Hydrolysates obtained by autohydrolysis-posthydrolysis of corncobs were detoxified with charcoal, concentrated, supplemented with nutrients, and fermented with Debaryomyces hansenii. After biomass removal, the fermented media contained 0.1137 kg of nonvolatile components (NVC)/kg of liquor, which corresponded mainly to xylitol (0.6249 kg/kg of NVC) but also to minor amounts of inorganic components (measured as ashes), proteins, nonfermented sugars (xylose and arabinose), uronic acids, arabitol, and other nonvolatile components (ONVC). The media were subjected to further processing (sequential stages of adsorption, concentration, ethanol precipitation, concentration, and crystallization) to obtain food-grade xylitol. Adsorption experiments were carried out at various solid-to-liquor ratios. Under selected conditions (1 kg of charcoal/15 kg of liquors), the xylitol content increased to 0.6873 kg/kg of NVC, and almost total decoloration was achieved. The resulting liquor was concentrated by evaporation to increase its NVC content to 0.4032 kg/kg of liquor (corresponding to a xylitol concentration of 0.280 kg/kg of liquor), and ethanol was added to precipitate a part of the NVC (mainly proteins, but also uronic acids, ashes, and other nonvolatile compounds). Refined liquors (containing 0.7303 kg of xylitol/kg of NVC) were concentrated again, and ethanol was added (to reach 40-60% volume of the stream) to allow crystallization at -10 or -5 degrees C. Under selected conditions, 43.7% of xylitol contained in the initial fermentation broth was recovered in well-formed, homogeneous crystals, in which xylitol accounted for 98.9% of the total oven-dry weight. Material balances are presented for the whole processing scheme considered in this work.  相似文献

The potential of postharvest dip treatments with fludioxonil (FLU) (a synthetic analogue of the bacterial metabolite of pyrrolnitrin), in controlling postharvest decay caused by Penicillium digitatum and Penicillium italicum of citrus fruit was investigated in comparison with the conventional fungicide imazalil (IMZ). The ultrastructural changes of fruit epicuticular wax was investigated as a function of water dip temperature, and the possible role of these changes was related to residue accumulation under FLU treatment. Residues retained by fruit were determined as a function of fungicide concentration, dip temperature, and fruit storage conditions. Scanning electron microscopy analysis revealed that fruit dipping in water at 30 or 40 degrees C did not cause differences in cuticular wax's ultrastructure in comparison to control fruit, while treatments at 50, 55, or 60 degrees C caused the disappearance of wax platelets, resulting in relatively homogeneous skin surface, due to partial "melting" of epicuticular wax. Residues of FLU in fruit treated at 20 or 50 degrees C were significantly correlated with the doses of fungicide applied. When equal amounts of fungicide were employed, the residue concentrations were notably higher (from 2.6- to 4-fold) in fruit treated at 50 degrees C than in fruit treated at 20 degrees C. The dissipation rate of FLU in "Salustiana" and "Tarocco" oranges was lower in fruit subjected to treatment at 50 degrees C. The minimal FLU concentration for almost complete decay control in artificially wounded fruit during 7-d storage at 20 degrees C was 400 mg/L active ingredient (ai) in fruit treated at 20 degrees C and 100 mg/L ai in fruit treated at 50 degrees C. Results on nonwounded Tarocco oranges subjected to 3 weeks of simulated quarantine conditions at 1 degrees C, plus 6 weeks of standard storage at 8 degrees C and an additional two weeks of simulated marketing period (SMP) at 20 degrees C revealed that almost complete decay control with FLU applications of 100 mg/L at 50 degrees C and 400 mg/L at 20 degrees C resulted in ca. 0.8 mg/kg FLU fruit residues, in agreement with results on wounded citrus fruit. When equal concentrations and temperatures were applied, FLU treatments were as effective as IMZ. In vitro trials showed a low sensitivity to FLU against P. digitatum and P. italicum isolates. MIC values for the complete inhibition of mycelium growth were >or=100 microg/mL, while ED(50) values ranged from 0.1 to 1 microg/mL for P. digitatum and from 1 to >100 microg/mL for P. italicum. The latter result suggests that care should be taken to avoid exclusive application of FLU in a sustainable program for management of fruit decay. However, integrating fungicide application and hot water dip may reduce the possibility of selecting fungicide-resistant populations of the pathogen, by increasing the effectiveness of the treatment.  相似文献

