A male Japanese domestic cat with retarded growth in Hokkaido, Japan, showed
progressive motor dysfunction, such as ataxia starting at 3 months of age and tremors,
visual disorder and seizure after 4 months of age. Finally, the cat died of neurological
deterioration at 9 months of age. Approximately half of the peripheral blood lymphocytes
had multiple abnormal vacuoles. Magnetic resonance imaging showed bisymmetrical
hyperintensity in the white matter of the parietal and occipital lobes in the forebrain on
T2-weighted and fluid-attenuated inversion recovery images, and mild encephalatrophy of
the olfactory bulbs and temporal lobes. The activity of lysosomal acid β-galactosidase in
leukocytes was negligible, resulting in the biochemical diagnosis of GM1 gangliosidosis.
Histologically, swollen neurons characterized by accumulation of pale, slightly granular
cytoplasmic materials were observed throughout the central nervous system. Dysmyelination
or demyelination and gemistocytic astrocytosis were observed in the white matter.
Ultrastructually, membranous cytoplasmic bodies were detected in the lysosomes of neurons.
However, genetic analysis did not identify the c.1448G>C mutation, which is the single
known mutation of feline GM1 gangliosidosis, suggesting that the cat was affected with a
new variant of the feline disease. 相似文献

The pharmacological effects of the anesthetic alfaxalone were evaluated after
intramuscular (IM) administration to 6 healthy beagle dogs. The dogs received three IM
doses each of alfaxalone at increasing dose rates of 5 mg/kg (IM5), 7.5 mg/kg (IM7.5) and
10 mg/kg (IM10) every other day. Anesthetic effect was subjectively
evaluated by using an ordinal scoring system to determine the degree of neuro-depression
and the quality of anesthetic induction and recovery from anesthesia. Cardiorespiratory
variables were measured using noninvasive methods. Alfaxalone administered IM produced
dose-dependent neuro-depression and lateral recumbency (i.e., 36 ± 28 min, 87 ± 26 min and
115 ± 29 min after the IM5, IM7.5 and IM10 treatments, respectively). The endotracheal
tube was tolerated in all dogs for 46 ± 20 and 58 ± 21 min after the IM7.5 and IM10
treatments, respectively. It was not possible to place endotracheal tubes in 5 of the
6 dogs after the IM5 treatment. Most cardiorespiratory variables remained
within clinically acceptable ranges, but hypoxemia was observed by pulse oximetry for 5 to
10 min in 2 dogs receiving the IM10 treatment. Dose-dependent decreases in rectal
temperature, respiratory rate and arterial blood pressure also occurred. The quality of
recovery was considered satisfactory in all dogs receiving each treatment; all the dog
exhibited transient muscular tremors and staggering gait. In conclusion, IM alfaxalone
produced a dose-dependent anesthetic effect with relatively mild cardiorespiratory
depression in dogs. However, hypoxemia may occur at higher IM doses of alfaxalone. 相似文献

Identifying the mechanism of superconductivity in the high-temperature cuprate superconductors is one of the major outstanding problems in physics. We report local measurements of the onset of superconducting pairing in the high-transition temperature (Tc) superconductor Bi2Sr2CaCu2O8+delta using a lattice-tracking spectroscopy technique with a scanning tunneling microscope. We can determine the temperature dependence of the pairing energy gaps, the electronic excitations in the absence of pairing, and the effect of the local coupling of electrons to bosonic excitations. Our measurements reveal that the strength of pairing is determined by the unusual electronic excitations of the normal state, suggesting that strong electron-electron interactions rather than low-energy (<0.1 volts) electron-boson interactions are responsible for superconductivity in the cuprates. 相似文献

Several studies have indicated that viruses require a specific cytoskeletal structure for replication in host cells. In this study, we examined the role of actin fiber in the replication of canine distemper virus (CDV), belonging to the Morbillivirus genus of the family Paramyxoviridae. For this purpose, we used two actin depolymerizing agents, cytochalasin-D (C-D) and mycalolide-B (ML-B). In Vero cells, C-D disrupted actin fibers distributed in the cytosol, but peripheral actin fibers remained intact. On the other hand, ML-B completely disrupted the actin fibers distributed in both areas. Treatment of Vero cells with C-D or ML-B inhibited the replication of CDV. Double staining of CDV-infected Vero cells with antibody to N-protein and rhodamine-phalloidin revealed the presence of N-protein in mid-cytoplasm. However, the N-protein was specifically localized at the submembrane region in the presence of C-D, whereas it was clustered in the presence of ML-B. Viral mRNA levels of N- and H-proteins were rather increased by treatment with C-D or ML-B. The treatment with ML-B strongly inhibited N-protein expression, whereas C-D only slightly inhibited N-protein expression. These results suggest that actin microfilaments distributed in the cytoplasm and on the membrane region in host cells may have a different role in the process of CDV replication. 相似文献

Medical records of dogs with colorectal polyps were retrospectively reviewed, and clinical presentation of inflammatory colorectal polyps in miniature dachshunds was evaluated. Of 33 dogs found to have colorectal polyps, miniature dachshunds were markedly over-represented with 16 dogs (48%), of which 12 (75%) were found to have inflammatory polyps. Multiple polyps localized between the rectum and the descending colon was the most common finding in miniature dachshunds with inflammatory polyps. Twenty dogs (80%) out of 25 miniature dachshunds with inflammatory colorectal polyps responded to immunosuppressive therapy using prednisolone and cyclosporine. The results of this study indicate that miniature dachshunds are predisposed to develop inflammatory colorectal multiple polyps, for which immunosuppressive therapy may be a treatment option. 相似文献

Juvenile hormone (JH) is an insect-specific hormone that regulates molting and metamorphosis. Hence, JH signaling inhibitors (JHSIs) and activators (JHSAs) can be used as effective insect growth regulators (IGRs) for pest management. In our previous study, we established a high-throughput screening (HTS) system for exploration of novel JHSIs and JHSAs using a Bombyx mori cell line (BmN_JF&AR cells) and succeeded in identifying novel JHSIs from a chemical library. Here, we searched for novel JHSAs using this system. The four-step HTS yielded 10 compounds as candidate JHSAs; some of these compounds showed novel basic structures, whereas the others were composed of a 4-phenoxyphenoxymethyl skeleton, the basic structure of several existing JH analogs (pyriproxyfen and fenoxycarb). Topical application of seven compounds to B. mori larvae significantly prolonged the larval period, suggesting that the identified JHSAs may be promising IGRs targeting the JH signaling pathway. 相似文献
