To produce high-affinity monoclonal antibodies against pesticide carbofuran, and the develop immunochemical assays for people's health and environmental protection, the hapten 4-[[(2,3-dihydro-2,2-dimethyl-7-benzofuranyloxy) carbonyl]- amino]-butanoic acid (BFNB) of carbofuran was synthesized and Balb/c mice were immunized by the hapten-carrier (BFNB-bovine serum albumin, BFNB-BSA) conjugates. The splenocytes of immunized mice were fused with Sp2/0 cells and the cultural supernatants of hybridoma cells were screened by the indirect enzyme-linked immunoabsorbent assay (ELISA), based on BFNB-ovoalbumin conjugates (BFNB-OVA). Purified monoclonal antibody (McAb) was obtained from fluids of ascites, deposited by octanoic acid and ammonium sulfate. The affinity and the specificity of McAb were characterized by ELISA or indirect competitive ELISA. A hybridoma cell line (5D3) secreting anti-carbofuran McAb had been established. The titer of culture medium and ascites was up to 1:2.048 × 10^3 and 1:1.024 × 10^6, respectively, and the subtype of the McAb was IgG1. The affinity constant of the McAb was about 2.54 × 10×9 L mol^-1, with an IC50 value of 1.18 ng mL^-1 and a detection limit of 0.01 ng mL^-1. Cross-reactivity studies showed that the McAb was quiet specific for carbofuran, as among the four analogous compounds, they were all hardly recognized (4.59 × 10^-4% for 2,3-dihydro-2,2- dimethyl-7-benzofuranol and less than 3.0 × 10^4% for others). The prepared McAb had a very high affinity and specificity, and it could be used to develop ELISA for rapid determination of carbofuran.  相似文献

Meiosis is a critical stage of gametogenesis in which alignment and synapsis of chromosomal pairs occur, allowing for the recombination of maternal and paternal genomes. Here we show that FK506 binding protein (Fkbp6) localizes to meiotic chromosome cores and regions of homologous chromosome synapsis. Targeted inactivation of Fkbp6 in mice results in aspermic males and the absence of normal pachytene spermatocytes. Moreover, we identified the deletion of Fkbp6 exon 8 as the causative mutation in spontaneously male sterile as/as mutant rats. Loss of Fkbp6 results in abnormal pairing and misalignments between homologous chromosomes, nonhomologous partner switches, and autosynapsis of X chromosome cores in meiotic spermatocytes. Fertility and meiosis are normal in Fkbp6 mutant females. Thus, Fkbp6 is a component of the synaptonemal complex essential for sex-specific fertility and for the fidelity of homologous chromosome pairing in meiosis.  相似文献

An efficient supercritical fluid extraction (SFE) process with carbon dioxide (SFE-CO(2)) was developed for the extraction of natural vitamin E (V(E)) from wheat germ. Both the pretreatment of extracted wheat germ and extraction conditions were optimized to ensure maximal V(E) yield. The extraction was undertaken at the extracting pressure of 4000-5000 psi, the extracting temperature of 40-45 degrees C, and the carbon dioxide flow rate of 2.0 mL/min for 90 min. An optimized pretreatment of wheat germ was usually necessary with a particle size of 30 mesh and a moisture content of 5.1%. A yield comparison of V(E) and its isomers extracted by supercritical CO(2) with those by conventional solvent extraction suggested that this SFE process was a practical process prospectively superior to conventional solvent extraction to prepare V(E) from wheat germ.  相似文献

以三峡库区消落带抢救植物中华蚊母的种子为材料,测量其含水量、吸胀度及种子活力,在不同温度和光照条件下,观察其萌发特性以寻找最佳萌发条件,提高成活率.结果表明:中华蚊母种子千粒质量为2.303 3 g,平均长宽分别为0.483和0.236 cm; 种子含水率为9.909 7%; 种子吸水性较好,种皮通透性好; 种子最佳浸种时间为20 h,其活力百分数大于81%; 最佳萌发条件是:在pH=11和光照强度为43.60 μmol/(s·m2)的条件下,浸种温度40 ℃、培养温度为20 ℃.  相似文献

