On the basis of worm counts in naturally infected Angora-X goats, albendazole at either 3.8mg/kg repeated after 24 hours or as a single dose at 7.6mg/kg, was >99% effective in removing adult Ostertagia and Trichostrongylus spp. Small numbers of adult Haemonchus contortus, Cooperia sp. and Chabertia ovina were present in control but not treated animals. The single dose of 7.6mg/kg removed > 99% of adult Oesophagostomum venulosum whereas the repeated dose of 3.8mg/kg removed only 96%.  相似文献

ObjectiveTo investigate the effect of plasma concentrations obtained by a low dose constant rate infusion (CRI) of racemic ketamine or S-ketamine on the nociceptive withdrawal reflex (NWR) in standing ponies.Study designProspective, blinded, cross-over study.AnimalsSix healthy 5-year-old Shetland ponies.MethodsPonies received either 0.6 mg kg−1 racemic ketamine (group RS) or 0.3 mg kg−1 S-ketamine (group S) intravenously (IV), followed by a CRI of 20 μg kg−1minute−1 racemic ketamine (group RS) or 10 μg kg−1minute−1 S-ketamine (group S) for 59 minutes. The NWR was evoked by transcutaneous electrical stimulation of a peripheral nerve before drug administration, 15 and 45 minutes after the start of the bolus injection and 15 minutes after the end of the CRI. Electromyographic responses were recorded and analysed. Arterial blood was collected before stimulation and plasma concentrations of ketamine and norketamine were measured enantioselectively using capillary electrophoresis. Ponies were video recorded and monitored to assess drug effects on behaviour, heart rate (HR), mean arterial blood pressure (MAP) and respiratory rate.ResultsThe NWR was significantly depressed in group RS at plasma concentrations between 20 and 25 ng mL−1 of each enantiomer. In group S, no significant NWR depression could be observed; plasma concentrations of S-ketamine (9–15 ng mL−1) were lower, compared to S-ketamine concentrations in group RS, although this difference was not statistically significant. Minor changes in behaviour, HR and MAP only occurred within the first 5–10 minutes after bolus drug administration in both groups.ConclusionAntinociceptive activity in standing ponies, demonstrated as a depression of the NWR, could only be detected after treatment with racemic ketamine. S-ketamine may have lacked this effect as a result of lower plasma concentrations, a more rapid metabolism or a lower potency of S-ketamine in Equidae so further investigation is necessary.  相似文献

ObjectiveTo evaluate the anesthetic and cardiorespiratory effects of two doses of intramuscular (IM) xylazine/ketamine in alpacas, and to determine if tolazoline would reduce the anesthetic recovery time.Study designProspective randomized crossover study.AnimalsSix castrated male alpacas.MethodsEach alpaca received a low dose (LD) (0.8 mg kg−1 xylazine and 8 mg kg−1 ketamine IM) and high dose (HD) (1.2 mg kg−1 xylazine and 12 mg kg−1 ketamine IM) with a minimum of one week between trials. Time to sedation, duration of lateral recumbency and analgesia, pulse rate, respiratory rate, hemoglobin oxygen saturation, arterial blood pressure, blood-gases, and the electrocardiogram were monitored and recorded during anesthesia. With each treatment three alpacas were randomly selected to receive tolazoline (2 mg kg−1 IM) after 30 minutes of lateral recumbency.ResultsOnset of sedation, lateral recumbency and analgesia was rapid with both treatments. The HD was able to provide ≥30 minutes of anesthesia in five of six alpacas. The LD provided ≥30 minutes of anesthesia in three of six alpacas. Respiratory depression and hypoxemia occurred with the HD treatment during the first 10 minutes of lateral recumbency: two animals were severely hypoxemic and received nasal oxygen for 5 minutes. Heart rate decreased, but there were no significant changes in arterial blood pressure. Tolazoline significantly shortened the duration of recumbency with the HD.ConclusionsThe HD provided more consistent clinical effects in alpacas than the LD. Intramuscular tolazoline shortened the duration of lateral recumbency in alpacas anesthetized with the HD combination.Clinical relevanceBoth doses of the combination were effective in providing restraint in alpacas and the duration of restraint was dose dependent. Supplemental oxygen should be available if using the HD and IM administration of tolazoline will shorten the recovery time.  相似文献

