为了解华南地区猪群中猪流感病毒(SIV)的流行及其遗传变异情况,本研究从2016年~2017年广东、广西等地猪群236份猪肺脏病料组织和143份鼻拭子样品中分离鉴定得到3株SIV,全基因组测序和遗传演化分析结果显示,3个分离株均属于H1N1亚型欧亚类禽分支SIV,并且均与pdm09分支病毒株发生了重组。HA蛋白分子特征分析结果显示,A/Swine/Guangxi/NK/2016 HA蛋白第23位糖基化位点发生了缺失。3265份血清样品抗体监测结果显示,欧亚类禽H1N1、pdm09 H1N1和H3N2 SIV的血清抗体阳性率分别为27.53%、20.98%和34.85%。另外,0.64%的(21份)血清样品为H9N2亚型流感病毒抗体阳性,并且猪群中不同亚型和不同分支SIV之间混合感染的情况非常普遍。猪群中流感病毒血清抗体监测结果显示,EA H1N1、pdm09和H3N2亚型SIV HI抗体滴度最高均可达到1:1280,而H9N2亚型HI抗体滴度最高为1:160,表明H9N2 AIV虽然可以感染猪,但对猪还不适应。各月份的血清抗体阳性率分析显示,SIV的流行具有季节性,冬季(11月、12月和1月份)的流行最为严重。本研究可为华南地区猪群SI防控及疫苗株的筛选提供参考依据。  相似文献

猪流感病毒H1、H3、N1、N2亚型分型 RT-PCR方法的建立   总被引:1,自引:0,他引:1  
根据GenBank中H1N1和H3N2亚型猪流感病毒(SIV)血凝素(hemagglutinin,HA)、神经氨酸酶(neuraminidase,NA)和M基因保守序列,分别设计合成了5对特异性引物,利用RT-PCR技术对SIV的型和亚型进行鉴定。结果表明,该方法的型RT-PCR可以检测出104 EID50病毒量所提取的RNA;H1、H3、N1和N2的亚型RT-PCR均可以检测出104 EID50病毒量所提取的RNA。除每对特异性引物所对应的亚型外,对其他亚型及猪繁殖与呼吸综合征病毒(PRRSV)和猪瘟病毒(CSFV)的检测均为阴性,应用该方法对临床样品进行检测,其结果与病毒分离结果符合率为100%。结果表明,该方法特异性好、敏感性高,有望成为SIV的一种特异、敏感、快速的分型检测方法,为猪流感分子流行病学的调查奠定了良好的基础。  相似文献

本研究2012年底从辽宁省某屠宰场猪鼻咽拭子样品中分离到1株流感病毒,经HA—HI试验和RT—PCR鉴定为H1N1亚型猪流感病毒株,命名为A/swine/Liaoning/01/2012(H1N1),通过对病毒的8个基因片段克隆并测序,并利用分子生物学软件进行遗传演化分析。结果表明,分离株HA基因裂解位点附近的氨基酸序列为IPSIQSRjG,符合低致病力流感病毒的分子特征。全基因组进化树结果表明,分离株的8个基因片段与A/swine/Jiangsu/40/2011(H1N1)株核苷酸同源性最高,分离株处在类禽型H1N1亚型遗传进化分支上;由于类禽型H1N1猪流感病毒具有潜在感染人的潜力,在国外和国内均有感染人的报道,因此,辽宁省首次分离到该型猪流感病毒对全省养猪业和公共卫生安全具有重要意义,值得深入研究。  相似文献

Novel swine influenza virus subtype H3N1 in Italy   总被引:2,自引:0,他引:2  
To date, three subtypes of swine influenza viruses, H1N1, H1N2, and H3N2 have been isolated in Italy. In 2006, a novel swine influenza virus subtype (H3N1) was isolated from coughing pigs. RT-PCR performed on lung tissues, experimental infection in pigs with the novel isolate, and cloning the virus by plaque assay confirmed this unique H and N combination. The novel isolate was also antigenically and genetically characterized. Genetic and phylogenetic analysis showed that the complete HA gene of the H3N1 strain has the highest nucleotide identity to three Italian H3N2 strains, one isolated in 2001 and two in 2004, whereas the full length NA sequence is closely related to three H1N1 subtype viruses isolated in Italy in 2004. The remaining genes are also closely related to respective genes found in H1N1 and H3N2 SIVs currently circulating in Italy. This suggests that the novel SIV could be a reassortant between the H3N2 and H1N1 SIVs circulating in Italy.  相似文献

