Forty-three reticuloruminal epithelial receptors, with excitatory receptive fields within the reticulum, were isolated in 19 anaesthetised sheep. The responsiveness of these receptors to acetic, propionic, butyric and DL-lactic acids were assessed as well as their ability to be activated by rumen fluids obtained from sheep with induced ruminal lactic acidosis. Eighteen (41.9 per cent) receptors were excited by all three volatile fatty acids (VFAs) and 17 (39.5 per cent) responded to two VFAs. Butyric acid was the most potent volatile fatty acid eliciting responses in 41 (95.3 per cent) receptors. Acetic acid activated 33 (76.7 per cent) receptors and propionic acid excited 22 (53.5 per cent) receptors. DL-lactic acid only activated 13 (30.2 per cent) receptors, six of which required concentrations of 200 mM to be activated. Rumen fluids obtained when forestomach motility was impaired, activated 37 (81.1 per cent) of the 43 receptors, whereas rumen fluids obtained when forestomach motility was normal did not activate any receptors. Despite the high levels of lactic acid in rumen fluids which activated epithelial receptors, this acid was not responsible for receptor excitation.  相似文献

试验旨在应用Rusitec系统研究不同粒度日粮对绵羊瘤胃发酵特性的影响。试验分为两个处理组,日粮颗粒分别为5和0.45 mm,每个处理4个重复,适应期10 d,采样期2 d,测定不同时间发酵液总产气量、甲烷产量及发酵参数。结果显示,5 mm颗粒组的总产气量极显著高于0.45 mm颗粒组(P<0.01),甲烷产量不受粒度的影响(P>0.05);5 mm颗粒组投料后6 h的pH、6和9 h的乙酸、6 h的异丁酸、12 h的乙酸/丙酸、0和12 h的总挥发性脂肪酸、6和9 h的总氮均显著或极显著大于0.45 mm颗粒组(P<0.05;P<0.01),而5 mm颗粒组12 h的丙酸及3、6、9、12 h戊酸则显著或极显著小于0.45 mm组(P<0.05;P<0.01),不同粒度对其余指标没有显著影响(P>0.05)。由此可知,人工瘤胃试验中日粮的不同粒度可影响发酵特性,较大颗粒有较高的总产气量、乙酸、乙酸/丙酸、总挥发性脂肪酸和总氮含量。  相似文献

This experiment was aimed to investigate the effects of different particle sizes of feeds on rumen fermentation using the rumen simulation technique (Rusitec). There were two treatments (5 and 0.45 mm) with 4 replicates per group. After 10 days'adaptations,the fermentation parameters were determined for 2 days,and the total gas production,methane production and fermentation parameters were determined. The results showed that the total gas production of 5 nm group was extremely significantly higher than that of 0.45 nm group (P <0.01),while methane production of the two group was no significant difference (P >0.05).The ruminal fluid pH at 6 h,acetic acid at 6 and 9 h, isobutyric acid at 6 h, acetic acid to propionic acid ratio at 12 h,butyric acid at 6 h,total volatile fatty acids at 0,12 h and total nitrogen at 6 and 9 h in 5 mm group were significantly or extremely significantly greater than those in 0.45 mm group (P <0.05;P <0.01). While propionic acid at 12 h and valeric acid at 3,6,9 and 12 h in 5 mm group were significantly or extremely significantly lower than that in 0.45 mm group (P <0.05;P <0.01).Other parameters were not significantly affected by particle size (P >0.05). These results certified that the particle size of feeds in Rusitec did affected the rumen fermentation. Large size improved the gas production,acetic acid to propionic acid ratio,total volatile fatty acids and total nitrogen cotents.  相似文献

