Two potyvirus isolates, one from germplasm of yard-long bean (Vigna unguiculata ssp.sesquipedalis) introduced into the Netherlands, and another one from soybean plants (Glycine max) in Indonesia, were compared with two virus isolates of blackeye cowpea mosaic virus (BICMV) from the USA and a Moroccan isolate of cowpea aphid-borne mosaic virus (CAMV). It is proposed that all five isolates be now considered BICMV on the basis of host ranges, symptoms and serology. From our results, and a reassessment of the literature it is suggested to drop the name CAMV in favour of BICMV.Samenvatting Twee potyvirussen, de een in Nederland ingevoerd met genenmateriaal vanVigna unguiculata ssp.sesquipedalis en de ander uit planten van sojaboon (Glycine max) in Indonesië, werden vergeleken met twee isolaten van blackeye cowpea mosiac virus (BICMV) en een Marokkaans isolaat van cowpea aphid-borne mosaic virus (CAMV). Labonuory bioassay and field experiments were conducted to characterize the dose response of weeds to methyl iodide and methyl bromide as soil fumigants. The patterns in potency of both fumigants and in sensitivity of diffcretit weed species to the fumigants were distinguished with the use of logistic dose-response models. Similar to its response to methyl hromide fumigation. Amaranthus retrofleus L. was the most sensitive to methyl iodide fumigation. Cyperus rotundtis L. was the least sensitive to methyl iodide fumigation, whereas Portuloca oleracea L. was the least sensitive to methyl hromide. Lolium multiflorum Lam. Abutilon theophrasti Medik. Chenopodium album L. P. ateracea . Brassica kaber (D.C.) L.C. Wheeler and Cyperus escuden-tus L. were similar in sensitivity to methyl iodide. Methyl iodide was as potent as methyl bromide for A. retroflexus but more potent than methyl bromide for L. multiflorum , A. theophrasti. C. album. P. oleracea. B. kaber, C. esculentus and C. rotundus. The dose response for weeds in the field was similar to that obtained in laboratory bioassays. Under fieid conditions. 280 kg ha-1 methyl iodide killed all species tested except Solanum nigrum L Methyl iodide appears to be a suitable replacement for meihyl bromide because it can be used in situations simitar to methyl bromide fumigation, has superior efficacy against a broad spectrum of pests and has a low potential for degrading the earth's ozone lavers. 相似文献
Forage technology has been successfully introduced into smallholder cattle systems in Cambodia as an alternative feed source to the traditional rice straw and native pastures, improving animal nutrition and reducing labour requirements of feeding cattle. Previous research has highlighted the positive impacts of forage technology including improved growth rates of cattle and household time savings. However, further research is required to understand the drivers, challenges and opportunities of forage technology for smallholder cattle households in Cambodia to facilitate widespread adoption and identify areas for further improvement. A survey of forage-growing households (n = 40) in July–September 2016 examined forage technology adoption experiences, including reasons for forage establishment, use of inputs and labour requirements of forage plot maintenance and use of forages (feeding, fattening, sale of grass or seedlings and silage). Time savings was reported as the main driver of forage adoption with household members spending approximately 1 h per day maintaining forages and feeding it to cattle. Water availability was reported as the main challenge to this activity. A small number of households also reported lack of labour, lack of fencing, competition from natural grasses, cost of irrigation and lack of experience as challenges to forage growing. Cattle fattening and sale of cut forage grass and seedlings was not found to be a widespread activity by interviewed households, with 25 and 10% of households reporting use of forages for these activities, respectively. Currently, opportunities exist for these households to better utilise forages through expansion of forage plots and cattle activities, although assistance is required to support these households in addressing current constraints, particularly availability of water, if the sustainability of this feed technology for smallholder cattle household is to be established in Cambodia. 相似文献
The aim was to verify the effect of follicle‐stimulating hormone (FSH) supplementation to α‐MEM+ or TCM199+ media on the in vitro development of ovarian preantral follicles (PFs) derived from collared peccaries. Ovaries (n = 5 pairs) were collected and divided into fragments destined to control group (non‐cultured) or treatments that were cultured for 7 days. The PFs morphology, growth and activation were evaluated by classical histology. The immunohistochemistry markers Ag‐NOR and PCNA were used for nuclear proliferation analysis, and the picrosirius red labelling was used for ovarian extracellular matrix (ECM) evaluation. After 7‐day culture, only the TCM199+ treatment maintained the proportion of intact PFs similar to day 1 (63.2%), but no differences were found among treatments (p > .05). In addition, a significant increase in the growing follicles proportion was verified for all the treatments, indicating follicular activation (p > .05). By the Ag‐NOR analysis, only the TCM199+/FSH maintained the nuclear proliferation similar to the first day (p > .05). The picrosirius red staining revealed that the ECM remained intact in all the treatments (p > .05). We suggest the use of TCM199+ medium supplemented of FSH for the in vitro development of peccaries PFs under 7‐day culturing conditions. 相似文献
We conducted a study on 81 initially bulk-milk ELISA negative dairy herds taken from a random sample of Dutch dairy herds to evaluate variation in bulk-milk S/P ratios and to study reasons for bulk-milk conversion. These herds were repeatedly (3-month intervals) tested between April 2004 and August 2005 and serostatus of all animals had previously been established as negative (N), low-positive (LP) or high-positive (HP). Of these herds, herd- and test-related factors associated with variation in sample over positive (S/P) ratios were analysed using a multivariable linear-mixed model with ‘herd’ as random effect. In addition, changes of animal serostatus in converting herds were described. S/P ratios were calculated as the optical density of the bulk-milk sample minus the optical density of the negative serum control divided by the difference in optical density between the positive and negative serum control.
