Bacterial DNA and CpG ODN have both been shown to have immunostimulatory effects in mammals, activating APCs and inducing a potent Th1 type immune response. They have also been shown to have a strong adjuvant effect and up-regulate MHC class 2 expression in murine cells, augment human and murine NK cell lytic activity, activate human B cells and induce murine B cell proliferation. However, little work has been carried out with regard to their effects on the piscine immune system. Here it is shown that various CpG ODN induce proliferation of peripheral blood leucocytes, spleen and head kidney cells from rainbow trout although, at the range of concentrations tested CpG ODN 2133 lacked the ability to induce specific antibody production to a protein antigen.  相似文献

Horses are commonly vaccinated to protect against pathogens which are responsible for diseases which are endemic within the general horse population, such as equine influenza virus (EIV) and equine herpesvirus-1 (EHV-1), and against a variety of diseases which are less common but which lead to greater morbidity and mortality, such as eastern equine encephalomyelitis virus (EEE) and tetanus. This study consisted of two trials which investigated the antigenicity of commercially available vaccines licensed in the USA to protect against EIV, EHV-1 respiratory disease, EHV-1 abortion, EEE and tetanus in horses. Trial I was conducted to compare serological responses to vaccines produced by three manufacturers against EIV, EHV-1 (respiratory disease), EEE, and tetanus given as multivalent preparations or as multiple vaccine courses. Trial II compared vaccines from two manufacturers licensed to protect against EHV-1 abortion, and measured EHV-1-specific interferon-gamma (IFN-gamma) mRNA production in addition to serological evidence of antigenicity. In Trial I significant differences were found between the antigenicity of different commercial vaccines that should be considered in product selection. It was difficult to identify vaccines that generate significant immune responses to respiratory viruses. The most dramatic differences in vaccine performance occurred in the case of the tetanus antigen. In Trial II both vaccines generated significant antibody responses and showed evidence of EHV-1-specific IFN-gamma mRNA responses. Overall there were wide variations in vaccine response, and the vaccines with the best responses were not produced by a single manufacturer. Differences in vaccine performance may have resulted from differences in antigen load and adjuvant formulation.  相似文献

Supplementation of vitamin E has indications for improving cattle health, performance, and retail characteristics when included in grain-based diets. This experiment was conducted to determine performance and carcass characteristics of steers fed diets containing wet distillers grains with solubles (WDGS) and supplemented with vitamin E. Steers of mixed Bos indicus and Bos taurus breeding (n = 199; BW = 363 ± 32 kg) were blocked by BW and assigned to 1 of 4 supplemental vitamin E (VITE) treatments [0 (control), 125, 250, and 500 IU·steer(-1)·d(-1)], which were fed for the last 97 d of the feeding period. Two blocks were on feed 129 d, and 3 blocks were fed for 150 d. Steers were fed a dry-rolled corn-based finishing diet with 35% WDGS (DM basis). Individual BW were measured initially, the initial day of vitamin E supplementation, and the day of slaughter. Carcass weights were collected at slaughter, and carcass data were collected after a 36-h chill. Body weight and ADG were not affected by VITE (P ≥ 0.34). There was a tendency for a linear (P = 0.08) increase in carcass-adjusted BW with increasing VITE. Use of carcass-adjusted final BW resulted in a linear increase (P = 0.04) in ADG with increasing VITE. Pre-vitamin E and vitamin E feeding period DMI were not affected (P ≥ 0.24) by VITE, but there was a tendency (P = 0.08) for a linear increase in overall DMI with increasing VITE. No difference (P ≥ 0.29) occurred in G:F measures using BW gains, but G:F using carcass-adjusted BW gains resulted in a trend (P = 0.11) for G:F to increase linearly with increasing VITE. Hot carcass weights tended (P = 0.08) to increase linearly with increasing dietary vitamin E. Vitamin E supplementation resulted in no effects (P ≥ 0.13) on measured carcass characteristics. Calculated yield grades (YG) were also not affected (P ≥ 0.37). However, the distribution of calculated YG resulted in a quadratic effect (P = 0.02) for YG 3 with the control and 500 VITE being greater than the 2 intermediate amounts. However, the percentage of carcasses grading YG 3 or less were not affected by vitamin E supplementation (P = 0.64). No differences were observed in the distribution of quality grades based on marbling scores (P ≥ 0.57). Data from this study suggest that vitamin E supplemented above basal requirements during the last 97 d of the feeding period in finishing diets containing 35% WDGS results in slight to no effect on animal performance or carcass characteristics.  相似文献

