Two different aphid species,Myzus persicae (Sulzer) andHyalopterus pruni (Geoffroy) (Homoptera: Aphididae), were used to analyze their ability to transmit two different potyviruses,Potato virus Y (PVY) andPlum pox virus (PPV), to pepper (Capsicum annuum) andNicotiana benthamiana plants, respectively. In parallel experiments,M. persicae consistently transmitted both viruses with high efficiency, whereasH. pruni always failed to transmit either virus. This is in contrast to previous reports describingH. pruni as a vector of these potyviruses. Different aphid probing behavior among individual aphids of each species was obtained in
electrical penetration graph (EPG) experiments performed on pepper plants. This suggested thatH. pruni did not transmit these potyviruses due to behavioral differences during probing that impeded virus acquisition and/or inoculation.
It was found thatM. persicae usually makes its first probe within the first 2 min, whereasH. pruni individuals remained for more than 10 min on the plant before starting to probe. Furthermore,M. persicae individuals displayed their first intracellular puncture during the first minute of probing whereasH. pruni needed ∼ 15 min to penetrate the cell plasmalemma with their stylets. In addition, intracellular stylet punctures occurred
very frequently forM. persicae but was a rare event, never exceeding a single one, forH. pruni. The relevance of these findings for the epidemiological spread of potyviruses by different aphid species is discussed.

In a 14-week study, 1-year-old Aleppo pine seedlings were grownin two growth chambers. Seedlings were artificially hardenedby decreasing photoperiod and temperature. In each chamber halfof the seedlings were fertilized with nitrogen (8.4 mg seedling1).In order to determine the relative importance of the hardeningenvironment versus fertilization, each chamber was programmedto decrease night temperatures down to a low of 8 or 4°C.Chlorophyll fluorescence and frost hardiness was measured fivetimes during the experiment. A sample of seedlings from eachtreatment was exposed to an artificial frost at 5°Cand the freezing effects were assessed by measurements of chlorophyllfluorescence and visual evaluation of needle damage. Seedlingsincreased their frost hardiness during the experiment in allthe treatments but the ratio of variable to maximal chlorophyllfluorescence (Fv/Fm) measured before freezing did not vary duringthe experiment. This indicates that Aleppo pine maintains itsphotosynthetic ability during hardening in contrast to otherconiferous species from colder climates. The effect of nitrogenfertilization on frost hardiness was small in comparison withchamber effect. Nitrogen fertilization slightly delayed theacquisition of hardening in the coldest chamber. Seedlings inthe warmest chamber did not become fully resistant to 5°C,but in the coldest chamber, where night temperature reached4°C, all the seedlings were resistant to the frost. Severedamage caused by frost could be related to a rapid rise of minimalfluorescence (F0) but the best index of damage was the dropof Fv/Fm after freezing. 相似文献

Ochratoxin A is an important mycotoxin that can enter the human food chain in cereals, wine, coffee, spices, beer, cocoa, dried fruits, and pork meats. Coffee is one of the most common beverages and, consequently, it has a potential risk factor for human health related to ochratoxin A exposure. In this study, coffee and corresponding byproducts from seven different geographic regions were investigated for ochratoxin A natural occurrence by HPLC-FLD, nutritional characterization, and antioxidant activities by spectrophotometric assay. The research focused on composition changes in coffee during the processing step "from field to cup". Costa Rica and Indian green coffees were the most contaminated samples, with 13 and 11 microg/kg, respectively, while the Ethiopian coffee was the least contaminated, with 3.8 microg/kg of ochratoxin A. The reduction of ochratoxin A contamination during the roasting step was comparable for any samples that were considered under the recommended level of 4 microg/kg. Total dietary fibers ranged from 58.7% for Vietnam and 48.6% for Ivory Coast in green coffees and ranged from 58.6% for Costa Rica to 61.2% for India in roasted coffee. Coffee silverskin byproduct obtained from Ivory Coast was the highest, with 69.2 and 64.2% of insoluble dietary fibers, respectively. 相似文献

This study evaluated the effects of the mode of transportation on weight and metabolism of young bulls, which are generally transported by two systems: droving and truck. This experiment consisted of two phases: transportation and refeeding and involved transportation by droving and truck. Fifty young Nellore bulls were separated in equal numbers in both groups, droving group and truck group. In the transportation phase, animals were moved, in a 640-km journey. The refeeding phase consisted of 84 days and commenced after the animals arrived at the fattening farm. The effect of the transportation system on animals’ weight, metabolic hormones, and serum biochemistry was assessed after 28, 56, and 84 days. During the transportation phase, animals in the droving group lost 10.11 kg and animals in the truck group gained 13.1 kg. The truck group showed 20.5 kg of weight gain at the end of the refeeding phase. The highest triiodothyronine (T3) and thyroxine (T4) concentrations were observed on days 0 and 60 in the transportation phase in both droving and truck groups. The serum cortisol concentration was higher in the droving group than in the truck group on day 56 of the refeeding phase. The IGF-I concentrations were higher during the refeeding phase in the droving group than in the truck group. Our results showed that transportation by droving leads to increased weight loss and reduced further weight gain compared to transportation by truck and that serum concentrations of T4, cortisol, and insulin-like growth factor (IGF-I) hormones are altered by the transportation system. 相似文献

Long pentraxin 3 (PTX3) is a conserved pattern-recognition secreted protein and a host-defence-related component of the humoral innate immune system. The aim of the present study was to characterize swine PTX3 (SwPTX3) protein expression in influenza virus infected pigs. First, we performed in silico studies to evaluate the cross-reactivity of PTX3 human antibodies against SwPTX3. Secondly, we used in vitro analysis to detect SwPTX3 presence in swine bone marrow dendritic cells (SwBMDC) upon stimulation with different agents by Western blot and immunofluorescence. Finally, the levels of SwPTX3 were assessed in experimental infection of pigs with different strains of influenza virus. This is a novel study where the expression of SwPTX3 was evaluated in the context of a pathogen infection. The initial characterization of SwPTX3 in influenza virus infected pigs contributes to understand the role of PTX proteins in the immune response. 相似文献
