Recently we reported elimination of radioactivity from [14C]heptachlor from body stores of lactating ovines, mainly into excreta rather than milk, contrasting sharply with bovines. To further assess heptachlor metabolism and clearance by ovines, 12 fine-wool wether lambs (41 +/- 3 kg) housed in metabolism stalls were fed pelleted alfalfa hay (96%) plus molasses (3%) ad libitum and were dosed i.p. once with [14C]heptachlor (1.643 mg/kg body wt; sp. act. = .89 microCi/mg). Feces and urine were collected separately and quantitatively. Light mineral oil was mixed with feed (5 g/100 g) of six lambs and trans-stilbene oxide, an inducer of biotransformational enzymes, was administered i.p. (4 g/hd initially; 2 g/hd daily thereafter) through 20 d to three lambs given each mineral oil treatment, in 2 x 2 factorial arrangement. Feces, urine, blood, bile and body tissues were assayed for total 14C activity. Radioactivity (heptachlor and [or] metabolites) eliminated into excreta during 21 d amounted to 34 to 36% of dose administered, of which 67% appeared in urine and 33% in feces. Biological half-time for elimination into excreta was 11.3 d [Kel = -.061/d], similar to 11.7 d we reported for lactating ewes. Clearance from blood had T1/2 = 14 d. Neither mineral oil nor trans-stilbene oxide altered rate or route of 14C activity excreted or concentrations of 14C activity in blood. Results confirmed that ovines eliminate heptachlor much more rapidly than bovines.  相似文献

Ruminally protected choline (RPC) was evaluated in two experiments. In Exp. 1, beef steers (n = 160; average initial BW = 350.9 kg) were fed a 90% concentrate diet with either 0, .25, .5, or 1.0% RPC (DM basis) for 112 to 140 d. Feeding .25% RPC increased ADG 11.6% compared with 0% RPC, but responses diminished with increasing RPC level (cubic response, P < .10). Daily DMI was not affected by RPC level, but feed:gain was improved 6.8% with .25% RPC compared with 0% RPC, and responses diminished with increasing RPC level (cubic response, P < .10). Carcass yield grade increased linearly (P < .10) as RPC level increased, but marbling score was lower for all three RPC-containing diets than for the 0% RPC diet (quadratic response, P < .05). In Exp. 2, 20 Suffolk lambs (initial BW = 29.8 kg) were fed an 80% concentrate diet for 56 d with the same RPC levels as in Exp. 1. Serum triglycerides (TG) and cholesterol (CLSTRL) were measured in weekly blood samples, and intensive blood samples were collected on d 28 and 56 to evaluate serum insulin (INS), GH, and NEFA. For the 56-d feeding period, ADG responded quadratically (P < .10) to RPC level, but DMI and feed:gain were not affected. Serum INS and NEFA concentrations increased linearly (P < .05) and serum GH responded cubically (P < .05) to RPC level on d 28, but no differences were noted on d 56. Serum TG concentrations in weekly samples increased linearly (P < .10) with RPC level, but, averaged over all weeks, serum CLSTRL concentrations did not differ (P > .10) among treatments. Quantities of carcass mesenteric (P < .05) and kidney fat (P < .10) increased linearly, but longissimus muscle and liver fat contents did not differ (P > .10) among RPC levels. Supplementing RPC in high-concentrate diets improved performance, but results were not consistent among RPC levels or between cattle and sheep. Potential effects of RPC might be mediated through alterations in fat metabolism and(or) metabolic hormones related to fat metabolism.  相似文献

