BackgroundCanine herpesvirus type 1 (CaHV-1) infects dogs and is associated with neonatal deaths and reproductive, ocular, neurological, and respiratory problems. In Brazil, reports of CaHV-1 have been restricted to the southeast and south regions, particularly in municipalities in the state of Rio Grande do Sul.ObjectivesTo assess the presence and variability of CaHV-1 in canine populations in the state of Pará, North Brazil.MethodsBiological samples from 159 dogs from 4 municipalities in the State of Pará were evaluated using polymerase chain reaction and phylogenetic analyses, with the target being the viral enzyme, thymidine kinase.ResultsCaHV-1 was detected in 13 dogs (8.2%), with 2 animals being from the municipality of Santa Bárbara do Pará, 8 from Algodoal Island, 2 from Salinópolis, and one from Capanema. The study sequences revealed 100% identity among themselves and 64% to 100% identity with the other nucleotide sequences from Australia, Brazil, United Kingdom, and United States, including 100% identity with the 2002 isolate from Australia. The 1996 isolate from France was grouped in a branch that was different from the sequence of this study.ConclusionsThis study presents the first molecular detection of CaHV-1 in dogs from the Amazon region in northern Brazil. The nucleotide identity between the strains and cytosine insertion in the sequences isolated in this study suggests at least 2 strains of CaHV-1 circulating in Brazil (Pará and BTU-1). 相似文献

The presence of illicit drugs and their metabolites in surface waters has to be considered a new type of hazard, still unknown, for the aquatic ecosystem, due to the potent pharmacological activities of all the illicit drugs. Our research was therefore aimed at evaluating the impact of illicit drugs on the aquatic fauna, till now still undervalued. To this aim, we verified the ability of the European eel (Anguilla anguilla), a well-known biomonitor of environmental contamination, to bioaccumulate cocaine, one of the most abundant illicit drugs found in surface waters. Silver eels were exposed to a nominal cocaine concentration of 20?ng/l for 1?month; at the same time, control, carrier, and post-exposure recovery groups were made. Brains, gills, liver, kidney, muscle, gonads, spleen, digestive tract, and sections of dorsal skin were assayed by high-pressure liquid chromatography. Cocaine was found in the tissues of the treated eels and, at low concentrations, in almost all tissues of post-exposure recovery eels. These results indicate that cocaine is able to accumulate into the eel tissues; its presence suggests potential risks for eels since cocaine could affect their physiology and contribute to their decline, and for humans consuming contaminated fish. 相似文献

Zinc (Zn) is a plant nutrient; however, at elevated levels it rapidly becomes phytotoxic. In order to obtain insight into the physiological background of its toxicity, the impact of elevated Zn2+ concentrations (1 to 10 μM) in the root environment on physiological functioning of Chinese cabbage was studied. Exposure of Chinese cabbage (Brassica pekinensis) to elevated Zn2+ concentrations (≥ 5 μM) in the root environment resulted in leaf chlorosis and decreased biomass production. The Zn concentrations of the root and shoot increased with the Zn2+ concentration up to 68‐fold and 14‐fold, respectively, at 10 μM compared to the control. The concentrations of the other mineral nutrients of the shoot were hardly affected by elevated Zn2+ exposure, although in the root both the Cu and Fe concentrations were increased at ≥ 5 µM, whereas the Mn concentration was decreased and the Ca concentration strongly decreased at 10 µM Zn2+. The uptake and metabolism of sulfur and nitrogen were differentially affected at ≥ 5 µM Zn2+. Zn2+ exposure resulted in an increase of sulfate uptake and the activity of the sulfate transporters in the root, and in enhanced total sulfur concentration of the shoot, which could be ascribed partially to an accumulation of sulfate. Moreover, Zn2+ exposure resulted in an up to 6.5‐fold increase in water‐soluble non‐protein thiol (and cysteine) concentration of the root. However, nitrate uptake by the root and the nitrate and total nitrogen concentrations of the shoot were decreased upon Zn2+ exposure, demonstrating the absence of a mutual regulation of the uptake and metabolism of sulfur and nitrogen at toxic Zn levels. Evidently, elevated Zn2+ concentrations in the root environment did not only disturb the uptake, distribution and assimilation of sulfate, it also affected the uptake and metabolism of nitrate in Chinese cabbage. 相似文献