本试验旨在建立一种测定瘤胃液中甲酸、乙酸、丙酸、异丁酸、丁酸、异戊酸和戊酸的气相色谱-质谱联用方法。样品选用湘东黑山羊和荷斯坦奶牛的瘤胃液,18000 r/min离心10 min,取上层清液。将上层清液与25%的偏磷酸按照9∶1(v∶v)的比例混合均匀,静置0.5 h后,4℃条件下18000 r/min离心10 min,取上清液过0.22μm滤膜,进行气相色谱-质谱分析。以DB-FFAP毛细管柱进行分离,甲酸、乙酸、丙酸、异丁酸、丁酸、异戊酸和戊酸在9 min内达到基线分离。结果表明:这7种组分色谱峰保留时间的相对标准偏差均小于0.5%,峰面积的相对标准偏差均小于5.0%。甲酸、乙酸、丙酸、异丁酸、丁酸、异戊酸和戊酸分别在0.8~121.0μg/mL、0.3~3081.0μg/mL、0.4~1165.2μg/mL、0.4~275.1μg/mL、0.2~543.0μg/mL、0.5~129.9μg/mL和0.4~271.5μg/mL内线性关系良好,线性相关系数均达到0.998以上,检出限(3倍信噪比)在0.069~0.252μg/mL。山羊和奶牛瘤胃液样品的加标回收率分别为91.8%~103.8%和78.7%~95.2%。本试验建立的测定瘤胃液中甲酸以及其他6种挥发性脂肪酸的气相色谱-质谱联用方法选择性强、样品用量少、操作简单,可为反刍动物瘤胃碳水化合物代谢及甲烷生成的相关研究提供技术支撑。  相似文献

A rapid quantitative method for the gas chromatographic determination of volatile fatty acids in aqueous media is described. It includes the use of formic acid for the elimination of “ghost peaks” and of iso-valeric acid as an internal standard. The method has been used for the examination of bovine rumen fluid and of silage.Statistical treatment of the result shows that the fluid from lactating cows has a significantly higher content of butyric acid than has the fluid from dry cows; such a difference could not be demonstrated as to acetic, propionic and valeric acid.Some experience from the determination of the volatile fatty acids in silage is also reported.  相似文献

为研究饲料中添加胍基乙酸对牛瘤胃体外发酵参数的影响,试验选取3只身体健康、2岁左右、体重为(200~250)kg、安装瘤胃瘘管的锦江黄牛供采集瘤胃液。采取体外产气法对添加不同水平胍基乙酸(0.1%、0.2%、0.3%、0.4%)体外发酵培养,测定24 h培养液中pH、氨态氮(NH3-N)、挥发性脂肪酸(VFA)、微生物蛋白(MCP)浓度及干物质消化率等瘤胃发酵指标。结果表明:(1)随着胍基乙酸添加量的增加,0.4%添加组产气量显著提高40.82%(P<0.05)。胍基乙酸添加组的pH显著低于空白对照组(P<0.05),但均在正常范围内;NH3-N浓度随着添加剂量的增加呈先降低后升高趋势,其中0.1%、0.2%添加组显著低于空白对照组(P<0.05),分别降低了10.55%、15.29%。瘤胃微生物蛋白含量随着胍基乙酸添加量的增加而增加,0.4%添加组显著提高了1.41%(P<0.05)。干物质消化率(IVDMD)随着胍基乙酸添加量的增加先增加后减少,其中0.4%添加组显著提高了11.24%(P<0.05)。(2)胍基乙酸组挥发性脂肪酸总量、乙酸、丙酸、丁酸有上升趋势,并且0.4%添加组显著高于空白对照组(P<0.05),其中TVFA提高了6.58%,丙酸含量提高了9.08%。但是乙酸/丙酸各组无显著差异(P>0.05)。综上所述,胍基乙酸添加可以促进瘤胃发酵功能。  相似文献

建立了程序升温气相色谱法测定羊瘤胃液中的挥发性脂肪酸。样品经过滤离心,偏磷酸溶液酸化后,直接进样分析。结果表明:该方法实现了羊瘤胃液中乙酸、丙酸和正丁酸直接快速分离,方法的精密度、回收率较高。  相似文献

