SPF鸡胚次代成纤维细胞涂片为抗原,建立了检测REV抗体的间接免疫荧光法(IFA)。确定了待检血清稀释度1:64和兔抗鸡IgG荧光抗体的稀释度1:32的工作浓度。应用该IFA方法对人工感染REV的20只30日龄SPF鸡进行了检测,接种后4天时均呈阴性反应,7天时8只鸡呈阳性反应,14天20只全部呈阳性的反应。通过对NDV、IBDV、ALV、MDV、CIAV、  相似文献

将鸡传染性支气管炎病毒肾型 T 株 S1 基因c D N A 连接于pc D N A3 的 Hind I I I与 Ba m H I位点之间构建含有 C M V 启动子及 B G Hpoly A 信号序列的鸡传染性支气管炎病毒 S1 蛋白真核表达质粒。实验证明, S P F 鸡肌注免疫后血清 Ig G 抗体逐渐升高,至第35 日龄左右达到高峰,攻毒后血清 Ig G 抗体先下降而后升高,血清 Ig G 抗体升高幅度不及 I B 油苗免疫组。质粒 D N A 免疫鸡攻毒后有40 % 的鸡可耐过强毒的攻击,说明 S1 基因在体内获得了表达并使鸡产生了一定的免疫力。  相似文献

Dot—ELISA间接法检测鸡新城疫病毒(NDV)的研究   总被引:2,自引:0,他引:2  
常规方法分离纯化鸡IgG,免疫羊制备羊抗鸡IgG二抗,以辣根过氧化物酶(HRP)标记制备酶标记物的工作浓度为1:1000;选用硝酸纤维素(NC)膜作固相载体,建立检测NDV抗原的斑点间接酶联免疫吸附试验诊断方法。检测NDV鸡胚毒40份,阳性检出率100%;人工感染非免疫鸡的气管组织、肺组织各47份,检出率为93.6%;人工感染SPF鸡气管组织、肺组织各40份,检出率100%。与传染性支气管炎病毒(IBV)、减蛋综合症病毒(EDSV)、传染性法氏囊病病毒(IBDV)无交叉反应。试验显示该方法简便、特异、快速、结果直观,便于生产应用  相似文献

本研究以猪流行性腹泻病毒细胞培养物中提纯的抗原,组织毒兔获得的高兔血清建立了间接ELISA检测PEDV抗体水平。结果表明:包被怕浓度为1:20000,酶标羊抗猪IgG浓度选用1:250为测定抗PEDVIgG抗体的最达工作浓度。田间检测结果与临床发病预期结果一致。  相似文献

用纯化的伊氏锥虫变异表面糖蛋白(VSG)免疫大白鼠制备出抗血清,经饱和硫酸铰盐析,DE-52纤维素离子交换层析和Sepharose4B亲和层析提取VSG特异性多克隆IgG抗体(Abl)。用Abl免疫大白鼠和家兔制备出抗独特型血清。ELISA抑制试验检测结果表明,8份大白鼠抗独特型血清的抑制率高于25%。用正常大白鼠IgG致敏的醛化绵羊红细胞吸收兔抗独特型血清除去异种抗体,采用Abl-Sepharose4B亲和层析,可从每毫升兔抗独特型血清中获得97.4微克抗独特型抗体(Ab2)。经ELISA抑制试验检测表明,提取的兔抗独特型抗体(Ab2)对Abl与VSG的结合反应可产生明显的抑制效应,能识别Abl的抗原结合部位,其配位的空间构型与VSG表位具有相似性。  相似文献