Primiparous Angus x Gelbvieh (n = 36) rotationally crossed beef cows (initial BW = 487.9 +/- 10.5 kg, body condition score = 5.5 +/- 0.02) were utilized to determine effects of supplemental safflower seeds high in linoleic (76% 18:2) or oleic (72% 18:1) acid on cow BW change, body condition score, milk production and composition, calf weight gain, cow serum metabolites, and metabolic hormones. On d 3 postpartum, cows were randomly assigned to one of three isonitrogenous dietary supplements with equal total quantity of TDN: corn-soybean control supplement (n = 12); high-linoleate safflower seeds (n = 12); or high-oleate safflower seeds (n = 12). Safflower-seed supplements were formulated to provide 5% DMI as fat. Supplements were individually fed from d 3 postpartum through 90 d postpartum. Cows had ad libitum access to native grass hay (7.8% CP), trace-mineralized salt, and water. Date of parturition was evenly distributed across treatments with all cows calving within 14 +/- 0.8 d. There were no differences (P = 0.65) in total OM intake among treatments. Although cow BW change did not differ (P = 0.33) by treatment, supplementation influenced cow body condition score (P = 0.02) with linoleate-supple-mented cows in higher (P = 0.005) condition overall than oleate-supplemented cows (5.1 +/- 0.06 vs 4.9 +/- 0.06). Twenty-four-hour milk production did not differ (P = 0.68) among treatments. Percentage milk fat was not different at d 30; however, at d 60 and d 90 percentage milk fat was greater (P ( 0.05) in control and oleate-supplemented cows than in linoleate-supplemented cows. Calf BW gains (P = 0.27) and adjusted 205-d weights (P = 0.48) were not affected by supplement treatment. Supplementation did not influence serum concentrations of glucose (P = 0.38), NEFA (P = 0.61), GH (P = 0.29), IGF-I (P = 0.81), insulin (P = 0.26), or IGF-I binding proteins (P > or = 0.11). Days to conception did not differ (P = 0.40) among treatments. Although overall productivity of the primiparous cows and their calves was not altered by safflower-seed supplementation, differential effects were noted between supplements. Oleate supplementation increased percentage milk fat at d 60, and cow body condition score was lower than in linoleate-supplemented cows. Linoleate-supplemented cows had greater body condition scores by 90 d postpartum than either corn-soybean- or oleatesupplemented cows.  相似文献

Effects of supplemental degradable (DIP) and undegradable (UIP) intake protein on forage intake, BW change, body condition score (BCS), postpartum interval to first estrus, conception rate, milk production and composition, serum metabolites and metabolic hormones, and calf gain were determined using 36 primiparous Gelbvieh x Angus rotationally crossed beef cows. On d 3 postpartum, cows (average initial BW = 495 +/- 10 kg and BCS = 5.5 +/- 0.1) were randomly assigned to one of three dietary supplements (12 cows/treatment). Date of parturition was evenly distributed across treatment (average span of calving date among treatments = 2.4 +/- 2.5 d). Individually fed (d 3 through 120 postpartum) dietary supplements were 0.82 kg of corn and 0.23 kg of soybean meal per day (DIP), the DIP + 0.12 kg of blood meal and 0.13 kg of corn gluten meal per day (DIP + UIP), and 0.82 kg of corn, 0.07 kg of blood meal, and 0.08 kg of corn gluten meal per day in an isonitrogenous replacement of soybean meal (UIP IsoN). Cows had ad libitum access to native grass hay (8.5% CP) and trace-mineralized salt. Total OM intake was greater (P = 0.06) for DIP + UIP than UIP IsoN cows. At 30 d postpartum, DIP + UIP cows produced more milk than UIP IsoN, with DIP being intermediate; however, at 60 d postpartum, DIP + UIP and DIP cows were not different, but both had greater milk production than UIP IsoN (treatment x day interaction; P = 0.08). A treatment x day interaction (P = 0.06) for BCS resulted from DIP + UIP cows having the greatest BCS at 60, 90, and 120 d d postpartum and DIP having greater BCS than UIP IsoN cows only on d 60 postpartum. Serum insulin concentrations were highest (treatment x day interaction; P = 0.09) for DIP + UIP cows at 30 d postpartum but did not differ among treatment thereafter. Serum insulin-like growth factor-binding protein (IGFBP)-2 (34 kDa) and -3 (40 and 44 kDa) were greatest (P < 0.0003) for DIP cows. Serum urea-N concentrations were greater (P < 0.01) in DIP + UIP cows than in either DIP or UIP IsoN cows. However, postpartum interval to first estrus, conception rate, and calf weaning weights were unaffected (P = 0.35, 0.42, and 0.64, respectively) by treatment. Although UIP in addition to or in replacement of DIP affected milk production and blood metabolites, the productivity of these primiparous beef cows was not altered. Thus, the type of supplemental protein does not seem to influence productivity of primiparous beef cows in production systems with conditions similar to our experimental conditions.  相似文献