利用体外产气法研究添加不同剂量延胡索酸二钠对绵羊瘤胃发酵的影响。采用单因素完全随机设计,发酵底物以可溶性淀粉和纤维素为碳源,以胰蛋白胨为氮源,对照组不添加延胡索酸二钠,试验1、2、3组分别添加占发酵底物干物质1.0%、2.0%和3.0%的延胡索酸二钠。结果显示,与对照组相比,添加1%的延胡索酸二钠(试验1组)可显著(P<0.05)提高瘤胃的总挥发性脂肪酸、乙酸、丙酸、丁酸产量以及体外产气量和干物质降解率;添加2%的延胡索酸二钠(试验2组)可显著(P<0.05)提高瘤胃的总挥发性脂肪酸、乙酸、丙酸、丁酸产量和干物质降解率;添加3%的延胡索酸二钠组(试验3组)的瘤胃氨氮浓度、总挥发性脂肪酸和乙酸产量、产气量、干物质降解率与对照组差异不显著(P>0.05),但丙酸产量显著增加(P<0.05)。综上表明,添加1%和2%的延胡索酸二钠会显著提高绵羊瘤胃发酵性能,但添加3%的延胡索酸二钠,瘤胃发酵能力有所降低。  相似文献

Digestive activities were studied in test calves (n = 12) in relation to age and feed intake. The calves were isolated after birth from adult cows. Since the 14th day of age, milk replacers were fed with concentrate feed mixture TG and alfalfa hay, i.e. the calves were in the period of plant feeding. The investigation lasted from the end of milk feeding (50th day) to the age of six months. Overall health condition was studied clinically. The characteristics of digestive activities studied in rumen fluid at weekly--monthly intervals were pH, ammonia content, total content of volatile fatty acids, content of particular volatile fatty acids, incidence and number of infusoria. At the age of 50 days, actual pH of rumen fluid was on the average 6.07 and it ranged from 5.84 to 6.76 in the next period. Ammonia content was the highest at the age of 50 days (21.49 mmol/l rumen fluid), then it dropped to 9.83 mmol/l rumen fluid at the age of 180 days. The average total concentration of volatile fatty acids (C2-C5) made 117.00 mmol/l during the first examination, it increased to 132.98 mmol/l at the age of 84 days, and it dropped to 94.16 mmol/l rumen fluid at the age of six months. A similar tendency was found in the volatile fatty acids. The infusoria were found out only at the age of 120 days--their numbers were 155 000 per ml; at the age of 180 days their numbers rose to 368 000 per ml rumen fluid. 为研究牧草物候期对其脂肪酸组成和放牧牦牛乳中共轭亚油酸(CLA)含量的影响及其相关关系,本研究在甘肃省甘南牧区选取平均体重为(234.9±10.5) kg的10头带犊甘南牦牛作为研究对象(自然放牧),分别于牧草返青期、盛草期、枯草期采集试验牦牛采食牧草、乳和瘤胃液,分别测定瘤胃液、牧草中脂肪酸及乳脂中共轭亚油酸组成,并分析其相关关系,以期探明牧草物候期对牦牛瘤胃液和牧草中脂肪酸组成与放牧牦牛乳中共轭亚油酸含量的影响及其变化规律。结果表明:物候期显著影响了牧草中月桂酸、肉豆蔻酸、棕榈酸、硬脂酸、亚油酸、亚麻酸、总脂肪酸、多不饱和脂肪酸、多不饱和脂肪酸/总脂肪酸(P<0.05),盛草期牧草中月桂酸、肉豆蔻酸、棕榈酸、亚油酸、亚麻酸、总脂肪酸、多不饱和脂肪酸、多不饱和脂肪酸含量/总脂肪酸均显著高于返青期和枯草期(P<0.05);物候期显著影响了牦牛乳中共轭亚油酸异构体c9t11、共轭亚油酸异构体t10c12和共轭亚油酸总含量(P<0.05),盛草期放牧牦牛乳中共轭亚油酸异构体c9t11、共轭亚油酸异构体t10c12和共轭亚油酸总含量显著高于返青期和枯草期(P<0.05);物候期显著影响了牦牛瘤胃pH值、乙酸、丙酸、乙酸/丙酸、异丁酸、丁酸、异戊酸、戊酸、总挥发酸和反式11-油酸含量(P<0.05),盛草期牦牛瘤胃乙酸、丙酸、丁酸、戊酸总挥发酸和反式11-油酸含量显著高于返青期和枯黄期(P<0.05);牧草粗蛋白、粗脂肪、无氮浸出物、单宁酸、总脂肪酸和瘤胃液pH值与放牧牦牛乳中CLA含量显著正相关(P<0.05),粗纤维和粗灰分与放牧牦牛乳中CLA含量显著负相关(P<0.05),多不饱和脂肪酸(PUFAs)和瘤胃总挥发酸(TVFA)与放牧牦牛乳中CLA含量无显著相关关系(P>0.05)。  相似文献