应用提取纯化的抗IBDVIgG免疫BALB/c小鼠,取其脾细胞与SP2/0细胞在PEG作用下融合,应用ELISA法检测筛选,经有限稀释法克隆2次,获得2株(2B6株,5F4株)分泌抗IBDV独特型抗体的杂交瘤细胞株,其能诱生BALB/c小鼠产生高效价的含抗IBDV独特型抗体的腹水。用此独特型抗体分别与福氏完全佐剂、福氏不完全佐剂按1∶1比例乳化制备成抗IBDV独特型抗体疫苗,免疫接种SPF鸡和普通京白公鸡,然后用IBDV野毒株经点眼和滴鼻方式攻毒,保护率分别为8/8、14/14,从而证实抗独特型抗体疫苗有潜在的研究和应用价值。  相似文献

禽流感病毒(AIV)感染的鸡胚尿囊液经差速离心后,再经蔗糖密度梯度离心,提纯AIV,纯化的AIV经NP40处理并反复冻融,即为AIELISA抗原,用该抗原包被聚苯乙烯微量反应板。将健康鸡IgG提纯后免疫兔,制备兔抗鸡IgG,用过碘酸钠法制备辣根过氧化物酶标记的兔抗鸡IgG。确立了间接酶联免疫吸附试验(ELISA)检测禽流感抗体的最适工作条件,即:抗原包被浓度1.9~3.8μg/ml,每孔100μl,4℃冰箱过夜;用含0.5%牛血清白蛋白(BSA)的磷酸盐缓冲液,37℃封闭60分钟;待检血清最佳稀释度为180~1640,作用60分钟;酶标抗体作12000稀释,作用60分钟。根据对52份SPF鸡血清的检测结果制定了判定标准。  相似文献

本试验对1日龄感染鸡传染性贫血病毒(CIAV)雏鸡接种新城疫(ND)疫苗及其强毒攻击后外周血液免疫球蛋白IgG、IgM、IgA含量和HI抗体滴度的动态变化进行研究。结果发现,CIAV感染雏鸡ND疫苗免疫后,其外周血液IgG、IgM、IgA含量和HI抗体滴度,于接种ND疫苗后7 ̄28d较未感染CIAV的免疫对照雏鸡明显减少,表明CIAV感染雏鸡对ND疫苗免疫的体液免疫应答功能明显降低。ND强毒攻击后  相似文献

本试验将卡介菌多糖核酸应用于新城疫疫苗免疫雏鸡,观察其抗氧化和对鸡体免疫功能的影响。将200只1日龄AA肉鸡随机分为5组,每组40只,分别为卡介菌多糖核酸高(0.12 mg/只)、中(0.06 mg/只)、低(0.03 mg/只)剂量组,黄芪多糖组(阳性对照,0.06 mg/只)和空白对照组。每组鸡7日龄时免疫新城疫疫苗并同时按分组要求注射药物,连续7 d,空白对照组以生理盐水代替。分别于用药后5、10、15和20 d分批剖杀鸡,采取胸腺、脾脏、法氏囊和颈静脉血,测定免疫器官系数,鸡新城疫病毒(NDV)抗体滴度,血浆IgG、IgM、超氧化物歧化酶(SOD)、脂质过氧化物(LPO)和谷胱甘肽过氧化物酶(GSH-Px)的含量。结果表明,卡介菌多糖核酸能显著提高鸡血清中SOD和GSH-Px含量,降低LPO含量(P<0.05),具有良好的抗氧化功能;能显著提高受试鸡的免疫器官系数、NDV抗体滴度、血清中IgG和IgM含量(P<0.05),具有良好的免疫佐剂效应。  相似文献

表达新城疫病毒融合蛋白基因禽痘病毒重组体的构建   总被引:1,自引:0,他引:1  
通过RT-PCR扩增新城疫病毒F基因,利用禽痘病毒复合启动子、SV40 PolyA终止信号序列构建F基因表达盒。将F基因表达盒与loxp-GFP-loxP序列串联插入禽痘病毒重组臂,构建了禽痘病毒转移质粒载体。在脂质体介导下转染CEF,获得了带有绿色荧光信号的重组病毒rFPVFGFP。通过二次转染,利用Cre-loxP位点特异性重组将重组病毒基因组的GFP自动去除,结果获得了只含新城疫病毒F基因表达盒的重组禽痘病毒rFPVF。试验结果表明,rFPVF复制稳定,表达的F蛋白具有免疫原性,能够刺激鸡只产生抗新城疫病毒的中和抗体。  相似文献

