Viral encephalopathy and retinopathy disease caused by betanodavirus, genus of the family Nodaviridae, affects marine, wild and farmed species including sea bass, one of the most important farmed species in Europe. This work describes a reliable and sensitive indirect ELISA assay to detect betanodavirus in biological samples using a polyclonal antiserum (pAb 283) against the 283/I09 virus strain, the most common red‐spotted grouper nervous necrosis virus (RGNNV) genotype in the Mediterranean area, and a capture‐based ELISA using a monoclonal antibody (mAb 4C3) specific to a common epitope present on the capsid protein. Using adsorbed, purified VERv preparation, the detection limit of indirect ELISA was 2 μg mL?1 (3 × 105 TCID50 per mL), whereas for capture‐based ELISA, the sensitivity for the antigen in solution was 17 μg mL?1 (35 × 105 TCID50 per mL). The capture‐based ELISA was employed to detect VERv in brain homogenates of in vivo infected sea bass and resulted positive in 22 of 32 samples, some of these with a high viral load estimates (about 1.1 × 108 TCID50 per mL). The ELISA system we propose may be helpful in investigations where coupling of viral content in fish tissues with the presence of circulating VERv‐specific IgM is required, or for use in samples where PCR is difficult to perform.  相似文献

Fish nodavirus (betanodavirus), a viral pathogen responsible for viral nervous necrosis (VNN) was isolated from infected Asian sea bass (Lates calcarifer). The distribution, clearance and expression of nodavirus vaccine, on the basis of DNA vaccine (pFNCPE42 DNA‐pcDNA3.1) construction, were analysed in tissues of the Asian seabass by PCR, RT‐PCR, ELISA and Immunohistochemistry. Fish immunized with a single intramuscular injection of 20 μg of the pFNCPE42‐DNA vaccine showed a significant increase in the serum antibody level in the 3rd week after vaccination, compared to control eukaryotic expression vector pcDNA3.1 vaccinated fish. Results from PCR studies indicated that the vaccine‐containing plasmids were distributed in heart, intestine, gill, muscle and liver 10 days after vaccination. Clearance of pFNCPE42‐DNA vaccine was studied at 10, 25, 50, 75 and 100 days of post vaccination (d p.v). At 100 days p.v. pFNCPE42‐DNA was cleared from muscle of vaccinated sea bass. In vitro and in vivo expression of fish nodavirus capsid protein gene (FNCP) was determined by fluorescent microscopy. Asian seabass was immunized with pFNCPE42‐DNA vaccine at a dose of 20 μg per fish and were challenged with betanodavirus by intramuscular injection. The vaccinated seabass was protected from nodaviral infection and 77.33% of relative percent survival (RPS) was recorded.  相似文献

The effects of different salinities (0, 2, 4, 6 and 10‰) on food intake, growth, food conversion, and body composition of the freshwater catfish Mystus vittatus (Bloch) were studied. Under a restricted feeding schedule daily intake of food was found to be salinity dependent. Fish reared in 10‰ consumed more Tubifex tubifex, converted less efficiently and displayed poor growth as compared to individuals reared in fresh water. Fish flesh production decreased from 483 g (fresh water) to 177 g (10‰ salinity) as the salinity was increased. Water content of the fish was found to decrease with increase in salinity, while maximum ash (25.56%) and fat (42.25%) were exhibited by fish reared in 10‰ salinity.  相似文献

The spermatozoa of various marine teleosts (sea bass, Dicentrarchus labrax; sea bream, Sparus auratus) or fresh water teleosts (trout, Salmo gairdneri; pike, Esox lucius; guppy, Poecilia reticulata) were diluted in media of different salinities. Motility, morphological changes and fertilizing ability were the criteria used in judging the effects of such treatments. The medium best adapted to dilution of sea-fish sperm had a salinity of about 20‰. Sperm motility was increased and prolonged, and fertilization rate was significantly improved (P<0.05) for the sea bass at a 11000 dilution. For freshwater fish (trout and pike), an extender with about 7‰ salinity increased motility time and fertility ability (P<0.01) as compared to freshwater. After dilution in fresh water the structure of trout spermatozoa was considerably altered (rupture of plasma membrane and mitochondrial swelling). When spermatozoa were diluted in the extender, there were no significant structural changes in trout, but alteration occurred in the mid-piece of the guppy spermatozoon. It is concluded that fresh water or sea water are not the best media for the practice of artificial insemination in freshwater or marine fish. Investigations should be carried out to define the best extender for use in techniques of artificial insemination in fish produced in aquaculture.  相似文献