Sixteen bulk-milk conversions in 12 dairy herds were seen with few indications of serological conversion in lactating cattle except for one herd in which recrudescence of infection appeared likely in nine cows. The effect of HP serostatus on bulk-milk S/P ratio was 2–3 times stronger compared with LP serostatus. In addition, bulk-milk S/P-ratio increased when the proportion of HP animals between 1 and 60 days in milk increased and decreased when the average milk-production level of the herd increased. Besides these herd-related factors, the use of different ELISA-testkits between test rounds had a significant effect on the S/P-ratio in bulk-milk samples. 相似文献
A 4-month old, 200 kg, grey warmblood colt presented for a firm, non painful mass on the distal medial aspect of the left third metatarsus. Excisional biopsy revealed a diagnosis of haemangiosarcoma. Equine haemangiosarcoma is uncommon and only limited reports of successful treatment are available. The prognosis for survival is therefore considered to be poor. After two separate incidences of recurrence with incomplete excision of the tumour, intralesional treatment with cisplatin without excision or debulking was performed on three separate occasions. Intralesional cisplatin injection was performed at monthly intervals for three treatments. Four years post treatment with cisplatin, the horse remained in remission. This case report describes the diagnostic and treatment challenges for successful treatment of a primary haemangiosarcoma on the distal limb of a warmblood foal using intralesional cisplatin chemotherapy. 相似文献
Samenvatting Een methode werd ontwikkeld om de resistentie van in de kas geteelde tomateplanten tegen tomatemozaïek virus (ToMV) te bepalen. Bladeren van een vatbare en een resistente cultivar werden afgesneden en geïnoculeerd met ToMV. Na 6, 10 en 17 dagen werden de geïnoculeerde bladeren getoetst op de aanwezigheid van virus met ELISA en door inoculatie van bladeren vanNicotiana glutinosa. Met beide toetsmethoden kon de virustoename in de vatbare cultivar al vroeg na inoculatie duidelijk worden aangetoond. In de bladeren van de resistente cultivar was een zeer kleine hoeveelheid virus pas laat na de inoculatie aantoonbaar. Met deze methode is het mogelijk om de resistentie tegen ToMV te bepalen, tevens zaad te winnen en landbouwkundige eigenschappen te evalueren, zonder de plant te infecteren. 相似文献
A disorder of unknown aetiology appearing as silvering on squash (Cucurbita pepo) leaves has become prevalent in south Florida. Typical symptoms, observed after 2 weeks of exposure to adults of Bemisia tabaci , the sweet potato whitefly (SPWF) include vein clearing followed by silvering of whole leaves. Fruits from affected plants are blanched or streaked longitudinally. The silverleaf syndrome was not mechanically transmissible when crude extracts of symptomatic leaves were used as inoculum. Two sizes of double-stranded RNA (dsRNA) of c. 4.6 and 4.2 kilobase pairs (kbp) were consistently observed in leaf extracts of caged, whitefly-infested, symptomatic plants. In contrast, caged plants not exposed to whiteflies showed no symptoms and contained no detectable dsRN A. In addition, squash plants exposed to SPWF colonies from California neither developed silverleaf symptoms nor contained any dsRNA. Double-stranded RNA could also be readily detected in whitefly-infested yellow summer squash and zucchini from the field. However, asymptomatic eggplant and watermelon infested with SPWF from nearby fields did not contain detectable dsRNA. Dot-spot hybridization assays using an RNA-specific probe detected homologous sequences in both adults and nymphs of SPWF that induced silverleaf symptoms but not in adults and nymphs of SPWF that could not induce silverleaf symptoms. These results, along with the non-endogenous nature of the detected dsRNA suggest that the dsRNA is or is associated with the causal agent of whitefly-mediated leaf silvering in squash. 相似文献
