In general, avian influenza (AI) vaccines protect chickens from morbidity and mortality and reduce, but do not completely prevent, replication of wild AI viruses in the respiratory and intestinal tracts of vaccinated chickens. Therefore, surveillance programs based on serological testing must be developed to differentiate vaccinated flocks infected with wild strains of AI virus from noninfected vaccinated flocks in order to evaluate the success of vaccination in a control program and allow continuation of national and international commerce of poultry and poultry products. In this study, chickens were immunized with a commercial recombinant fowlpox virus vaccine containing an H5 hemagglutinin gene from A/turkey/Ireland/83 (H5N8) avian influenza (AI) virus (rFP-H5) and evaluated for correlation of immunological response by hemagglutination inhibition (HI) or agar gel immunodiffusion (AGID) tests and determination of protection following challenge with a high pathogenicity AI (HPAI) virus. In two different trials, chickens immunized with the rFP-H5 vaccine did not develop AGID antibodies because the vaccine lacks AI nucleoprotein and matrix genes, but 0%-100% had HI antibodies, depending on the AI virus strain used in the HI test, the HI antigen inactivation procedure, and whether the birds had been preimmunized against fowlpox virus. The most consistent and highest HI titers were observed when using A/turkey/Ireland/83 (H5N8) HPAI virus strain as the beta-propiolactone (BPL)-inactivated HI test antigen, which matched the hemagglutinin gene insert in the rFP-H5 vaccine. In addition, higher HI titers were observed if ether or a combination of ether and BPL-inactivated virus was used in place of the BPL-inactivated virus. The rFP-H5 vaccinated chickens survived HPAI challenge and antibodies were detected by both AGID and HI tests. In conclusion, we demonstrated that the rFP-H5 vaccine allowed easy serological differentiation of infected from noninfected birds in vaccinated populations of chickens when using standard AGID and HI tests.  相似文献

Irregular plagues of house mice, Mus musculus, incur major economic impacts on agricultural production in Australia. The efficacy of zinc phosphide (ZnP), the only registered broadacre control agent for mice, is reported as increasingly variable. Have mice become less sensitive over time or are they taking a sub-lethal dose and developing aversion? In this laboratory study, the sensitivity of mice (wild caught; outbred laboratory strain) was assessed using oral gavage of a range of ZnP concentrations. The estimated LD50 values (72–79 mg ZnP/kg body weight) were similar for each mouse group but are significantly higher than previously reported. The willingness of mice to consume ZnP-coated grains was determined. ZnP-coated grains (50 g ZnP/kg grain) presented in the absence of alternative food were consumed and 94% of wild mice died. Mice provided with alternative food and ZnP-coated wheat grains (either 25 or 50 g ZnP/kg grain) consumed toxic and non-toxic grains, and mortality was lower (33–55%). If a sublethal amount of ZnP-coated grain was consumed, aversion occurred, mostly when alternative food was present. The sensitivity of wild house mice to ZnP in Australia is significantly lower than previously assumed. Under laboratory conditions, ZnP-coated grains coated with a new higher dose (50 g ZnP/kg grain) were readily consumed. Consumption of toxic grain occurred when alternative food was available but was decreased. Our unambiguous findings for house mice indicate a re-assessment of the ZnP loading for baits used for control of many rodents around the world may be warranted.  相似文献

Opencast coalmine effluent contains higher concentrations of Cd, Cr, Co, Ni and Pb. Biofiltration of these metals has been demonstrated successfully with the help of aquatic macrophytes i.e., E. crassipes, L. minor and A. pinnata. Experiments revealed E. crassipes reduced highest concentration of heavy metals followed by L. minor and A. pinnata on 20th days retention period. Plant tissue analysis revealed higher accumulation of metals in roots than leaves. Highly significant correlations have been noted between removal of heavy metals in effluent and their accumulation in roots and leaves of the experimental sets. Translocation factor also revealed lower transportation of metals from root to leaves. Reduction in chlorophyll and protein content was noted with the accumulation of heavy metals. N, P and K analysis in plant tissues indicated continuous decrease in their concentration with increasing metal concentration. Negative and significant correlations between metal accumulation and N, P and K concentrations in plant tissues showed adverse effects of heavy metals. Surface runoff, erosion, compaction and the leaching of potential pollutants from land can degrade the soil resource and damage the water environment, reducing crop yields, causing loss of valuable nutrients and organic matter, together with increasing flood risk. Increasingly, it is recognized that scientific information must be translated into practical tools to change practices and protect the soil and water resource. Working alongside agencies in Scotland, we applied a suite of simple, transparent, rule-based models to identify areas most at risk of exporting sediment and pollutants that may degrade water quality based on field-scale (1:25,000) soil maps. The maps have been used by Scottish Environment Protection Agency, Scottish Water and Scottish Government to assess soil risks to waters from field to regional scale. The work brought scientists together with policy makers, agencies and the water industry to pool their knowledge to apply these practical tools for decision-making. It highlights the need to apply existing knowledge to answer salient questions. All three examples described show that providing the right type of information, which is based on fundamental soils data, can directly influence the implementation of policies, investment and monitoring decisions and provide evidence in support of government. However, this requires both researchers and agency scientists to develop skills as knowledge brokers and to normalize the use of soil data in everyday agency settings.  相似文献

The change in soil carbon (C) concentration, soil pH and major nutrients for approximately 1,000 topsoil sampled from on-farm experimental sites over a thirty-year period from 1950 to 1980 in north-east Scotland are summarized. This period coincided with increased agricultural intensification, which included regular liming and fertilizer additions. During 2017, 37 of these sites were resampled and reanlaysed. While pH and extractable phosphorus (P) and potassium (K) increased over this period, there was no detectable change in the percentage loss on ignition. Composite soil samples were taken by auger from a depth of 0–15 cm and compared with the corresponding archived samples collected at the initiation of each experiment. Analysis of these resampled soils indicated no significant change in soil carbon (C), although soil pH, extractable magnesium (Mg) and K and Nitrogen (N) concentrations were significantly greater (p < .001) but extractable soil P concentration was significantly less (p = .015) compared with the original samples. Even though measuring C concentration alone is a poor indicator of overall changes in soil C stocks, it does provide a relative quick “early warning” of C losses that would justify a more comprehensive measure of stocks.  相似文献
