Increasing nitrogen deposition due to human activity might have a serious impact on ecosystem functions such as the nitrogen transformations conducted by microbes. We therefore focused on nitrous oxide (N2O) production as an indicator of soil microbial activity. The rates of N2O emission from the forest floor were measured every two weeks in two forest stands in the central part of Japan: a red pine stand at Kannondai and a deciduous stand at Yasato. Nitrogen deposition rates by throughfall were 30.6 kg N ha?1 y?1 at Kannondai and 15.7 at Yasato. The rates of N2O emission ranged from 0.5 to 14.2 µg N m?2 h?1 (mean 4.5) at Kannondai and from 0.2 to 7.0 µg N m?2 h?1 (mean 2.3) at Yasato. The N2O emission rate showed significant positive relationships with soil temperature and nitrogen deposition during the preceding two weeks. The annual emission rates of N2O were 0.38 kg N ha?1 y?1 at Kannondai and 0.20 at Yasato. As a the annual nitrogen deposition, these rates were 1.23% at Kannondai and 1.27% at Yasato.  相似文献

Dynamic tests of nailed plywood–timber joints are conducted under harmonic vibrations from 2 to 7 Hz. The principal results are as follows: under dynamic loading, nailed plywood–timber joints may break in low-cyclic bending fatigue failure of nails besides the other failure modes typical under static loading. The dynamic response of nailed plywood–timber joints is clearly dependent upon both the input frequency and the acceleration. These responsive characteristics arise from the nonlinear load–slip relationships and the characteristic cyclic stiffness degradation of nailed joints; that is, the cyclic degradation of the equivalent linear stiffness decreases the resonant frequencies of the same joints, which results in a transition of dynamic responses. It indicates that frequency components of seismic waves resonant to the frequencies corresponding to safety-limit stiffness of nailed joints may lead them to critical failures, even if the accelerations do not exceed the accelerations equivalent to the static damage-limit resistance.  相似文献

Isolates of Rhizoctonia solani AG2-2 obtained from turf with symptoms of large-patch disease of warm-season turfgrasses were compared with known AG2-2 isolates belonging to cultural types IIIB and IV. Some isolates that were previously identified as type IV have been separated here and named LP isolates. Comparisons among isolates were based on cultural morphology, hyphal growth rate, pathogenicity and restriction fragment length polymorphism (RFLP) analysis in the nuclear encoded ribosomal DNA (rDNA) genes. The cultural characteristics of LP isolates varied from those of types IIIB and IV. LP isolates did not show distinct sclerotial formation and zonation, and the colour of their mycelia and pigment deposition was dark brown. LP isolates had slower hyphal growth rates than types IIIB and IV, with an optimum temperature of 25°C compared with 28°C for types IIIB and IV. LP isolates were less virulent on radish but highly virulent on zoysia grass when compared with isolates of types IIIB and IV. Genomic DNA was digested separately with Eco RI, Ban III, Xba I and Sal I, and probed with cloned rDNA from Alternaria alternata in Southern hybridizations. LP isolates had one RFLP pattern, while both IIIB and IV possessed four different patterns each. Cluster analysis of RFLPs showed that R. solani AG2-2 is divided into three genetic subgroups, consisting of the IIIB, IV and LP isolates, respectively. The polymerase chain reaction (PCR) amplified rDNA internally transcribed spacer (ITS) regions of the IIIB, IV and LP isolates had the same length but produced different restriction patterns when digested with Msp I and Taq I. These results indicate that there are three cultural types in R. solani AG2-2, namely IIIB, IV and LP.  相似文献

Rhizoctonia -diseased specimens were collected from various host species growing in or near maize fields in different geographic regions of the Philippines. A greater range of host species, with varying types of disease symptoms, was found in Mindanao than in Luzon. Fifty-two isolates belonged to anastomosis group AG1-IA and caused banded leaf and sheath blight in maize ( Zea mays ), but they showed considerable variation in virulence. The most and least virulent isolates recovered from maize were both collected from Mindanao. Isolates from necrotic spots/foliar blight of durian and coffee, which were collected from the same region, showed the lowest lesion heights. UPGMA-SAHN clustering analysis from RAPD fingerprint data of 30 haplotypes of R. solani AG1-IA isolates from the Philippines and Japan resolved seven groups of AG1-IA at the 75% similarity level. Variation among isolates from upland crops seemed to be partially correlated with geographical origin and virulence. In the case of paddy rice isolates from Japan and the Philippines, some were closely related, with over 75% similarity, suggesting a common origin. In PCR-RFLP analysis of the rDNA internal transcribed spacer region, no polymorphism was observed among the AG1-IA isolates but they were differentiated from subgroups AG1-IB and AG1-IC using the endonucleases Eco RI, Mbo I and Hin fI.  相似文献

Increased atmospheric N deposition could suppress plant litter decomposition, due to the P limitation for soil microorganisms in Japanese forested Andisols with a high P sorption capacity. To explore this possibility, we used a laboratory incubation experiment to study the influence of N addition on β-d-glucosidase and polyphenol oxidase activities, which are important for cellulose and lignin degradation, respectively, in an Andisol with larch (Larix kaempferi) leaf litter. The addition of N increased the β-d-glucosidase activity, whereas it decreased the polyphenol oxidase activity in the soil. However, the addition of both N and P increased the polyphenol oxidase activity in the soil, suggesting the possibility of; (1) an inferior competitive ability of polyphenol oxidase-producing microorganisms under nutrient-rich conditions and; of (2) their P limitation through competition in the Andisol.  相似文献

