East Coast fever (ECF) causes considerable mortality and production losses in the Tanzania smallholder dairy sector and limits the introduction of improved dairy breeds in areas where the disease is present. The infection and treatment method (ITM) was adopted by smallholder dairy farms for ECF immunisation in Hanang and Handeni districts of Tanzania. This study recorded incidence rates for ECF and other tick-borne diseases (TBDs) for ECF-immunised and non-immunised cattle between 1997 and 2000. Approximately 80% of smallholder households from both sites (n?=?167) participated in this longitudinal study, with immunisations carried out at the request of the livestock owners. Efficacy of ITM for preventing ECF cases in these crossbred dairy cattle was estimated at 97.6%, while that for preventing ECF deaths was 97.9%. One percent of the cattle developed clinical ECF as a result of immunisation. Since ECF immunisation permits a reduction in acaricide use, an increase in other TBDs is a potential concern. Sixty-three percent of farmers continued to use the same acaricide after immunisation, with 80% of these reducing the frequency of applications. Overall, 78% of farmers increased the acaricide application interval after immunisation beyond that recommended by the manufacturer, resulting in annual savings in the region of USD 4.77 per animal. No statistical difference was observed between the immunised and non-immunised animals in the incidence of non-ECF TBDs. However, immunised animals that succumbed to these diseases showed fewer case fatalities. A 2‐year whole‐farm system study compared the accumulation, utilization and nutritive value of grass in spring‐calving grass‐based systems differing in stocking rate (SR) and calving date (CD). Six treatments (systems) were compared over two complete grazing seasons. Stocking rates used in the study were low (2·5 cows ha?1), medium (2·9 cows ha?1) and high (3·3 cows ha?1), respectively, and mean CDs were 12 February (early) and 25 February (late). Each system had its own farmlet of eighteen paddocks and one herd that remained on the same farmlet area for the duration of the study. Stocking rate had a small effect on total herbage accumulation (11 860 kg DM ha?1 year?1), but had no effect on total herbage utilization (11 700 kg DM ha?1 year?1). Milk and milk solids (MS; fat + protein) production per ha increased by 2580 and 196 kg ha?1 as SR increased from 2·5 to 3·3 cows ha?1. Milk production per ha and net herbage accumulation and utilization were unaffected by CD. Winter feed production was reduced as SR increased. Increased SR, associated with increased grazing severity, resulted in swards of increased leaf content and nutritive value. The results indicate that, although associated with increased milk production per ha, grazed grass utilization and improved sward nutritive value, the potential benefits of increased SR on Irish dairy farms can only be realized if the average level of herbage production and utilization is increased. 相似文献

The addition of yeast cell wall (YCW) mannan fractions or low concentrations of antibiotics to the diet of broilers positively affects gut health by improving intestinal cell morphology and improves feed efficiency and performance; however the exact mechanisms are unclear. Based on these production responses, the objective of this study was to compare the effects of supplementing YCW and bacitracin methylene disalicylate (BMD) in the diet on mRNA levels in the jejunum of 6-week-old broilers.
2. Dietary treatments were a maize-soya control diet and the control diet with the addition of YCW or BMD. Birds (n = 7) from each dietary treatment were randomly selected and killed at d 42. Whole jejunum (with serosa) samples were collected for RNA isolation. Gene expression analysis was performed using the AffymetrixGeneChip Chicken Genome Array (Santa Clara, CA, USA).
3. Supplementation with YCW resulted in 928 genes that were significantly changed (456 down-regulated, 472 up-regulated) and supplementation with BMD resulted in 857 genes that significantly changed (408 down-regulated, 449 up-regulated). In addition, 316 genes were significantly changed by both YCW and BMD (146 down-regulated, 170 up-regulated).
4. BMD increased the expression of genes involved in lipid and carbohydrate metabolism and decreased expression of genes associated with T-helper cell pathways. Gene expression profiles from birds fed on diets containing YCW showed changes on a genomic level that correspond to slower gut cell turnover and therefore increased energy preservation for growth.