旨在了解河南省猪流感病毒的流行情况及其遗传进化和基因组特征。2018年4月,从河南省某一出现疑似流感症状猪群中采集鼻拭子样品150份用于分离病毒,对分离病毒的全基因组进行序列测定和分析。同时感染6周龄BALB/c小鼠,研究其对小鼠的致病性。结果显示,获得1株H1N1亚型病毒[命名为A/swine/Henan/NY20/2018(H1N1)]。遗传进化表明,其HANA基因属于欧亚类禽H1N1分支,PB2、PB1、PANPM基因属于2009甲型H1N1分支,NS基因属于经典H1N1分支。HA蛋白的裂解位点序列为PSIQSR↓GL,具有低致病性流感病毒的分子特征,在小鼠肺和鼻甲有效复制并能引起肺组织病理学变化。本研究分离到1株3源重排H1N1亚型病毒,对小鼠呈现一定致病力,提示应进一步加强对SIV的监测。  相似文献

旨在进一步了解山东省猪流感的流行情况及其病原特征,笔者于2019年春季,在山东省泰安某屠宰场采集130份猪鼻拭子,进行病毒分离鉴定;并对分离病毒进行全基因组测序和分子特征分析;用禽H9N2亚型标准抗原联合血凝抑制方法检测2018—2019年从山东省8个地区猪场采集的1 527份猪血清样品中的猪流感病毒抗体。结果显示:分离到1株H9N2亚型流感病毒,命名为A/swine/Shandong/TA009/2019(H9N2)。分离病毒与A/environment-air/Kunshan/NIOSH-BL20/2018(H9N2)和A/environment-air/Kunshan/NIOSH-BL25/2018(H9N2)遗传关系最近,其基因片段的核苷酸相似性均在99.5%以上。分离病毒的HANA基因属于Y280-like分支,PB2和M基因属于G1-like分支,PB1、PANPNS基因属于SH/F98-like分支。分离病毒HA蛋白裂解位点处的氨基酸序列为“PSRSSR/GL”,符合低致病性禽流感病毒的分子生物学特性。HA蛋白的216位为L,具有结合人源唾液酸α 2,6-Gal的能力。血清学分析结果显示,9份血清中H9N2抗体呈阳性,其总阳性率为0.59%。综上:本研究分离到1株猪源H9N2亚型流感病毒,并在猪血清中检测到H9N2抗体,提示应加强对猪流感的流行情况及其病原特征的持续监测。  相似文献

The objective of this study was to explore the epidemic situation and pathogenic characteristics of swine influenza virus (SIV) in Shandong Province. In the spring of 2019, 130 swine nasal swab samples were collected from a slaughterhouse in Tai'an city, Shandong Province for virus isolation and identification. The whole genome of isolated virus was sequenced and analyzed. Meanwhile, 1 527 swine serum samples were collected from swine farms in 8 regions of Shandong province and their anti-SIV antibody were detected by HI assay using standard avian H9N2 antigen. The results showed that a H9N2 subtype influenza virus strain was isolated and named as A/swine/Shandong/TA009/2019(H9N2). The homology analysis showed that the isolated virus had close genetic relationship with A/environment-air/Kunshan/NIOSH-BL20/2018(H9N2) and A/environment-air/Kunshan/NIOSH-BL25/2018(H9N2), and the nucleotide homology of the gene fragments were above 99.5%. Phylogenetic analysis results demonstrated that HA and NA genes of the isolated virus belong to the Y280-like lineage, PB2 and M genes belong to the G1-like lineage, and PB1, PA, NP and NS genes belong to the SH/F98-like lineage. The cleavage site in HA protein is “PSRSSR/GL”, which was in accordance with the molecular biological characteristics of low pathogenic avian influenza virus.The position 216 of HA protein is L, and it has the ability to bind human-derived sialic acid α 2,6-Gal. The results of HI showed that 9 among 1 527 serum samples were positive with a positive rate of 0.59%. The isolated virus was swine-derived H9N2 virus, and serological investigations revealed that H9N2 subtype virus infection was present in swine herds in Shandong Province. The results of this study suggest that continuous surveillance of the SIV epidemiological situation and its pathogenic characteristics should be strengthened.  相似文献