The volatile oil of the bark of Cinnamomum zeylanicum was extracted by means of supercritical CO2 fluid extraction in different conditions of pressure and temperature. Its chemical composition was characterized by GC-MS analysis. Nineteen compounds, which in the supercritical extract represented >95% of the oil, were identified. (E)-Cinnamaldehyde (77.1%), (E)-beta-caryophyllene (6.0%), alpha-terpineol (4.4%), and eugenol (3.0%) were found to be the major constituents. The SFE oil of cinnamon was screened for its biological activity about the formation of melanin in vitro. The extract showed antityrosinase activity and was able to reduce the formation of insoluble flakes of melanin from tyrosine. The oil also delayed the browning effect in apple homogenate. (E)-Cinnamaldehyde and eugenol were found to be mainly responsible of this inhibition effect.  相似文献

It is likely that in the near future sequence information from sequencing programmes and EST libraries will generate an abundance of genic microsatellite markers. This study is focused on the assessment of their likely impact and performance vis-à-vis their genomic counterparts. Microsatellites from two sources were used to assess the genetic diversity in 56 old and new varieties of bread wheat on the UK Recommended List. A set of 12 microsatellite markers generated from genomic libraries and 20 expressed sequence tag (EST)-derived microsatellites were used in the study, and the performance of both marker sets assessed. The EST-derived or genic microsatellites delivered fingerprints of superior quality, amplifying clear products with few stutter bands. Diversity levels as revealed bygenic microsatellites are similar to the few published results. The PIC values for the genic markers were generally lower than those calculated for the genomic microsatellites, though advantages of both marker classes for variety identification applications are discussed. Application of urban refuse compost to agricultural soil could help to solve municipalities' problems related to the increasing production of waste only if soil property improvement and environmental conservation can be demonstrated. The use of low-pressure tractor tyres is another proposal in modern agriculture for reducing soil compaction. This study thus aimed to detect the effects of both compost and low-pressure tractor tyres on soil loss, runoff, aggregate stability, bulk density, penetrometer resistance and maize (Zea mays L.) yield. A 3-year field experiment was carried out on a hilly (15% slope) clay loam soil in central Italy. Twelve plots (200 m2 each) were monitored with tipping-pot devices for runoff and soil erosion measurement. Treatments were: compost addition (64 Mg ha−1), mineral fertilisation, use of low-pressure tyres, use of traditional tyres, with three replicates, in a fully randomised block design. Compost was applied once at the beginning of the experiment. Runoff reduction due to compost ranged between 7 and 399 m3 ha−1 during seasons, while soil erosion was reduced between 0.2 and 2.4 Mg ha−1. Mean weight diameter (MWD) of stable aggregates, measured on wheel tracks, increased by 2.19 mm, then progressively decreased. Compost significantly increased bulk density by 0.08 Mg m−3 due to its inert fraction content. This effect was less evident in the second and third year, probably due to harrowing. Maize yields were slightly, but significantly, reduced in composted plots by 1.72 Mg ha−1 in the third year. Low-pressure tyres significantly reduced soil loss in the third year by 1 Mg ha−1. Furthermore, they did not significantly influence runoff volumes and soil structural stability. Low-pressure tyres or compost addition were singly able to prevent an increase in penetrometer resistance due to agricultural machinery traffic. Low-pressure tyres increased the maize yield during the 3 years and the difference (0.4 Mg ha−1) became significant in the third year. In conclusion, results show the positive lasting effect of compost in ameliorating soil physical properties and reducing runoff and soil erosion. Low-pressure tyres appear justifiable both for the observed increase of grain production and reduction of soil compaction. This latter effect is, nevertheless, masked by compost addition which is also able to reduce penetrometer resistance. Further research is required to explain the causes of the slight inhibition of grain yield observed when compost was compared with mineral fertilisation.  相似文献

