Isolates of Actinobacillus (Haemophilus) pleuropheumoniae were studied by restriction endonuclease fingerprinting (REF) analysis using the enzymes BamHI and HindIII. Restriction fragments were resolved by polyacrylamide gel electrophoresis and visualized by silver staining. Except for serotypes 1 and 9, reference strains of A. pleuropneumoniae serotypes 1 to 10 had clearly distinguishable REF profiles. Analysis of REF profiles of southern Ontario field isolates revealed limited heterogeneity amongst isolates of serotype 1 or serotype 5. The REF profiles of the serotype 7 isolates studied showed greater variation. Heterogeneity could not be correlated with the presence of plasmids nor with antibiotic resistance. Limited heterogeneity could also be detected amongst REF profiles of A. pleuropneumoniae isolates recovered from a closed herd suggesting that there is a small amount of genetic variation within clonal populations. 相似文献

Some aspects of the reproductive biology of Charybdis feriatus (Linnaeus) were investigated to identify suitable techniques for broodstock management and seed production. Likewise, factors such as ablation, water depth and light requirements affecting survival or reproductive performance were tested. Production of megalops in tanks and juveniles in net cages installed in earthen ponds was conducted. Wild‐caught berried females produced a significantly higher number of zoeae per gram body weight (BW) of the female (3300±600) than captive spawners (867±58). Ablated and unablated crabs spawned after a month and ovaries of both had oocytes in all developmental stages after spawning, indicating that ablation was not necessary. Broodstock survived higher when stocked in 1 m‐deep water and kept in dark conditions compared with shallow (0.5 m depth) water or ambient lighting. There were six zoea and one megalopa stage. Megalops were produced (survival of 2–22% in 1 tonne or 23–55% in 3 L tanks) when methods for the mud crab Scylla serrata (Forsskål) were used, but feeding with Artemia started only at the Z4 stage. Survival of megalops after 1 month was higher when stocked in net cages installed in an earthen pond (32–82%) than when reared continuously in land‐based tanks (5–11%). 相似文献

OBJECTIVE: To determine the impact of successive days of endurance exercise on select serum chemistry values in conditioned Alaskan sled dogs. DESIGN: Prospective cohort study. ANIMALS: 10 conditioned Alaskan sled dogs. PROCEDURES: All dogs ran 160 km/d for 5 consecutive days. Serum was obtained prior to exercise and immediately after each exercise run; all samples were obtained before dogs were fed. Serum electrolyte, mineral, protein, total bilirubin, urea nitrogen, creatinine, and cardiac troponin-I concentrations and serum alkaline phosphatase, alanine aminotransferase, creatine kinase, and aspartate aminotransferase activities were measured. Data were analyzed by means of analysis of covariance for a randomized complete block design with dog as a blocking variable, time as a covariate, and distance run as the treatment of interest. Least square mean values were compared with values obtained prior to exercise, and linear and quadratic contrasts were examined. RESULTS: Serum globulin concentration was low prior to exercise (mean +/- SD, 2.2 +/- 0.3g/dL) and progressively decreased as exercise continued. Exercise was associated with increases in serum chloride, urea nitrogen, and cardiac troponin-I concentrations and serum alanine aminotransferase, creatine kinase, and aspartate aminotransferase activities and with progressive decreases in serum potassium, total protein, and albumin concentrations. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that multiple successive days of endurance exercise resulted in mild aberrations in serum chemistry variables in conditioned sled dogs. Changes likely reflected the metabolic stresses of prolonged endurance exercise as well as dietary composition. Hypoglobulinemia in resting, conditioned sled dogs may reflect the immunosuppressive or catabolic effects of intense endurance training. 相似文献

Banana and plantain are among the most important food crops in developing countries but production is threatened by increasing
virulent forms of Fusarium oxysporum f. sp. cubense. Chemical control is not economically effective and,therefore, breeding programs are necessary. Traditional field studies
of new genotype resistance to this disease are time-consuming and destructive. Therefore,we developed a rapid and non-destructive
procedure to differentiate field-grown banana resistant from susceptible clones. This procedure implicates application of
culture filtrates of Fusarium oxysporum f. sp. cubense race 1 onto banana leaves. The relationship between duration of the fungal in vitro incubation, and the fungal culture fresh mass, the culture filtrate absorbency, and the Gross Michel (susceptible cultivar)leaf
lesion area (after application of the culture filtrate) were similar and at 24day-incubation the highest values of the recorded
indicators were observed. A comparison between Gross Michel and FHIA-01(resistant) was also performed. The most relevant differences
between cultivars were observed at 48 hours after application of the culture filtrate, and in the middle-aged leaves. The
position of the culture filtrate application in the leaf limb (distal, middle, proximal) was not determinant. A wider comparison
among banana cultivars confirmed previous results informed by other researchers using different systems to study this plant-fungus
interaction. Such a confirmation validates the effectiveness of the procedure described here to select rapid and non-destructively
banana resistance to this disease at field level.

This study investigates the effect of forest age (20-, 50-, 160-year-old, and primeval forest) on oribatid mite communities on the bark of oak trees in the National Park Pusza Białowieża in eastern Poland. We hypothesized that oribatid mite diversity on bark peaks at forests of intermediate age and that the number of parthenogenetic species of oribatid mites is highest in young stands. In contrast to these hypotheses, the diversity, density, number of juveniles, community structure and the mode of reproduction of oribatid mite species were not significantly affected by forest age. None of the oribatid mite species occurred exclusively on trees of a specific age. The results suggest that oribatid mite communities on the bark of trees are minimally affected by tree harvesting regimes. In contrast to oribatid mites in soil, communities on bark appear to be less sensitive to disturbances. 相似文献

Although many age-related changes have been described in the nervous system of different species, few authors have specifically studied the topic. Knowledge of such changes is essential to veterinary pathologists, who must distinguish the lesions of specific pathologic processes from those arising as a result of normal aging. The brains of 20 old dogs, ranging in age from 8 to 18 years, were compared with those of 10 young dogs using routine staining techniques (hematoxilin and eosin, periodic acid-Schiff), special staining techniques (periodic acid-methenamine silver stain), and immunohistochemical techniques to detect glial fibrillary acid protein, neurofilaments, ubiquitin, and beta-amyloid. Changes affected meninges and choroid plexuses, meningeal and parenchymal vessels, neurons, and glial cells. Of special interest was the presence of polyglucosan bodies, cerebrovascular amyloid deposition, senile plaques, and ubiquitinated bodies. Some of the age-related changes found, particularly lipofuscin, polyglucosan bodies, and beta-amyloid protein deposition, may play a role in the pathogenesis of the canine cognitive dysfunction syndrome. The traditional whedo system involves the digging of large ditch-like ponds that fill seasonally with flood water, trapping fish which are subsequently harvested during the dry season. Although whedos are well described in the South of Benin, basic information necessary for their management in the North is lacking. Since 1998, the commune of Malanville has experienced a kind of ‘blue revolution’ because up to now, approximately 500 whedos (known locally as tschifi dais) have been constructed covering a total surface area of 9.3 ha, thus reflecting the high acceptance of the practice among the local population. Despite the fast development this system has received little attention from national development planners and international agencies, but in view of the disastrous state of the river Niger's fishery, sustainable aquaculture, fully relying on local species should be supported to release pressure on natural fish stocks. Instead of focussing all efforts on the introduction of modern aquaculture, it is important to enhance the existing system because of the knowledge and acceptance by the local population.