Swine influenza virus (SIV) and Mycoplasma hyopneumoniae (Mhp) are widespread in farms and are major pathogens involved in the porcine respiratory disease complex (PRDC). The aim of this experiment was to compare the pathogenicity of European avian-like swine H1N1 and European human-like reassortant swine H1N2 viruses in na?ve pigs and in pigs previously infected with Mhp. Six groups of SPF pigs were inoculated intra-tracheally with either Mhp, or H1N1, or H1N2 or Mhp+H1N1 or Mhp+H1N2, both pathogens being inoculated at 21 days intervals in these two last groups. A mock-infected group was included. Although both SIV strains induced clinical signs when singly inoculated, results indicated that the H1N2 SIV was more pathogenic than the H1N1 virus, with an earlier shedding and a greater spread in lungs. Initial infection with Mhp before SIV inoculation increased flu clinical signs and pathogenesis (hyperthermia, loss of appetite, pneumonia lesions) due to the H1N1 virus but did not modify significantly outcomes of H1N2 infection. Thus, Mhp and SIV H1N1 appeared to act synergistically, whereas Mhp and SIV H1N2 would compete, as H1N2 infection led to the elimination of Mhp in lung diaphragmatic lobes. In conclusion, SIV would be a risk factor for the severity of respiratory disorders when associated with Mhp, depending on the viral subtype involved. This experimental model of coinfection with Mhp and avian-like swine H1N1 is a relevant tool for studying the pathogenesis of SIV-associated PRDC and testing intervention strategies for the control of the disease.  相似文献

This paper reports on a serological and virological survey for swine influenza virus (SIV) in densely populated pig areas in Spain. The survey was undertaken to examine whether the H1N2 SIV subtype circulates in pigs in these areas, as in other European regions. Six hundred sow sera from 100 unvaccinated breeding herds across Northern and Eastern Spain were examined using haemagglutination inhibition (HI) tests against H1N1, H3N2 and H1N2 SIV subtypes. Additionally, 225 lung samples from pigs with respiratory problems were examined for the presence of SIV by virus isolation in embryonated chicken eggs and by a commercial membrane immunoassay. The virus isolates were further identified by HI and RT-PCR followed by partial cDNA sequencing. The HI test on sera revealed the presence of antibodies against at least one of the SIV subtypes in 83% of the herds and in 76.3% of the animals studied. Of the 600 sow sera tested, 109 (18.2%), 60 (10%) and 41 (6.8%) had SIV antibodies to subtype H1N2 alone, H3N2 alone and H1N1 alone, respectively. Twelve H3N2 viruses, 9 H1N1 viruses and 1 H1N2 virus were isolated from the lungs of pigs with respiratory problems. The analysis of a 436 nucleotide sequence of the neuraminidase gene from the H1N2 strain isolated further confirmed its identity. Demonstrably, swine influenza is still endemic in the studied swine population and a new subtype, the H1N2, may be becoming established and involved in clinical outbreaks of the disease in Spain.  相似文献

In early 2007, H2N3 influenza virus was isolated from a duck and a chicken in two separate poultry flocks in Ohio. Since the same subtype influenza virus with hemagglutinin (H) and neuraminidase (N) genes of avian lineage was also identified in a swine herd in Missouri in 2006, the objective of this study was to characterize and compare the genetic, antigenic, and biologic properties of the avian and swine isolates. Avian isolates were low pathogenic by in vivo chicken pathogenicity testing. Sequencing and phylogenetic analyses revealed that all genes of the avian isolates were comprised of avian lineages, whereas the swine isolates contained contemporary swine internal gene segments, demonstrating that the avian H2N3 viruses were not directly derived from the swine virus. Sequence comparisons for the H and N genes demonstrated that the avian isolates were similar but not identical to the swine isolates. Accordingly, the avian and swine isolates were also antigenically related as determined by hemagglutination-inhibition (HI) and virus neutralization assays, suggesting that both avian and swine isolates originated from the same group of H2N3 avian influenza viruses. Although serological surveys using the HI assay on poultry flocks and swine herds in Ohio did not reveal further spread of H2 virus from the index flocks, surveillance is important to ensure the virus is not reintroduced to domestic swine or poultry. Contemporary H2N3 avian influenza viruses appear to be easily adaptable to unnatural hosts such as poultry and swine, raising concern regarding the potential for interspecies transmission of avian viruses to humans.  相似文献

为了对供港猪群中的猪流感流行情况进行分析,从华南地区供港猪群中用无菌棉拭子采集鼻腔粘液样品,采用鸡胚接种方法,从供港猪群中分离出了2株不同亚型的猪流感病毒株,经国家流感中心鉴定分别为H1N1和H3N2亚型。本研究设计了猪流感常见亚型的HA和NA分型特异性引物,建立了猪流感型特异性RT-PCR检测方法;对分离鉴定的2株猪流感病毒和禽流感H5N1 HI检测抗原进行了RT-PCR检测,并对其部分HA和NA基因进行克隆测序分析。对供港猪群的血清检测结果表明:供港猪群中H1N1和H3N2亚型抗体阳性率分别为26.87%、38.26%,禽流感H5N1和H9N2亚型抗体阳性率均为0%。  相似文献

