Background
In order to improve the biological control agent (BCA) efficacy, stress factors threatening the viability of microorganisms during spray application need to be determined. The effect of spray mixture temperature and exposure time on Trichoderma harzianum T 22 and Bacillus amyloliquefaciens QST713 viability were tested. Concurrently the combined effect of mechanical and thermal stress effect on BCA viability were tested at two initial spray mixture temperatures (14 and 25 °C) by simulating a spray application using airblast sprayers featured by different tank capacity and a spray liquid circuit (without and with hydraulic agitation system). To assess the BCA microorganism viability, spray mixture samples were collected at time intervals along trials and plated to count the colony forming units (CFU).Results
The critical temperature threshold that inhibited BCA viability was 35 °C with 30 min of exposure. The sprayer type, the initial temperature of the spray mixture and the temperature increment during the trials significantly decreased the number of CFU recovered. When simulating a spray application, the spray mixture temperature increase rate was determined mainly by the residual amount of spray mixture in the tank. Even if the tank capacity does not substantially affect the final temperature reached by the spray mixture, the higher residual spray mixture in bigger tanks can expose the BCAs for a longer time to critical temperatures.Conclusions
Experimental trials allowed us to identify the effect of factors affecting the viability of tested BCAs, providing information about the actual chance to guarantee the biological efficacy of BCA treatments. © 2023 The Authors. This paper describes the development of a systems based model to characterise farmers’ decision-making process in information-intensive practices, and its evaluation in the context of Precision Agriculture (PA). A participative methodology was developed in which farm managers decomposed their process of decision-making into brief decision statements along with associated information requirements. The methodology was first developed on a university research farm in Denmark and further revised during testing on a number of research and commercial farms in Indiana, USA. Twenty-one decision-analysis factors were identified to characterise a farm manager’s decision-making process. Then, a general data flow diagram (DFD) was constructed that describes the information flows “from data to decision”. Illustrative examples of the model in the form of DFDs are presented for a strategic, a tactical and an operational decision. The model was validated for a range of decisions related to operations by three university farm managers and by five commercial farmers practicing PA for cereal, corn and soybean production in Denmark and in Indiana, USA. 相似文献

The effect of pH and temperature on the microbial reductive transformation of pentachloronitrobenzene (PCNB), an organochlorine fungicide, was investigated with a mixed fermentative/methanogenic culture developed from a contaminated estuarine sediment. Culture series were incubated at a temperature range from 4 to 45 degrees C at pH 6.9+/-0.1 and at a pH range from 2.7+/-0.1 to 7.6+/-0.1 at 22 degrees C. Significant differences were observed in terms of biotransformation rate, extent, and products as a function of temperature. Incubation at different pH values resulted in differences in biotransformation rate and extent, but not in terms of products formed. PCNB (3 microM) was transformed to pentachloroaniline (PCA) in all culture series. However, sequential dechlorination of PCA was observed only at a temperature range from 4 to 35 degrees C and at a pH range from 6.2+/-0.1 to 7.6+/-0.1. The highest PCA dechlorination rate was observed at 22 degrees C and at pH 7.6+/-0.1. The effect of temperature on the PCA dechlorination rate was modeled using an Arrhenius relationship, which accounts for both enzyme activation and deactivation. The dechlorination of PCA and chlorinated aniline intermediates was simulated using a branched-chain Michaelis-Menten model, and kinetic constants were determined. 相似文献

Crocus sativus stigmas are one of the widely known spices (saffron) and consist of unusually polar carotenoids. Alzheimer's disease is characterized pathologically by deposition of amyloid beta-peptide (Abeta) fibrils. Oxidation is thought to promote Abeta fibril formation and deposition. To identify agents inhibiting the pathogenesis of Alzheimer's disease, we examined in vitro the antioxidant properties of extract of C. sativus stigmas and its effect on Abeta(1-40) fibrillogenesis. The antioxidant properties were determined by measuring the ferric-reducing antioxidant power and Trolox-equivalent antioxidant capacity, while its effects on Abeta-aggregation and fibrillogenesis were studied by thioflavine T-based fluorescence assay and by DNA binding shift assay. The water:methanol (50:50, v/v) extract of C. sativus stigmas possesses good antioxidant properties, higher than those of tomatoes and carrots, and inhibited Abeta fibrillogenesis in a concentration and time-dependent manner. The main carotenoid constituent, trans-crocin-4, the digentibiosyl ester of crocetin, inhibited Abeta fibrillogenesis at lower concentrations than dimethylcrocetin, revealing that the action of the carotenoid is enhanced by the presence of the sugars. Our findings suggest the possible use of C. sativus stigma constituents for inhibition of aggregation and deposition of Abeta in the human brain. 相似文献