In ruminants, nutrition is one of the exogenous inputs affecting reproductive function at different levels of the hypothalamic-hypophyseal-gonadal axis. However, the exact mechanisms or even the identification of the signalling metabolic compounds by which nutrition affects reproductive function still need further clarification. The role of static body condition (BC) and its interaction with a short-term protein supplementation (PL), on secretion of metabolic hormones [growth hormone (GH), insulin and insulin-like growth factor-1 (IGF-1)], as well as on secretion of LH and progesterone (P4) was evaluated in sheep. Twenty-four Rambouillet ewes divided into two groups, with lower (LBC) and higher body condition (HBC), were randomly assigned within BC to one of two PL levels: low (LPL, 24% of crude protein; 14 g/animal/day), and high (HPL, 44% of crude protein; 30 g/animal/day). The secretion of GH, insulin, IGF-1 and LH was evaluated on day 10 of the oestrous cycle; appearance and timing of oestrous behaviour were previously detected using rams. Progesterone secretion was evaluated on day 13 of the same cycle. No differences were found (p > 0.05) between PL groups on serum GH concentrations during the sampling period (overall mean of 4.0 +/- 0.3 ng/ml), but a trend for lower values in HBC sheep was found (3.6 +/- 0.4 vs 4.4 +/- 0.4 ng/ml, p = 0.06). A BC effect was observed (p < 0.05) on serum IGF-1 level, with higher values in HBC sheep (p < 0.05). Neither BC nor PL affected (p > 0.05) secretion of LH and the number of corpora lutea, nor serum P4 and insulin concentrations. Results indicate a predominance of the static component of nutrition on sheep metabolic hormone responses, GH and IGF-1, with no effect of short-term PL on secretion of pituitary and ovarian hormones as well as luteal number and activity.  相似文献

Breeding ewes in drylot were fed pelleted complete diets with 3% cottonseed meal (CSM) or 7% dried, gamma-irradiated sewage solids (DGSS) for 4 yr. Cytochrome P-450 (P-450) content and enzyme activities for xenobiotics biotransformations were assayed in livers after 2 yr and in livers, kidneys and ileal tissue after 4 yr. Dietary DGSS caused no increase in P-450 and few changes in activities of oxidative, hydrolative and conjugative biotransformational enzymes. Consumption of DGSS for 4 yr caused slight enlargement of spleens (1.1-fold, P less than .10) and ovaries (1.3-fold, P less than .10), but no change in size of livers, kidneys, hearts, adrenals and thyroids (P greater than .10), nor liver vitamin A levels (P greater than .10). Of 22 refractory lipophilic residues assayed in abdominal adipose tissue, few were detected and of those detected DGSS caused none to exceed normal levels. Dietary DGSS increased (P less than .01) Fe in livers 1.5-fold and in spleens 5.6-fold, and increased Cu in livers 1.3-fold (P less than .01) and in kidneys 1.2-fold (P less than .05). Dietary DGSS increased Cd levels in livers (P less than .01) but not in kidneys or spleens (P greater than .10); yet all Cd levels were within ranges for livestock fed conventional feed. Dietary DGSS caused no increase (P greater than .10) in levels of Ag, Ca, Cr, Hg, K, Mg, Mn, Na, Ni, P, Pb or Zn in livers, kidneys or spleens. There were no histopathological lesions of toxicosis except mild hemosiderosis of spleens. Consumption of a diet with 7% DGSS throughout 4 yr caused no hazardous accumulation of toxic elements and little, if any, evidence of toxicosis.  相似文献

Acanthocephalan and spargana parasites were identified within a body wall mass during exploratory surgery in a wild green tree snake. Acanthocephalan parasites have not previously been reported in this species. Background

Heifers not used as breeding stock are often implanted with steroids to increase growth efficiency thereby altering hormone profiles and potentially changing the environment in which ovarian follicles develop. Because bovine granulosa cell culture is a commonly used technique and often bovine ovaries are collected from abattoirs with no record of implant status, the objective of this study was to determine if the presence of an implant during bovine granulosa cell development impacts follicle stimulating hormone-regulated steroidogenic enzyme expression. Paired ovaries were collected from 16 feedlot heifers subjected to 1 of 3 treatments: non-implanted (n = 5), Revalor 200 for 28 d (n = 5), or Revalor 200 for 84 d (n = 6). Small follicle (1 to 5 mm) granulosa cells were isolated from each pair and incubated with phosphate buffered saline (n = 16) or 100 ng/mL follicle stimulating hormone (n = 16) for 24 h.

