BACKGROUND: Since the discovery of Agrilus planipennis Fairmaire (emerald ash borer) in 2002, researchers have tested several methods of chemical control. Soil drench or trunk injection products containing imidacloprid are commonly used to control adults. However, efficacy can be highly variable and may be due to uneven translocation of systemic insecticides. The purpose of this study was to determine whether sectored xylem anatomy might influence imidacloprid distribution in tree crowns. RESULTS: Imidacloprid equivalent concentrations were higher in leaves from branches in the plane of the injection point (0°) than in leaves from branches on the opposite side of the injection point (180°). Leaves from branches 90° to the right of injection points had higher imidacloprid equivalent concentrations than leaves from branches 90° to the left of injection points. Leaves and shoots had higher imidacloprid equivalent concentrations than roots and trunk cores, indicating that imidacloprid moves primarily through the xylem. CONCLUSION: Imidacloprid equivalent concentration in leaves varied over time and in relation to injection points. It is concluded that ash trees have sectored ‘zigzag’ xylem architecture patterns consistent with sectored flow distribution. One of the key issues in compost research is to assess when the compost has reached a mature stage. The maturity status of the compost determines the quality of the final soil amendment product. The nematode community occurring in a Controlled Microbial Composting (CMC) process was analyzed with the objective of assessing whether the species composition could be used as a bio-indicator of the compost maturity status. The results obtained here describe the major shifts in species composition that occur during the composting process. Compared to terrestrial ecosystems, nematode succession in compost differs mainly in the absence of K-strategists and numerical importance of diplogastrids. At the beginning of the composting process (thermophilic phase), immediately after the heat peak, the nematode population is primarily built by bacterial feeding enrichment opportunists (cp-1) (Rhabditidae, Panagrolaimidae, Diplogastridae) followed by the bacterial-feeding general opportunists (cp-2) (Cephalobidae) and the fungal-feeding general opportunists (Aphelenchoididae). Thereafter, during the cooling and maturation stage, the bacterial-feeding-predator opportunistic nematodes (Mononchoides sp.) became dominant. Finally, at the most mature stage, the fungal-feeding Anguinidae (mainly Ditylenchus filimus) were most present. Both, the Maturity Index (MI) and the fungivorous/bacterivorous ratio (f/b ratio), increase as the compost becomes more mature (ranging, respectively, from 1 to 1.86 and from 0 to 11.90). Based on these results, both indices are suggested as potential suitable tools to assess compost maturity.  相似文献

Alfalfa cultivar development will be enhanced by breeding strategies whichutilize the full potential of autotetraploid population genetic structures. Thisstudy evaluates the effectiveness of an allelic selection scheme, which wasdeveloped to overcome limitations of inbreeding depression and to exploitgeneral and specific combining ability effects in autotetraploid populations.Allelic selection entails the minimization of non-additive genetic effects byselecting among full-sib families (F1) which are at uniform levels ofheterozygosity. Such F1 lines are developed by crossing individuals fromtwo unrelated random mating populations. Selected F1 lines wereintercrossed to form an improved population. Eight random matingpopulations of alfalfa were developed to study the effectiveness of allelicselection. Selection for increased dry matter yield resulted in alfalfapopulations with 38 percent greater yield than the parent populations. Twocycles of intercrossing, among selected F1 lines, did not dissipate the gainfrom selection. This result has important implications for synthetic cultivardevelopment in which a major limitation is the decline in productivity withadvancing generations of seed increase. A positively correlated response toselection for dry matter yield was observed for plant height and stemdiameter. The results of this research indicate that continued testing of theallelic selection scheme is warranted and could have a significant impact onthe breeding of autotetraploid alfalfa, particularly for synthetic cultivardevelopment.  相似文献

Twenty two RAPD and 22 ISSR markers were evaluated for their potential use in determination of genetic relationships in chickpea (Cicer arietinum L.) cultivars and breeding lines. We were able to identify six chickpea cultivars/breeding lines by cultivar-specific markers. All of the cultivars tested displayed a different phenotype generated either by the RAPD or ISSR primers. Though ISSR primers generated less markers than RAPD primers, the ISSR primers produced higher levels of polymorphism (% of polymorphic markers per primer) than RAPD primers. A high level of within cultivar homogeneity was observed in chickpea. Cultivars/breeding lines originating from a common genetic background showed closer genetic relationship. Chickpea lines with similar seed type(kabuli or desi) had a tendency to cluster together. This revised version was published online in August 2006 with corrections to the Cover Date.  相似文献