本试验旨在研究过瘤胃胆碱(RPC)对成年黔北麻羊瘤胃发酵及血浆生化指标的影响。以体况良好,年龄、体重[(35.0±1.2)kg]一致的成年黔北麻羊6只为试验动物,分为对照组和试验组,每组3个重复,每个重复1只羊。采用交叉试验设计,对照组饲喂基础饲粮,不含RPC;试验组补饲10 g/d的RPC。试验持续30 d,分2期,每期15 d;每期最后1 d采集瘤胃液测定p H、氨态氮浓度、微生物蛋白产量、挥发性脂肪酸(乙酸、丙酸、丁酸)浓度、纤维素酶(微晶纤维素酶、羧甲基纤维素酶、纤维二糖酶、木聚糖酶)活性,同时采血测定血浆葡萄糖、尿素氮、总蛋白、白蛋白、总胆固醇、甘油三酯、肌酐浓度。结果显示:1)RPC对山羊采食速度没有影响,对采食量没有显著影响(P0.05);2)RPC对瘤胃液p H、氨态氮浓度、微生物蛋白产量、挥发性脂肪酸浓度及4种纤维素酶活性影响较小,2组间差异均不显著(P0.05);3)RPC对血浆生化指标影响较小,2组间差异均不显著(P0.05)。由此可见,补饲10 g/d的RPC对成年黔北麻羊的采食量、瘤胃发酵参数和血浆生化指标没有显著影响。  相似文献

旨在探讨利用体外产气法研究不同瘤胃降解蛋白平衡日粮(RDPB,g·kg-1 DM)对瘤胃发酵、总产气量和甲烷产量的影响.试验选用美国Ankom产气系统,采用随机区组试验设计,配置6种不同瘤胃降解蛋白平衡日粮,分别为日粮1(-20 g· kg-1 DM),日粮2(-10 g· kg-1 DM),日粮3(0 g· kg-1 DM),日粮4(10g·kg-1DM),日粮5(20 g·kg-1 DM)和日粮6(30 g· kg-1 DM),每组2个重复,培养期为24 h.结果表明:(1)本研究中6种类型日粮发酵液的pH、乙酸/丙酸、产气量和CH4产生量表现为日粮3(P<0.01)或日粮4(P<0.01)值最低.(2)挥发性脂防酸乙酸、丙酸、丁酸以及氨氮的浓度随着瘤冒降解平衡值的增加而增加,表明改变日粮瘤胃降解蛋白平衡可以影响瘤胃发酵.而瘤胃降解蛋白平衡对总挥发性脂肪酸以及原虫数量无显著影响.(3)日粮瘤胃降解蛋白平衡值影响不同时间点甲烷的浓度和24 h甲烷产量.其中以日稂4(P<0.05)甲烷总产量最低.由此可见,在体外培养条件下,改变日粮RDPB影响了瘤胃发酵液pH、氨态氮(NH3-N)及挥发性脂肪酸(VFA)浓度、总产气量、CH4产生量,综合考虑其对瘤胃发酵和甲烷产量等指标的影响,体外产气条件下适宜的RDPB水平为0~10g·kg-1 DM.  相似文献

