Involvement of histone H2B monoubiquitination in the regulation of mouse preimplantation development

Histone H2B monoubiquitination (H2Bub1) plays an important role in developmental regulation in various vertebrate species. However, the role of H2Bub1 in mammalian preimplantation development remains unclear. In the present study, we examined the role of H2Bub1 in the regulation of mouse preimplantation development. Based on immunocytochemical analysis using an anti-H2Bub1 antibody, no H2Bub1 signal was detected in the metaphase chromosomes of unfertilized oocytes or the pronuclei of early 1-cell stage embryos, but a weak signal was observed in late 1-cell stage embryos. The signal increased after cleavage into the 2-cell stage, and thereafter a strong signal was observed until the blastocyst stage. To assess the significance of H2Bub1 in the regulation of preimplantation development, RNF20 (an H2B-specific ubiquitin E3 ligase) was knocked down using small interfering RNA (siRNAs). In embryos treated with siRNA, the levels of Rnf20 mRNA and H2Bub1 decreased
at the 4-cell and morula stages. Although these embryos developed normally until the morula stage, only one-third developed into the blastocyst stage. These results suggested that H2Bub1 is involved in the regulation of preimplantation development. 相似文献

This study was designed to obtain information on the residual influence of dietary monensin on ruminant fermentation, methanogenesis and bacterial population. Three ruminally cannulated crossbreed heifers (14 months old, 363 ± 11 kg) were fed Italian ryegrass straw and concentrate supplemented with monensin for 21 days before sampling. Rumen fluid samples were collected for analysis of short chain fatty acid (SCFA) profiles, monensin concentration, methanogens and rumen bacterial density. Post‐feeding rumen fluid was also collected to determine in vitro gas production. Monensin was eliminated from the rumen fluid within 3 days. The composition of SCFA varied after elimination of monensin, while total production of SCFA was 1.78 times higher than on the first day. Methane production increased 7 days after monensin administration ceased, whereas hydrogen production decreased. The methanogens and rumen bacterial copy numbers were unaffected by the withdrawal of monensin. 相似文献

We evaluated effects of dietary supplementation with astaxanthin (Ax)‐rich yeast, Phaffia rhodozyma (Xanthophyllomyces dendrorhous), on broiler chicken meat quality. Fourteen‐day‐old female Ross broilers were divided into three groups: control group, Ax‐free diet; Ax 10 group, 10 mg/kg Ax diet; and Ax 20 group, 20 mg/kg Ax diet for 28 days. At 42 days old, chickens were slaughtered, and then growth performance, meat quality and sensory attributes were analyzed. Compared with the control, a* values increased significantly after slaughter and 48 h postmortem for Ax 20 samples (P < 0.05) and for b* values in Ax 20 and Ax 10 groups (P < 0.05). Cooking loss decreased in the Ax 20 group (P < 0.05). After 120 h aging, contents of several free amino acids and total free amino acid content of Ax 20 group were significantly higher than the control (P < 0.05). In sensory evaluation, meat texture attributes improved significantly in the Ax 20 group (P < 0.01). No significant changes occurred in flavor attribute scores of meat soup from the Ax 20 group compared with the control even though most assessors preferred meat soup from the Ax 20 group. It would be preferable to use a reliable crop growth model for studies on climate change impact assessment. The objectives of this study was to evaluate simulation performance for two maize models, including CERES-Maize and IXIM models, included in the DSSAT model (version 4.6) in terms of phenology and yield. Two early maturing cultivars, Chalok#1 and Junda# 6, were grown under controlled environment in plastic houses at Suwon, Korea. Each cultivar, which was sown at four different date in 2013 and 2014, was subjected to four sets of temperature conditions including ambient (AT), AT+1.5°C, AT+3°C, and AT+5°C. In simulations of phenology under given conditions, the anthesis date and grain filling ratio were underestimated, especially when temperature was unusually high, e.g., in 2013. The maize models also had poor accuracy in grain yield, which resulted from the fact that these models had relatively large errors in simulation of kernel number and kernel weight under elevated temperature conditions. In addition, both models were not able to simulate the drastic decrease of kernel number due to heat stress around flowering periods. These results indicated that two maize models would need improvements in simulation of crop response to supra-optimal temperature before they would be used to assess the impact of the climate change on maize yield. General (GSI) and specific (SSI) selection indices were constructed for three open-mating populations of sugarcane (Saccharum
spp.) seedlings and their ratoon stages. Though specific indices were found to be the most effective in improving mean brix
yield in their own source populations, but the GSI was better to SSI in other populations as indicated by higher mean values
and more number of superior clones at selection and evaluation stages. This indicated the wider applicability of the GSI over
SSI. Inspite of better expected genetic advance through GSI in ratoon crop, the actual gain in brix yield attained, through
GSI, was better in seedlings.

Understanding the mechanisms of phosphorus (P)-use efficiency (PUE) may contribute to enhancing crop P nutrition because species growth variability at low-P is well known. The experiment was carried out to evaluate the response of kale genotypes to different P supply in randomized block design in three replications. Low-P supply led to a decrease in most parameters, whereas an increase was recorded in root growth parameters. Genotypes differed in shoot dry weight (DW), leaf area, root length and area, and shoot and root P content and concentration. Root traits significantly positively correlated with PUE. Genotypes Red Russian (RR) and IJK 17 were superior in terms of shoot DW production at low P supply, and had the highest uptake efficiency. Genotypes IJK 17 and 81 had the highest P utilization efficiency, while Vates blue curled (VBC) showed the lowest PUE. Genotypes had similar shoot P content and concentration at low P supply, but large PUE differences, implying the importance of P utilization efficiency. 相似文献