The sensitivity of CERES-Maize yield predictions to uncertainty in a set of soil-related parameters and solar radiation was evaluated in Pergamino, in the Argentine Pampas. Maize yields were simulated for Pergamino using a 31 years climatic record for a range of values of a group of important model input variables. The input variables considered (and the range evaluated) were: soil nitrogen content at sowing (from 20 to 80 Kg ha−1), soil organic matter content (from 1.75% to 4%), soil water storage capacity (from 150 to 200 mm), soil water content at sowing (from 50% to 100% of total available water), soil infiltration curve number (from 76 to 82) and daily solar radiation (from −20% to 12% of the historical values). Then, a sensitivity analysis using a combination of mathematical and graphical approaches was performed to evaluate the model response to changes in the values of the input variables considered. Moreover, a simplified method based on the evaluation of the model sensitivity at extreme values of the input variables is proposed to evaluate the model non-linear responses with a reduced number of runs. Under the scenario evaluated, representative of the typical maize productions systems of the Argentine Pampas, the model results showed higher sensitivity to changes in radiation (normalized sensitivity were −0.69 and 0.45 for rainfed and irrigated conditions, respectively) than for the soil variables (normalized sensitivity ranged from 0.20 to 0.28). The CERES-Maize model was found to have similar sensitivity for the different soil inputs. Furthermore, some of the variables evaluated (soil curve number, soil water content at sowing and radiation under rainfed conditions) showed an important non-linear response.  相似文献

Background, Aim and Scope   Polycyclic aromatic hydrocarbons (PAHs) are formed as a result of incomplete combustion and are among the most frequently occurring contaminants in soils and sediments. PAHs are of great environmental concern due to their ubiquitous nature and toxicological properties. Consequently, extensive research has been conducted into the development of methods to remediate soils contaminated with PAHs. Fenton's reagent or ozone is the most commonly studied chemical oxidation methods. However, the majority of remediation studies use soils that have been artificially contaminated with either one or a limited number of PAH compounds in the laboratory. Hence, it is essential to extend such studies to soils contaminated with multiple PAHs under field conditions. Objectives   The objective of this study is to investigate the capacity of Fenton's reagent and ozone to degrade PAHs in soils. The soils have been collected from a number of different industrial sites and, therefore, will have been exposed to different PAH compounds in varying concentrations over a range of time periods. The capacity of Fenton's reagent and ozone to degrade PAHs in industrially contaminated soils is compared to results obtained in studies using soils artificially contaminated with PAHs in the laboratory. Materials and Methods: Nine soil samples, contaminated with PAHs, were collected from five different industrial sites in Sweden. For the Fenton's reagent procedure, the pH of the soil slurry samples was adjusted to pH 3 and they were kept at a constant temperature of 70oC whilst H2O2 was added. For the ozone procedure, soil samples were mixed with 50% water and 50% ethanol and kept at a constant temperature of 45 oC. Ozone was then continually introduced to each soil sample over a period of four hours. Following the Fenton's reagent and ozone oxidation procedures, the samples were filtered to isolate the solid phase, which was then extracted using pressurized liquid extraction (PLE). The sample extracts were cleaned up using open columns and then analysed by gas chromatography-mass spectrometry (GC-MS). Results: The relative abundance of the detected PAHs varied between soils, associated with different industries. For example, low molecular weight (LMW) PAHs were more abundant in soil samples collected from wood impregnation sites and high overall PAH degradation efficiencies were observed in soils originating from these sites. In the contaminated soils studied, PAHs were more effectively degraded using Fenton's reagent (PAH degradation efficiency of 40-86%) as opposed to ozone (PAH degradation efficiency of 10-70%). LMW PAHs were more efficiently degraded, using ozone as the oxidizing agent, whereas the use of Fenton's reagent resulted in a more even degradation pattern for PAHs with two through six fused aromatic rings. Discussion: The degradation efficiency for both methods was largely dependent on the initial PAH concentration in the soil sample, with higher degradation observed in highly polluted soils. LMW PAHs are more susceptible to degradation than high molecular weight (HMW) PAHs. As a result of this the relative abundance of large (often carcinogenic) PAHs increased after chemical oxidation treatment, particularly after ozone treatment. Repeated Fenton's reagent treatment did not result in any further degradation of soil PAHs, indicating that residual soil PAHs are strongly sorbed. The effectiveness of the two oxidation treatment approaches differed between industrial sites, thus highlighting the importance of further research into the influence of soil properties on the sorption capacity of PAHs. Conclusions: This study demonstrates that the degree to which chemical oxidation techniques can degrade soil bound PAHs chemical degradation is highly dependent on both the concentration of PAHs in the soils and the compounds present, i.e. the various PAH profiles. Therefore, similarities in the PAH degradation efficiencies in the nine soil samples studied were observed with the two chemical oxidation methods used. However, the degradation performance of Fenton's reagent and ozone differed between the two methods. Overall, Fenton's reagent achieved the highest total PAH degradation due to stronger oxidation conditions. LMW PAHs showed higher susceptibility to oxidation, whereas high molecular weight (HMW) PAHs appear to be strongly sorbed to the soils and therefore less chemically available for oxidation. This study highlights the importance of including soils collected from a range of contaminated sites in remediation studies. Such soil samples will contain PAH contaminants of varying concentrations, chemical and physical properties, and have been aged under field conditions. In addition to the chemical and physical properties of the soils, these factors will all influence the chemical availability of PAHs to oxidation. Recommendations and Perspectives: We recommend including aged contaminated soils in chemical degradation studies. In future chemical remediation work, we intend to investigate the potential influence of the chemical and physical properties of PAHs and soil parameters potential influence on the chemical oxidation efficiency in aged contaminated soils. Due to the vast number of contaminated sites there is a great need of efficient remediation methods throughout the world. This study shows the difficulties which may be experienced when applying remediation methods to a variation of contaminated sites.  相似文献