试验旨在观察鸡传染性支气管炎病毒(infectious bronchitis virus,IBV)和鸡新城疫病毒(newcastle disease virus,NDV)之间的增殖干扰现象,分析在免疫过程中两种疫苗存在的免疫干扰,为确定疫苗的免疫程序提供依据。采用完全随机试验设计,选取10日龄鸡胚尿囊腔接种不同浓度的IBV、NDV以及不同浓度混合的IBV和NDV,收集尿囊液测定其血凝效价(HA);用不同浓度的IBV、NDV以及不同浓度混合的IBV和NDV分别免疫BALB/c小鼠及SPF雏鸡,收集血清,间接酶联免疫吸附法(ELISA)和血凝抑制(HI)试验测定IBV和NDV的抗体效价及抑制效价。结果表明,同胚培养时,无论先接种IBV后接种NDV还是先接种NDV后接种IBV或IBV和NDV同时接种,IBV对NDV均有干扰作用,而NDV对IBV没有干扰作用;接种IBV和NDV的小鼠,其产生IBV和NDV的抗体效价均低于单独免疫组,免疫的次序及免疫间隔时间对IBV和NDV的抗体效价均有影响,IBV对NDV的免疫效果具有较大的干扰作用。不同针混合免疫方式能提高IBV和NDV的抗体效价;接种IBV和NDV的雏鸡,免疫次序及免疫相隔时间对IBV和NDV的抗体效价均有影响,IBV对NDV的免疫效果具有较大的干扰作用;雏鸡和小鼠免疫血清HI试验数据表明,免疫次序及免疫间隔时间对IBV和NDV抗体的产生均有影响,但对NDV抗体的产生影响更明显,随着免疫间隔时间的增加,IBV和NDV的抗体水平呈增加趋势,雏鸡比小鼠更能敏感地反映出IBV对NDV的免疫干扰作用。混合注射时IBV和NDV的抗体水平均降低,IBV对NDV的免疫效果有干扰作用。因此,在同胚培养、小鼠及雏鸡的免疫中,IBV对NDV有干扰作用,而NDV对IBV无干扰作用。混合接种时IBV和NDV抗体效价均下降,IBV对NDV的免疫效果有干扰作用。免疫次序及免疫间隔时间对IBV和NDV抗体的产生均有影响,但随着免疫间隔的增加,IBV和NDV的抗体水平呈增加趋势。  相似文献

This experiment aimed to investigate the proliferation interference effects of the infectious bronchitis virus (IBV) and Newcastle disease virus (NDV),analyze the immune interference of two vaccines in the immune process,and provide a basis for determining vaccine immune program.This experiment was in a complete randomized design,10-day-old allantoic cavity of the chick embryo was inoculated with different concentrations of IBV and NDV and with a mixture of different concentrations of IBV and NDV,and then the allantoic fluid was collected for determination of the titer of hemagglutination (HA).Furthermore,BALB/c mice and SPF chicken were immunized with different concentrations of IBV and NDV and also with the mixed IBV and NDV with different concentrations,the blood of mice and chicken were collected,and antibody titer and inhibitory titer of IBV and NDV were determined by indirect enzyme linked immunosorbent assay (ELISA) and hemagglutination inhibition (HI) test.Experimental results showed that in the homeomorphic cultivation research,whether IBV and NDV were inoculated in a different order or inoculated at the same time,the inoculation of IBV behaved interference to the NDV,while the inoculation of NDV had no interference effects to IBV;Mice were inoculated with IBV and NDV,IBV and NDV antibody titers were lower than single immunized group,immune procedures and immune intervals often had effect on the titer of IBV and NDV antibody,IBV had interference on the immune effect of NDV,different needle mixed immunization can improve the antibody titer of IBV and NDV.In the group of SPF chicken inoculated with IBV and NDV,the order of the inoculation and the immune interval had effect on the titer of IBV and NDV antibody,IBV had a interference on the immune effect of NDV;In the immune serum inhibitory test,the data showed that the IBV and NDV antibody production were affected by the immune order of the inoculation and the interval,the effect on the NDV antibody was more obvious,but with the increase of the immune interval,the antibody level of IBV and NDV showed an increasing trend,the chicken was more sensitive than the mice to reflect the immune interference of IBV on NDV.IBVand NDV antibodies titer was also reduced in the groups that was inoculated with the mixed IBV and NDV,IBV had a interference on the immune effect of NDV.In the homeomorphic cultivation research and animal research,IBV behaved interference to the NDV,while NDV had no interference effects to IBV.IBV and NDV antibody titers decreased when mixed immunization,IBV had a interference on the immune effect of NDV.The order of the inoculation and the immune interval had effect on the antibody titer of IBV and NDV,but with the increase of the immune interval,the antibody level of IBV and NDV showed an increasing trend.  相似文献

