24 calves of the breed "Deutsche Schwarzbunte Rasse" were used for qualitative and quantitative investigations on the metabolism of fatty acids, particularly that of linoleic acid. The trial comprised 6 calves in 4 groups. The experimental rations consisted of powdered skim milk, oat meal, milk sugar, coco fat and varying proportions of linoleic acid (1.0%, 1.5%, 2.5%). The control group received the basal ration without supplements of linoleic acid. After a short period of adjustment the metabolic trial was started with 9 periods each lasting 7 days. The composition of fatty acids in the faecal lipids was analysed by gas chromatography. The studies led to the conclusion that the fatty acids contained in the faecal fat were not immediately derived from the dietary fat. The results of the trial also showed that the major proportion of all the fatty acids present in the faecal lipids consisted of only 4 fatty acids, viz. oleic acid, palmitic acid, lauric acid and myristic acid. Changes observed for the content of linoleic acid in faecal lipids were negligible so that the assumption may be made that all of the linoleic acid added was completely digested.  相似文献

Thirty-one duplicate and similar named rice germplasms of Bangladesh were studied to assess the genetic variation for the agro-morphological and physico-chemical traits and simple sequence repeat banding patterns during 2009–2012 at Bangladesh Rice Research Institute. The range of variations within the cultivar groups showed higher degree. The principal component analysis showed that the first five components with vector values 1 contributed 82.90% of the total variations. The cluster analysis grouped the genotypes into four clusters, where no duplicate germplasm was found. The highest number(11) of genotypes was constellated in cluster I and the lowest(3) in cluster II. The intra- and inter-cluster distances were the maximum in cluster I(0.93) and between clusters I and IV(24.61), respectively, and the minimum in cluster IV(0.62) and between clusters I and III(5.07), respectively. The cluster mean revealed that the crosses between the genotypes of cluster I with those of clusters II and IV would exhibit high heterosis for maximum good characters. A total of 350 alleles varied from 3(RM277) to 14(RM21) with an average of 7.8 per locus were detected at 45 microsatellite loci across the 31 rice accessions. The gene diversity ranged from 0.48 to 0.90 with an average of 0.77, and the polymorphism information content values from 0.44(RM133) to 0.89(RM206) with an average of 0.74. RM206, RM21, RM55, RM258 and RM433 were considered as the best markers on the basis of their higher polymorphism information content values. The dendrogram from unweighted pair-group method with arithmetic average clustering also classified the genotypes into four groups, where group IV comprised of 20 genotypes and group III of one genotype, but no duplicate was found. Finally, similar and duplicate named rice germplasms need to be conserved in gene bank as are distinct from each other.  相似文献

The nematicidal effect of Pseudomonas fluorescens, Paecilomyces lilacinus, Pichia guilliermondii and Calothrix parietina singly or in combination was tested against root-knot nematode, Meloidogyne incognita. Treatments with P. fluorescens and P. lilacinus caused mortality of M. incognita as 45% and 30% of juveniles after 48 h of exposures, respectively compared to water control in vitro. Under greenhouse conditions, all treatments reduced the disease severity and enhanced plant growth compared to untreated control. Application of P. fluorescens, P. lilacinus and P. guilliermondii Moh 10 was more effective compared to C. parietina. There was a negative interaction between C. parietina and either P. lilacinus or P. guilliermondii. Fresh and dry weight of shoots and roots of plants were significantly reduced as a result of infection with M. incognita, however application of biocontrol agents singly or in mix recovered this reduction. Moreover, they enhanced the growth parameters compared with the control. Our results proved that application of different biocontrol agents (P. fluorescens, P. lilacinus and P. guilliermondii) not only has a lethal effect on nematode, but also enhances the plant growth, supplying many nutritional elements and induction the systemic resistance in plants. Presence of C. parietina as a soil inhabitant cyanobacterium could antagonize biocontrol agents leading to the reduction of their practical efficiency in soil.  相似文献