Effect of rearing salinity on the performance of juvenile golden pompano Trachinotus ovatus (Linnaeus 1758) was studied under a laboratory condition. Fish growth, survival, RNA/DNA ratio, pepsin activity, α‐amylase activity and FCR were used as evaluation criteria. The growth and RNA/DNA ratio were significantly affected by the rearing salinity. High growth rate and RNA/DNA ratio were observed when fish were reared at the salinity of 34‰. The pepsin activity of fish was not significantly affected by the rearing salinity. However, the α‐amylase activity of fish was significantly affected by the rearing salinity. The α‐amylase activity of fish reared at the salinity of 10‰ was significantly lower than fish cultured at the salinity of 34‰. Rearing salinity can significantly affect the FCR of juvenile golden pompano. The FCR of fish cultured at the salinity of 10‰ was 5‐times higher than the FCR of fish reared at 34‰. Results from the present study indicate that juvenile golden pompano can be reared above 26‰ without affecting fish performance, and the salinity of 10‰ may be too low to rear juvenile golden pompano as fish growth, RNA/DNA ratio and α‐amylase activity were reduced.  相似文献

Viral encephalopathy and retinopathy (VER) is a serious neuropathological fish disease affecting in the Mediterranean aquaculture mainly European sea bass, Dicentrarchus labrax. It is well known that betanodaviruses are neurotropic viruses that replicate in nerve tissues, preferentially brain and retina. However, routes of entry and progression of the virus in the central nervous system (CNS) remain unclear. The role of four tissues—eye, oesophagus, gills and skin—as possible gateways of a betanodavirus, the redspotted grouper nervous necrosis virus (RGNNV), was investigated after experimental challenges performed on European seabass juveniles. The dispersal pattern of Betanodavirus at primarily stages of the disease was also assessed, using a real-time qPCR assay. The development of typical clinical signs of VER, the presence of characteristic histopathological lesions in the brain and retina and the detection of viral RNA in the tissues of all experimental groups ascertained that successful invasion of RGNNV under all experimental routes was achieved. Transneuronal spread along pathways known to be connected to the initial site of entry seems to be the predominant scenario of viral progression in the CNS. Furthermore, viraemia appeared only after the installation of the infection in the brain.  相似文献

Effects of salinity on the growth, survival, superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX) and sodium‐potassium adenosine triphosphatase (Na+K+‐ATPase) activities of juvenile golden pompano Trachinotus ovatus were studied under a laboratory condition. Experimental fish were reared at the salinities of 10‰, 18‰, 26‰ and 34‰ for 30 days. Growth and survival of juvenile golden pompano were significantly affected by the rearing salinity. Fish reared at 34‰ achieved the highest specific growth rate, while the highest survival was obtained when fish were cultured at the salinity of 26‰. The highest GPX activity was obtained when fish were cultured at 26‰, and the lowest GPX activity was observed when fish reared at 34‰ salinity. The SOD activities of fish reared at 18‰ and 34‰ were significantly higher than those reared at 10‰ and 26‰. The lowest of Na+K+‐ATPase activity was obtained in fish reared at 34‰, while the highest Na+K+‐ATPase activity was obtained when fish reared at 18‰. Results from present study indicate that juvenile golden pompano can be reared above 18‰ without sacrificing fish survival, and the best growth can be achieved when fish is reared at the salinity of 34‰. The salinity of 10‰ may be too low for juvenile golden pompano as the growth, survival and SOD activity were reduced.  相似文献