We evaluated the diagnostic significance of the N-terminal pro-B-type natriuretic peptide (NT-proBNP) concentration in asymptomatic cats with cardiac enlargement. The plasma NT-proBNP concentration was measured in 21 clinically healthy control cats, and 67 asymptomatic cats with cardiac enlargement defined as end-diastolic interventricular septum thickness (IVSd) and/or diastolic left ventricular posterior wall thickness (LVPWd) >0.6 cm, vertebla heart scale (VHS) >7.8, and/or left atria/aorta ratio (LA/Ao) >1.5. The plasma NT-proBNP concentration in the asymptomatic cats with cardiac enlargement (median: 662.0, range: 24.0-2,449.0 pmol/l) was significantly higher than that in the controls (24.0, 24.0-95.0 pmol/l, P<0.001). The plasma NT-proBNP concentration was significantly correlated with the VHS, LA/Ao, IVSd and LVPWd (r=0.578, P<0.001; r=0.462, P<0.001; r=0.563, P<0.001; and r=0.764, P<0.001, respectively). Receiver operating characteristic analysis showed a cut-off value of 95.0 pmol/l for the detection of asymptomatic cats with cardiac enlargement, sensitivity and specificity of 88.1 and 100%, respectively, and an area under the curve of 0.971. These results suggest that the determination of the plasma NT-proBNP concentration can be a useful screening test for asymptomatic cats with cardiac enlargement.  相似文献

To establish clinical markers for canine necrotising meningoencephalitis (NME) and to elucidate its pathogenesis, glial fibrillary acidic protein (GFAP) and anti-GFAP autoantibodies were measured in the cerebrospinal fluid (CSF) of 32 dogs with NME, 23 dogs with other inflammatory central nervous system (CNS) diseases, 27 dogs with miscellaneous CNS diseases and 25 healthy dogs, including five pugs. The dogs with NME had the highest levels of anti-GFAP autoantibodies. The diagnostic sensitivity and specificity of anti-GFAP autoantibodies for NME were 91 per cent and 73 per cent, respectively. Some of the dogs with NME and the healthy pugs, had high CSF concentrations of GFAP, suggesting a breed-specific fragility of astrocytes. The leakage of GFAP and the development of autoimmunity may be key to understanding the pathogenesis of NME.  相似文献

Isolates of an unidentified Rhizoctonia sp. (UR isolates) were obtained from creeping bentgrass and Kentucky bluegrass with reddish brown sheath and foliar rots. Because the UR isolates anastomosed with isolates of three varieties of Waitea circinata (var. oryzae, var. zeae, and var. circinata), colony morphology, hyphal growth rate at different temperatures, pathogenicity, sequence analysis of the internal transcribed spacers (ITS) region of ribosomal RNA genes (rDNA) were compared. The colony color of mature UR isolates was distinct from isolates of the other three varieties of W. circinata. In pathogenicity tests on creeping bentgrass, the severity of the disease caused by UR isolates was significantly higher than that caused by the three varieties of W. circinata. Sequence similarities of the rDNA-ITS region between UR isolates and between isolates within each variety were high (97–100%), but they were lower among isolates from UR and the varieties of W. circinata (88–94%). In a phylogenetic tree based on the rDNA-ITS sequences, UR isolates formed a cluster separate from each of the clusters formed by the three varieties of W. circinata. These results indicate that the UR isolates clearly differ from the three varieties of W. circinata. We therefore propose that the UR isolates be classified as new Rhizoctonia sp. that are closely related to W. circinata and that the disease on creeping bentgrass should be called Waitea reddish-brown patch disease (Sekikasshoku-hagusare-byo in Japanese).  相似文献

Exotic animals can cause genetic contamination of indigenous species if they escape and reproduce in wild populations. The loach Misgurnus anguillicaudatus and species related to it have been imported into Japan for commercial use. We collected live loach specimens from the central wholesale market in Tokyo. For the 451 specimens collected, ploidy status was examined by DNA content flow cytometry, and polyploid loaches with triploid, tetraploid, and other higher ploidy ranges were detected. Hypertriploid and hypertetraploid individuals were easily detected by flow cytometry using standard eudiploid, eutriploid, and eutetraploid controls, and the reproductive capacities of these hyperpolyploid males were then examined. Sperm of hypertriploid males did not exhibit active progressive motility, and major populations of spermatozoa or spermatozoon-like cells were detected in the triploid and hexaploid ranges. Motile haploid spermatozoa were very rare in sperm from hypertriploid males. Therefore, hypertriploid males were sterile, whereas hypertetraploid males produced fertile hyperdiploid spermatozoa with active progressive motility after contact with ambient water. Viable progeny occurred in crosses between normal wild-type diploid females and hypertetraploid males, but androgenotes induced by the fertilization of UV-irradiated eggs with the sperm of hypertetraploid males were inviable hyperdiploids. Cytogenetic analyses of those androgenotes indicated that hypertetraploid males should produce hyperdiploid spermatozoa with 2n?=?54 (i.e., four supernumerary microchromosomes).  相似文献