OBJECTIVE: To investigate alterations in peri-operative body temperatures and oesophageal-skin temperatures in isoflurane-anaesthetized rabbits following either ketamine-midazolam or ketamine-medetomidine induction of anaesthesia. ANIMAL POPULATION: Fifty client-owned rabbits, (25 male, 25 female) of different breeds anaesthetized for elective neutering (age range: 3-42 months; mass range: 1.15-4.3 kg). STUDY DESIGN: Randomized, blinded clinical study. METHODS: Pre-anaesthetic rectal temperature was measured. A 24 SWG catheter was placed in a marginal ear vein after local anaesthesia. Ketamine (15 mg kg(-1)) with medetomidine (0.25 mg kg(-1)) (group KMT) or with midazolam (3 mg kg(-1)) (group KMZ) was injected intramuscularly (IM). Following endotracheal intubation anaesthesia was maintained with isoflurane in oxygen. Carprofen (3 mg kg(-1)) and glucose saline (5 mL kg(-1) hour(-1)) were administered through the intravenous catheter. Room temperature and humidity, skin temperature (from tip of pinna) and oesophageal temperature were measured during anaesthesia. Ovariohysterectomy or castration was performed. Rectal temperature was taken when isoflurane was discontinued (time zero) and 30, 60 and 120 minutes thereafter. Atipamezole (0.5 mg kg(-1)) was administered IM to rabbits in group KMT at zero plus 30 minutes. Mass, averaged room temperature and duration of anaesthesia data were compared using a two-tailed t-test. Age, averaged room humidity, rectal temperature decrease, oesophageal temperature decrease and oesophageal-skin difference data were compared using a Kruskal-Wallis test. p < 0.05 was considered significant. RESULTS: The averaged oesophageal-skin temperature difference was significantly greater in group KMT [median 9.85 degrees C (range 6.42-13.85 degrees C)] than in group KMZ [4.38 degrees C (2.83-10.43 degrees C)]. Rectal temperature decreased over the anaesthetic period was not significantly different between the two groups; however, oesophageal temperature decrease was significantly less in group KMT [1.1 degrees C (-0.1-+2.7 degrees C)] than in group KMZ [1.4 degrees C (0.6-3.1 degrees C)]. CONCLUSIONS: Oesophageal-skin temperature difference is larger in rabbits anaesthetized with ketamine-medetomidine combination than ketamine-midazolam. CLINICAL RELEVANCE: The oesophageal temperature in rabbits anaesthetized with ketamine-medetomidine and isoflurane decreases significantly less than in animals anaesthetized with ketamine-midazolam and isoflurane, during anaesthesia.  相似文献

外源性H2S在诸多方面对中枢神经系统具有保护性作用,本研究通过预试验确定了外源性H2S的给药时间和剂量,将48只SD大鼠随机均分为4组,分别为对照组(CP组)、低剂量组(LP组,7 μmol/kg NaHS)、中剂量组(MP组,14 μmol/kg NaHS)、高剂量组(GP组,28 μmol/kg NaHS)。在盐酸氯胺酮麻醉前20 min,3个剂量组分别腹腔注射7、14、28 μmol/kg NaHS,观察外源性H2S对大鼠体温、心率、呼吸频率、血氧饱和度及血压等生理指标的干预效果。试验结果表明,与对照组相比,3个剂量组体温和血氧饱和度浓度均显著升高(P<0.05),心率和血压均显著降低(P<0.05),但呼吸频率呈先减慢后加快趋势,且这些变化呈显著的剂量依赖性。  相似文献

ObjectiveTo evaluate the influence of premedication with tramadol on xylazine–ketamine anaesthesia in young pigs.Study designProspective, randomized, blinded cross-over study.AnimalsTen young Niger hybrid pigs: mean weight 6.1 ± 0.6 kg.MethodsPigs were anaesthetized twice. Xylazine (2.5 mg kg?1), ketamine (25 mg kg?1) and atropine (0.04 mg kg?1) were administered by intramuscular (IM) injection, 5 minutes after either tramadol (5 mg kg?1)) (treatment XKT) or saline (treatment XKS). Time to loss of righting reflex (TLRR), duration of antinociception, duration of recumbency (DR) and recovery times (RCT) were recorded. Quality of induction of anaesthesia including ease of endotracheal intubation was assessed using a subjective ordinal rating score of 1 (worst) to 4 (best). Heart, pulse and respiratory rates, arterial oxygen saturations and rectal temperatures were determined over 60 minutes. Antinociception was assessed by the pigs’ response to artery forceps applied at the interdigital space. Data were compared with Student's t-test, Mann–Whitney's test or analysis of variance (anova) for repeated measures as appropriate and are presented as mean ± standard deviation.ResultsThe quality of anaesthetic induction was significantly better and duration of antinociception significantly longer (p < 0.05) in treatment XKT (3.1 ± 0.7 score; 43.7 ± 15.5 minutes) than in treatment XKS (2.8 ± 0.6 score; 32.0 ± 13.3 minutes). TLRR, DR and RCT did not differ significantly (p > 0.05) between treatment XKT (2.1 ± 0.8, 65.8 ± 17.0 and 13.2 ± 6.7 minutes) and treatment XKS (2.1 ± 1.3, 58.0 ± 14.8 and 10.3 ± 5.6 minutes). Physiological measurements did not differ between the treatments.Conclusion and clinical relevanceTramadol improved the quality of anaesthetic induction and increased the duration of antinociception in xylazine–ketamine anaesthetized young pigs without increasing duration of anaesthesia, nor causing additional depression of the physiological parameters measured.  相似文献