Mass mortalities of the European sea bass, Dicentrarchus labrax (L.), occurred in different ongrowing units in Greece. A presumptive diagnosis of viral nervous necrosis (VNN) was made on the basis of the light microscopic observations of a vacuolating encephalopathy and retinopathy. Positive peroxidase reaction with VNN antiserum confirmed nodavirus as the causative agent. Usually recorded in larvae and juvenile stages of different marine fish species, VNN is described in adult sea bass and sea bass reared in floating cages in the Mediterranean for the first time. The horizontal transmission of the disease has been strongly suspected. Histological analyses clearly demonstrate that cells other than nerve cells are also infected. Some epidemiological aspects of the disease are described and their implications for the establishment of prophylactic guidelines are discussed.  相似文献

The giant freshwater prawn, Macrobrachium rosenbergii, is a species with a high commercial value in aquaculture. Two experiments were performed to determine the effects of salinities on the osmoregulation, growth and molting cycles of M. rosenbergii during growout. The first experiment was designed to determine whether these animals are capable of adapting to the changes in salinity seen in salinity intrusions in tropical deltas, with an incremental increase in salinity of 3‰ per day from 0‰ to 30‰ Haemolymph osmolality was rapidly regulated up to salinities of 15‰ , whereas animals conformed at higher salinities. The second experiment determined the growth, moulting cycle, osmolality, muscle water content and mortality during a 4‐month experiment at 0‰, 15‰ or 25‰ salinity. The weight gains in 0‰ and 15‰ were not significantly different and were comparable to the growth rates achieved in production farms with body mass increases of 2.6 and 2.3‐fold their initial body mass, respectively, after 4 months. The 25‰ group suffered from low growth, high mortality and a significantly lower moulting frequency. These data show that this species can be reared in brackish water up to 15‰, allowing for farming in the large areas impacted by salt water intrusions in tropical deltas.  相似文献

海刺猬(Glyptocidaris crenularis)是海胆中经济价值较高的一种,因其性腺品质较高,具有很大的市场潜力。然而,其生长发育却极易受到环境因素的影响,这也就制约了海刺猬人工养殖的开展。在适温(18.5~23.6℃)条件下,对41‰、38‰、35‰、32‰、29‰、26‰、23‰、20‰、17‰九个盐度下,海刺猬的活力、摄食和存活状况进行了初步的观察和分析。实验结果表明,盐度会对海刺猬的活力、摄食和存活产生影响。在盐度为29‰、32‰、35‰组中海刺猬的状态良好,活力较强;盐度过高或过低,都会抑制海刺猬的摄食。而在出现死亡的各组中,高盐度组(38‰、41‰)出现死亡现象的时间早于低盐组(17‰),同时其存活率低于低盐组。因此,相对于高盐度来说,海刺猬具有一定的低盐耐受力。研究表明,在适温(18.5~23.6℃)条件下,海刺猬的最适盐度为29‰~32‰。  相似文献

A study was performed to examine the effects of salinity on water quality, fish performance, carcass composition and haemato‐biochemical parameters in juvenile meagre, Argyrosomus regius. Fish (5.0 g) were stocked in fibreglass tanks at four salinity levels: 8‰, 16‰, 24‰ and 32‰, and fed a pelleted diet (47/17 protein/lipid) for 56 days. Results indicated that the growth, feed utilization, carcass composition and haemato‐biochemical parameters of meagre gradually improved with the increase in salinity up to 24‰ and then significantly (p ≤ .05) decreased at 32‰. The survival per cent showed a significant decrease when A. regius exposed to 8‰ salinity. An improvement with 32%, 47% and 34.1% of FCR, protein productive value and energy utilization was detected at 24‰ compared with 8‰ salinity respectively. The highest content of protein and the lowest of lipids were recorded in fish carcass at 24‰ compared with the opposite trend at 8‰ salinity. The 24‰ salinity treatment exhibited the highest value of haemoglobin (4.9 g/dl) and the lowest ratio (0.73) of albumin/globulin. The serum total protein, albumin and globulin were significantly higher at 24‰ and 32‰ salinity than those at 8‰ and 16‰ salinity groups. These findings indicate that 24‰ salinity level might be the best for meagre.  相似文献

