A long-term study was carried out in 11 dairy herds in the Khon Kaen province of northeast Thailand between August 2001 and November 2004. The objective was to investigate seroprevalence dynamics of Neospora caninum infection in the herds and to demonstrate patterns of seroconversion in individual cattle. Each herd was visited once a year, in total four times, and sera from cattle > 3 months of age and farm dogs as well as a sample from the bulk milk were collected. All samples were analysed for presence of specific antibodies by an N. caninum iscom ELISA. The overall percentage of antibody-positive cattle was constant and varied only between 10 and 13% over the 4 years, but the variation in within-herd seroprevalence between herds was substantial. Two herds had > or = 20% seropositive animals at all samplings and consistently high bulk milk OD, whereas two herds had no seropositive animal at the last two samplings and low bulk milk OD. Five herds had a decreasing trend of within-herd seroprevalence, whereas the remaining six herds had a higher portion of test-positive individuals at the end of the study. A total of 424 individuals were sampled more than once; 344 (81%) and 32 (8%) were consistently antibody-negative and antibody-positive, respectively. The proportions of animals that changed from being seronegative to seropositive and from being seropositive to seronegative between the years were 3.9-4.6% and 19-39%, respectively. Apparent vertical and horizontal transmission rates were 58% (95% CI; 44-71%) and 5% (95% CI; 3-7%), respectively. In conclusion, the overall percentage of N. caninum antibody-positive cattle was constant over the years, but the within-herd seroprevalence varied substantially between the herds. Stormwater bioretention systems are widely used to treat diffuse infiltration of runoff from paved surfaces and roofs. Substantial questions remain about the hydraulic performance and the accumulation of pollutants in systems over the long term. Data of metal accumulation of systems with operational times >10 years currently is limited. This study deals with the accumulation of metals in a variety of long-term operational bioretention systems (11–22 years) to derive further operation recommendations for the water authorities.
Materials and methods
The hydraulic conductivity of the bioretention systems in field was measured using a double ring infiltrometer. Media soil samples from 22 diverse designed systems were collected across the surface and at intervals up to a depth of 65 cm to determine the spatial accumulation of Zn, Cu, Pb and Cd. Leaching experiments of selected bioretention media soils were derived to assess the metal leachability by water.
Results and discussion
The hydraulic performance of most bioretention systems still met the technical guidelines of Germany even after long-term operation. Considerable metal accumulation occurred in the topsoil (0–20 cm). Median concentrations of all metals are highest at the soil surface (0–10 cm), decreasing with increasing depth. High concentrations were determined at the inflow points of the runoff waters, whereas concentrations at more than 1.5 m distance from the inflow were only slightly increased compared to the initial soil concentrations. Leachability tests have shown that most of the metals deposited in bioretention soils are only slightly water soluble. No concentrations exceeding the threshold values of the German Soil Contamination Ordinance for the pathway soil to groundwater could be determined.
Conclusions
The hydraulic conductivity of the bioretention systems is given even well after long-term operation. Most of the metal accumulation is concentrated in the top 20 cm; concentrations decrease rapidly and mostly reach background/initial concentrations after depths of 30 cm. The water-soluble metals are all below the trigger values of the German Soil Act. This underlines the strong retention capacity of long-term bioretention systems after long-term operational times.
Pancreas disease (PD) caused by salmonid alphavirus (SAV) severely affects salmonid aquaculture during the seawater phase. To characterize immune cells in target tissues for SAV infection, heart, pancreas and pyloric caeca were analysed from two groups of fish adapted to seawater for 2 and 9 weeks. The sections were scored for the relative abundance of cells expressing MHC class II, IgM, CD3, CD8 or neutrophil/granulocyte markers using immuno‐histochemical techniques. In general, necrosis of tissue was more severe in fish infected at 2 weeks post‐seawater transfer (wpt) compared with those infected at 9 wpt. At 9 wpt, there were higher numbers of MHC II+ cells in heart, pancreas and pyloric caeca, IgM+ cells in heart and pancreas, and CD3+ cells in pancreas compared to those infected at 2 wpt. The majority of the immune cells infiltrating PD‐affected tissues were MHC II+ and CD3+ cells suggesting that antigen‐presenting cells and T lymphocytes are the main types of immune cells responding to SAV infection. All the investigated cell types were also observed in pyloric caeca of infected fish, suggesting that this tissue may play a role in the immune response to SAV. 相似文献
Predicting growth is critical in aquaculture, but models of growth are largely missing for mud crab species. Here, we present the first model of natural growth in juvenile and adult mud crabs Scylla serrata from East Africa using a stepwise growth function based on data on intermoult periods and growth at moult from field mark‐recapture, pond and laboratory studies. The results showed a sigmoid growth pattern in carapace width and suggest that S. serrata in East Africa will reach 300 g and sexual maturity ~9.9 months after settlement, and a commercial size of 500 g after 12.4 months. Analyses of the literature identified several issues with the common praxis to compare standard growth measures between aquaculture studies with different initial size or growing periods. Using the new growth function to estimate the proportional difference between modelled and obtained growth as an alternative method, we show that growth rates of S. serrata cultured in cage systems, which are dominant in East Africa, was <40% of the estimated natural growth and growth obtained in pond systems. The analysis also indicated that growth rates of S. serrata in Southeast Asia was over 50% higher compared with similar culture systems in East Africa, and that different species of mud crabs had large differences in growth rates. This study shows that growth in the present mud crab aquaculture systems in East Africa is below their expected potential. Further work is needed to identify the factors behind this observation. 相似文献
