While numerous studies have evaluated intestinal phytase activity in terrestrial monogastric species, little work has been conducted investigating the enzyme's activity in finfish. This experiment was conducted to determine if hybrid striped bass Morone saxatilis × M. chrysops possess intestinal phytase activity, and to perform a preliminary characterization of this activity in terms of pH, metal ion dependence and substrate kinetics. Hybrid bass do exhibit low levels of intestinal phytase activity. The activity has an acidic pH optimum between 3.5 and 4.5. Low concentrations of Mn2+ slightly enhance this activity. The divalent cations Mg2+ and Zn2+ were shown to either have no effect on the enzyme activity or to be inhibitory, particularly at high concentrations. The present results indicate that intestinal phytase activity in hybrid bass does follow Michaelis-Menten kinetics, with estimated Km and Vmax values of 2.5 mM phytate and 4.8 units mg−1 protein, respectively. The physiologic significance of the enzyme's presence in hybrid striped bass is questionable due to the low activity as compared to terrestrial monogastrics, the acidic pH optimum and the low affinity the enzyme appears to have for phytate. These observations also raise the question of whether the intestinal phytase activity detected in hybrid bass is merely a manifestation of non-specific acid phosphatase activity. Further investigation is needed to determine the practical significance of phytase activity in hybrid bass in terms of phytate phosphorus utilization. Additional experiments should also be conducted which evaluate intestinal phytase activity in other important aquacultured finfish species. The reported high phenolic levels in sorghum have led an interest from sorghum breeding programs in developing and identifying germplasms with high phenolic levels, which require screening a large number of samples to find those with the highest levels. Since wet chemistry screening methods are slow, expensive, and destructive, the use of NIR calibration curves could be an alternative. The objectives of this project were to determine the variation range in total phenols, condensed tannins, and 3-deoxyanthocyanidin levels in a diverse set of sorghum genotypes and to assess the predictive value of NIR curves to estimate these compounds in sorghum. A calibration curve to estimate each compound was developed and validated with an independent validation set. Calibration curve correlations for total phenols, condensed tannins, and 3-deoxyanthocyanidins were r = 0.98, 0.97, and 0.99, respectively. Correlations between NIR-predicted values and reference values in the validation set were significant for total phenols (r = 0.93), condensed tannins (r = 0.81) and 3-deoxyanthocyanidin (r = 0.82). These indicated that sufficient variation for these compounds existed within sorghum and that NIR calibration curves could be used to rapidly and non-destructively predict total phenols, condensed tannins, and 3-deoxyanthocyanidins concentrations in whole grain sorghum.  相似文献

Genetically modified (GM) cassava is currently being developed to address problems of diseases that threaten the food security of farmers in developing countries. The technologies are aimed at smallholder farmers, in hopes of reducing the vulnerability of cassava production to these diseases. In this paper we examine barriers to farmers’ voice in the development of GM cassava. We also examine the role of a translational research process to enhance farmers’ voice, to understand the sources of vulnerability farmers in a group in Kenya’s Coast face, and to determine if their concerns are consistent with those of the scientists in agriculture addressing farmers’ needs. A two-way communication participatory process provided insights into the complex vulnerability context of farmers, their primary concerns with processing and markets of cassava in order to improve livelihoods, the lack of networks with two way communication flows, and the lack of information on GM technologies. The translational research engaged farmers and scientists in an iterative process where scientists are learning what farmers need, and farmers are learning about the potential benefits and risks from GM technologies, while at the same time expressing their concerns.  相似文献

In the spring of 2013, project leaders who received funding from the John Templeton Foundation’s program “Can GM Crops Help to Feed the World?” met in England to discuss progress on funded projects and to identify common objectives and research interests. The collection of essays in this special symposium is one outcome of that meeting. This introduction provides background on the symposium’s theme of understanding the challenges to smallholder farmers having a voice. Farmer voice is important not only in debates about genetically modified crops but also for policies, technologies and other efforts designed by interests seeking ostensibly to improve the livelihoods of smallholder farmers.  相似文献

The reproduction of wild-caught Farfantepenaeus paulensis in captivity has been successfully achieved since 1983. However, year-round supply of wild broodstock is costly and unpredictable. This study compares the reproductive performance of animals of similar size (females from 32–33 g and males from 19–21 g) but from different sources: estuary-caught juveniles reared indoors for 9 mo and wild-caught adults from shallow coastal waters. Groups of 20 males and 30 unilaterally eyestalk-ablated females were maintained for 60 d in four maturation tanks under controlled environmental conditions. A higher number of eggs per spawning event (78,414 ± 34,491) and total egg production (4,469,600) were observed for the wild females. Nevertheless, captive females produced a higher number of nauplii (1,788,259) due to their higher percentage of fertilized spawns. Therefore, within the size range of this study, the reproductive performance of estuary-caught F. paulensis juveniles over-wintered indoors was found to be equivalent to that of wild-caught females from shallow coastal waters.  相似文献

Abstract— A population of 57 blue or fawn Doberman Pinschers was studied to determine the incidence of colour dilution alopecia in this breed. Forty two dogs had hair loss, typical of colour dilution alopecia. The diagnosis was confirmed in 11 dogs by skin biopsy and in 6 dogs by light microscopic examination of plucked hairs. Key diagnostic features included large macromelanosomes in the hair shafts, free clumps of melanin in the lumens of hair follicles, and numerous perifollicular melanophages. An hypothesis on the pathogenesis of colour dilution alopecia is proposed. Résumé— Une population de 57 Pinschers à robe bleue ou feu a étéétudiée pour déterminer la fréquence de l'alopécie par dilution de couleur dans cette race. 42 chiens ont présenté une perte de poils caractéristique de l'alopécie par dilution de couleur. Le diagnostic a été confirmé chez 11 chiens par biopsie cutanée et chez 6 chiens par l'examen au microscope optique des poils épilés. Les caractéristiques diagnostiques comprennent la présence de grands macromélanosomes dans les racines des poils, de manchons libres de mélanine dans la lumière des follicules pileux et de nombreux mélanophages périfolliculaires. Une hyhpothèse sur la pathogénie de l'alopécie par dilution de couleur est proposée. Zusammenfassung— Untersucht wurden 57 blaue und rehfarbene Dobermänner, um das Vorkommen der Farbmutanten-Alopezie bei dieser Rasse feststellen zu können. 42 dieser Hunde zeigten den für diese Erkrankung typischen Haarausfall. Bei 11 Patienten wurde die Diagnose durch Hautbiopsie und bei 6 durch lichtmikroskopische Untersuchung von ausgezupften Haaren gesichert. Die charakteristichen Bilder waren Makromelanosome in den Haarschäften, freies zusammengeballtes Melanin im Lumen der Haarfollikel und zahlreiche perifollikuläre Melanophagen. Eine Hypothese für die Pathogenese dieser Erkrankung wird aufgestellt. Resumen Se estudió una población de 57 perros de raza Doberman y color gris o azul para determinar la incidencia de la alopecia por dilución de color en esta raza. 42 perros presentaban pérdidas de pelo típicas de la alopecia por dilución de color. En 11 perros el diagnóstico se confirmó mediante biopsia y en 6 mediante examen microscópico del pelo. Las características diagnósticas de este proceso consistían en la presencia de macromelanosomas en las vainas de los pelos, y en la presencia de acúmulos de melanina libre en el interior de los folículos pilosos y de melanófagos perifoliculares.
