Summary Inheritance of resistance to beet necrotic yellow vein virus (BNYVV) was studied in segregating F2 and backcross families obtained from crosses between resistant plants of the sugar beet selection Holly-1-4 or the wild beet accession Beta vulgaris subsp. maritima WB42 and susceptible parents. Greenhouse tests were carried out, in which seedlings were grown in a mixture of sand and infested soil. Virus concentrations of BNYVV in the rootlets were estimated by ELISA. To discriminate resistant and susceptible plants, mixtures of normal distributions were fitted to log10 virus concentrations, estimated for segregating F1, F2 and BC populations of both accessions. The hypothesis that Holly-1-4 contained one single dominant major gene was accepted. For WB42, results fitted with the hypotheses that resistance was based on either one (or more) dominant major gene(s) showing distorted segregation, or two complementary dominant genes, which are both required for resistance. Beet cyst nematodes (BCN, Heterodera schachtii), Cercospora beticola, and rhizomania, caused by the beet necrotic yellow vein
virus (BNYVV) and vectored by the soil-borne fungus Polymyxa betae, are the most serious diseases of sugar beet ( Beta vulgaris
subsp. vulgaris). The wild Beta species of section Procumbentes are known to be completely resistant to H. schachtii, C. beticola
and P. betae. Alien monosomic additions (2n=19), plants of cultivated beet (2n=18) carrying different individual chromosomes
of B. procumbens (2n=18) or B. patellaris (2n=36), were tested in greenhouse experiments for resistance to these pathogens.
Gene(s) conferring full resistance to the beet cyst nematode in B. patellaris are located on chromosome 1.1, and the other
tested chromosomes of B. patellaris are not involved in the expression of resistance. Artificial inoculation under greenhouse
conditions, with in vitro produced inoculum of C. beticola and spot-percentage rating of the disease intensity, showed that
the high level of resistance that was observed in the wild species B. procumbens and B. patellaris was not found in any of
the monosomic additions tested. It was suggested that genes on various chromosomes of the wild species are needed to express
full resistance, and that the chromosomes of group 7 of B. patellaris and chromosome 7 of B. procumbens have the largest effect.
The greenhouse tests for resistance to P. betae in B. patellaris derived monosomic additions showed that the addition families
of group 4.1 have a strong partial resistance, while the addition families of group 8.1 appeared to be completely resistant
to the pathogen. Resistance to P. betae in the two wild species as well as in the two resistant addition types did not exclude
infection with BNYVV, but resulted in a considerable reduction of the virus concentration. It was concluded that resistance
to the vector would complement virus resistance, and may provide a more effective and durable control of rhizomania.

Summary Three Cucumis sativus L. (CS) lines adapted to USA growing conditions were used as female parents in crosses to one line each of seven Cucumis sativus var. hardwickii (R.) Alef. (CH) accessions used as male parents to determine the relative combining ability of the CH accessions for six horticultural characters. The 21 F1 progenies were grown in a randomized complete block design in two locations and evaluated for fruit number, lateral branch number, fruit length, fruit length/diameter ratio, number of female flowering nodes, and days to anthesis. General combining ability (GCA) was significant for all characters in each location. Specific combining ability (SCA) was significant for all characters except days to anthesis in one location and lateral branch number in both locations. Significant location × combining ability interactions were also evident for several characters. Cucumber (Cucumis sativus L.) is a major cucurbit vegetable species whose genetic base has been drastically reduced during its domestication. The crop’s
narrow genetic base (3–12% DNA polymorphism) has resulted from the use of limited genetic material and intense selection during
plant improvement. Recently, however, interspecific hybridization has been successful in Cucumis via mating of C. hystrix Chakr. and C. sativus, which resulted in the amphidiploid C. hytivus. We report herein a marker-assisted strategy for increasing genetic diversity in cucumber through introgression backcrossing
employing C. hytivus. The comparatively late-flowering but high-yielding, indeterminate, monoecious line WI 7012A (P1; donor parent) derived from a C. hytivus × C. sativus-derived line (long-fruited Chinese C. sativus cv. Beijingjietou) was initially crossed to the determinate, gynoecious C. sativus line WI 7023A (P2; recurrent parent 1), and then advanced backcross generation progeny (BC2) were crossed with the gynoecious indeterminate line WI 9-6A (P3; recurrent parent 2). More specifically, a single F1 individual (P1 × P2) was backcrossed to P2, and then BC progeny were crossed to P2 and P3, where marker-assisted selection (MAS) for genetic diversity (8 mapped and 16 unmapped markers; designated Sel) or no selection
(designated NSel) was applied to produce BC3P2 (Sel) and BC3P3 (Sel), and BC2P2 (NSel) and BC2P2S1 (NSel) progeny. Relative vegetative growth, number of lateral branches (LB), days to flowering (DF), yield (fruit number),
and fruit quality [as measured by length:diameter (L:D) and endocarp:total diameter (E:T) ratios] were assessed in parents
and cross-progeny. DF varied from ~20 (BC3P2Sel) to ~25 days (BC2P3Sel) among the populations examined, where progeny derived from P2 possessed the shortest DF. Differences in cumulative yield among the populations over six harvests were detected, varying
from ~8 fruits per plant in BC3P2 (Sel) to ~39 fruits per plant in BC2P3 (Sel). Although the vigorous vegetative growth of line P1 was observed in its backcross progeny, highly heterozygous and polymorphic backcross progeny derived from P3 were comparatively more vigorous and bore many high-quality fruit. The antigenic evolution of influenza A (H3N2) virus was quantified and visualized from its introduction into humans in 1968 to 2003. Although there was remarkable correspondence between antigenic and genetic evolution, significant differences were observed: Antigenic evolution was more punctuated than genetic evolution, and genetic change sometimes had a disproportionately large antigenic effect. The method readily allows monitoring of antigenic differences among vaccine and circulating strains and thus estimation of the effects of vaccination. Further, this approach offers a route to predicting the relative success of emerging strains, which could be achieved by quantifying the combined effects of population level immune escape and viral fitness on strain evolution. 相似文献