Intake camshaft equipped with continuously adjustable cam phasing units is applied to a 486Q gasoline engine. Phasing strategies are researched through engine cycle simulation to achieve superior engine performance. The simulation results show that at high load and low and moderate speed of the engine,phasing adjustment can lead to 6% to 8% increase in power performance because phasing advance contributes to suppressing intake backflow at IVC(intake valve closed). Cam phases obtained from numerical optimization have good agreement with engine test. At moderate load,cycle simulations show that phasing advance can increase residual gas coefficient from 9% to 20%,and reduce NO emission by 80%,BSFC(brake specific fuel consumption) by 2% at a demonstration operation. With regard to fuel economy and emissions,adjustment and optimization of intake phasing at moderate load are further verified by engine test. At low load,technical approaches to enhance combustion stability,reduce HC emission and improve cold start are investigated. The effectiveness of suppressing exhaust backflow into intake duct by phasing retard is testified by simulation. Phasing changes at the operating condition are optimally obtained by engine test for improvement of combustion stability.  相似文献

Axial bearing capacity of three times cold-formed thin-walled lipped channel short column with holes was studied and calculated by finite element software ANSYS. The comparative analysis was carried out to the ANSYS, Chinese code and North America code to make out the influence of hole on the bearing capacity of the column. The results show that distortional buckling appears in the failure mode of axial compression column. Effective width method in Chinese code indirectly considers the impact of distortional buckling, and it has a good agreement with the results of North America code. Holes obviously decrease the axial bearing capacity of the column, and the variation is not according with conventional axial compression member. Opening reduction to the carrying capacity of three times cold-formed thin-walled lipped channel should be emphasised in design. Reduction factor can be taken as K=2.41×10 -5λ 2-3.78×10 -3λ+9.59×10 -2 to cross-section of the article.  相似文献

Aiming at the overhead camshaft valve mechanism,the theoretical and numerical solutions of camshaft load torque are calculated respectively by a derived formula and transient finite element analysis. The two solutions show good agreement,thus the correctness of the transient finite element model is verified. By combining with camshaft instantaneous constraint modal analysis and vibration response with consideration of gas pressure,it is found the position of camshaft which easily excites resonance and the main influence factors of camshaft vibration. Then the effect of the width of bearing seat on the camshaft vibration is investigated and the results show that the vibration of camshaft is mainly bending vibration,appropriate width of bearing seat can effectively control the camshaft vibration.  相似文献

通过利用当地的玉米秸秆为主要材料与土壤、草炭分别按不同的比例配制的人工营养土与土壤做比较,通过物理化学的方法检测所配人工营养土的营养成分、物理化学特性的好坏,并在日光温室内对番茄进行试验,研究所配制的人工营养土对番茄的生长发育、产量和各种品质的影响。研究结果表明,人工营养土栽培可以在很大程度上改善番茄植株的产量。在本试验所做的四个处理中,处理B:土:玉米秸秆=1:2+鸡粪(15kgm-3)和处理D:草炭:玉米秸秆=1:2+鸡粪(15kgm-3)的处理效果较好,考虑到经济和节省资源等因素,建议在生产中应用处理B:土:玉米秸秆=1:2+鸡粪(15kgm-3)。  相似文献

【目的】为筛选出适合北京地区无土栽培模式种植并推广的高品质鲜食番茄品种。【方法】以主栽鲜食番茄品种普罗旺斯为对照品种,对无土栽培模式下8个鲜食番茄品种植株的田间长势、抗病性进行观察评价,同时对8个品种番茄果实的果色、果形指数、单果质量、硬度、可溶性固形物、可溶性糖、可滴定酸、糖酸比、维生素C含量和番茄红素含量等外观、风味和营养品质指标进行分析。【结果】京番701、瑞粉882植株长势较旺,京番701、瑞粉882、京采6号田间较抗病毒病,京番309、京采6号果实转色较早;各品种果形指数都在0.77~0.87之间,为近圆形,属于较优果形;京番701和瑞粉882果实为大果,其余品种为中小果。京番309、瑞粉882、京采6号果实硬度较低;甜脆脆、原味1号、京番308、京番309可溶性固形物含量较高;甜脆脆、京番701、京番308可溶性糖含量较高;普罗旺斯、京采6号、瑞粉882可滴定酸含量较低;普罗旺斯、京番308和京采6号糖酸比较高;京番309、京番701、原味1号维生素C含量较高;瑞粉882、甜脆脆、原味1号番茄红素含量较高。依据各指标在鲜食番茄品种比较中的重要程度不同赋予不同的权重,进行综合...  相似文献