The effects of rearing volume on on‐growing European sea bass performance and stress parameters were investigated for the first time in a pilot aquaculture farm. Fish were held under the same initial stocking densities in triplicate net‐pen cages of different sizes (1.4, 45 and 252 m3) for a period of 8 months. Results showed significant differences among the experimental groups in most parameters tested, with better performance in the two larger rearing volumes. In particular, growth rate showed a linear association with rearing volume, being 0.68 g day?1 for the large cage group, 0.56 g day?1 and 0.32 g day?1 for the medium and the small groups respectively. The feed conversion ratio and per cent survival (%) were also better in the large cage group. Fish reared in the small and medium‐sized cages showed higher plasma cortisol concentrations than those reared in large cages, which showed low basal cortisol concentrations. Additionally, after an acute chasing stress challenge, fish in the large and small groups, but not the medium group, showed increased cortisol concentrations. Differences also occurred in the ratio of the expression of cortisol receptors, namely the mineralocorticoid (mr) and glucocorticoid receptors (gr). In specific, the ratio of mr to grmRNA expression in the liver was higher in fish reared in the small cages. These findings verify that experimental scale significantly affects experimental results and is a critical factor for the interpretation of results.  相似文献

Abstract.— Juvenile white bass Morone chrysops (age 158 d, 110.4 mm total length) were reared at salinity levels of 0, 4, 8, 12, 16, and 20 parts per thousand (ppt). Fish (10/tank) were stocked in 64-L tanks with four replicates/treatment ( N = 40heatment) and fed a pelleted feed daily. Fish were sampled semimonthly for 70 d. Fish reared at salinities of 12 ppt and below were significantly ( P ≤ 0.05) larger (mean = 151.5 mm total length; range 146.8–155.7 mm) than fish reared at salinities of 16 and 20 ppt (mean 133.7 mm total length; range 131.5–135.8 mm) after 70 d. Survival was also higher ( P ≤ 0.05) among fish reared at salinities of 12 ppt or lower (mean 85.6%; range 77.5%–92.5%) compared to those reared at the two highest salinities (42.5% survival at 16 ppt; 5.0% survival at 20 ppt). Thus, although typically found in freshwater habitats, white bass can survive and grow in brackish water.  相似文献

The aim of this study was to examine species‐specific differences in pre‐ and post‐stress concentrations of haematological, metabolic and hormonal parameters in two Mediterranean fish species, one with relatively high (European sea bass, Dicentrarchus labrax) and the other with low (meagre, Argyrosomus regius) basal and post‐stress blood cortisol concentrations, in relation to different water temperatures. Fish were reared in net‐pen sea cages and exposed to an identical acute stressor (crowding and chasing with a net for 5 min) in three different periods of the year. Results indicated that inter‐specific differences occurred in most of the examined parameters. In addition, within each species, differences between sampling periods existed in plasma cortisol, glucose, osmolarity, blood pH and muscle glycogen concentration. Glucose, lactate and osmotic pressure showed a constant pattern of change, with maxima at half an hour post stress in both species, while cortisol, and blood and muscle pH, were significantly altered only in E. sea bass. No alterations were observed in liver and muscle glycogen concentrations in E. sea bass, while a significant reduction was evident only in the coldest temperature in meagre. It is concluded that there are important species‐specific differences in the magnitude of hormonal and metabolic response to acute stress and that both basal and post‐stress blood cortisol concentrations are affected by the sampling period.  相似文献

