ABSTRACT:     Formaldehyde (FA)-induced denaturation of myofibrillar proteins and its prevention were investigated by means of measuring the solubility, adenosine triphosphatase (ATPase) activity, and thermal gel formability of myofibrils and surimi proteins in the presence and absence of free amino acids and glutathione, reduced form. The addition of FA decreased the solubility of myofibrils in 0.5 M NaCl at pH 7.0 and 0°C depending on its concentration and incubation time. The solubility decrease was completely inhibited by the presence of equal, twofold, and threefold amounts of cysteine (Cys), glutathione, and histidine (His) to the amount of FA, respectively. Myofibrillar Ca-ATPase was markedly activated at the initial phase and then decreased later by the addition of FA. The K-ATPase was inactivated with an increase in the amount of FA. The FA-induced changes in both ATPase activities were inhibited in the presence of Cys and His. Thermal gel formability of surimi paste increased only in a short period after the addition of a low concentration of FA. Practically, FA inhibited the thermal gelation and setting effect through the inactivation of transglutaminase. In the presence of Cys, His or glutathione, a strong elastic surimi gel was produced because FA-induced detrimental effects were inhibited.  相似文献

We used an up‐to‐date, a high accuracy CO2 manipulation system to investigate the sensitivity of organisms to CO2 acidification, rearing marine calcifiers under elevated CO2 in running water. We evaluated the effects of elevated partial pressures of carbon dioxide (pCO2) in seawater on larvae of the commercially important marine gastropod Ezo abalone Haliotis discus hannai. In larval Ezo abalone, no effect of exposure to <1100 μatm pCO2 seawater was observed in fertilization, malformation, or mortality rates until 15 h after fertilization. However, compared to control larvae in seawater (450 or 500 μatm pCO2), the fertilization rate and the hatching rate (15 h after fertilization) decreased with increased pCO2 exposure (1650 and 2150 μatm pCO2) and the malformation rate increased significantly, with the larval shell length being smaller 75 h after hatching. These results suggest that ocean acidification will potentially impact the marine population of Ezo abalone as a human food source in the future.  相似文献