Antibody-forming cells (AFCs) against Newcastle disease virus (NDV) and their immunoglobulin (Ig) class were demonstrated by a double immuno-enzyme histochemical technique. The AFCs were stained and quantified in spleen sections of chickens euthanatized at day 7 postexposure to the Roakin strain of NDV. The sections were incubated with NDV to determine the specificity of the AFCs. Bound virus was subsequently visualized with a primary monoclonal antibody (MAb), a secondary horseradish peroxidase-conjugated MAb, and 3-amino-9-ethylcarbazole as substrate. IgM and IgA were stained with MAbs and an alkaline phosphatase (AP)-conjugated secondary antibody. IgG class antibodies were demonstrated with an AP-conjugated rabbit serum. The final substrate for the three Igs was naphthol AS-MX-phosphate and fast blue BB. About 64-159/mm2 AFCs against NDV were detected. Of these virus-binding cells, about 55% produced IgM, 37% produced IgG, and the remaining 8% produced IgA.  相似文献

The humoral immune response and immunity conferred in chicks were compared following separate and combined oral vaccination with F strain of Newcastle disease virus (NDV) and HP1 strain of fowl pox virus. The haemagglutination inhibition (HI) antibody titre against NDV and passive haemagglutination (PHA) antibody titre against fowl pox virus were comparable in two respective groups. The serum IgG concentration increased significantly after the second vaccination in all the groups. The NDV vaccine induced significantly higher IgG production as compared to fowl pox virus vaccine. There was no significant difference in serum IgG concentration produced by combined vaccine and separate F strain vaccine. The protection afforded by combined and separate vaccinations did not vary significantly against challenge with virulent strains of NDV and fowl pox virus at different stages.  相似文献

Red blood cells (RBC) become sensitized following the elution of strain 575 Newcastle disease virus (NDV). The neuraminidase (NA) in the haemagglutinin (HA)-sialic acid configuration is inactive. The HA on sensitized RBC agglutinates normal RBC. The sialic acid on normal RBC initiated reactivation of the NA-a newly described function. Then normal-sensitized RBC agglomerates separated at 37 degrees C in the irreversible agglutination-separation (AS) reactions. With separation the AS products. HN spikes (150-200 kDa) and altered NDV, which contain fewer HN spikes than intact allantoic NDV, were removed from the sensitized RBC and the NDV membrane. Extraction of HN spikes from the membrane required more sialic acid than the removal of AS products from RBC. Thus 2 reactions were delineated for the orderly removal. Amounts of each released AS product suggest the source of the HN spikes. AS reactions and ether treatment of NDV increased the HA titres up to 19.2 fold. HA-sialic acid configurations were estimated by the amounts of normal RBC agglutinated by sensitized RBC and also by agglutination with fetuin. Elution of B1 vaccine, HN spikes from ether-treated NDV as well as AS products separated on sephadex resulted in incompletely sensitized RBC; fewer configurations were titrated with normal RBC; all failed to respond to anti-NA antibody. In contrast, sensitized RBC or suspensions of 575 NDV, but not the B1 vaccine strain, responded to both anti-NA and anti-HA antibody. Sensitization, slow elution, and responding to anti-NA antibody, which was accompanied by fluorescent foci on sensitized RBC, required intact NDV and the infrequent HA-sialic acid configuration. The NA was inactive for the HA-sialic acid configuration but cleaved fetuin, indicating substrate specificity. An inactive NA would allow time for fusion and NDV penetration rather than elution by an active NA early in NDV infection.  相似文献

