通过富集方式和干燥方法的单因素实验,最陡爬坡试验和响应面设计,优化辣木鲜叶γ-氨基丁酸(γ-aminobutyric acid,GABA)的富集工艺。建立的辣木鲜叶中GABA最适富集条件为辣木鲜叶在45℃下,真空处理34 h,然后50℃干燥。该条件处理的辣木GABA含量为(19.70±0.16)mg/g,是对照的2.63倍。  相似文献

A new cytotoxic thiodepsipeptide, verrucosamide (1), was isolated along with the known, related cyclic peptide thiocoraline, from the extract of a marine-derived actinomycete, a Verrucosispora sp., our strain CNX-026. The new peptide, which is composed of two rare seven-membered 1,4-thiazepane rings, was elucidated by a combination of spectral methods and the absolute configuration was determined by a single X-ray diffraction study. Verrucosamide (1) showed moderate cytotoxicity and selectivity in the NCI 60 cell line bioassay. The most susceptible cell lines were MDA-MB-468 breast carcinoma with an LD₅₀ of 1.26 µM, and COLO 205 colon adenocarcinoma with an LD₅₀ of 1.4 µM. Also isolated along with verrucosamide were three small 3-hydroxy(alkoxy)-quinaldic acid derivatives that appear to be products of the same biosynthetic pathway.  相似文献   

Shikonin reverses HGF-induced resistance to gefitinib in lung cancer cells HCC827

AIM: To investigate the effect of shikonin on reversing hepatocyte growth factor(HGF)-induced resistance to gefitinib in lung cancer HCC827 cells, and to explore its possible mechanisms.METHODS: The gefitinib-resistant HCC827 cells induced by HGF were treated with shikonin and gefitinibthe alone or in combination. The inhibition rates of cell viability were determined by MTT assay. The invasive ability of HCC827 cells with HGF-induced resistance to gefitinib was determined by Transwell assay. The protein levels of epithelial-mesenchymal transition (EMT) and related signaling pathway in the HCC827 cells were detected by Western blot.RESULTS: The results of MTT assay showed that the cell activity of HCC827 cells was significantly inhibited by shikonin in a dose dependent manner. The IC₅₀ of shikonin in HCC827 cells was 3.06 μmol/L. And the IC₅₀ of gefitinib in HCC827 cells was 0.51 μmol/L. Under the condition of combined treatment with shikonin and gefitinib in the presence of HGF (20 μg/L), the IC₅₀ of gefitinib was 7.36 μmol/L, significantly lower than that treated with gefitinib alone (P<0.01), so did the result of the cell migration (P<0.01). HGF induced EMT, while shikonin reversed this effect. The protein expression level of p-AKT was significantly up-regulated by HGF, while markedly down-regulated treatment with shikonin and gefitinib compared with gefitinib alone (P<0.01).CONCLUSION: Shikonin reverses HGF-induced resistance to gefitinib in lung cancer HCC827 cells, and the mechanism may be likely related to the preventon of EMT and the inhibition of HGF-induced activation of p-AKT signaling pathway.  相似文献

将鸡传染性贫血病毒M9905株VP1、VP2基因分别或同时克隆到杆状病毒转移载体pFastBacDUAL中，然后转化到DHIOBAC感受态细胞中与Bacmid杆状病毒穿梭载体进行转座重组，最后将重组子转染Sf9昆虫细胞，得到分别或同时含VP1、VP2基因的重组杆状病毒rBacVP1、rBacVP2及rBacVP1—2。PCR扩增结果证实VP1、VP2基因重组到杆状病毒基因组中；SDS-PAGE电泳分析和间接免疫荧光试验结果表明VP1、VP2基因在重组病毒中得到了表达。  相似文献

针对伪狂犬病毒gD基因的不同片段设计三对引物，分别进行PCR扩增用于猪伪狂犬病的诊断，扩增的三个片段的长度分别为262bp、217pb以及1203bp的全基因。通过比较发现，这三种PCB诊断方法均具有很高的特异性和敏感性，值扩增gD基因内部262bp的PCR诊断方法种具有突出优点，其退火与延伸合成一步，其操作可于1h内完成，敏感性更高，更适于猪伪狂犬病的快速诊断。  相似文献

In a feeding trial lasting for months Halothane-positive fattening pigs were put on a diet containing different concentrations of vitamin E and selenium. After a Myostress application vitamin E and selenium were additionally injected in order to examine stress resistance (CK-test) and regenerative capacity (histological examination of Musculus longissimus dorsi) of skeletal muscle. The study revealed that neither high dietary levels of vitamin E/selenium nor a vitamin E/selenium treatment applied after stress are capable of preventing stress related myopathy nor can they influence its healing up favourably.  相似文献   

Cryptococcal osteomyelitis in a dog

Osteomyelitis caused by Cryptococcus neoformans was diagnosed in a three-year-old male Irish wolfhound. The case details and radiological findings of radiolucency and periosteal reaction of the left third metatarsal bone had lead to an initial diagnosis of a malignant bone tumour. Biopsy revealed a fungal granuloma and culture identified the causal organism. A four month course of ketaconazole was instigated and the lameness resolved. Twelve months after the end of treatment there was no recurrence of the problem.  相似文献