本试验旨在通过体外法研究脂多糖(LPS)对瘤胃发酵的影响。选取4头健康、体况相近、安装有永久性瘤胃瘘管的荷斯坦奶牛用于瘤胃液的采集。试验分为对照组(不添加LPS)和试验组(添加LPS 100 000 EU/m L),分别在发酵2、4、8、12、24 h后取样,测定发酵液p H及挥发性脂肪酸、氨态氮、微生物蛋白浓度。结果表明:随着发酵时间的延长,对照组与试验组发酵液p H逐渐下降,发酵液总挥发性脂肪酸、氨态氮、微生物蛋白浓度及乙酸、丙酸、丁酸含量逐渐增加,对照组与试验组之间均无显著差异(P0.05),但在8、24 h试验组发酵液p H与对照组相比有降低的趋势(0.05≤P0.10),4、8、24 h试验组氨态氮浓度与对照组相比有降低的趋势(0.05≤P0.10)。结果显示,在体外发酵条件下,添加LPS具有降低发酵液p H以及氨态氮浓度的趋势,但这一影响并不显著。  相似文献

In order to confirm earlier fragmentary results, the effect of defaunation and refaunation of the rumen on the fermentation pattern and flow of N-components in the proximal duodenum of two sheep was investigated. Defaunation had no effect on acetic acid as a proportion of the total volatile fatty acids in the rumen, while the proportions of propionic acid increased with a concomitant decrease in butyrate. Refaunation resulted in lower acetic acid and higher butyric acid proportions. The concentration of ammonia N in the rumen was clearly decreased after defaunation, already indicating an effect of the elimination of protozoa on nitrogen metabolism in the rumen. Defaunation also increased significantly the flow of total N, non ammonia N and individual and total amino acids in the proximal duodenum. Defaunation resulted in higher bacterial growth efficiency, significantly in one sheep, but the decrease after refaunation was statistically significant for both sheep. Determination of rumen digestibility of organic matter and acid detergent fibre (ADF) revealed lower values in the absence of the protozoa, while total digestibility was only influenced to a much lower extent. This indicated a shift of digestion from rumen to the lower digestive tract. Finally, earlier work is discussed in the light of the present findings.  相似文献

本试验以3只健康的卡拉库尔羊为瘤胃液供体,采集瘤胃液进行体外发酵培养,试验设计为基础日粮中分别添加不同来源样组:对照组(G0)、10%胀果甘草粗提物组(G1)、10%刺果甘草粗提物组(G2)、10%刚果甘草粗提物(G3)、10%甘草原样(G4);分别添加不同水平甘草提取物组:0%组(Z0)、0.5%组(Z1)、1%组(Z2)、3%组(Z3)、5%组(Z4)、7%组(Z5),研究其对瘤胃发酵参数:pH、挥发性脂肪酸(乙酸、丙酸、丁酸)、产气量、氨态氮浓度(NH3-N)的影响。结果显示,本试验添加的G1、G2、G3、G4组在一定程度上均可以降低瘤胃产气量和乙酸产量;添加不同剂量的甘草粗提取物时,Z3组的添加剂量使瘤胃pH在正常范围,同时对产气量无显著影响,乙酸/丙酸比值为最低。  相似文献

试验旨在利用体外厌氧发酵培养技术研究不同硒源及不同硒水平的添加对奶牛瘤胃挥发性脂肪酸(VFA)含量的影响。试验按照3×4完全随机试验设计,第一因素为硒源,即亚硒酸钠、硒代蛋氨酸和酵母硒3种,第二因素为硒添加量,即每毫升培养液中设0μg、0.1μg、0.2μg和0.3μg共4个添加水平,其中0μg/mL添加水平为对照组,共计10个处理组,每个处理组均进行3h、6h、9h、12h、24h和48h的体外批次培养,每个处理组设3个重复。试验结果表明,体外培养液中添加3种不同来源的硒对乙酸、丁酸、TVFA的影响组间差异显著(P<0.05),对丙酸、乙酸丙酸比影响较小(P>0.05),其中以添加酵母硒的乙酸、丙酸、丁酸、TVFA的效果最好,添加硒代蛋氨酸次之;体外培养液中添加0.3μg/mL硒,可显著提高乙酸、丁酸、TVFA的含量(P<0.05),对丙酸及乙酸丙酸比影响组间差异不显著,但均以0.3μg/mL硒组的效果较好。  相似文献