High protein distiller's dried grains (HP‐DDG) are co‐products of ethanol production that uses prefractionation technology. An 8‐week growth trial was conducted to investigate the effect of partial replacement of soybean meal (SBM) by three levels of HP‐DDG (30%, 40% and 50%) on growth performance, physiological parameters and histological changes of the intestine of European sea bass, Dicentrarchus labrax. The results indicated that an increased dietary level of HP‐DDG of more than 30% significantly increases growth performance and improved the FCR of sea bass. In addition, replacement of SBM by HP‐DDG enhanced feed intake efficiency and the health status of fish. Haematology and serum biochemistry (haemoglobin (Hb), red blood cells (RBCs), white blood cells (WBCs), packed cell volume (PCV %) and humeral immune parameters including total protein, albumin, globulin, cholesterol, lysozyme activity and total antioxidant capacity significantly increased with increase HP‐DDG inclusion levels. The findings of this study indicated that HP‐DDG is a good complementary protein source for inclusion aquaculture diets and levels above 30% as a replacement of SBM did not compromise growth performance and physiological parameters of sea bass while enhancing some important indices of health status in this species.  相似文献

Identification of B-cell epitopes on the betanodavirus capsid protein   总被引:1,自引:0,他引:1  
The pepscan procedure was used to identify betanodavirus B-cell epitopes recognized by neutralizing mouse monoclonal antibodies (MAbs) and serum samples obtained from sea bass, Dicentrarchus labrax, naturally infected with betanodavirus. Pepscan was performed with a panel of thirty-four 12-mer synthetic peptides that mimicked the entire betanodavirus capsid protein. Sea bass serum samples reacted strongly with three regions of the capsid protein comprising amino acid residues 1-32, 91-162 and 181-212. The latter region was also recognized by neutralizing MAbs and coincided with a region of high antigenic propensity identified by an antigen prediction algorithm. These data suggest that a region of the betanodavirus capsid protein spanning amino acid residues 181-212 may represent a neutralization domain that could potentially be used to inform the development of nodavirus vaccines and immunodiagnostic reagents.  相似文献

Asian seabass (Lates calcarifer) has been recognized as an economically important aquaculture species which can be adapted to and cultivated in wide range of salinities. The number of freshwater intensive seabass farms in Thailand is increasing annually. Here, we first describe the susceptibility of Asian seabass, which were cultured in freshwater, to Streptococcus inae (SI) and their pathological changes. Three isolates of putative SI were identified using a combination of standard biochemical assays and species‐specific PCR prior subjected to in vivo challenge. Accumulated mortalities of the fish which received 107 CFU fish?1 of either SI1J, SI SGSA or SI2J were 90%, 90% and 100% at 7 days‐post infection (dpi), respectively, and mortalities increased sharply between 3 and 5 dpi. Clinical signs such as erratic swimming and opaque eyes were identified from a few infected fish, while most died rapidly without any abnormal signs. Histopathological manifestations were observed in the multiple organs (kidney, liver and brain). Haemorrhage, hyperhemia, cellular degeneration and inflammatory cells infiltration were commonly found within the internal organs. Notably, the formation of numerous encyst‐like lesion aggregated by eosinophilic cells, resembling macrophages, were typically found in the brain of the infected fish. Summarily, this study first revealed that freshwater reared Asian seabass is highly susceptible to SI infection and haemorrhagic septicaemia was a major pathological change that could be found in the infected fish.  相似文献

Betanodaviruses are the causative agents of the disease known as viral nervous necrosis (VNN) or viral encephalopathy and retinopathy (VER) in a variety of marine and freshwater fish species. The aim of this study was to demonstrate experimental infection of an isolate of betanodavirus (RGNNV genotype) in freshwater fish, Gambusia affinis, for elucidation of transmission mechanism and potential use as a laboratory model. Morbidity and mortality rate was significantly higher by injection route of infection as compared to immersion by bath and resembled the natural infection of juvenile marine fish. The fish in disease affected group showed severe neurological disorders accompanied by extensive vacuolar degeneration and mild to moderate neuronal necrosis of the brain in comparison to control. Amplification of ~?427 bp product in the variable region of the coat protein gene of betanodavirus was achieved by RT-PCR with 100% sequence homology to RGNNV genotype.  相似文献