Résumé— Les réponses cutanées à l'exposition solaire (radiations UV: 290–400 nm) et l'effet protecteur de deux sortes d'écrans solaires vis-à-vis des coups de soleil et du bronzage ont étéétudié chez des chiens nus descendants de chiens nus Méxicains par trois méthodes (colorimétrie, comptage des mélanocytes DOPA positifs et histologie cutanée). Après la premiére exposition aux UV, alors qu'aucune modification de couleur de la peau (érythème) n'était constatée, les modifications histologiques étaient patentes. Ensuite, après une exposition prolongée, les modifications de couleur étaient nettes (bronzage) et corrélées avec le nombre de mélanocytes DOPA-positifs. Après le troisième jour d'exposition, les mélanocytes étaient retrouvés dans tout l'épiderme des sites non protégés par les écrans solaires et aucune modification de la couleur ni des nombres de mélanocytes n'a été observé dans les zones protégées par les écrans solaires. Histologiquement, seules des modifications mineures ont été notées après 3 à 6 jours d'irradiation UV et il existait des diférences significatives selon la valeur de protection des différents écrans solaires. Par conséquent les écrans solaires protègent efficacement la peau de chiens nus des coups de soleil et du bronzage induits par les expositions solaires. [Kimura, T., Doi, K. Resumen— Se estudiaron las respuestas cutáneas a la exposición solar (radiación UV 290–400 nm) y los efectos protectores de dos tipos de pantallas solares sobre las quemaduras y bronceados solares en descendientes pelones de perros Pelones Mejicanos utilizando tres métodos distintos [colorimetría, histología y contaje de melanocitos positivos a la dihidroxifenilalanina (DOPA)]. Después de la primera exposición a la radiación UV no se detectó cambio en la coloración de la piel (eritema) aunque si se observaron alteraciones histológicas. Posteriormente, tras sucesivas exposiciones, los cambios de coloración se hicieron visibles (bronceado) y se halló correlación con el número de melanocitos DOPA-positivos. En el tercer dia de exposición, se detectaban melanocitos en toda la epidermis de las zonas desprotejidas pero no se observó ningún cambio de coloración ni aumento de melanocitos en las zonas protejidas por pantallas solares. Histológicamente sólo se detectaron ligeros cambios en la piel protejida al cabo de 3 y 6 dias de radiación UV y existian diferencias entre zonas protejidas con pantallas de diferentes valores SPF. Concluimos que las pantallas solares protegieron la piel de los perros pelones de las quemaduras y bronceado solares. [Kimura, T., Doi, K. Zusammenfassung— Es wurden die Hautreaktionen auf Sonnenbestrahlung (UV-Strahlen von 290–400 nm) und die Schutzwirkung von 2 Arten von Sonnenschutzmitteln auf Sonnenbrand und Verfärhungsreaktionen bei haarlosen Abkümmlingen von Mexikanischen Nackthunden mittels dreier Methoden untersucht (Colorimeter, Dihydroxyphenylalanin (DOPA)-positiver Melanozytenzählung und Hauthistologie). Nach der ersten Exposition gegenüber UV-Strahlung konnten histologiesche Veränderungen festgestellt werden, obwohl keine Veränderung der Hautfarbe (Erythem) sichtbar war. Bei der darauffolgenden Exposition traten Farbveränderungen auf (Verfärbungen) und korrelierten mit den Veränderungen der Zahl der festgestellten DOPA-positiven Melanozyten. Am dritten Tag der Exposition konnten Melanozyten in der Epidermis an nicht durch Sonnenschutz bedeckten Stellen festgestellt werden, während an den son-nengeschützten Stellen weder Farbveränderungen noch Melanozytenvermehrung stattfanden. Histologisch konnten auch nach 0 bis 6 Tagen Bestrahlung nur geringgradige Veränderungen in der behandelten Haut ermittell werdem. Es bestanden Unterschiede zwischen Stellen mit Sonneschutzmitteln verschiedener Lichtschutzfaktoren. Sonnenschutzmittel schützen also die Haut von haarlosen. Hurden gegen Sonnenbrand und Verfärbungereaktionen, die durch Sonnenbestrahlung ausgelöst wird. Abstract— Skin responses to solar exposure (UV radiation: 290–400 nm) and protective effects of two kinds of sunscreens on sunburn and suntan reactions were examined in hairless descendants of Mexican hairless dogs using three methods [colorimeter, dihydroxyphenylalanin (DOPA)-positive melanocyte count and skin histology]. After the first exposure to UV irradiation, while skin color change (erythema) was not detected, histologic changes were apparent. Thereafter, with subsequent exposure, color changes were apparent (tanning) and correlated with changes in numbers of DOPA-positive melanocytes detected. By the third day of exposure, melanocytes were detected throughout the epidermis in sites unprotected by sunscreens but no color change nor increase in melanocytes was noted in sunscreen-protected sites. Histologically, only minor changes were noted in treated skin after 3 and 6 days of UV irradiation, and there were differences between sites treated with sunscreens of different SPF values. Thus, sunscreens effectively protected the skin of hairless dogs from sunburn and suntan reactions induced by solar exposure.  相似文献

Three late-lactation Holstein cows were used to determine the effects of environmental temperature on performance and energy partitioning. Each cow was housed in a respiratory chamber for 30 consecutive days and exposed to three different conditions of environmental temperature: (i) 20°C and 20°C (20°C), (ii) 25°C and 20°C (25°C), (iii) 30°C and 25°C (30°C) during the day and night, respectively. The temperature was switched in an interval of 10 days. Humidity in the chamber was maintained at 55–65% through the entire experimental period. The daily mean as well as morning and evening rectal temperatures of Holstein cows increased linearly ( P  < 0.05) as chamber temperature increased. There was a significant linear reduction in dry matter (DM) intake ( P  < 0.05) and an increase in DM digestibility ( P  < 0.05). The response in milk yield, however, was not affected by heat stress. There were no significant differences among treatments for intake energy, heat production, net energy for lactation and net energy for gain. This results of this study disagreed with the assumption that late lactation cows gave priority to increasing body tissue at the expense of milk production under thermal stress.  相似文献