Emmer wheat (Triticum dicoccon Schrank, 2n = 4x = 28) consists in a hulled wheat; its cultivation has been drastically reduced during the last century as a consequence of
its low yield. Recently, its agronomic and nutritive values, as well as the increase of popularity of organic agriculture,
have led to a renewed interest making its cultivation economically viable in the marginal lands with an increase of the cultivated
areas. In Italy, it mainly survives in few marginal lands of central and southern Italy, where local varieties, adapted to
the natural environment from where they originate, are used; moreover, some selected lines have also been developed. In the
present work, agro-morphological and qualitative traits, together with molecular analyses of 20 emmer accessions consisting
of Italian landraces, breeding lines, and cultivars, were performed. The field experiments were conducted for two consecutive
years (2001/2002–2002/2003) in two locations: Viterbo in central Italy, and Foggia in south Italy. The analyzed emmer wheat
accessions showed a good amount of genetic variability for both evaluated agro-morphological and molecular traits. This study
illustrates an increase in earliness, GY, TW, TKW, and YI going from landraces, breeding lines to cultivars, while the variability

The rates of sap flow and xylem vessel features were studied in two-year-old nongrafted and grafted avocado (Persea americana Mill.) trees. Daily sap flow rates were measured with heat and balance stem gauges in clonal Duke 7 (D7) and Toro Canyon (TC) trees and ‘Hass’ clonal scions grafted onto clonal D7 (H/D7) and TC (H/TC) rootstocks. Vessel features as size, number and total vessel area were determined histologically in the stem of the scion and rootstock and the roots of the grafted trees. Significant differences in the sap flow rate were found among the rootstocks, where D7 had a 29% higher sap flow rate than did TC (grafted and nongrafted trees). There were no differences among xylem vessel features in the stems of any of the varieties. However in the roots, D7 had wider and fewer vessels then TC do. Also, D7 had a 19% higher total vessel area than TC. These results suggest that the differences in water consumption of ‘Hass’ on different rootstocks may be associated with differences in the efficiency of the roots to absorb water across conductive tissue which may be linked to differences in the area of xylem vessels in the root. 相似文献

Genetic diversity was evaluated by RAPD markers and morpho-agronomic characters for a total of 42 accessions of Barberton daisy (Gerbera jamesonii) consisting of 29 commercial and 13 wild accessions. A total of 74 polymorphic bands were obtained employing a set of 12 primer pairs. The average genetic similarity coefficient for the 42 accessions, evaluated by Jaccard index was 0.55 ranging from 0.28 to 1.00. The genetic structure found among Barberton daisy accessions was evaluated by hierarchic classification analyses and UPGMA modeling, revealing six clusters of genotypes where two of them include the wild accessions and the remaining four including commercial material, except for wild genotype number 9. Shannon (H′) index was calculated using the molecular markers to investigate the genetic variation among the Gerbera accessions and showed higher values for the commercial cluster in comparison to the values obtained for the individuals from the non-commercial cluster, namely 0.34 versus 0.27, respectively. Therefore, both calculated indices (Jaccard and Shannon) indicated the presence of higher genetic variation among commercial accessions in comparison to the cluster representing non-commercial accessions, suggesting that genetic breeding programs may focus on commercial accessions to recombine interesting genotypes with commercially important and marketing-desired characteristics. 相似文献

The use of plant water status indicators such as midday stem water potential (Ψstem) and maximum daily trunk shrinkage (MDS) in irrigation scheduling requires the definition of a reference or threshold value, beyond which irrigation is necessary. These reference values are generally obtained by comparing the seasonal variation of plant water status with the environmental conditions under non-limiting soil water availability. In the present study an alternative approach is presented based on the plant’s response to water deficit. A drought experiment was carried out on two apple cultivars (Malus domestica Borkh. ‘Mutsu’ and ‘Cox Orange’) in which both indicators (Ψstem and MDS) were related to several plant physiological responses. Sap flow rates, maximum net photosynthesis rates and daily radial stem growth (DRSG) (derived from continuous stem diameter variation measurements) were considered in the assessment of the approach. Depending on the chosen plant response in relationship with Ψstem or MDS, the obtained reference values varied between −1.04 and −1.46 MPa for Ψstem and between 0.17 and 0.28 mm for MDS. In both cultivars, the approach based on maximum photosynthesis rates resulted in less negative Ψstem values and smaller MDS values, compared to the approaches with sap flow and daily radial stem growth. In the well-irrigated apple trees, day-to-day variations in midday Ψstem and MDS were related to the evaporative demand. These variations were more substantial for MDS than for midday Ψstem. 相似文献
