Lanthanide nanomaterials are considered a less toxic alternative to quantum dots for bioimaging applications. This study evaluated the cytotoxicity of terbium (Tb)-doped gadolinium oxide (Gd(2)O(3)) and dysprosium oxide (Dy(2)O(3)) nanoparticles exposed to human (BEAS-2B) and mouse (L929) cell lines at a concentration range of 200-2000?μg/ml for 48 h. Two assay methods were utilized-WST-8 assay (colorimetric) based on mitochondrial metabolic activity and Pico-Green assay (fluorescence), which measures total DNA content. The authors' data showed that Tb-doped Gd(2)O(3) nanoparticles were consistently more toxic than Tb-doped Dy(2)O(3) nanoparticles. However, exposure to these nanomaterials caused a decrease in proliferation rate for both cell lines rather than a net loss of viable cells after 48 h of exposure. Additionally, there was some degree of discrepancy observed with the two assay methods. For the mouse L929 cell line, the WST-8 assay yielded consistently lower proliferation rates compared to the Pico-Green assay, whereas the opposite trend was observed for the human BEAS-2B cell line. This could arise because of the differential effects of these nanoparticles on the metabolism of L929 and BEAS-2B cells, which in turn may translate to differences in their postexposure proliferation rates. Hence, the Pico-Green assay could have an advantage over the WST-8 assay because it is not skewed by the differential effects of nanomaterials on cellular metabolism.  相似文献

Asymmetric positioning of the mitotic spindle before cytokinesis can produce different-sized daughter cells that have distinct fates. Here, we found an asymmetric division in the Caenorhabditis elegans Q neuroblast lineage that began with a centered spindle but generated different-sized daughters, the smaller (anterior) of which underwent apoptosis. During this division, more myosin II accumulated anteriorly, suggesting that asymmetric contractile forces might produce different-sized daughters. Indeed, partial inactivation of anterior myosin by chromophore-assisted laser inactivation created a more symmetric division and allowed the survival and differentiation of the anterior daughter. Thus, the balance of myosin activity on the two sides of a dividing cell can govern the size and fate of the daughters.  相似文献

The effects of feeding Artemia nauplii enriched with or without poly‐β‐hydroxybutyrate (PHB) and/or highly unsaturated fatty acids (HUFA) on Siberian sturgeon (Acipenser baerii) larvae were investigated. Feeding larvae with PHB‐enriched nauplii (PHB, PHB + HUFA) decreased the growth performance of the larvae. PHB affected the body composition by increasing the lipid content of the whole body and decreasing total saturated, monoenoic, n3, n6 and decosahexanoeic acid (DHA) in the larvae, indicating that the PHB addition affected lipid metabolism. A high activity of pepsin was observed in the digestive extracts of PHB treatments (PHB, PHB + HUFA), while PHB suppressed amylase activity in the intestine of the larvae. Based on molecular analysis, PHB changed the microbial community in the distal intestine of the larvae. The highest counts of goblet cells were observed in the HUFA‐containing treatments (HUFA, PHB + HUFA), indicating that HUFA addition may improve the mucosal barrier defence system. The overall quality of the larvae was evaluated by exposing them to different salinities and ammonia stress levels. PHB decreased survival rates in these challenges. Our results show that optimal PHB doses for bio‐encapsulation into Artemia remain to be determined for further application at the earliest larval stages of sturgeon.  相似文献

Syrup formation in refrigerated doughs is a problem since it reduces the doughs’ shelf life. Microbial exogenous xylanases associated with wheat kernels were found to play a role in this syruping phenomenon. Using xylanase-producing microorganisms isolated from wheat kernels, we investigated their potency to induce syruping in dough. Growth of the fungal xylanase producer Fusarium sp. (102 colony forming units (CFU)/g dough) and the bacterial xylanase producer Paenibacillus sp. (104 CFU/g dough) in synthetic media and their respective addition to wheat dough could not bring about a significant amount of syruping. However, when these species were grown on moist wheat kernels and an extract of these kernels containing both the organisms and its xylanases was made and added to dough, intensive syruping was noted. This effect was primarily attributed to the xylanases present in the extract. These findings suggest that the involvement of xylanase-producing microorganisms in the syruping phenomenon is situated prior to harvest. Additional quantitative analyses of microbial biomass present on wheat kernels revealed that the fungi in particular could be correlated to higher microbial exogenous xylanase activities on wheat. Our results indicate that the syruping is linked to fungal xylanase production on the wheat kernels in the field.  相似文献

