Triticale (×Triticosecale Wittmack) holds great potential as a source for biomass production for industrial applications in Central Europe and hybrid breeding in particular appears promising owing to the higher vigour of hybrids compared to lines. In this study, a set of 178 winter triticale genotypes, including 91 hybrids, their 10 male and 23 female parental lines, as well as 54 varieties were evaluated for biomass yield and other agronomically relevant traits in 2 years at five locations in Germany. We observed a large variation of dry biomass yield as well as significant genotypic variances and high heritabilities for all traits. For the hybrids, a moderate correlation was observed between biomass and plant height and between biomass and grain yield. Mid‐parent heterosis of biomass yield ranged from ?13.6 to 16.5% with an average of 4.8%, and the maximum commercial heterosis was 9.1%. Taken together, our results illustrate the potential of hybrid breeding of triticale for biomass yield to diversify our portfolio of crops for biomass production.  相似文献

Haemolysin is one type of virulence factor that assists in the pathogenesis of Escherichia coli. Currently, hemolytic activity in E. coli has been attributed to haemolysin genes found in either uropathogenic or enterohemorrhagic E. coli. Both haemolysins are classified as RTX toxins because they both have repeats in toxin domains and share similar operon organization, sequence homology, and mechanisms of action. Haemolytic avian E. coli isolates, however, lack either E. coli haemolysin gene. To investigate the avian E. coli haemolysin, a genomic library was made from an avian pathogenic E. coli. A haemolytic clone that was isolated was shown to contain homology with sheA, an E. coli K- 12 gene which causes haemolysis when present in high copy number. The cloned haemolysin gene, hlyE, lacked the conserved amino acid sequence and accessory genes common to all RTX toxins. DNA hybridizations and polymerase chain reaction amplifications showed that the nucleotide sequences homologous to hlyE were not present in a collection of three O157: H7 E. coli, five haemolytic canine uropathogenic E. coli, one haemolytic O26 E. coli, and three haemolytic avian pathogenic E. coli. Thus we have identified a new E. coli haemolysin distinct from the RTX haemolysins and have shown that some avian pathogenic E. coli possess a haemolysin with no apparent homology to hlyE or RTX haemolysins.  相似文献

Selection within families can be conducted as family deviation (FDS) or strict within‐family selection (WFS). Our objectives were to (i) investigate two breeding schemes combining selection among families with FDS or WFS and (ii) compare the optimum allocation of test resources for these breeding schemes. We focused on selection among S1 families and doubled haploid (DH) lines within S1 families and used Monte Carlo simulations to determine the selection gain (Δ?), its standard deviation (SDΔ?), and the average coefficient of coancestry among the selected DH lines (). For breeding schemes focusing only on within‐family selection, as employed in animal breeding, the maximum Δ?, its SDΔ?, and were larger for FDS than for WFS. However, for breeding schemes combining among‐ and within‐family selection, as employed commonly in plant breeding programmes, the maximum Δ?, SDΔ?, and were almost equal for FDS and WFS. Furthermore, the optimum allocation of test resources was similar for FDS and WFS. We conclude that FDS and WFS are equally suited for short‐ and long‐term success in breeding schemes where among‐family selection is followed by within‐family selection.  相似文献

The phase 1 (fliC) and phase 2 (fljB) Salmonella flagella genes were analyzed by restriction fragment length polymorphism (RFLP)-polymerase chain reaction (PCR) to aid in the identification of different Salmonella serotypes. Twenty-four phase 1 flagellin and eight phase 2 flagellin genes could be differentiated among each other with restriction endonucleases Sau3A and HhaI in RFLP-PCR analysis. These flagellin genes comprise the major antigenic formulas for 52 serotypes of Salmonella sp., which include the common serotypes found in poultry and other important food animal species. With the knowledge of the O antigen composition determined from conventional O serotyping, 90% of the Salmonella serotypes could be identified by this double restriction enzyme RFLP analysis of fliC and fljB genes. This RFLP-PCR flagellar typing scheme was successfully applied to the identification of serotype for 112 Salmonella isolates obtained from poultry environment. There was a significant correlation between RFLP-PCR and conventional serotyping (chi-square, P < 0.001). Overall, PCR-RFLP proved to be a fast, accurate, and economical alternative approach to serotyping Salmonella sp.  相似文献

Staphylococcus aureus is an important opportunist that can cause superficial to life-threatening illnesses in a variety of animal species. In poultry, this organism has been implicated in osteomyelitis, synovitis, and cellulitis. Whereas most infections can be treated with antibiotics, because of the organism's propensity to acquire antimicrobial resistance, it is important to continually monitor antibiotic susceptibilities of clinical isolates. We surveyed 77 clinical poultry S. aureus isolates, collected from 1998 to 2000, for susceptibilities to a panel of 18 antimicrobial agents. Thirty-six percent of isolates were susceptible to all antibiotics. Forty-three and 16% of avian S. aureus were resistant to one and two antibiotics respectively. Staphylococcus aureus isolates were commonly resistant to tetracycline (40%; minimal inhibitory concentration [MIC]90 > 32 microg/ml), lincomycin (19%; MIC90 > 32 microg/ml), erythromycin (12%; MIC90 > 8 microg/ml), and kanamycin (8%; MIC90 < 128 microg/ml). All S. aureus isolates were susceptible to chloramphenicol, gentamicin, streptomycin, nitrofurantion, linezolid, quinupristin/dalfopristin, vancomycin, and the production antimicrobials virginiamycin, salinomycin, and flavomycin. A periodic assessment of antimicrobial susceptibilities of important avian pathogens like S. aureus will be important in helping the clinician's choice of antibiotic to control infection.  相似文献

