[目的]研究细胞分裂素对红掌组培苗繁殖速度及质量的影响。[方法]以继代培养第6代的愈伤组织为试验材料,探讨了ZT、TDZ、6-BA 3种细胞分裂素及其浓度对不定芽分化及变异的影响。[结果]在使用浓度2.0~8.0 mg/L的范围内,随着细胞分裂素浓度的增加,正常不定芽的分化数及叶片数呈下降趋势,而异常不定芽分化逐渐增多,且随着培养时间的延长,这种影响变得更加明显;3种细胞分裂素的培养效果具有显著差异,ZT和6-BA添加浓度分别在2.0和4.0 mg/L以下时,其正常不定芽分化较多,不良变异率较少,而TDZ在0.5 mg/L时已显著抑制了正常不定芽的分化,同时易产生异常不定芽。[结论]3种细胞分裂素作用的强弱依次为TDZZT6-BA;在红掌愈伤组织继代培养诱导不定芽过程中,应避免使用高浓度细胞分裂素。 相似文献

The pituitary is an important endocrine tissue of the vertebrate that produces and secretes many hormones. Accumulating data
suggest that several types of cells compose the pituitary, and there is growing interest in elucidating the origin of these cell
types and their roles in pituitary organogenesis. Therein, the histogenous cell line is an extremely valuable experimental tool
for investigating the function of derived tissue. In this study, we compared gene expression profiles by microarray analysis and
real-time PCR for murine pituitary tumor-derived non-hormone-producing cell lines TtT/GF, Tpit/F1 and Tpit/E. Several genes are
characteristically expressed in each cell line: Abcg2, Nestin, Prrx1,
Prrx2, CD34, Eng, Cspg4 (Ng2),
S100β and nNos in TtT/GF; Cxcl12, Raldh1,
Msx1 and Twist1 in Tpit/F1; and Cxadr, Sox9,
Cdh1, EpCAM and Krt8 in Tpit/E. Ultimately, we came to the following conclusions: TtT/GF cells
show the most differentiated state, and may have some properties of the pituitary vascular endothelial cell and/or pericyte.
Tpit/F1 cells show the epithelial and mesenchymal phenotypes with stemness still in a transiting state. Tpit/E cells have a
phenotype of epithelial cells and are the most immature cells in the progression of differentiation or in the initial
endothelial-mesenchymal transition (EMT). Thus, these three cell lines must be useful model cell lines for investigating pituitary

同性间动物体色的变异是一种普遍现象,在自然选择和性选择中起到重要的作用,并与其采用的繁殖策略密切相关。为了检验雄性胎生蜥蜴腹鳞颜色的分化机制和影响因素,于2012年5—6月,选取黑龙江省大兴安岭地区的一个胎生蜥蜴种群进行实验。结果表明:该胎生蜥蜴种群雄性个体的腹鳞主要分为橙红、桔黄和淡黄3种颜色,且在个体形态上(除了前后肢和腹鳞数外)存在明显的差异,推测腹鳞颜色的分化与年龄有关。温度和光照对腹鳞颜色具有间接的影响,在低温或无光条件下,3种颜色的雄性胎生蜥蜴的体重显著下降,腹鳞的颜色变暗。在此基础上,讨论了雄性胎生蜥蜴腹鳞颜色作为一种通讯信号所具有的潜在生态学意义。 相似文献

将美花石斛原球茎培养于含有不同生长调节物质的SH培养基上,结果发现,不同生长调节物质对美花石斛原球茎分化的不同阶段产生不同的影响.含有香蕉提取物和BAP2.0 mg/L+KT2.0 mg/L+NAA 1.0 mg/L的SH培养基对鲜重增加影响较显著;白萝卜汁对叶片分化有一定的促进作用;含有酵母提取物的SH培养基对增殖芽和不定根分化效果最好;马铃薯、香蕉和椰子汁对生根分化影响较大.建议在美花石斛原球茎不同分化阶段,根据其生长和分化需要,继代培养于相应的培养基上,以获得优质美花石斛组培苗. 相似文献

[目的]为运用现代生物技术开发利用重瓣铁线莲提供参考。[方法]以重瓣铁线莲的茎段为外植体,在MS基本培养基中添加0.2mg/L IAA,0.5、1.0、1.52、.0 mg/L的6-BA和0.1、0.2、0.4、0.6、0.81、.0 mg/L的2,4-D,观察重瓣铁线莲愈伤组织的诱导情况,统计诱导率。[结果]各处理诱导出的愈伤组织均为致密型愈伤组织,很少进行分裂分化,故在增殖培养中需提高植物生长物质的浓度。培养基MS+1.0 mg/L 6-BA+0.8 mg/L 2,4-D+0.2 mg/L IAA的诱导效果最好,不仅形成的愈伤组织量多,而且诱导率高达90.0%。在重瓣铁线莲愈伤组织诱导中,植物生长物质6-BA、2,4-DI、AA的适宜比例为5∶4∶1。[结论]重瓣铁线莲茎段愈伤组织适宜的诱导培养基为:MS+1.0mg/L 6-BA+0.8 mg/L 2,4-D+0.2 mg/L IAA。 相似文献

Allozyme variation was assessed in a collection of pearl millet cultivars derived from three different regions of traditional
cultivation of this cereal in Tunisia and West African countries. Using starch gel electrophoresis, 11 loci corresponding
to seven enzyme staining systems were identified. Important within-cultivar genetic diversity was detected. Genetic diversity
within Tunisian cultivars was similar and was more pronounced than for West African ones. This result could be explained by
the low level of the human selection characterizing the Tunisian cultivars. Most of the total genetic diversity is maintained
within subregions suggesting little genetic differentiation. Cluster analysis did not reveal separate clusters for the various
subregion cultivars. However the cluster analysis should help in the selection of interesting genotypes for future breeding

Summary Studies of intermediate wild-cultivated plants suggested that the Indica-Japonica differentiation of cultivars of Oryza sativa L. has taken place with the domestication of wild plants. The wild progenitor, which is considered to be the Asian form of O. perennis
Moench, shows no trace of differentiation into two such types as the Indica and Japonica. To elucidate the potentiality of the wild progenitor to evolve the two types when domesticated, the selfed progenies from an Indica × wild and a japonica × wild cross were examined with regard to three characters known to be useful for distinguishing between the two types. The data showed that Japonica-like plants were derived from the Indica × wild cross and Indica-like plants from the Japonica × wild cross. The Indica-Japonica differentiation was considered to have resulted from selection of incipient domesticates having such potentiality under different climatic conditions; the dynamics was discussed. 相似文献