Background  

At depths below 10 m, reefs are dominated by blue-green light because seawater selectively absorbs the longer, 'red' wavelengths beyond 600 nm from the downwelling sunlight. Consequently, the visual pigments of many reef fish are matched to shorter wavelengths, which are transmitted better by water. Combining the typically poor long-wavelength sensitivity of fish eyes with the presumed lack of ambient red light, red light is currently considered irrelevant for reef fish. However, previous studies ignore the fact that several marine organisms, including deep sea fish, produce their own red luminescence and are capable of seeing it.  相似文献

We have sought to understand the molecular mechanisms by which dissolved organic matter (DOM) forms and soil organic matter (SOM) degrades in upland peaty gley soil under grass. Pyrolysis mass spectrometry (Py-MS) and pyrolysis gas chromatography mass spectrometry (Py-GC/MS) were applied to characterize the DOM collected from lysimeters and its parent SOM. The macromolecular organic matter in the litter and fermentation (Lf) horizon of the soil consists primarily of little decomposed lignocellulose from grass, whereas the humus (Oh) horizon is characterized by an accumulation of selectively decomposed lignocellulose material, microbial metabolites and bound fatty acids. The mineral horizon produced a relative enrichment of furan structures derived from microbial reworking of plant polysaccharides but virtually no lignin signals. A series of exceptional long chain C43 to C53 fatty acids with odd over even predominance, probably derived from mycobacteria, were also identified in the Oh horizon. Side-chain oxidation and shortening, increase of carboxyl functionality and selective removal of syringyl (S) > guaiacyl (G) > p-hydroxyphenyl (P) lignin units were the main reactions when lignin degraded. Compared with SOM, the DOM shows a large accumulation of more oxidized lignin and aromatic structures, especially those containing carboxylic and dicarboxylic acid functionalities and with shorter side-chain length. The polysaccharide-type compounds in the DOM were more modified (greater abundance of furan structures in pyrolysis products), and had significantly lower molecular weight and more diverse polymeric structures than did those in soils. Increased temperature and rainfall appeared to result in greater relative abundance of lignin degradation products and aromatic compounds in DOM.  相似文献

Seven marine cyanobacteria were isolated from two regions of the Gulf of Thailand and evaluated by the agar diffusion method for antibacterial activity. Inhibitory compound was purified from the crude methanol extract and its structure was elucidated based on extensive spectroscopic analysis, including IR, 1D and 2D NMR spectra as well as mass spectrometry. A novel antimicrobial compound produced by the marine cyanobacterium Leptolyngbya sp. LT19 was identified to be a 2‐hydroxyethyl‐11‐hydroxyhexadec‐9‐enoate which has so far never been reported in microorganisms. Biological assays revealed that this novel compound exhibited antibacterial activities against the Gram‐negative, persistent shrimp pathogens, Vibrio harveyi and V. parahaemolyticus with minimal inhibitory concentration of 250–1000 and 350–1000 μg mL?1 respectively.  相似文献

Twenty young healthy dogs weighing from 9.8 to 28.6 kg were studied by M-mode echocardiography. Parameters were measured and statistically evaluated to determine whether a correlation to body surface area existed. A statistically significant correlation to body size was found for the aortic, left atrial, left ventricular, septal, and posterior wall dimensions and the mitral valve amplitude of motion. In addition, normal values not correlated to body surface area are presented with their means and standard deviations. These values include velocity of circumferential fiber shortening, ejection time, percent systolic thickening of septum and posterior wall, percent change in minor diameter, selected dimension ratios, and mitral valve velocities  相似文献

To determine the effects of lungworm infection on energy metabolism and rate of weight gain, five 3-mo-old male Friesian calves were infected orally twice each week with 640, third-stage larvae of Dictyocaulus viviparus (D.v.) over an 8-wk period. Infected calves were matched with uninfected controls on the basis of similar rates of feed consumption and weight gain during the acclimation period before infection. Infected calves were fed 2 kg of concentrates daily (88% DM), about 8.5 Mcal/d. Controls each received approximately 250 g less, about 7.5 Mcal/d. Similar amounts of hay (5.6 to 5.8 Mcal/d) were provided to all calves. Clinical, serum chemical, hematological and parasitological criteria, weight gain and utilization of energy were monitored on a weekly basis. Serum chemical and hematological analyses and clinical examinations of infected animals revealed signs typical of lungworm infection. Fecal and sputum sample examinations for infected calves were positive for D.v. larvae and ova, respectively. Control animals gained approximately 80 g.animal-1.d-1 more than infected calves. Lungworm infection had no significant effect on digestibility of energy or protein. Metabolizability of energy ingested was somewhat higher in the infected calves due to a higher dietary concentrate to roughage ratio. Utilization of metabolizable energy and protein tended to be less efficient for infected animals. Results showed that D.v.-infected calves need more feed for gain than do uninfected animals. This extra requirement is due to an increased maintenance requirement and probably to a reduced protein retention from digested protein.  相似文献
