Amplified fragment length polymorphism (AFLP) analysis has been used to measure genetic diversity in perennial ryegrass (Lolium perenne L.) and to relate intra- and interpopulation variation to breeding history. Cluster analysis of AFLP data from contrasting
populations showed features consistent with the origins of these varieties. Significant differences in intrapopulation diversity
were detected and partial separation of different cultivars was observed. Restricted base cultivars, derived from small numbers
of foundation clones, were suitable for this type of study, allowing near complete discrimination of closely related cultivars.
Analysis of bulked samples was based on the pooling of genomic DNA from 20 individuals from 6 selected populations. Cluster
analysis of AFLP data from bulked samples produced a phenogram showing relationships consistent with the results of individual
analysis. AFLP profiling provides an important tool for the detection and quantification of genetic variation in perennial
ryegrass.

Summary Pathogenicity patterns of 42Mycosphaerella graminicola (Septoria tritici) isolates secured from the major wheat growing regions in Israel, were assessed on seedlings of 16 bread and durum wheats. The spring bread wheat cultivar Titan (CI12615), the winter bread wheats Bezostaya 1, Kavkaz, NE7060 and the durum wheats Nursit 163 and Zenati Bouteille, all exhibited a high level of resistance. Significant cultivar × isolate interactions were recorded for isolates from the different regions in relation to the response of the 16 cultivars organized in 6 reponse classes to the test isolate ISR8036. Number of genes for resistance in the 16 cultivars was estimated, based on the assumption of a gene-for-gene relationship. Twelve complementary genes were hypothesized in the 42M. graminicola isolates × 16 wheat cultivars matrix. There was considerable variation in virulence frequencies between regions and between locations within the same region. The overall frequency of Bezostaya-Kavkaz virulence was low throughout the country (5–7%). The frequency of designated hypothesized virulence genes VST OLAF, VST COLOTANA, VST IAS 20 in Israel, was high in most locations. The use of doubled haploids improves the efficiency of cultivar development in many crops and can be helpful in genetic and molecular studies. The major problem with this approach is the low efficiency of green plant regeneration. We describe here an efficient method for inducing embryos and regenerating green plants directly from isolated microspores of durum wheat cv. ‘Jennah Khetifa’. Tillers from donor plants were pretreated in 0.3 m mannitol and were stored at 4°C at various times: 3, 5, 6, 7, 8, 10 and 12 days. Our results showed clearly that the novel pretreatment combined mannitol 0.3 m and cold for 7 days had a strong effect on the number of embryos produced and regenerated green plants. Under this condition 13 475 mature embryos were produced from 2 693 500 microspores. Moreover, 85 green plants were obtained. High green plants regeneration frequency was recorded. As an average 11.55 green plants were produced per 100 000 microspores (about the equivalent of six plants per spike). Therefore, this study showed clearly that our results are the best ones published until now in durum wheat. 相似文献

The Pi‐ta gene in rice confers resistance to strains of the blast pathogen Magnaporthe grisea (Herbert) Borr. (anamorph Pyricularia oryza Cav.) containing the corresponding avirulence gene AVR‐Pita in a gene‐for‐gene fashion. The Pi‐ta gene is a typical nucleotide‐binding site type resistance gene. Nucleotide sequences distinguishing the resistant Pi‐ta and susceptible pi‐ta alleles were previously identified and used for developing DNA markers for a resistant Pi‐ta haplotype and three susceptible pi‐ta haplotypes. In the present study, the existence of the Pi‐ta gene in 141 rice germplasm accessions was rapidly determined using these markers, and the results were confirmed by inoculating rice germplasm with an M. grisea strain containing AVR‐Pita. The Pi‐ta gene was found in accessions from several major rice producing countries, including China, Colombia, Japan, Vietnam, the Philippines, Iran and the United States. The usefulness of DNA markers for rapid determination of the genotype of rice germplasm was thus demonstrated. The Pi‐ta gene also was found in rice cultivar known to contain the Pi‐ta2 gene, although the allelic relationship of these genes remains to be determined. The presence of the Pi‐ta gene in landrace cultivars in several different geographical locations, the Philippines and Vietnam, other indica rice cultivars in China and Colombia suggest that the Pi‐ta gene may have spontaneously originated in indica rice cultivars. These results are useful for incorporating the Pi‐ta gene into advanced breeding lines by marker‐assisted selection for rice breeding programmes worldwide. 相似文献