Results

Granulosa cells of implanted heifers treated with follicle stimulating hormone produced medium concentrations of progesterone similar (P = 0.22) to non-implanted heifers, while medium estradiol concentrations were increased (P < 0.10) at 28 and 84 d compared to non-implanted heifers indicating efficacy of treatment. Additionally, real-time PCR analysis in response to follicle stimulating hormone treatment demonstrated a decrease in steroidogenic acute regulatory protein (P = 0.05) mRNA expression in heifers implanted for 84 d and an increase in P450 side chain cleavage mRNA in granulosa cells of heifers implanted for 28 (P < 0.10) or 84 d (P < 0.05) compared to non-implanted females. However, no difference in expression of 3-beta-hydroxysteroid dehydrogenase (P = 0.57) and aromatase (P = 0.23) were demonstrated in implanted or non-implanted heifers.

Conclusions

These results indicate follicles which develop in the presence of high concentrations of androgenic and estrogenic steroids via an implant tend to demonstrate an altered capacity to respond to follicle stimulating hormone stimulation. Thus, efforts should be made to avoid the use of implanted heifers to study steroidogenesis in small follicle granulosa cell culture systems.  相似文献

Ninety-nine multiparous Brahman-influenced (1/4 to 3/8 Brahman) cows were managed to achieve low (BCS = 4.3 +/- 0.1; n = 50) or moderate (BCS = 6.1 +/- 0.1; n = 49) body condition (BC) to determine the influence of bovine somatotropin (bST) on estrous characteristics, reproductive performance, and concentrations of serum GH and plasma NEFA. Beginning 32 d postpartum, cows within each BC were assigned randomly to treatment with or without bST. Non-bST-treated cows received no treatment, and treated cows were administered bST (Posilac, 500 mg s.c.) on d -35, -21, and -7 before initiation of the breeding season. On d -7, all cows received an intravaginal, controlled internal drug-releasing (CIDR) device. On d 0 (initiation of the 70-d breeding season), the CIDR were removed and cows received prostaglandin F2alpha (PGF2alpha). Blood samples were collected from the median caudal vein of the cows at each bST treatment and at d -28 and 0. Estrous behavior was monitored by radiotelemetry during the first 30 d of the breeding season. Growth hormone was increased (P < 0.05) in low and moderate BC cows treated with bST. The percentage of cows detected in estrus during the first 30 d of the breeding season was decreased (P = 0.05) for low BC (64%) compared with moderate BC (82%) cows. The interval to first estrus tended (P = 0.07) to be shorter in low BC-bST-treated cows (3.7 +/- 1.9 d) than in moderate BC-bST-treated cows (9.6 +/- 1.8 d). During the first 30 d of the breeding season, cows in low BC had a decreased (P = 0.02) number of mounts received and increased (P = 0.001) quiescence between mounts compared with cows in moderate BC. The number of mounts received was reduced (P = 0.04) in bST-treated cows. More (P = 0.02) cows treated with bST became pregnant during the first 3 d of the breeding season compared with non-bST-treated cows. The cumulative first-service conception rate tended (P = 0.07) to be greater for bST-low BC cows than non-bST-treated cows in low or moderate BC. On d 0, NEFA were greater (P < 0.05) in bST-treated vs. non-bST-treated cows. Low BC and bST reduced the intensity of behavioral estrus in postpartum Brahman-influenced cows. However, bST increased the first-service conception rate during the first 30 d of breeding and pregnancy rates during the first 3 d of breeding in postpartum Brahman-influenced cows.  相似文献

The local effects of isolated magnetic adatoms on the electronic properties of the surface of a superconductor were studied with a low-temperature scanning tunneling microscope. Tunneling spectra obtained near magnetic adsorbates reveal the presence of excitations within the superconductor's energy gap that can be detected over a few atomic diameters around the impurity at the surface. These excitations are locally asymmetric with respect to tunneling of electrons and holes. A model calculation based on the Bogoliubov-de Gennes equations can be used to understand the details of the local tunneling spectra.  相似文献