To examine the effect of storage temperature on the emission of methane (CH4) and carbon dioxide (CO2) and on recovery of nutrients from raw dairy manure slurry (RS) and slurry digested in a methane digester (DS), both slurries were stored in closed 100 L steel tanks under psychrophilic conditions (5, 10, 15 and 20°C) for a 150 day period. As the storage temperature increased, total methane emission increased in both types of slurry. The amount of methane emitted per unit of volatile solids of the RS and DS was 0.19 L/g and 0.10 L/g, respectively. The respective carbon dioxide emissions were 0.20 L/g for RS and 0.12 L/g for DS at 20°C of storage temperature. At temperatures greater than 15°C, the methane concentration in the emitted gas remained more than 40% of the total gas. During the experimental period, in excess of 90% of the total Kjeldahl nitrogen in these slurries was recovered (91.4–93.7% for RS and 93.7–98.4% for DS) after storage, and ammonium nitrogen was recovered in excess of 100% (100.1–143.2% for RS and 106.7–143.2% for DS storage tanks) because of the mineralization of organic nitrogen in the influent. These results indicate that manure slurry characteristics and storage temperature have significant impacts on methane emission. It can be concluded that on typical farms located in northern Japan, methane emission from manure storage tanks during late fall, winter and early spring may be negligible, because of manure temperatures less than 10°C. During late spring, summer and early fall, methane emissions can be substantially reduced by using underground storage to maintain lower manure temperatures.  相似文献

A 7-year 6-month-old, castrated male Shiba dog presented with a 1-month history of lethargy, anorexia, vomiting, and frequent watery diarrhea. Weight loss, hypoalbuminemia, anemia, and leukocytosis were detected at the first visit. The dog was diagnosed with non-responsive enteropathy (NRE) based on clinical and histopathological examinations. Since the dog did not respond to the immunosuppressive drugs, fecal microbiota transplantation (FMT) was performed during the treatment with chlorambucil. A single endoscopic FMT into the cecum and colon drastically recovered clinical signs and clinicopathological abnormalities and corrected dysbiosis in the dog. No recurrence or adverse events were observed. The present case report suggests that FMT, possibly together with chlorambucil, might be a treatment option for NRE in Shiba dogs that have poorer prognosis compared with other dog breeds.  相似文献

The Japanese sand lance (Ammodytes personatus) population in the Seto Inland Sea shows large fluctuations every few years. Its recruitment to the fishery resources is much more dependent on the survival rate of 0-age fish than is the case for other fish species because 0-age fish directly recruit to next-year spawning adults. Since the 0-age fish population depends on its larval population, which is easily influenced by natural and human impacts, high larval mortality due to predation by adults and high dispersion of larvae by wind-driven currents from spawning grounds to other locations can have very important effects on stock fluctuations. A numerical model of population dynamics of the sand lance in the eastern Seto Inland Sea was developed that focuses on wind-induced transport and adult predation of the larvae. The model successfully simulated the two-year periodic fluctuation and was consistent with actual fluctuations in fishery catches. It suggests that the various coefficients given in the model are appropriate to explain the actual population dynamics of the sand lance. The model shows that adult predation on the larvae plays a very important role in the two-year periodic fluctuation, and the maximum predation rate is estimated to be ten individuals per day. Wind-induced transport modifies the impact of this predation. Variances in the two-year periodic fluctuation become smaller with weaker westerly wind velocity. The appropriate transport rates from Bisan Strait and the Harima-nada Sea are estimated to be 0.04 d-1 and 0.01 d-1, respectively.  相似文献

On the basis of Japanese long-line fishery data during 1970–1988, anticlockwise migration routes of albacore in the North Pacific are newly proposed. The annual migration route for mature albacore is described as a closed ellipse with a centre at 20°N and 170°E, and is wider in El Niño years than non-El Niño years associated with an appearance of a cold-water region in the central and south-western North Pacific. Immature albacore also have an anticlockwise migration route in winter which extends from 25°N to 35°N and from 130°E to 180°E, when the Kuroshio has a relatively straight path. However, the migration does not persist when the Kuroshio takes a large meander path.  相似文献
