This study was carried out to assess the effects of a blend of herbal essential oils (namely Thymus vulgaris, Mentha piperita, Rosmarinus offisinalis and Anethum graveolens) and different dietary lipid sources on the performance, ileal bacterial enumeration, intestinal histological alterations and immune responses in laying hens. For this purpose, a total of 150 laying hens were randomly allocated to six experimental treatments with five replicates of five birds each. Dietary treatments consisted of three levels of a mixture of essential oils (0, 100 and 200 mg/kg) and two sources of lipid (soybean oil and fish oil containing different ratios of n‐6 to n‐3 fatty acids) fed to the animals during an 80‐days feeding trial. Findings indicated that dietary administration of fish oil not only increased significantly the spleen relative weight (p < 0.01) and the antibody titre against Newcastle virus (p < 0.05) but also led to reductions in liver relative weight (p < 0.05) and feed conversion ratio (p < 0.05). Moreover, the mixture of herbal essential oils brought about declines in hepatic relative weight, heterophile to lymphocyte ratio (p < 0.05) and intestinal pathogen populations (p < 0.01). Mention must also be made of the improvements it made in spleen weight (p < 0.01), antibody titres against SRBC (p < 0.01) and Newcastle virus (p < 0.05), villus height to crypt depth ratio (p < 0.01), goblet cell numbers (p < 0.05), lamina propria lymphatic follicle diameters (p < 0.01) and feed conversion ratio (p = 0.06). It may be claimed that the enhancements observed in the performance of laying hens fed fish oil and 200 mg/kg of the blend of essential oils could have potentially been associated with improved intestinal health indices as well as cellular and humoral immune responses. 相似文献
This study compared ensiled sorghum (ES) from two varieties (first and second cuts) with corn silage (CS) for chemical analysis, total-tract nutrient digestibility, ruminal variables and blood biochemistry metabolites in sheep. Five experimental silages representing first cut ES var. Pegah (PS1), second cut ES var. Pegah (PS2), first cut ES var. Speedfeed (SS1), second cut ES var. Speedfeed (SS2) and CS were assessed for chemical analysis, silage fermentation and anti-quality compounds. Diets consisted of lucerne hay, silage and a premix (at the ratio of 53.5:46:0.5 on a dry matter [DM] basis). Feed consumption, total-tract nutrient digestibility, microbial nitrogen supply (MNS), ruminal variables and blood biochemistry metabolites were determined in a Latin square design of five periods with five male sheep. Compared to sorghum, CS had lower (p < 0.01) concentrations of ash, water-soluble carbohydrates and crude protein, but higher starch, lignin (sa), non-structural carbohydrates and apparent nutrient digestibility. The measurements of pH, lactic acid, NH3-N and volatile fatty acids (VFA) confirmed that all silages were well-preserved. The anti-quality compound levels in the silages were below the safe limits for ruminants. No differences were observed in feed consumption (p > 0.05) and total-tract digestibility of DM and nutrients (p > 0.05). Rumen pH, NH3-N, VFA concentrations and acetate to propionate ratio showed no difference among treatments (p > 0.05). Sheep fed on sorghum silage showed a decrease (p < 0.01) in total protozoa, and particularly in the Entodiniae population. There were no differences in cellulolytic bacteria counts (p > 0.05) among treatments. Urinary N, MNS and blood biochemistry metabolites showed no difference among treatments (p > 0.05). Silages from both cuts of sorghum had comparable total-tract nutrient digestibility to CS, therefore, these varieties could be used as a more drought resilient silage source in the feeding of sheep. 相似文献
This study evaluated the effect of specific calcium and magnesium ratios at two hardness values on reproductive performance and offspring quality of Macrobrachium rosenbergii brood‐females. Hatchery water containing 110.5 mg/L CaCO3 hardness was considered baseline as it has been used successfully to hatch and rear larvae and was used as the control treatment. At each hardness value of 150 and 190 mg/L CaCO3, four experimental water treatments were made in triplicate. Each treatment was adjusted using soluble salts (CaCl2·H2O, MgSO4·7H2O, and MgCl2·6H2O) to provide the hardness and calcium to magnesium ratios of 0:20, 20:80, 50:50, and 80:20 needed. Each of the experimental tanks (140 L) were stocked with six females and one male. The results indicated that both hardness and Ca:Mg ratios affect brood‐female reproductive qualitative parameters such as intermolt period, egg hatchability, egg dry weight, and egg‐clutch somatic index (ESI) parameters (P < 0.05) but not fecundity and eggs per spawn. The results revealed that brood‐females at 150 mg/L hardness showed greater reproductive performance than at 190 and 110.5 mg/L hardness. The findings also demonstrated that the treatment 50Ca50Mg at 150 mg/L hardness with 38.8 mg/L calcium and 12.9 mg/L magnesium had optimum reproductive performance and offspring quality for M. rosenbergii brood‐females. 相似文献
A lysimeter experiment was conducted to investigate the effect of water table management (WTM) on distribution of soil salinity