Forty-four reticuloruminal epithelial receptors were tested with rumen fluids obtained from 12 sheep before they were intraruminally infused with 4.0M acetic acid (8 sheep) or 4.0M butyric acid (4 sheep; preinfusion rumen fluid) and with rumen fluids obtained at the onset of ruminal stasis (abolition rumen fluid). The preinfusion rumen fluids from the 8 acetic acid-infused sheep (mean pH, 6.55) contained 1.7 mM nondissociated volatile fatty acids (VFA)/L and excited none of the 25 receptors tested. Preinfusion rumen fluids from the 4 butyric acid-infused sheep (mean pH, 6.98) contained 0.3 mM nondissociated VFA/L and also did not evoke responses in any of the 19 receptors tested. Abolition rumen fluids from sheep treated with acetic acid excited 17 of the 25 receptors tested and contained 89.4 mM nondissociated VFA/L, of which nondissociated acetic acid comprised 85.0 mM/L. Abolition rumen fluids from sheep treated with butyric acid activated 14 of the 19 receptors tested and contained 61.1 mM nondissociated VFA/L, of which 38.7 mM/L was nondissociated butyric acid. Preinfusion rumen fluids whose pH values were adjusted to that of abolition rumen fluids with HCl contained nondissociated VFA levels ranging from 16.3 mM/L (acetic acid-treated sheep) to 20.6 mM/L (butyric acid-treated sheep) and elicited responses in 4 of 30 receptors tested. The contents of volatile fatty acids in the rumen fluid were studied in calves at an early (seven weeks of age) and traditional weaning term (nine weeks of age). Weaning at the age of seven weeks enabled to save 16.2 kg of milk replacer per calf; there was also a lower content of concentrates in the starter feed with a supplement of Amylastim. The health condition of the calves was good in both groups. The average daily weight gains for the period from the second to the ninth week of age were 0.550 kg in the early weaned calves and 0.690 kg in the calves weaned at a normal time. Early weaning had a positive influence on the development of rumen metabolism. Calves weaned at the age of seven weeks, compared with those weaned at the age of nine weeks, had much higher concentrations of VFA (p less than 0.05) in rumen fluid (at the age of seven weeks: 130.49 mmol per litre vs. 111.53 mmol per litre; at the age of eight weeks: 119.74 mmol per litre vs. 96.98 mmol per litre). Early weaned calves had the statistically significantly (p less than 0.05) higher proportions of propionic acid, butyric acid, i-butyric acid, and valeric acid. 试验旨在研究饲粮中添加不同水平的甘露寡糖(MOS)对荷斯坦奶牛围产后期瘤胃微生物区系的影响。选取60头荷斯坦奶牛,随机分4组,每组15头。对照组饲喂基础饲粮,低、中、高剂量组分别在基础饲粮基础上添加5、10、15 g/(头·d)MOS。结果显示,随着MOS添加量的增加,奶牛产后21 d瘤胃液中微生物蛋白含量、总挥发性脂肪酸(TVFA)浓度以及乙酸、丙酸、丁酸和异丁酸浓度和乙丙比均线性上升(P<0.05)。厚壁菌门和蓝藻菌门相对丰度呈线性下降(P<0.01),而淋溶菌门相对丰度线性上升(P<0.05)。研究在20个样品中共检测到147个属,其中11个属与MOS添加量显著相关,主要来自厚壁菌门,且与瘤胃液中TVFA浓度和乙酸摩尔比例具有较强的相关性。研究表明,围产期奶牛日粮中添加一定量的MOS可能能通过调节瘤胃微生物菌群结构来提高纤维的瘤胃降解率,进而改善奶牛生产性能,最适添加量为15 g/(头·d)。  相似文献