Tomato fruits can be contaminated by saprophytic strains of Alternaria alternata which is the reason for the frequent occurrence of Alternaria toxins like alternariol, alternariol monomethylether or tenuazonic acid in these types of products. It was shown earlier that alternariol is a colonization factor for tomatoes. In the current analysis two different tomato genotypes were analysed by untargeted comprehensive two-dimensional gas chromatography mass spectrometry (GC×GC-MS). This analysis revealed clear differences in the metabolic profiles which were paralleled by differences in resistance towards Alternaria colonization. One of the genotypes was more resistant against A. alternata infection and contained high amounts of chlorogenic acid in contrast to the other genotype which was sensitive against infection. In in vitro analysis, chlorogenic acid reduced alternariol biosynthesis during the first days of growth of A. alternata. Expression analysis of the alternariol polyketide synthase gene, a key gene in the biosynthesis of alternariol, also revealed a temporal reduction in its expression in the first phases of growth. However by chromatographic analysis it could be demonstrated that chlorogenic acid was degraded over time. This degradation leads to a relief of inhibition resulting in an only temporal inhibition of alternariol biosynthesis. In vivo colonization experiments revealed that chlorogenic acid reduces colonization of tomatoes by A. alternata in a concentration dependent manner, which however is partly counteracted by the addition of alterariol. 相似文献

FT-IR microspectroscopy and light microscopy were used to investigate pork muscle musculus semitendinosus samples, taken from three animals, that were subjected to brine salting at different concentrations (0.9, 3, 6, and 9% NaCl). Differences in spectral characteristics and in microstructure were observed in meat from animals differing in initial pH and varying salt concentrations. The FT-IR data displayed changes in the amide I region from 1700 to 1600 cm(-1). This spectral range was analyzed by principal component analysis (PCA) and partial least-squares regression (PLSR). These methods revealed correlations between the spectral data and the different animals, salt content, moisture content, pH value, and myofiber diameter. A shrinking share of alpha-helical components was related to an increase in salt concentration in the muscle. At the same time, a greater share in nonhydrogenated C=O groups (1668 cm(-1)) was related to an increase in salt concentration in the meat. The share of aggregated beta-strands differed with respect to the different animals but was not influenced by salt concentration. The meat at higher pHs (>6) had less aggregated beta-strands than that at lower pHs (5.6-5.7). It could be demonstrated that simultaneous with changes in microstructure, pH value, salt, and moisture content were alterations in the protein amide I region as measured by FT-IR microspectroscopy, revealing a relationship between these biophysical and chemical parameters and secondary protein structure attributes. 相似文献
We used a prospective seroepidemiological approach to investigate endemic abortion in a dairy herd in Arequipa, Peru, and its association with Neospora caninum and bovine viral-diarrhoea virus (BVDV) infections. Between January 2002 and March 2004, 1094 pregnancies were confirmed in 538 cows. Of these, 137 pregnancies (13%) in 121 cows ended in abortion. The serological status to N. caninum was assessed using a single serological screening, whereas BVDV status was assessed at the herd level through consecutive samplings of young stock.
Cox proportional-hazards models were used to estimate the effect of N. caninum and BVDV on the hazard of early (between day 42 and day 100 in gestation), and late (after day 100) abortions, respectively. Serological status to N. caninum was included as a dichotomous variable, and the effect of BVDV estimated at the herd level, as a time-dependent seasonal effect. Because data from repeated pregnancies were included, we considered possible lack of independence between observations and included frailty effects into the models. Our models also considered the possible confounding by parity and animal origin.
Only multiparity was associated with the hazard of early abortion (HR = 2.8 compared to nulliparous heifers). N. caninum seropositivity significantly affected the hazard of late abortion, but interacted with parity. The HRs for Neospora-positive animals were 6.4, 3.7 and 1.9, respectively, for nulliparous heifers, first-lactation cows and multiparous cows. Evidence of BVDV circulating (or not) among the young stock was not associated with abortions, but few cows in this herd were susceptible to incident infection. 相似文献
A study was undertaken to determine the prevalences of Cryptosporidium and Giardia in dairy calves less than 6 months of age in Norway. Faecal samples were collected from a total of 1386 calves, between 3 and 183 days of age, in 136 dairy farms from three different areas of Norway. Faecal samples were examined for Cryptosporidium oocysts and Giardia cysts after concentration and immunofluorescent staining.
Giardia was found in 93% (127 out of 136) of the farms and in 49% (679 out of 1386) of the calves. Cryptosporidium was found in 53% (72 out of 136) of the farms and in 12% (167 out of 1386) of the calves. The level of Giardia and/or Cryptosporidium was low in the majority of the infected calves.
Infection peaked in the age group 2–3 months for both Cryptosporidium and Giardia. The prevalences of both parasites were higher in samples taken during winter than in samples taken during summer, and statistically significant differences were found when prevalences in different age groups of calves were compared between the three areas. A significantly lower prevalence of Cryptosporidium was found in calves housed in shared pens that were thoroughly washed more than three times a year than in calves from pens washed less often. For Giardia there was a trend for decreasing intensity of infection with increasing age in the sampled calves. For Cryptosporidium there was a trend for increasing herd prevalence with increasing number of calves in the herd, but this trend was not statistically significant.