Light is an important resource that crops and weeds compete for and so increased light interception by the crop can be used as a method of weed suppression in cereal crops. This research investigated the impact of altered availability of photosynthetically active radiation (PAR) (from crop row orientation or seeding rate) on the growth and fecundity of Lolium rigidum. Wheat and barley crops were sown in an east–west (EW) or north–south (NS) direction, at a high or low seeding rate, in three field trials in 2010 and 2011 (at Merredin, Wongan Hills and Katanning, Western Australia). The average PAR available to L. rigidum in the inter‐row space of EW crops compared with NS crops was 78% to 91% at crop tillering, 39% to 56% at stem elongation, 28% to 53% at boot/anthesis and 41% to 59% at grain fill. Reduced PAR in the EW crop rows resulted in reduced L. rigidum fecundity in five of the six trials (average of 2968 and 5705 L. rigidum seeds m?2 in the EW and NS crops). Availability of PAR was not influenced by seeding rate, but the high seeding rate reduced fecundity in three of the six trials (average of 3354 and 5092 seeds m?2 in the crops with high and low seeding rate). Increased competitive ability of crops (through increased interception of PAR or increased crop density) was highly effective in reducing L. rigidum fecundity and is an environmentally friendly and low cost method of weed suppression.  相似文献

The role of disulfide bridges in the folding of Aspergillus niger phytase pH 2.5-optimum (PhyB) was investigated using dynamic light scattering (DLS). Guanidinium chloride (GuCl) at 1.0 M unfolded phytase; however, its removal by dialysis refolded the protein. The thiol reagent tris(2-carboxyethyl)phosphine (TCEP) reduces the refolding activity by 68%. The hydrodynamic radius (R(H)) of PhyB phytase decreased from 5.5 to 4.14 nm when the protein was subjected to 1.0 M GuCl concentration. The active homodimer, 183 kDa, was reduced to a 92 kDa monomer. The DLS data taken together with activity measurements could indicate whether refolding took place or not in PhyB phytase. The correlation between molecular mass and the state of unfolding and refolding is a very strong one in fungal phytase belonging to histidine acid phosphatase (HAP). Unlike PhyA phytase, for which sodium chloride treatment boosted the activity at 0.5 M salt concentration, PhyB phytase activity was severely inhibited under identical condition. Thus, PhyA and PhyB phytases are structurally very different, and their chemical environment in the active site and substrate-binding domain may be different to elicit such an opposite reaction to monovalent cations.  相似文献

The present study was undertaken to investigate the effect of soil contamination with heavy metals on physico-biochemical properties of tomato fruit. The deleterious effect of soil contamination on the quality and quantity of tomato plant yield was determined. Harvested tomato fruit characteristics including fruit length, diameter, volume and fresh and dry weights, total soluble solids, titratable acidity, lycopene and carbohydrates contents were measured in tomato fruits cultivated in soil contaminated with industrial effluents. In addition, some secondary metabolites (total phenols and flavonoids), micro- and macronutrients were also detected. Residual heavy metals (Cd, Co, Ni and Pb) were examined in the harvested fruits. The obtained results were compared with those of tomato fruits cultivated in non-contaminated soil. Results demonstrate that soil contamination with heavy metals have a negative effect on tomato fruits characteristics, titratable acidity, total soluble solids, lycopene, ascorbic acid, micro-elements and carbohydrates content. Moreover, fruits originated from plants grown in contaminated soil possess high phenols and flavonoids contents and higher heavy metals content compared to control fruits. It is recommended that fruits cultivated in that area not to be eaten by large quantities, to avoid excessive accumulation of heavy metals in the human body.

Rhizobium-legume symbioses are important for their nitrogen input, but salinity and elevated temperature in arid and semi-arid areas limit their effectiveness, and therefore plant growth and productivity. Sixteen Rhizobium strains isolated from root nodules of Leucaena trees grown in different geographical areas of Egypt varied in their degree of tolerance to salinity and in their symbiotic effectiveness with Leucaena leucocephala under saline conditions. Three strains were tolerant to >3% NaCl. L. leucocephala grown in the greenhouse at concentrations of NaCl up to 1.0% and inoculated either with strain DS 78 or strain DS 158 displayed significantly better growth than those plants grown at the same levels of salinity and inoculated with reference strain TAL 583. Although nine of the Rhizobium strains grew at 42  °C, their mean generation times were lengthened two- to fourfold. When daylight growth temperatures were elevated from 30  °C to 42  °C, nodule number and mass, nitrogenase activities and shoot top dry weight of plants inoculated with strains DS 78, DS 157 and DS 158 significantly increased, whereas these parameters decreased in plants inoculated with strain TAL 583. Weed seeds are introduced to agronomic systems naturally or through human-mediated seed dispersal, and introduced seeds have a high chance of being resistant to selective, in-crop herbicides. However, colonisation (invasion) rates for a weed species are usually much lower than rates of seed dispersal. The current research investigated colonisation of a winter annual wheat cropping system in Western Australia by a range of winter or summer annual weed species. The weed seeds were sown (at 100 seeds/m2) directly before seeding the crop in 2016 and allowed to grow in the following 3 years of wheat. Selective herbicides were not applied, to simulate growth of weed populations if the initial seed had been resistant to herbicide. Bromus diandrus, Hordeum leporinum, Rumex hypogaeus, Sonchus oleraceus, Polygonum aviculare, Lolium rigidum, Citrullus amarus and Tribulus terrestris colonised the crop, while Dactyloctenium radulans, Chloris truncata and Salsola australis failed to establish over 3 years. The most successful weed was B. diandrus, with a plant density of 1,170/m2 by the third year and seed production of 67,740/m2. The high density of B. diandrus reduced wheat density by 76% in the third year and reduced average yield by 36%. Lolium rigidum reduced average yield by 11%, and the other weed species did not affect crop yield. Further research is required on the invasiveness of these species in other regions, but it is clear that the spread of B. diandrus to new areas or the introduction of resistant B. diandrus seeds via contaminated grain should be avoided.  相似文献