and annual alfalfa (Medicago scutellata) yield. Subirrigations with three levels of water table namely, 0.5 (WT0.5), 0.7 (WT0.7), and 1.0 m (WT1.0) and a free drainage
(FD) conventional irrigation treatment were selected for this study. All treatments were arranged in a complete randomized
block design with three replicates. The results of this study indicated that the average soil electrical conductivity of the
saturated extract (ECe) in the root zone gradually increased and exceeded the designated crop threshold value (4 dS/m) after
the first forage harvest in subirrigated lysimeters. A higher salt accumulation was observed at the WT0.5 treatment. The average
dry matter yield of annual alfalfa in WT0.5 and WT0.7 treatments was found to be 52 and 73% higher compared with the control

The antifungal activity of onion (Allium cepa L.) on two important dermatophytes, Trichophyton rubrum and Trichophyton mentagrophytes, with special reference to morphological aspects was studied. Growth of both fungi was found to be strongly inhibited by aqueous onion extract (AOE) as a dose-dependent manner. The extract showed fungicidal effect for both fungi at concentrations >3.12% (v/v). The fungus T. mentagrophytes was more affected by the onion as compared to T. rubrum at all concentrations used. Morphological effects of onion exposure were examined in correlation with fungal growth. Corresponding to the growth inhibition, light and electron microscopy observations revealed morphological anomalies in hyphal compartments. The results demonstrated that AOE targets the cell membrane of the fungi as breaking down of both inner and outer membranes with consequent extrution of materials into the surrounding medium. Cytoplasmic membranes and other membranous structures of organelles, such as nuclei and mitochondria, were also disrupted. In correlation to the fungal growth, morphological alterations occurred to a less content for T. rubrum compared with T. mentagrophytes. The hyphae of T. rubrum were found to be mainly affected by converting to resistant forms, i.e., chlamidospores as a consequence of phenotype switching response to AOE. Plasmolysis accompanied by an almost complete depletion and disorganization of cytoplasmic structures were found to be the final event which led to cell death. Ultrastructural evidences obtained from this study strongly support that morphological changes of T. rubrum and T. mentagrophytes caused by AOE are associated with its fungistatic and fungicidal activities. With respect to the morphological results and the preliminary data on fungal biochemistry, a mechanism of action by interacting of AOE with thiol (-SH) groups present in essential compartments of the fungal cells was postulated. 相似文献
BACKGROUND: Adenoviruses are used extensively to deliver genes into mammalian cells, particularly where there is a requirement for high-level expression of transgene products in cultured cells, or for use as recombinant viral vaccines or in gene therapy. In spite of their usefulness, the construction of adenoviral vectors (AdV) is a cumbersome and lengthy process that is not readily amenable to the generation of large collection of clones. METHODS: In this project, to delete E1 gene in adenovirus, an adenoviral plasmid containing lateral sites of E1 region of adenovirus was made and recombination in the 293A cells between the homologous region of this linearized plasmid and the adenovirus genome resulted in the formation of the complete adenoviral recombinant. RESULTS: This recombination resulted in loss of E1 region and we constructed a recombinant adenovirus type 5 vector that E1 gene was deleted by homologous recombination. CONCLUSION: Homologous recombination is more easy and fast technique in the production of AdV. 相似文献
The aim of this study was to investigate the possibility of improving the growth and physiological indices of plant and fish by adding different levels of ascorbic acid (vitamin C) to water in the aquaponic system using Nile tilapia (Oreochromis niloticus) and saffron plant (Crocus sativus). 240# fish (12.5?±?0.21 gr) and 120# saffron corms (2.8?±?0.12 gr) were randomly assigned to 15 experimental units and underwent treatments of adding 0 (control), 2, 4, 6, and 8 mg L?1 ascorbic acid to water, every 6 days, for 8 weeks. The fish final weight and subsequently other growth performance indices increased in the treatment of 4 mg L?1 vitamin C compared to other treatments (P?≤?0.05). Some growth performances of saffron plants such as saffron production, in treatment of 6 mg L?1, were significantly higher than the control group and reached from 17.34?±?0.27 mg flower?1 in the control group to 25.4?±?1.61 mg flower?1 in treatment of 6 mg L?1. Measuring the serum biochemical parameters of the fish showed that, in the treatment of 8 mg/L ascorbic acid, the cortisol content in the blood reached its maximum (21.49?±?2.42 µg dL?1). The trypsin activity in proximal intestine and mid-intestine significantly increased in treatments of 2, 4, and 6 mg L?1, respectively. The current experiment showed that, by adding 4–6 mg/L ascorbic acid to the aquaponic system water (every 6 days), the optimal levels of Nile tilapia and saffron plant growth performances would occur.