Objective The purpose of this study was to determine the cardiovascular effects of sevoflurane in calves. Study design Prospective experimental study. Animals Six, healthy, 8–12‐week‐old Holstein calves weighing 80 ± 4.5 (mean ± SEM) kg were studied. Methods Anesthesia was induced by face‐mask administration of 7% sevoflurane in O2. Calves tracheae were intubated, placed in right lateral recumbency, and maintained with 3.7% end‐tidal concentration sevoflurane for 30 minutes to allow catheterization of the auricular artery and placement of a Swan‐Ganz thermodilution catheter into the pulmonary artery. After instrumentation, administration of sevoflurane was temporarily discontinued until mean arterial pressure was > 100 mm Hg. Baseline values were recorded and the vaporizer output increased to administer 3.7% end‐tidal sevoflurane concentration. Ventilation was controlled to maintain normocapnia. The following were recorded at 5, 10, 15, 30 and 45 minutes after collection of baseline data and expressed as the mean value (± SEM): direct systolic, diastolic, and mean arterial blood pressures; cardiac output; mean pulmonary arterial pressure; pulmonary arterial occlusion pressure, heart rate; and pulmonary arterial temperature. Cardiac index and systemic and pulmonary vascular resistance values were calculated using standard formulae. Arterial blood gases were analyzed at baseline, and at 15 and 45 minutes. Differences from baseline values were determined using one‐way analysis of variance for repeated measures with post‐hoc differences between mean values identified using Dunnet's test (p < 0.05). Results Mean time from beginning sevoflurane administration to intubation of the trachea was 224 ± 9 seconds. The mean end‐tidal sevoflurane concentration at baseline was 0.7 (± 0.11)%. Sevoflurane anesthesia was associated with decreased arterial blood pressure at all sampling times. Mean arterial blood pressure decreased from a baseline value of 112 ± 7 mm Hg to a minimum value of 88 ± 4 mm Hg at 5 minutes. Compared with baseline, arterial pH was decreased at 15 minutes. Pulmonary arterial blood temperature was decreased at 15, 30 and 45 minutes. Arterial CO2 tension increased from a baseline value of 43 ± 3 to 54 ± 4 mm Hg (5.7 ± 0.4 to 7.2 ± 0.3 kPa) at 15 minutes. Mean pulmonary arterial pressure was increased at 30 and 45 minutes. Pulmonary arterial occlusion pressure increased from a baseline value of 18 ± 2 to 23 ± 2 mm Hg at 45 minutes. There were no significant changes in other measured variables. All calves recovered from anesthesia uneventfully. Conclusion We conclude that sevoflurane for induction and maintenance of anesthesia was effective and reliable in these calves and that neither hypotension nor decreased cardiac output was a clinical concern. Clinical relevance Use of sevoflurane for mask induction and maintenance of anesthesia in young calves is a suitable alternative to injectable and other inhalant anesthetics.  相似文献

Traditional upland livestock grazing is declining worldwide, leading to concerns about possible impacts on biodiversity. Although monitoring of protected areas often focuses on rare species, management changes also affect currently common species and vegetation structure. “Biodiversity” is a concept rather than a simple variable, and monitoring it requires indicators that are widely applicable and appropriate to context. This paper presents a novel knowledge-driven approach to developing a selection of biodiversity indicators that can then be rapidly and objectively measured within rangeland environments. Stakeholder and professional opinion on likely changes in biodiversity following reductions in sheep grazing was elicited using a workshop-based process. Potential variables suitable for use as biodiversity indicators were developed and professional opinion on their usefulness sought. A number of indicators were then tested in a natural experiment field study on the impact of reduced sheep grazing in Scotland. In the field study, red deer abundance appeared to increase where sheep grazing was reduced. It was therefore necessary to use estimates of both sheep and deer abundance as explanatory variables. In agreement with the professionals’ predictions, dwarf shrub abundance and vegetation height were greater where sheep grazing had been reduced, after taking into account differences in deer. In contrast to the professionals’ predictions, the field results showed rough grasses and dead material were less abundant where sheep had been reduced, with deer also having an impact on dead material. The professionals were unsure of the effects of reduced sheep grazing on vegetation mosaic structure; the field results suggested that reduced sheep grazing leads to a reduction in structural heterogeneity, but that deer had the greater impact in this case. Several other predictions showed nonsignificant differences in relation to reduced sheep grazing. The participatory method and some of the field methods presented are cost effective and widely applicable.  相似文献