Three subtypes (H1N1, H1N2, and H3N2) are currently diffused worldwide in pigs. The H1N2 subtype was detected for the first time in Italian pigs in 1998. To investigate the genetic characteristics and the molecular evolution of this subtype in Italy, we conducted a phylogenetic analysis of whole genome sequences of 26 strains isolated from 1998 to 2010. Phylogenetic analysis of HA and NA genes showed differences between the older (1998-2003) and the more recent strains (2003-2010). The older isolates were closely related to the established European H1N2 lineage, whereas the more recent isolates possessed a different NA deriving from recent human H3N2 viruses. Two other reassortant H1N2 strains have been detected: A/sw/It/22530/02 has the HA gene that is closely related to H1N1 viruses; A/sw/It/58769/10 is an uncommon strain with an HA that is closely related to H1N1 and an NA similar to H3N2 SIVs. Amino acid analysis revealed interesting features: a deletion of two amino acids (146-147) in the HA gene of the recent isolates and two strains isolated in 1998; the presence of the uncommon aa change (N66S), in the PB1-F2 protein in strains isolated from 2009 to 2010, which is said to have contributed to the increased virulence. These results demonstrate the importance of pigs as mixing vessels for animal and human influenza and show the presence and establishment of reassortant strains involving human viruses in pigs in Italy. These findings also highlighted different genomic characteristics of the NA gene the recent Italian strains compared to circulating European viruses.  相似文献

The knowledge of the genome constellation in pandemic influenza A virus H1N1 2009 from different countries and different hosts is valuable for monitoring and understanding of the evolution and migration of these strains. The complete genome sequences of selected worldwide distributed influenza A viruses are publicly available and there have been few longitudinal genome studies of human, avian and swine influenza A viruses. All possible to download SIV sequences of influenza A viruses available at GISAID Platform (Global Initiative on Sharing Avian Influenza Data) were analyzed firstly through the web servers of the Influenza Virus Resource in NCBI. Phylogenetic study of circulating human pandemic H1N1 virus indicated that the new variant possesses a distinctive evolutionary trait. There is no one way the pandemic H1N1 have acquired new genes from other distinguishable viruses circulating recently in local human, pig or domestic poultry populations from various geographic regions. The extensive genetic diversity among whole segments present in pandemic H1N1 genome suggests that multiple introduction of virus have taken place during the period 1999-2009. The initial interspecies transmission could have occurred in the long-range past and after it the reassortants steps lead to three lineages: classical SIV prevalent in the North America, avian-like SIV in Europe and avian-like related SIV in Asia. This analysis contributes to the evidence that pigs are not the only hosts playing the role of "mixing vessel", as it was suggested for many years.  相似文献

本试验针对猪流感病毒(swine influenza virus,SIV)NP基因保守区域设计并合成6条引物,建立了SIV的环介导等温扩增(LAMP)检测方法,并进行了特异性、敏感性和重复性试验。结果显示,该方法可特异性检测H1N1、H1N2、H3N2、类禽H1N1亚型SIV及甲型H1N1流感病毒,但不能检测猪繁殖与呼吸综合征病毒、猪细小病毒、猪瘟病毒、猪伪狂犬病病毒、猪圆环病毒2型、日本乙型脑炎病毒;该方法的最低检测量为100拷贝/μL质粒DNA。结果表明建立的LAMP方法具有较高的敏感性和特异性,可用于SIV的快速检测。  相似文献

This study presents the results of the virological surveillance for swine influenza viruses (SIVs) in Belgium, UK, Italy, France and Spain from 2006 to 2008. Our major aims were to clarify the occurrence of the three SIV subtypes – H1N1, H3N2 and H1N2 – at regional levels, to identify novel reassortant viruses and to antigenically compare SIVs with human H1N1 and H3N2 influenza viruses. Lung tissue and/or nasal swabs from outbreaks of acute respiratory disease in pigs were investigated by virus isolation. The hemagglutinin (HA) and neuraminidase (NA) subtypes were determined using standard methods. Of the total 169 viruses, 81 were classified as ‘avian‐like’ H1N1, 36 as human‐like H3N2 and 47 as human‐like H1N2. Only five novel reassortant viruses were identified: two H1N1 viruses had a human‐like HA and three H1N2 viruses an avian‐like HA. All three SIV subtypes were detected in Belgium, Italy and Spain, while only H1N1 and H1N2 viruses were found in UK and Northwestern France. Cross‐hemagglutination inhibition (HI) tests with hyperimmune sera against selected older and recent human influenza viruses showed a strong antigenic relationship between human H1N1 and H3N2 viruses from the 1980s and H1N2 and H3N2 human‐like SIVs, confirming their common origin. However, antisera against human viruses isolated during the last decade did not react with currently circulating H1 or H3 SIVs, suggesting that especially young people may be, to some degree, susceptible to SIV infections.  相似文献