Background:As a member of the Paramyxoviridae group,Newcastle disease virus(NDV) is the key causative agent of Newcastle disease(ND) that attacks chickens,turkeys and other avian birds.Surviving birds showed lower feed utilization,growth performance or egg production,which results in severe economic losses.The purpose of this study was to determine the effect of different doses of NDV immunization on growth performance,plasma variables and immune response of broiler chickens.Methods:A total of 480 one-day-old Arbor Acres broilers were randomly administrated with 0,4,6 or 8 doses of NDV at 12 d and 28 d,respectively.Each group consisted often replicates with 12 birds each.Growth performance and organ weight were recorded.Plasma concentration of glucose,total protein,cholesterol,triglycerides and nonesterified fatty acid was determined using commercial kits.The concentration of plasma corticosterone and insulin was measured using commercially available radio immune assay kits.Serum antibody titer and peripheral blood lymphocyte proliferation were also recorded.Results:The results showed that NDV decreased body weight gain(BWG),and increased Feed:Gain ratio at 1-21 d at all doses(P 0.05).Plasma insulin concentration was lower in all immunization groups after the first immunization at12 d(P 0.01).The rest of the plasma indexes were not affected by NDV immunization,including glucose,total protein,cholesterol,triglycerides,nonesterified fatty acid,heterophil/lymphocyte ratio,as well as the proliferation of peripheral blood lymphocyte(P 0.05).Compared with the control group,NDV treatment elevated NDV antibody titer at 10 d after the first inoculation(P 0.05),and at d 5,9 and 13 after the second inoculation(P 0.05).Repeated NDV inoculation had no deleterious impacts on body composition at 42 d,and nutrient accretion rates at 8-42 d(P0.05).Conclusions:In conclusion,NDV challenge decreased BWG and feed efficiency in earlier stage of growth.However,NDV treatment at 6 doses down-regulated the Feed:Gain ratio by 6.36%throughout the whole growing period.These data suggest that appropriate lower doses of NDV inoculation increase feed efficiency of broiler chickens.  相似文献

Two experiments were conducted to determine the effect of breeder vaccination program and maternal antibody on the efficacy of Newcastle disease immunization of 1-day-old chicks. Experimental protocol was the same for both. In the first experiment, broilers were from breeders that were 32 weeks old, and in the second experiment, broilers were from breeders 50 weeks old. Breeders received three live Newcastle disease virus (NDV) vaccines and either a killed vaccine at 18 weeks or continual live boosting at 60-to-70-day intervals through lay. Broilers were vaccinated at 1 day of age with a commercial coarse-spray machine; they were bled, sera were examined for antibody against NDV, and broilers were challenged with virulent NDV at 2, 4, and 6 weeks of age. In the first experiment, maternal antibody was higher in chicks from the younger breeders given the inactivated vaccine, and in the second experiment maternal antibody was higher in chicks from older breeders given continual live vaccines. Higher antibody in 1-day-old broilers resulted in fewer vaccine-induced reactions, less vaccine virus shed, and decreased duration of vaccine-induced immunity from coarse-spray vaccination.  相似文献