5. In conclusion, supplementation with BMD or YCW had similar influences on the number of differentially expressed genes in the jejunum. Biological functions common to both YCW and BMD with positive activation scores included antiviral response and antimicrobial response. Genes that were affected by BMD or YCW classified into both different and common biological functions and pathways related to improved metabolism and health in the jejunum. 相似文献
Using a nonionic, alkyl polyoxyethylene surfactant (X-77) in aqueous solutions, sessile droplet spreading, pinning, evaporation, contraction, and post-evaporation deposits are characterized. X-77 is widely used in the agricultural field as a spreader/adherent, intended to optimize pathenogenic agent coverage. Using a single droplet size under monitored temperature conditions, we control humidity, substrate hydrophobicity, and surfactant concentration to mimic varying agricultural conditions. For hydrophilic surfaces, the droplet spreads, reaching and retaining a maximum, stationary size. At this stage, a ring accretion occurs at the maximum spread diameter. During the final stage, the water film retracts, resulting in deposition of small islands of surfactant residue inside the ring. At lower concentrations of surfactant, we discover ring formations that break-up into "ring islands" at late-stage evaporation, accompanied by a distribution of the smaller islands in the interior portion of the substrate contact area. These are promoted by higher relative humidity. At higher concentrations, only a solid ring of surfactant remains, post-evaporation. Increasing surfactant concentration tends to increase the mean of the interior island size and to broaden the overall island size distribution. On sufficiently hydrophobic surfaces, surfactant-laden droplets do not evidence pinning, ring formations, or post-evaporation interior islands. Interestingly, lower humidity increases spreading at higher surfactant concentrations. Such pattern formations of surfactant deposit are reported for the first time and are of significance in projecting how surfactants such as X-77 distribute pesticides or other chemicals on leaf surfaces. 相似文献
Cattle feedyards can impact local environments through emission of ammonia and dust deposited on nearby land. Impacts range
from beneficial fertilization of cropland to detrimental effects on sensitive ecosystems. Shortgrass prairie downwind from
an adjacent feedyard on the southern High Plains of Texas, USA changed from perennial grasses to annual weeds. It was hypothesized
that N enrichment from the feedyard initiated the cascade of negative ecological change. Objectives were to determine the
distribution of soil nitrogen and estimate N loading to the pasture. Soil samples were collected from 119 locations across
the pasture and soil total N (TN), nitrate-N and ammonium-N (AN) determined in the top 30 cm. Soil TN concentration decreased
with distance downwind from the feedyard from 1.6 ± 0.2 g kg−1 at 75 m to 1.2 ± 0.05 g kg−1 at 582 m. Nitrate-N concentration decreased within 200 m of the feedyard and changed little at greater distances. Ammonium-N
concentration decreased linearly (P < 0.001) with increasing distance from the feedyard from 7.9 ± 1.7 mg kg−1 within 75 m from the feedyard to 5.8 ± 1.5 mg kg−1 at more than 550 m from the feedyard; however, distance only explained 12% of the variability in AN concentration. Maximum
nitrogen loading, from 75 to 106 m from the feedyard, was 49 kg ha−1 year−1 over 34 years and decreased with distance from the feedyard. An estimate of net dry deposition of ammonia indicated that
it contributed negligibly to N loading to the pasture. Nitrogen enrichment that potentially shifted vegetation from perennial
grasses to annual weeds affected soil N up to 500 m from the feedyard; however, measured organic and inorganic N beyond that
returned to typical and expected levels for undisturbed shortgrass prairie.
Organisations acting to conserve and protect species across large spatial scales prioritise to optimise use of resources. Spatial conservation prioritization tools typically focus on identifying areas containing species groups of interest, with few tools used to identify the best areas for single-species conservation, in particular, to conserve currently widespread but declining species.
Objective
A single-species prioritization framework, based on temporal and spatial patterns of occupancy and abundance, was developed to spatially prioritize conservation action for widespread species by identifying smaller areas to work within to achieve predefined conservation objectives.
Methods
We demonstrate our approach for 29 widespread bird species in the UK, using breeding bird atlas data from two periods to define distribution, relative abundance and change in relative abundance. We selected occupied 10-km squares with abundance trends that matched species conservation objectives relating to maintaining or increasing population size or range, and then identified spatial clusters of squares for each objective using a Getis-Ord-Gi* or near neighbour analysis.
Results
For each species, the framework identified clusters of 20-km squares that enabled us to identify small areas in which species recovery action could be prioritized.
Conclusions
Our approach identified a proportion of species’ ranges to prioritize for species recovery. This approach is a relatively quick process that can be used to inform single-species conservation for any taxa if sufficiently fine-scale occupancy and abundance information is available for two or more time periods. This is a relatively simple first step for planning single-species focussed conservation to help optimise resource use.
We developed a TaqMan real-time polymerase chain reaction (PCR) assay for the quantitative detection of Theileria equi from the in vitro-cultured parasite and field blood samples collected from horses living in Ghana and Brazil. The detection limit for the assay was determined to be 1.5 parasites/microl per sample, and the quantitative capacity was demonstrated using the in vitro-cultured parasite. For field applications, the real-time PCR assay was compared to a previously established nested PCR assay used as the gold standard for the real-time PCR assay. Of 65 field blood samples, 46 samples were T. equi-positive in the nested PCR assay, while the real-time PCR assay also detected the parasite in all 46 of the nested PCR-positive samples but did not detect T. equi in the remaining 19 negative blood samples. This quantitative real-time PCR assay provides a valuable tool for fast laboratory diagnostic assessment of T. equi infection in horses. 相似文献
In this study, we developed two immunochromatographic tests (ICTs), which are nitrocellulose membrane-based immunoassays for the convenient and rapid serodiagnosis of bovine babesiosis caused by Babesia bovis (BoICT) and Babesia bigemina (BiICT). The efficacy of two ICTs was evaluated using 13 positive sera from experimentally infected cattle with B. bovis or B. bigemina. Clear results showed that the BoICT and ELISA detected antibodies in sera collected from 14 to 93 days post-infection, while BiICT and ELISA detected from 13 to 274 days post-infection. In additon, non-infected cattle, Neospora caninum, and Cryptosporidium parvum were negative in two ICTs. To evaluate the field utility of the ICTs, we tested 186 field bovine sera collected from cattle living in Yanbian (China) and Mato Grosso do Sul (Brazil). The results of ICTs were compared to those of classical serodiagnostic methods, enzyme-linked immunosorbent assay (ELISA) and the indirect immunofluorescence assay (IFAT). The overall concordances of BoICT were determined as 92.5 and 90.3% when the results of ELISA and IFAT were set as the reference standards, respectively. In contrast, those of BiICT showed 96.8 and 92.5% relative to the results of standard ELISA and IFAT, respectively. Conventional and rapid diagnosic devices for bovine babesiosis may provide a valuable tool in clinical and field applications. 相似文献