As pigs are susceptible to infection with both avian and human influenza A viruses, they have been proposed to be an intermediate host for the adaptation of avian influenza viruses to humans. In April 2006, a disease caused by highly pathogenic porcine reproductive and respiratory syndrome virus (PRRSV) occurred in several pig farms and subsequently overwhelmed almost half of China with more than 2,000,000 cases of pig infection. Here we report a case in which four swine H9N2 influenza viruses were isolated from pigs infected by highly pathogenic PRRSVs in Guangxi province in China. All the eight gene segments of the four swine H9N2 viruses are highly homologous to A/Pigeon/Nanchang/2-0461/00 (H9N2) or A/Wild Duck/Nanchang/2-0480/00 (H9N2). Phylogenetic analyses of eight genes show that the swine H9N2 influenza viruses are of avian origin and may be the descendants of A/Duck/Hong Kong/Y280/97-like viruses. Molecular analysis of the HA gene indicates that our H9N2 isolates might have high-affinity binding to the alpha2,6-NeuAcGal receptor found in human cells. In conclusion, our finding provides further evidence about the interspecies transmission of avian influenza viruses to pigs and emphasizes the importance of reinforcing swine influenza virus (SIV) surveillance, especially after the emergence of highly pathogenic PRRSVs in pigs in China.  相似文献

We have completed the genetic characterization of all eight gene segments for four low pathogenic avian influenza (LPAI) viruses. The objective of this study was to detect the presence of novel signatures that may serve as early warning indicators of the conversion of LPAI viruses to high pathogenic avian influenza (HPAI) viruses. This study included three H5N2 and one H5N3 viruses that were isolated from live poultry imported into Singapore as part of the national avian influenza virus (AIV) surveillance program. Based on the molecular criterion of the World Organisation for Animal Health (OIE), sequence analysis with the translated amino acid (aa) sequence of the hemagglutinin (HA) gene revealed the absence of multibasic aa at the HA cleavage site, identifying all four virus isolates as LPAI. Detailed phylogenetic tree analyses using the HA and neuraminidase (NA) genes clustered these isolates in the Eurasian H5 lineage, but away from the HPAI H5 subtypes. This analysis further revealed that the internal genes clustered to different avian and swine subtypes, suggesting that the four isolates may possibly share their ancestry with these different influenza subtypes. Our results suggest that the four LPAI isolates in this study contained mainly avian signatures, and the phylogenetic tree for the internal genes further suggests the potential for reassortment with other different circulating avian subtypes. This is the first comprehensive report on the genetic characterization of LPAI H5N2/3 viruses isolated in South-East Asia.  相似文献

从天津地区不同猪场分离到6株H1N1亚型猪源流感病毒(SIV)。根据GenBank发表的H1N1亚型 SIV的核蛋白(NP)、基质蛋白(M)及非结构蛋白(NS)基因序列,分别设计3对引物,将RT-PCR产物克隆至pMD18-T载体,进行测序分析。遗传进化分析结果表明:A/swine/Tianjin/TJ2/2005(H1N1)与A/swine/Tianjin/TJ4/2006(H1N1)的NP、M及NS基因核苷酸序列在遗传进化树中均与A/swine/Guangdong/33/2006(H1N1)位于同一分支上,属于古典型H1N1猪谱系;A/swine/Tianjin/TJ3/2006(H1N1)与A/swine/Tianjin/TJ8/2006(H1N1)的NP、M及NS基因核苷酸在遗传进化树中均与A/Dunedin/2/2000(H1N1)组成一个大分支,可能起源于人谱系;A/swine/Tianjin/TJ6/2009(H1N1)与A/swine/Tianjin/TJ7/2009(H1N1)NP、M及NS基因核苷酸序列在遗传进化树中均与A/swine/Jiangsu/s15/2011(H1N1)位于同一分支上,属于类禽H1N1猪谱系。本试验对6株H1N1亚型SIV的NP、M及NS全基因序列进行分析,在一定程度上揭示了天津地区H1N1亚型SIV的基因进化与流行情况。  相似文献
