Mannheimia granulomatis是导致牛、羊呼吸道传染病的一种病原菌.为快速、准确诊断由该菌导致的疫病,从基因库中获得M.granulomdtis的基因序列,再从M.granulomatis的基因序列中获得与其他细菌包括溶血性曼氏杆菌和多杀性巴氏杆菌等不同的基因片段,设计引物,建立了特异性好、敏感性较高的M.granulomatis的PCR诊断方法.结果表明,其特异性为100%,最小检出量为5×104cfu/mL~5×103cfu/mL M.granulomatis或1/10个单个菌落.  相似文献

Several experiments were performed to investigate the physiology of seawater acclimation in the striped bass, Morone saxatilis. Transfer of fish from fresh water (FW) to seawater (SW; 31–32 ppt) induced only a minimal disturbance of osmotic homeostasis. Ambient salinity did not affect plasma thyroxine, but plasma cortisol remained elevated for 24h after SW transfer. Gill and opercular membrane chloride cell density and Na+,K+-ATPase activity were relatively high and unaffected by salinity. Average chloride cell size, however, was slightly increased (16%) in SW-acclimated fish. Gill succinate dehydrogenase activity was higher in SW-acclimated fish than in FW fish. Kidney Na+, K+-ATPase activity was slightly lower (16%) in SW fish than in FW fish. Posterior intestinal Na+,K+-ATPase activity and water transport capacity (Jv) did not change upon SW transfer, whereas middle intestinal Na+,K+-ATPase activity increased 35% after transfer and was correlated with an increase in Jv (110%). As salinity induced only minor changes in the osmoregulatory organs examined, it is proposed that the intrinsic euryhalinity of the striped bass may be related to a high degree of “preparedness” for hypoosmoregulation that is uncommon among teleosts studied to data.  相似文献

Summary Plants were grown from seed of two-rowed wild barley, Hordeum spontaneum, and six-rowed brittlerachised barley. H. agriocrithon, collected in Ladakh, north-western India. Whereas the H. spontaneum remained true to type, segregation for morphological characters was observed in progeny rows grown from heads of H. agriocrithon plants. The H. agriocrithon heads also showed segregation for a biochemical character, the polypeptide pattern of the endosperm storage protein fraction (hordein). The H. agriocrithon seed therefore originated from natural hybridization between cultivated H. vulgare and weedy H. spontaneum. Crosses of H. vulgare and H. spontaneum gave progeny which resembled H. agriocrithon and showed similar hordein polypeptide segregation patterns. The results indicate that six-rowed brittle-rach ised barleys from the Himalayas have a similar origin to forms found in the Middle East, and that H. agriocrithon does not play a direct role in the evolution of barley.  相似文献

Stomata regulate gas exchange and are distributed across the leaf epidermis with characteristic spacing. Arabidopsis stomata are produced by asymmetric cell divisions. Mutations in the gene TOO MANY MOUTHS (TMM) disrupt patterning by randomizing the plane of formative asymmetric divisions and by permitting ectopic divisions. TMM encodes a leucine-rich repeat-containing receptor-like protein expressed in proliferative postprotodermal cells. TMM appears to function in a position-dependent signaling pathway that controls the plane of patterning divisions as well as the balance between stem cell renewal and differentiation in stomatal and epidermal development.  相似文献

Three known bromophenols, 2,3-dibromo-4,5-dihydroxybenzylaldehyde (1), 2,2′,3-tribromo-3′,4,4′,5-tetrahydroxy-6′-hydroxymethyldiphenylmethane (2) and bis(2,3-dibromo-4,5-dihydroxylbenzyl) ether (3), and one new one, 5,5″-oxybis(methylene)bis(3-bromo-4-(2′,3′-dibromo-4′,5′-dihydroxylbenzyl)benzene-1,2-diol) (4), were isolated from an extract of the red alga, Vertebrata lanosa. The antioxidant activity of these four bromophenols was examined using one biochemical and two cellular assays: Oxygen Radical Absorbance Capacity (ORAC), Cellular Antioxidant Activity (CAA) and Cellular Lipid Peroxidation Antioxidant Activity (CLPAA) assays. Compound 2 distinguished itself by showing potent activity, having a better antioxidant effect than luteolin in both the CAA and CLPAA assays and of quercetin in the CLPAA assay. Although several bromophenols are known to be potent antioxidants in biochemical assays, this is the first time their cellular antioxidant activity has been demonstrated.  相似文献

Sir:- In June 1982, possums (Trichosurus vulpecula) from the Waitahuna forest area near Lawrence in Central Otago were examined for endoparasites (181 possums) and ectoparasites (125 possums). The alimentary tract was separated into stomach, small intestine and large intestine. These sections were washed thoroughly through a 60 mesh sieve and total parasite counts and identification were carried out. The fur from the base of the forelegs along the back and around the base of the tail was digested in 20% KOH and the ectoparasites were recovered. There was no evidence of pathogenicity ascribable to the parasites found. A summary of results is presented in Table I.  相似文献

The properties of a white wheat bread could be changed by adding normal or heat‐treated barley flour in small amounts (2 and 4%) to a white wheat bread recipe. Differences regarding gelatinization as well as retrogradation properties were found when analyzing the two flours in model systems. The heat‐treated flour was fully gelatinized due to prior time, temperature, and pressure treatment and could therefore absorb larger amounts of water than the other flours. In gelatinized model systems with 40% flour (dwb), the heat‐treated barley flour contained less retrograded amylopectin as compared with normal barley flour after storage for up to 14 days, whereas no differences were found with 20% flour (dwb). However, stored breads showed an increased retrogradation of amylopectin (as measured by differential scanning calorimetry [DSC]) when 2% pretreated barley flour was added as compared with addition of 2% normal barley flour. On the other hand, there were no significant differences at the 4% level. Addition of either of the barley flours resulted in less firm breads during storage as compared with the control breads. Increased water absorption in barley flour and thus increased water content in the breads or different water‐binding capacities of the flour blends could explain these results. The present study indicated that water had a stronger influence on bread firmness than the retrogradation of amylopectin. This conclusion was based on breads with pretreated barley flour being less firm than breads with normal barley flour, although the retrogradation, as determined by DSC, was higher.  相似文献