The disposition and effect of benazepril and its active metabolite, benazeprilat, were evaluated in cats using a pharmacokinetic/pharmacodynamic model. Cats received single 1 mg/kg doses of intravenous 14C-benazeprilat and oral 14C-benazepril.HCl, and single and repeat (eight daily) oral administrations of 0.25, 0.5 and 1.0 mg/kg nonlabelled benazepril.HCl. The pharmacokinetic endpoints were plasma concentrations of benazepril and benazeprilat, and recovery of radioactivity in faeces and urine. The pharmacodynamic endpoint was plasma angiotensin-converting enzyme (ACE) activity. Benazeprilat data were fitted to an equation corresponding to a single-compartment model with a volume equal to the blood space (Vc = 0.093 L/kg). Within this space, benazeprilat was bound nonlinearly to ACE, which was mainly tissular (89.4%) rather than circulating (10.6%). Free benazeprilat was eliminated quickly from the central compartment (t1/2 approximately 1.0 h; Cl approximately 0.125 L/kg/h), elimination being principally biliary ( approximately 85%) rather than urinary ( approximately 15%). Nevertheless, inhibition of ACE was long-lasting (t1/2 16-23 h) due to high affinity binding of benazeprilat to ACE (Kd approximately 3.5 mmol/L, IC50 approximately 4.3 mmol/L). Simulations using the model predict a lack of proportionality between dose of benazepril, plasma benazeprilat concentrations and effect due to the nonlinear binding of benazeprilat to ACE. For example, increasing the dose of benazepril (e.g. above 0.125 mg/kg q24 h) produced only small incremental inhibition of ACE (either peak effect or duration of action).  相似文献

Testes of 36 normal New Zealand white rabbits (8, 15, 18, 26, and greater than 52 weeks of age) were examined by light and electron microscopy. The incidence and number of affected tubules were determined for spermatid giant cells, focal tubular hypospermatogenesis, cytoplasmic swelling of spermatogonia, intracytoplasmic vacuoles in seminiferous epithelium, and tubular dilatation. Spermatogenesis commenced at 15-18 weeks of age and was present in all rabbits by 18 weeks. Spermatid giant cells occurred in 96% of rabbits 15 weeks of age and older. Focal hypospermatogenesis was present in 14-57% of testes once active spermatogenesis began. Ninety-seven percent of testes in all age groups combined had spermatogonial swelling. Infrequent dilated seminiferous tubules were present in five rabbits. Ultrastructurally, spermatid giant cells were round cells with multiple nuclei that appeared to arise by widening of narrow intercellular bridges that normally connect spermatogenic epithelial cells. Pale-staining spermatogonia consisted of cytoplasmic and nuclear swelling with disruption of plasma and nuclear membranes. Tubules with spermatogonial swelling were more numerous in 15- and 18-week-old rabbits. There were no significant differences in incidence or extent of spermatid giant cells, focal hypospermatogenesis, cytoplasmic vacuoles, or tubular dilatation between age groups after spermatogenesis commenced. Although the cause of these changes is not known, they were frequent findings in normal rabbits 15 weeks of age and older.  相似文献

Computer-automated sperm-head morphometry was used in this study to determine the effects of cryopreservation on red deer sperm-head morphometry. Epididymal sperm samples were collected from 40 mature stags and were divided. One portion was diluted at room temperature in a Tris-citrate egg yolk medium, containing 6% glycerol. A microscope slide was prepared from single extended sperm samples prior to freezing. The remainder of each sample was frozen in nitrogen vapours. After thawing, sperm smears were prepared as described above. All slides were air dried and stained with Hemacolor. The sperm-head dimensions for length, width, area, perimeter and shape factor (length/width), for a minimum of 135 spermatozoa were determined for each slide by means of the Sperm-Class Analyser (SCA). Firstly, our results show that cryopreservation substantially reduced (p < 0.001) sperm motility and plasma membrane and acrosome integrities. In addition, sperm heads were significantly smaller in cryopreserved spermatozoa than in the companion extended samples for area (32.05 microm2 vs 32.56 microm2; p < 0.05), length (8.46 microm vs 8.53 microm; p < 0.0001) and shape factor (1.833 vs 1.849; p < 0.0001) for all stags. These differences were found within 29 of 40 stags (75%) for at least three of the morphometric parameters. The individual variability (CV) of sperm head measurements from extended samples was negatively correlated (p < 0.005) with the per cent of change in sperm head measurements after cryopreservation for area (r = -0.465), width (r = -0.483) and perimeter (r = -0.375). Thus, the lower the sperm head variability in the extended samples, the greater the sperm change as a consequence of the cryopreservation. These results suggest that the variability (heterogeneity) in sperm head dimensions of individual stags may be a good indicator of sperm freezability.  相似文献