Cv. Star Ruby grapefruit (Citrus paradisi Macf.) were subjected to a 3-min dip in water at room temperature (20 degrees C) or at 50 degrees C with or without 25, 50, or 100 mg/L azoxystrobin (AZX). Then, the fruits were subjected to cold quarantine at 2 degrees C and 90-95% relative humidity (RH) for 3 weeks and then stored for 5 weeks at 8 degrees C and approximately 85% RH and for another 2 weeks at 20 degrees C and 80% RH to simulate a 2-week marketing period (SMP). No AZX residues were detected in the albedo and pulp following treatments at 20 or 50 degrees C, the total amount of residues being recovered from the flavedo tissue. There was a relationship between the AZX uptake in fruit and the amount of fungicide employed at 20 or 50 degrees C. When AZX was applied to the fruit at 25 mg/L at 20 degrees C, the residue level averaged 0.11 mg/kg (active ingredient, whole fruit basis). This residue concentration increased by 50 and 75% when the application rate increased from 25 to 50 or 100 mg/L, respectively. A similar pattern of accumulation was detected in fruit subjected to treatments at 50 degrees C. However, treatments at 50 degrees C produced residue levels higher than the treatments at 20 degrees C, with increases ranging from 63 to 84%, for the same concentration. Storage conditions did not affect the amount of AZX residues in the fruit. Treatment at 50 or 100 mg/L at 20 degrees C reduced the incidence of moderate to severe chilling injury (CI). Water dips at 50 degrees C reduced the incidence and severity of CI to a very low extent, with no additional advantages when hot water was used in combination with AZX. Treatments with 50 or 100 mg/L of AZX at 20 degrees C produced beneficial effects in decay control similar to those of 25 mg/L AZX at 50 degrees C or hot water alone. Better results were achieved with 50 or 100 AZX at 50 degrees C, providing complete control of decay during cold storage and with negligible decay after SMP. It was concluded that when AZX was applied at 50 degrees C, low doses of fungicide and minimal residue levels in fruit were required to control the postharvest decay of grapefruit. This treatment does not impair fruit quality and offers very interesting prospects for large scale application, due to the reduced potential toxicity of AZX to nontarget organisms and to the environment.  相似文献

A number of N2-alkyl and N2-acyl derivatives of guanosine 5'-phosphate (GMP) have been synthesized and tested for their synergistic effect with monosodium L-glutamate (MSG), the prototypical substance imparting umami taste to savory-based foods. Capacities to enhance the taste intensity of MSG (gamma values) were estimated through subjective comparisons of MSG/nucleotide mixtures in water with appropriate solutions of MSG alone. Assuming beta = gamma[nucleotide]/gamma[IMP], beta values of the N2-substituted GMPs were found in the range 1.2-5.7. Such values appear to be related to the chain length of the substituent in the 2-position of the purine nucleus and dependent on the replacement of a CH 2 group with an S atom and/or with an alpha-CO group. These findings indicate that the exocyclic NHR group of the guanine moiety is actively implicated in the synergism between GMP derivatives and MSG. Theoretical calculations suggest that an anti conformation is probably assumed by ribonucleotide molecules interacting with umami receptors.  相似文献

Tacle and Clara [Monreal clementine (Citrus clementina Hort. ex Tan) x Tarocco orange (Citrus sinensis L. Osbeck)] are two new triploid citrus hybrids developed by the CRA-Istituto Sperimentale per l'Agrumicoltura (Acireale, Italy). The fruits are easy-peeling and juicy, have a pleasant taste like the Tarocco orange, and are sweet like the Monreal clementine. In addition, a distinctively attractive characteristic of these mandarin-like fruits is the red-pigmented flesh caused by the presence of anthocyanins. This study reports the juice quality attributes of fresh fruits harvested at different ripening stages and of cold-stored fruits kept for 104 days at 6+/-1 degrees C and 90-95% relative humidity. Physico-chemical analyses showed that the fresh-fruit juice yield ranged between 39 (Tacle) and 41% (Clara); these values were 11-14% lower after 104 days of storage. Vitamin C content in the Clara juice was decisively higher than that in the Tacle juice. Juice anthocyanins and other polyphenols increased during cold storage. These results show that low-temperature storage enhances the functional attributes of Tacle and Clara fruit juices.  相似文献
