In the present study, we are reporting permeability and pharmacokinetics of nobiletin in rat plasma and brain, using a validated reverse phase high performance liquid chromatographic method. Protein precipitation method was used for the extraction of nobiletin and coumarin (IS) from rat plasma and brain tissue. The system was run in isocratic mode with mobile phase consisting of potassium dihydrogen ortho-phosphate (pH 4.5; 0.04 mM) and acetonitrile in ratio of 50:50, v/v. The total chromatographic run time was 9.0 min. The method was proved to be accurate and precise at linearity range of 0.05–10 μg/mL with a correlation coefficient (r) of ≥ 0.994 in rat plasma and ≥ 0.995 in rat brain. The intra- and inter-day precision and accuracy values are found to be within the assay variability limits as per the FDA guidelines. Nobiletin was found stable in the battery of stability studies viz., bench-top, auto-sampler, freeze/thaw cycles and long term storage in a freezer at − 70 ± 10 °C. Maximum concentrations of nobiletin in both plasma and brain were observed at 1 h after single oral dosing (50 mg/kg). The maximum concentration in plasma and brain were 1.78 and 4.20 μg/mL, respectively. The AUC0–t in plasma and brain were 7.49 and 20.66 μg·h/mL, respectively. The mean elimination half life (t1/2) in plasma and brain were 1.80 and 11.42 h, respectively. The Parallel Artificial Membrane Permeability Assay (PAMPA) permeability of nobiletin was found to be high at both pH 4.0 and 7.0.  相似文献

The production of energy from windmills designed specifically to operate in the low wind velocities that usually prevail in India during the main irrigation season is estimated to be reasonably economical for irrigating small farms from open wells. The economics would improve if irrigation was practiced all year round, even in the hot summer season. The calculations made here are expected to be valid for the windier 10 percent of the locations identified from available wind speed records. Other, windier locations are expected to be identified when more wind speed data are collected in the future. Governmental policies along lines similar to those for rural electrification may be needed to support research and development efforts to optimize the designs of windmills and to promote their use in rural areas.  相似文献

Vermicomposting of fly ash has been attempted, using red earthworm, Eisenia foetida. Fly ash, which was obtained from thermal power station, was mixed with cowdung in different proportions (20, 40, 60 and 80%). These mixtures were used as feed for earthworms, and after 30 days, vermicast recovery, worm zoomass and numbers of juveniles produced were recorded. A total of six runs each of 30 days were conducted during the whole study. Concentrations of heavy metals in different mixtures of fly ash–cowdung, before and after vermicomposting and in the earthworms used in the study were also estimated. Results show maximum output of vermicasts and maximum number of juveniles produced was in reactors with 40% fly ash while maximum weight gain by earthworm was in 20% fly ash vermireactors. Performance of vermireactors up to 60% fly ash was more or less similar but at 80% fly ash, there is a marked reduction in overall performance of the reactors. Chemical analysis of different samples of fly ash–cowdung mixtures prior to vermicomposting revealed high concentrations of zinc, chromium, lead, nickel and copper. Chemical analysis of vermicomposted samples showed 30–50% reduction in heavy metals up to 60% fly ash and 10–30% reduction in 80% fly ash. Metal analysis of earthworms revealed considerable bioaccumulation of heavy metals in their body. The Present study indicates the feasibility of E. foetida for mitigating the toxicity of metals and up to 60% fly ash–cowdung mixtures can be used for sustainable and efficient vermicomposting.  相似文献

Seventy-four 9-week old female C57BL/6J mice housed in a conventional facility were manipulated to induce experimental autoimmune encephalomyelitis, among which 26 developed clinical signs including lethargy, absence of defecation, and abdominal distension. By gross necropsy examination, there was distension of the cecum and colon with fecal impaction. By histologic examination, there was severe ulcerative and proliferative typhlocolitis. Fecal ELISA confirmed the presence of toxins A and B of Clostridium difficile. Alteration in immune status of the immunocompetent mice, due to stress caused by experimental manipulation or autoimmune disease, may have led to intestinal dysbiosis, followed by opportunistic infections resulting in C. difficile-associated disease. This report brings to light the occurrence of the disease in immunocompetent laboratory mice during experimental manipulations associated with alteration in immune status, and it discusses potential hazards associated with conventional housing within a hospital-associated research institute.  相似文献

The study was carried out to analyze the genetic variability for different growth parameters in poplar clones at the age of 2 and 3 yr in the nursery. Forty-nine exotic and indigenous clones of poplar were evaluated for eight morphological traits. Clones were planted in randomized block design (RBD) with three replications with four clones in a block in each treatment. Observations for different characters were recorded on six selected competitive clones per genotype. Results showed a high interclonal variability for most parameters. Statistically significant differences among clones indicated that the majority of study characters are controlled by genetic factors, specific to each clone. Highly significant genotypic difference supported by wide range of variation of mean and range values were observed for the characters under study. Significant and positive correlation was observed between diameter at breast height (DBH) and plant height. Leaf lamina length showed positive and significant correlation with petiole length, total leaf length, leaf width, and L/B ratio. Total leaf length showed positive and significant correlation with leaf width and L/B ratio. High estimates of heritability (in a broad sense) were observed for almost all characters in the study. High genetic advance expressed as percent of mean was recorded for petiole length (60.90), followed by plant height (60.78) and collar diameter (44.19) at 2 yr. At the age of 3 yr, genetic advance was found maximum for petiole length (60.05), followed by collar diameter (47.62) and plant height (45.29). The efforts for selecting new clones and their field-testing must continue under a long-term improvement plan so that the best clones can be recommended for plantations and hybridization programs.  相似文献

Johne’s disease (paratuberculosis) is an economically important disease of cattle worldwide. The disease is caused by Mycobacterium avium subsp. paratuberculosis (MAP), a fastidious gram-positive bacterium. PCR is increasingly used in diagnostic laboratories for the detection of MAP in fecal samples given the rapid test turnaround time and sensitivity and specificity comparable to fecal culture. However, efficient extraction of DNA for sensitive detection of MAP by PCR is affected by the complex lipid-rich cell wall of MAP and the presence of PCR inhibitors in feces. We evaluated a high-throughput nucleic acid extraction method (MagMAX core nucleic acid purification kit with mechanical lysis module) in conjunction with an hspX gene PCR for the detection of MAP from bovine fecal samples, which resulted in correct identification of all negative (13 of 13) and positive (35 of 35) proficiency test samples obtained from the National Veterinary Services Laboratories. In addition, all 6 negative and 50 of 51 positive diagnostic specimens tested were categorized correctly.  相似文献