To date, most research on freshwater cyanotoxin(s) has focused on understanding the dynamics of toxin production and decomposition, as well as evaluating the environmental conditions that trigger toxin production, all with the objective of informing management strategies and options for risk reduction. Comparatively few research studies have considered how this information can be used to understand the broader ecological role of cyanotoxin(s), and the possible applications of this knowledge to the management of toxic blooms. This paper explores the ecological, toxicological, and genetic evidence for cyanotoxin production in natural environments. The possible evolutionary advantages of toxin production are grouped into two main themes: That of “competitive advantage” or “physiological aide”. The first grouping illustrates how compounds produced by cyanobacteria may have originated from the need for a cellular defence mechanism, in response to grazing pressure and/or resource competition. The second grouping considers the contribution that secondary metabolites make to improved cellular physiology, through benefits to homeostasis, photosynthetic efficiencies, and accelerated growth rates. The discussion also includes other factors in the debate about possible evolutionary roles for toxins, such as different modes of exposures and effects on non-target (i.e., non-competitive) species. The paper demonstrates that complex and multiple factors are at play in driving evolutionary processes in aquatic environments. This information may provide a fresh perspective on managing toxic blooms, including the need to use a “systems approach” to understand how physico-chemical conditions, as well biological stressors, interact to trigger toxin production.  相似文献

Abstract

AIM: To investigate and characterise an inborn error of metabolism in a dog with skeletal and ocular abnormalities.

METHODS: A 2.5-year-old small male Miniature Poodle-like dog was presented with gross joint laxity and bilateral corneal opacities. Clinical examination was augmented by routine haematology, serum chemistry, radiographs, pathology, enzymology and molecular genetic studies. Euthanasia was requested when the dog was 3 years of age because of progressively decreasing quality of life.

RESULTS: Radiology revealed generalised epiphyseal dysplasia, malformed vertebral bodies, luxation/subluxation of appendicular and lumbosacral joints with hypoplasia of the odontoid process and hyoid apparatus. These clinical and radiographic findings, together with a positive urinary Berry spot test for mucopolysaccharides, and metachromatic granules in leucocytes, were indicative of a mucopolysaccharidosis (MPS), a lysosomal storage disease. Histological lesions included vacuolation of stromal cells of the cornea, fibroblasts, chondrocytes, macrophages and renal cells. The brain was essentially normal except for moderate secondary Wallerian-type degeneration in motor and sensory tracts of the hind brain. Dermatan sulphate-uria was present and enzymology revealed negligible activity of N-acetylgalactosamine-4-sulphatase, also known as arylsulphatase B, in cultured fibroblasts and liver tissue. A novel homozygous 22 base pair (bp) deletion in exon 1 of this enzyme's gene was identified (c.103_124del), which caused aframe-shift and subsequent premature stop codon. The “Wisdom pure breed-mixed breed” test reported the dog as a cross between a Miniature and Toy Poodle.

CONCLUSIONS: The clinicopathological features are similar to those of MPS type VI as previously described in dogs, cats and other species, and this clinical diagnosis was confirmed by enzymology and molecular genetic studies. This is an autosomal recessively inherited lysosomal storage disease.

CLINICAL RELEVANCE: The prevalence of MPS VI in Miniature or Toy Poodles in New Zealand and elsewhere is currently unknown. Due to the congenital nature of the disorder, malformed pups may be subject to euthanasia without investigation and the potential genetic problem in the breed may not be fully recognised. The establishment of a molecular genetic test now permits screening for this mutation as a basis to an informed breeding policy.  相似文献

Myophosphorylase deficiency (glycogen storage disease Type V) has been diagnosed in Charolais calves in New Zealand. It is manifested as exercise intolerance and recumbency, usually at several weeks of age, due to an inability to mobilise glucose, as glucose phosphate, from glycogen. Affected calves may rise and walk after a rest to again become affected on further exercise. There is rhabdomyolysis and after repeated or severe attacks the calf may remain recumbent and need to be euthanised. The mutant phosphorylase gene has been described. We have established a PCR-RFLP test based on a published method, and have confirmed the diagnosis in affected calves and heterozygous status of sires, dams and other related individuals belonging to an extended family. In addition, several other non-related heterozygotes have been identified in that and another herd. There appears to have been importations of the gene from both England and America.  相似文献

AIM: To describe a disease of muscle in Charolais calves and confirm the putative diagnosis of inherited myophosphorylase deficiency.

METHODS: Variously stained paraffin sections of muscle prepared from affected calves were used to describe the lesions. A polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) test was developed and applied to affected calves, their sires, dams and other individuals.

RESULTS: The lesions were those of rhabdomyolysis of skeletal muscles and sub-sarcolemmal spaces in normal fibres. The PCR-RFLP test confirmed the expected mutation for phosphorylase deficiency of Charolais cattle in two affected calves. In addition, sires, dams and other closely-related individuals of four affected calves tested as heterozygous for the mutation. Other apparently unrelated animals also tested as heterozygous.

CONCLUSIONS: The diagnosis of myophosphorylase deficiency was confirmed. The PCR-RFLP test is suitable for use in controlling this recessively-inherited disorder as it can diagnose heterozygous individuals that are otherwise clinically normal.  相似文献