Germplasm conserved as seeds in genebanks requires regular regeneration. In this process, selection and genetic drift may cause loss of genetic diversity from accessions. In the case of selfing crops, separation of distinct lines into different accessions may be an efficient strategy to avoid these negative effects. In order to evaluate the applicability of this method for collection management, knowledge about the level of intra-accession genetic diversity is required. By means of AFLP analysis intra-accession variation was investigated in two cultivars, two landraces and two wild populations of ex situ conserved barley germplasm. In the total sample of 216 individuals analysed (36 per accession), 22 genotypes were observed based on 104 polymorphic loci. The number of genotypes detected ranged from 1 to 3 per accession, except for a Nepalese landrace that revealed 12 genotypes. An UPGMA cluster analysis grouped the genotypes unambiguously into the accession they belonged to and genotypes within accessions were generally found to be closely related. In order to determine the repeatability of the results obtained, 11 individuals belonging to 4 genotypes from the Nepalese landrace were scored for a second set of AFLP markers. Matrices of genetic distances calculated for the two AFLP datasets were found to be highly correlated (r = 0.9346, P < 0.001). Separation of genotypes into different accessions was considered a relevant option only for the Nepalese landrace. Analysis of molecular variance indicated that this accession could be well divided into 8 distinct lines. Further implications of the results for genebank practices are discussed. 相似文献

Effects of chlorpyrifos in freshwater model ecosystems: the influence of experimental conditions on ecotoxicological thresholds 总被引:7,自引:0,他引:7
Three experiments were conducted to determine the impact of the insecticide chlorpyrifos (single applications of 0.01 to 10 microg AI litre(-1)) in plankton-dominated nutrient-rich microcosms. The microcosms (water volume approximately 14 litres) were established in the laboratory under temperature, light regimes and nutrient levels that simulated cool 'temperate' and warm 'Mediterranean' environmental conditions. The fate of chlorpyrifos in the water column was monitored and the effects on zooplankton, phytoplankton and community metabolism were followed for 4 or 5 weeks. The mean half-life (t1/2) of chlorpyrifos in the water of the test systems was 45 h under 'temperate' conditions and about 30 h under 'Mediterranean' environmental conditions. Microcrustaceans (cladocerans and copepod nauplii) were amongst the most sensitive organisms. All three experiments yielded community NOEC (no observed effect concentrations) of 0.1 microg AI litre(-1), similar to those derived from more complex outdoor studies. Above this threshold level, responses and effect chains, and time spans for recovery, differed between the experiments. For example, algal blooms as an indirect effect from the impact of exposure on grazing organisms were only observed under the 'Mediterranean' experimental conditions. The relatively simple indoor test system seems to be sufficient to provide estimates of safe threshold levels for the acute insecticidal effects of low-persistence compounds such as chlorpyrifos. The robustness of the community NOEC indicates that this threshold level is likely to be representative for many freshwater systems. 相似文献