Immobilizing materials such as lherzolite could reduce metal bioavailability but the effectiveness of lherzolite on the extractability and bioavailability of cadmium (Cd) and zinc (Zn) is rarely investigated. We conducted a greenhouse experiment to investigate the effect of 5% application of lherzolite to a contaminated soil on the chemical fractionation of Cd and Zn and their uptake by radish (Raphanus sativus L.) and Japanese mustard spinach (Brassica rapa L. var. perviridis). Both plants were grown in a highly contaminated (with Cd and Zn) sandy loam soil. Plants were cultivated consecutively three times in the same pots. After the third cultivation, soil samples were collected and analyzed by sequential extraction procedure into five operationally defined fractions (F1—exchangeable, F2—carbonate-bound, F3—oxides-bound, F4—bound with organic matter, and F5—residual). Addition of lherzolite to soil decreased 50% of exchangeable (F1) Cd but it increased the carbonate (F2), oxide (F3), and organic (F4) fraction Cd. For Zn, application of lherzolite resulted into the reduction of both F1 (87%) and F2 (33%) fractions but it increased the F3, F4, and F5 fractions. The reduction in exchangeable fraction of Cd and Zn in the soil resulted in higher plant growth and lower concentrations of both Cd (64% to 92%) and Zn (78% to 99%) in plant tissues of both plant species grown. We may thus conclude that application of lherzolite resulted into lower availability of these metals in the soil leading to lower uptake of Cd and Zn by plant roots, lower toxicity, and ultimately higher plant growth.  相似文献

The present study was conducted to examine the comparative efficacy of potassium simplex optimization medium (KSOM) and North Carolina State University (NCSU)-23 medium supplemented with beta-mercaptoethanol (beta-ME) and amino acids (AA) on the developmental competence of porcine in vitro fertilized (IVF) embryos. Four experiments were conducted. KSOM and NCSU-23 medium were used to culture porcine parthenogenetic (Exp. 1) and IVF (Exp. 2) embryos. KSOM and NCSU-23 were equally effective in supporting porcine parthenogenetic and IVF embryo development from the 1-cell stage to blastocysts. The NCSU-23 medium (Exp. 3) and KSOM (Exp. 4) were supplemented with amino acid (AA; 5 microl/ml non-essential amino acids + 10 microl/ml essential amino acids) and/or 10 microM beta-mercaptoethanol (beta-ME). The quality of blastocysts from Exp. 3 and 4 was evaluated by counting the number of total cells and determining the ratio of the inner cell mass (ICM) to trophoectoderm (TE) cells. Supplementing with AA and beta-ME or beta-ME alone in NCSU-23 produced significant (p<0.05) differences in terms of rate of cleavage to the 2- to 4- cell (80.8 to 85.4% vs. 73.6%) and blastocyst (30.4 to 30.5 vs. 23.5%) stages and the number of TE (51.4 to 53.8 vs. 35.8) and total cells (67.2 to 71.2 to 48.8) over the control group. On the other hand, supplementing KSOM with AA and/or beta-ME produced significant (p<0.05) differences in terms of rate of cleavage to the 2- to 4-cell (78.8% vs. 67.7%) and morula (57.8% vs. 46.3%) stages and the number of ICM (18.6 to 19.2 vs. 11.6) and total cells (62.8 to 70.6 vs. 42.8) over control group. In conclusion, our study demonstrates that both KSOM and NCSU-23 medium supplemented with AA and beta-ME and/or only beta-ME alone are superior to normal KSOM and NCSU-23 for porcine IVF embryo culture in terms of embryo developmental competence and quality.  相似文献