Cultivated Chinese wheat germplasm has been a valuable genetic resource in international plant breeding. Patterns of gliadin among cultivated Chinese accessions are unknown, despite the proven value and potential novelty. The objective of this work was to analyse the diversity within improved Chinese wheat germplasm. The electrophoretic banding patterns of gliadin in winter wheat cultivars and advanced lines were determined by acid-polyacrylamide gel electrophoresis. For 148 leading commercial cultivars and promising advanced lines used in this study, 48 patterns were identified, 29 corresponding to ω -gliadin, nine to γ -gliadin, five to β -gliadin and five to α -gliadin. The most frequent patterns were A6 in ω ; B in γ ; B in β and A in the region of α . A total of 116 band types appeared in the 148 samples: 94 accessions had unique gliadin types, and 22 gliadin, types while not unique, were found in 54 accessions. The gliadin patterns of Chinese wheat cultivars and lines greatly differed from the patterns of wheat lines from other countries. Three patterns, E, J, H, M, N and O in the ω -zone had not been reported previously. Three wheat zones of China, the Northern Winter Wheat Region, the Yellow and Huai Valley River valleys Winter Wheat Region and the Southwestern Winter Wheat Region, showed different frequencies in their gliadin patterns. Summary Narrow-leafed lupin or lupine (Lupinus angustifolius L.) is one of the three species of the genus Lupinus that grows naturally in Galicia (northwestern Spain). In this region, with more than one million of cattle heads, almost
20% of Spanish total, there is no cultivation of any protein legume for feed. Lupins are widely used as a source of protein
and energy in livestock feed and would contribute to the supply of vegetable protein at low cost. A 2-year study of natural
narrow-leafed lupin germplasm was conducted under winter Mediterranean conditions in northwestern Spain. Fifty-nine wild populations
and two cultivars developed in Poland were evaluated for major morphological, phenological, and agronomical characteristics.
The objectives were to assess the agronomic potential of this germplasm for the future production and breeding, and to know
the relationships among populations studied. Lupin populations showed significant differences for most of the traits studied.
Genotype × environment interaction was significant for plant height, green and dry mass, pod length, seeds per pod, seed length
and width, and water permeability of seed coat, indicating that some genotypes were better fitted to a specific environment
than others.
Three narrow-leafed lupin populations (LUP-0098, LUP-0138 and LUP-0200) were identified with suitable agronomical characteristics
for their possible future cultivation in the region and breeding purposes. The accession LUP-0138 presented the best behaviour
for seed production. These results open the way for the winter narrow-leafed lupin to become a promising crop in the future

Italian and Tunisian durum wheats from different eras of breeding were assessed for the presence of a gene for brittle rachis. Nine of 15 Italian durum landraces had brittle rachides. Strampellis achievement was the release of the well-known variety, Senatore Cappelli, which was derived from a Tunisian landrace, Jenah Rhettifah, which has a brittle rachis. Rachides of two Tunisian landraces were also brittle. Since the 1950s, 16 accessions were released as selections from crosses or mutagenesis involving Senatore Cappelli. Seven of these accessions have brittle rachides. F2 segregation in intercrosses of Senatore Cappelli with other durum accessions with brittle rachides indicated a common allele for brittle rachis. Segregation of the F2 from Senatore Cappelli/ANW 10A and the F2 of Senatore Cappelli/ANW 10B showed that the gene for brittle rachis of Senatore Cappelli was allelic to the brittle allele of the Br-B1 locus on chromosome 3B. Senatore Cappelli was presumably the only source of brittle rachis used in Italian breeding programmes. The genes for brittle rachis have been retained in the gene pool of durum wheat, suggesting that the brittle rachis character is not associated with an appreciable yield loss in modern farming systems in Mediterranean environments. 相似文献

The most common reasons, which cause the deterioration of structures in China, are reinforcement corrosion and freeze thaw cycles of concrete. Based on the summarizing of research works of the effect of reinforced concrete deterioration on structures, it is indicated that the structural durability research should be carried out either in view of material science or that of structural science, and should be also considered the interaction of several factors with uncertain nature instead of the determined individual factor analysis. The structural durability of serviceable should be paid much more attention while researching the durability of structural capacity.
