We characterized the adventitious rooting response of Eucalyptus globulus Labill. to various concentrations of calcium, nitrogen, phosphorus, iron, manganese, zinc, boron and copper. The parameters analyzed were percent rooting, root number, root length and mean rooting time. Root number and root length were significantly affected by mineral nutrition, whereas mean rooting time and rooting percentage seemed to be closely related to auxin availability. Root number was affected by calcium, nitrogen source and zinc, whereas root length was influenced by concentrations of phosphorus, iron and manganese, and by nitrogen source. Based on these results, we evaluated various combinations of several concentrations of these minerals in each rooting phase. Cuttings that were rooted in an optimized mineral nutrient medium and acclimatized to ex-vitro conditions for two months showed significantly higher survival after transplanting and drought stress than cuttings rooted in basal medium and treated in the same way.  相似文献

Psychotria umbellata Vell. (Rubiaceae), a Brazilian coastal woody species, produces umbellatine (also known as psychollatine), an analgesic indole alkaloid. An in vitro embryogenic regeneration protocol capable of yielding alkaloid-accumulating plants was developed. Rhizogenic calli, which were obtained from stem segments derived from rooted apical cuttings, were cultured on Murashige and Skoog's (MS) medium containing either 1 mg l(-1) NAA (naphthalene acetic acid) and no kinetin, or 5 mg l(-1) NAA + 1 mg l(-1) kinetin. Calli did not accumulate umbellatine. Segments of rhizogenic callus were cultured on complete MS medium with various concentrations of kinetin and sucrose. Plant regeneration was best in the light with 0.25 mg l(-1) of kinetin and 1.5% sucrose. After 3 months of acclimatization in soil mixture, plant survival was 81%. Leaves of 10-month-old regenerated plants yielded umbellatine concentrations equivalent to those of adult forest-grown plants.  相似文献

Adventitious rooting is essential for vegetative propagation of woody species. We studied the effects of auxin and light on the development of adventitious roots in cuttings obtained from seedlings of Eucalyptus saligna Smith and E. globulus Labill in an attempt to characterize the adventitious rooting process and identify factors controlling rhizogenesis. Root development was scored as rooting percentage, root density (roots per rooted cutting), mean rooting time and root length. In both species, rooting time was reduced in the presence of auxin. Cuttings from 2-month-old E. saligna seedlings were responsive to lower auxin concentrations than comparable cuttings from E. globulus seedlings. Cuttings from 3-month-old E. saligna seedlings rooted promptly and rooting was not significantly affected by light conditions. In contrast, rooting of cuttings from 3-month-old E. globulus seedlings exhibited recalcitrant behavior and no roots were formed if illuminated during the root formation phase. Effective root regeneration of E. globulus cuttings was obtained by a 4-day exposure to 10 mg l(-1) IBA and culture in darkness during the root formation step. Loss of rooting capacity with seedling age was more pronounced in E. globulus than in E. saligna. The possibility of switching adventitious rooting off and on by manipulating light regime and exogenous auxin supply in E. globulus, and the constitutive nature of rooting in E. saligna may provide useful models for examining the rooting process at the biochemical and molecular levels in Eucalyptus.  相似文献

The extraction of induced oleoresin from Pinus elliottii through bark streaking supplies products to the food, pharmaceutical and chemical industries. Resin flow is a typical defense response in conifers and is controled by various environmental factors. Although many studies have focused on oleoresin production in temperate forests, particularly in response to bark beetle attacks, relatively little is known about the physiological regulation of sustainable resin exudation in subtropical planted forests, in which commercial exploitation of this product is favored by the absence of significant pest insect populations. Therefore, the annual oleoresin yield of approximately 2660, 28-year-old trees from different sites was monitored for two consecutive years in southern Brazil. Plain wounding and wounding followed by application of a commercial resin stimulant paste containing the ethylene precursor 2-chloroethylphosphonic acid (CEPA) were used as reference treatments. Different concentrations of auxin, paraquat, yeast extract, and salicylic acid (isolated or in combination with CEPA) were also tested. Higher resin yields were associated with spring and summer, whereas winter had lower but significant yields. Auxin and salicylic acid stimulated the production of oleoresin relative to commercial paste in at least one concentration. Replacement of CEPA with auxin or salicylic acid yielded equivalent oleoresin production, indicating a comparable or complementary effect of these molecules in the process, as well as having implications for new stimulant paste formulations.  相似文献

Eucalyptus globulus Labill and hybrids thereof have low lignin content, favoring cellulose extraction, but are often recalcitrant to clonal propagation. This work analyzed biochemical and morphological changes during adventitious rooting of mini-cuttings of E. globulus × maidenni obtained from mini-stumps cultured in drip fertigated sand bed or intermittent flooding tray commercial propagation systems. Morphological (% rooting, root number and length, mean rooting time) and biochemical parameters (peroxidase activity, total phenolic content and flavonoid content) were monitored to characterize the rooting phases. All of the rooting parameters were equivalent in both systems, indicating comparable efficiency of both methods in clonal propagation. Kinetic profiles of biochemical parameters were also similar, although the activity of peroxidases was an order of magnitude higher and the phenolic content about three times lower in cuttings derived from intermittent flooding-grown mini-stumps than in those derived from sand bed-grown mini-stumps. Taken together, results suggest that rooting phases were similar in both systems: induction before day 5, formation from day 5 to 15, and elongation from day 15 to 45. These data may contribute to the development of rooting phase-specific mineral nutrient solutions to maximize clonal propagation and plant survival.  相似文献

The production of oleoresin from Pinus trees is a relevant economic activity in developing countries, since it provides terpenes used in the chemical industry. The oleoresin production by trees of Pinus elliottii in southern Brazil was investigated. Trees of approximately 28 years of age were individually analyzed for oleoresin yield using the bark streak method of wounding for resin tapping. Plain wounding and wounding followed by application of commercial resin stimulant paste, composed of an ethylene precursor (CEPA) and sulphuric acid, were used as reference treatments. The optimum concentrations of CEPA and sulphuric acid in the stimulant paste were investigated in relation to oleoresin yield, as well as the shape of wound and tree diameter. The replacement of CEPA with auxin in the resin stimulant paste was also evaluated. The results showed that both horizontal and V-shaped streaks are equivalent for oleoresin yield. Trees with larger diameter at breast height, defined as the trunk diameter 1.4 m above the ground (22–23.5 cm) yielded more oleoresin than their smaller counterparts (18–19.5 cm). Oleoresin yield was equivalent with stimulant pastes containing between 1.5 and 4.5% CEPA. Trees exposed to stimulant pastes containing from 3 to 4.5% CEPA had average year oleoresin yield above 5 kg. The use of 20% sulphuric acid in the stimulant paste resulted in higher oleoresin yields. Auxin (2,4-D at 1 mM) was effective in replacing CEPA in the stimulant paste, particularly in smaller DBH trees or after 1 year of application in larger DBH individuals. In conclusion, cost reductions on resin tapping can be achieved by adjusting stimulant paste components and managing subsets of trees in specific fashion based on DBH ranges.  相似文献