ObjectiveTo study the anaesthetic and cardiorespiratory effects of intramuscular (IM) administration of different combinations of romifidine and ketamine in cats.Study designProspective, randomized, cross-over experiment.AnimalsSeven healthy adult cats weighing (mean ± SD) 3.4 ± 0.7 kg and aged 4.6 ± 3.2 years.MethodsAnimals received romifidine 100 μg kg?1 with ketamine 7.5 (R100/K7.5) and 10 mg kg?1 (R100/K10), romifidine 200 μg kg?1 with ketamine 5 (R200/K5), 7.5 (R200/K7.5) and 10 mg kg?1 (R200/K10) by IM injection. The time required to perform orotracheal intubation (IT) was measured and the ease of intubation assessed. The onset of anaesthesia (OA), duration of anaesthesia (DA) and anaesthesia recovery times (AR) were measured. Analgesia and muscle relaxation scores were recorded every 5 minutes for 60 minutes after OA. Heart rate, systolic arterial pressure, arterial haemoglobin saturation, respiratory rate, end-tidal carbon dioxide and oesophageal temperature were also measured.ResultsThe IT, OA and DA were not significantly different between the treatments. The analgesia and muscle relaxation scores were similar between all treatments at most time points. The cardiorespiratory variables were not significantly different between the treatments in most cases. The adverse effects were dose dependent and similar to those previously described for other combinations of α2-agonists and ketamine.Conclusions and clinical relevanceAnaesthesia produced by the studied combinations of romifidine and ketamine may only be reliable when conducting brief and noninvasive procedures in cats. The OA times were slower and the DA shorter than those reported for other alpha-2 agonists combined with ketamine. A dose-related increase in the intensity of the anaesthetic effects could not be demonstrated in this study.  相似文献

This study quantitatively investigated the analgesic action of a low-dose constant-rate-infusion (CRI) of racemic ketamine (as a 0.5 mg kg−1 bolus and at a dose rate of 10 μg kg−1 min−1) in conscious dogs using a nociceptive withdrawal reflex (NWR) and with enantioselective measurement of plasma levels of ketamine and norketamine. Withdrawal reflexes evoked by transcutaneous single and repeated electrical stimulation (10 pulses, 5 Hz) of the digital plantar nerve were recorded from the biceps femoris muscle using surface electromyography.Ketamine did not affect NWR thresholds or the recruitment curves after a single nociceptive stimulation. Temporal summation (as evaluated by repeated stimuli) and the evoked behavioural response scores were however reduced compared to baseline demonstrating the antinociceptive activity of ketamine correlated with the peak plasma concentrations. Thereafter the plasma levels at pseudo-steady-state did not modulate temporal summation. Based on these experimental findings low-dose ketamine CRI cannot be recommended for use as a sole analgesic in the dog.  相似文献

ObjectiveTo compare racemic ketamine and S-ketamine as induction agents prior to isoflurane anaesthesia.Study designProspective, blinded, randomized experimental study.AnimalsThirty-one healthy adult goats weighing 39-86 kg.MethodsGoats were premedicated with xylazine (0.1 mg kg?1) intravenously (IV) given over 5 minutes. Each goat was assigned randomly to one of two treatments for IV anaesthetic induction: group RK (15 goats) racemic ketamine (3 mg kg?1) and group SK (16 goats) S-ketamine (1.5 mg kg?1). Time from end-injection to recumbency was measured and quality of anaesthetic induction and condition for endotracheal intubation were scored. Anaesthesia was maintained with isoflurane in oxygen for 90 minutes. Heart rate, invasive arterial blood pressure, oxygen saturation, temperature, end-tidal carbon dioxide and isoflurane were recorded every 5 minutes. Arterial blood samples were taken for analysis every 30 minutes. Recovery time to recurrence of swallowing reflex, to first head movement and to standing were recorded and recovery quality was scored. Two-way repeated measures anova, Mann-Whitney and a Mantel-Cox tests were used for statistical analysis as relevant with a significance level set at p < 0.05.ResultsInduction of anaesthesia was smooth and uneventful in all goats. There was no statistical difference between groups in any measured parameter. Side effects following anaesthetic induction included slight head or limb twitching, moving forward and backward, salivation and nystagmus but were minimal. Endotracheal intubation was achieved in all goats at first or second attempt. Recovery was uneventful on all occasions. All goats were quiet and needed only one or two attempts to stand.Conclusions and clinical relevanceS-ketamine at half the dose rate of racemic ketamine in goats sedated with xylazine and thereafter anaesthetised with isoflurane induces the same clinically measurable effects.  相似文献