In the present study the structural properties of potato protease inhibitor 1 (PI-1) were studied as a function of temperature to elucidate its precipitation mechanism upon heating. A cDNA coding for PI-1 from cv. Bintje was cloned and expressed in Pichia pastoris. Using the recombinant PI-1 it was suggested that PI-1 behaves as a hexameric protein rather than as a pentamer, as previously proposed in the literature. The recombinant protein seems either to have a predominantly unordered structure or to belong to the beta-II proteins. Differential scanning calorimetry analysis of PI-1 revealed that its thermal unfolding occurs via one endothermic transition in which the hexameric PI-1 probably unfolds, having a dimer instead of a monomer as cooperative unit. The transition temperature for the recombinant PI-1 was 88 degrees C. Similar results were obtained for a partially purified pool of native PI-1 from cv. Bintje.  相似文献

The runoff of methylmercury (MeHg) from forested catchments to surface waters has been identified as a potentially significant input of MeHg to the aquatic ecosystem. Little, however, is known of the processes which control the transfer of MeHg from soils to surface waters. This study investigated the potential terrestrial sources of MeHg in runoff by sampling profiles of soil solution chemistry and determining the flux of water through those profiles into two tributaries on the Svartberget Catchment in northern Sweden. One study profile was from the podzol soil that covers most of the catchment area. The other profiles were taken in the riparian zone of each of the two tributaries. Soil solution was extracted from the soils by centrifugation. High catchment soil solution concentrations of MeHg (>1 ng l?1) occur in the surface layers of the soil, but overland flow on the catchment is rare. MeHg concentrations in the podzol profile dropped to less than 0.2 ng l?1 in the mineral soil just 5 cm below the mor layer. In the riparian soil profiles sampled in October, MeHg concentrations were higher (ca. 0.4 ng l?1), but in a July sampling the concentrations in the riparian profiles were comparable to those in the podzol (i.e. < 0.2 ng l?1). Very high concentrations of MeHg were found in the streambank sphagnum mosses (>2 ng l?1) partially submerged within the stream. The concentrations of MeHg observed under the podzol soil were insufficient to sustain the concentration of MeHg in runoff from the forested subcatchments where podzol profiles cover more than 70% of the surface area. The only sources of additional MeHg that lie along major runoff flow pathways are the riparian soils and mosses on the streambanks. It is therefore hypothesized that output of MeHg from the forest areas of the catchment is controlled by the biogeochemical processes in the riparian zone.  相似文献

A low-cost, accurate and sensitive passive measurement method for ozone has been developed and tested. The method is based on the reaction of ozone with indigo carmine which results in colourless reaction products which are detected spectrophotometrically after exposure. Coated glass filters are mounted in a short polystyrene badge-type vessel in which the diffusion path is restricted by a Teflon membrane filter. From wind tunnel experiments good concentration-independent linearity has been found. The detection limit of the method is about 23 ppbv for 1 hour exposure. Tests with interfering oxidants, such as peroxyacetyl nitrate and NO2, indicated no significant effects for common ambient conditions. No humidity effect was found when the relative humidity varied from 20 to 80%. The uptake rate of the sampler is dependent on wind velocity. In general an average air velocity of approximately 0.5 m/s is necessary to ensure a stable collection rate of ozone. This implies the necessity to measure wind speed during exposure or to place the samplers in the wind stream of a small fan. The method is suitable for hourly to daily mean measurements of ambient ozone. Under field conditions an accuracy of 11 ± 9% (40 experiments) was established in comparison to a continuous UV photometric monitor.  相似文献
