Effects of adding different concentrations of melatonin (10?7, 10?9 and 10?11 M) to maturation (Experiment 1; Control, IVM + 10?7, IVM + 10?9, IVM + 10?11) and culture media (Experiment 2; Control, IVC + 10?7, IVC + 10?9, IVC + 10?11) were evaluated on in vitro bovine embryonic development. The optimal concentration of melatonin (10?9 M) from Experiments 1–2 was tested in both maturation and/or culture media of Experiment 3 (Control, IVM + 10?9, IVC + 10?9, IVM /IVC + 10?9). In Experiment 1, maturated oocytes from Control and IVM + 10?9 treatments showed increased glutathione content, mitochondrial membrane potential and percentage of Grade I blastocysts (40.6% and 43%, respectively). In Experiment 2, an increase in the percentage of Grade I blastocysts was detected in IVC + 10?7 (43.5%; 56.7%) and IVC + 10?9 (47.4%; 57.4%). Moreover, a lower number and percentage of apoptotic cells in blastocysts were observed in the IVC + 10?9 group compared to Control (3.8 ± 0.6; 3.6% versus 6.1 ± 0.6; 5.3%). In Experiment 3, the IVC + 10?9 treatment increased percentage of Grade I blastocysts with a lower number of apoptotic cells compared to IVM /IVC + 10?9 group (52.6%; 3.0 ± 0.5 versus 46.0%; 5.4 ± 1.0). The IVC + 10?9 treatment also had a higher mRNA expression of antioxidant gene (SOD 2) compared to the Control, as well as the heat shock protein (HSPB 1) compared to the IVM + 10?9. Reactive oxygen species production was greater in the IVM /IVC + 10?9 treatment group. In conclusion, the 10?9 M concentration of melatonin and the in vitro production phase in which it is used directly affected embryonic development and quality. 相似文献

We evaluated the optimal level of alfalfa inclusion in soybean hull-based diets. In Exp. 1, 20 Holstein steers (319 kg of BW) were used in a complete block design. Treatments included a soybean hull mix (95.7% soybean hulls, 3% molasses, 0.5% urea, 0.8% mineral mix; DM basis) fed alone (100:0) or with 10.4, 20.7, or 30.9% (DM basis) coarsely chopped alfalfa hay (90:10, 80:20, and 70:30, respectively) or alfalfa alone (0:100). Diets were fed once daily at 1.75% (DM basis) of BW. In some cases, orts were present, which caused DM, OM, and NDF intakes to decrease (linear, P < 0.05) as alfalfa was added to the diets. Digestibilities of DM, OM, and NDF decreased linearly (P < 0.05) as alfalfa was added to the diets, but quadratic responses (P < 0.05) indicated that positive associative effects occurred between soybean hulls and alfalfa. Liquid dilution rates increased (linear, P < 0.05) with alfalfa additions to the diets and also demonstrated positive associative effects between soybean hulls and alfalfa (quadratic, P < 0.05). Solid passage rates were similar for 100:0 and 0:100 but were increased (quadratically and cubically, P < 0.05) when combinations of soybean hulls and alfalfa were fed. In Exp. 2, in vitro NDF digestibilities were measured for soybean hulls, alfalfa, and a blend of 85% soybean hulls and 15% alfalfa, each with no N source or supplemented with casein or urea to ascertain the effects of protein from alfalfa on digestibility. Disappearances were increased (P < 0.05) by addition of urea or casein, but no interactions between substrate and N supplement were observed. Addition of 30% alfalfa to diets consisting primarily of soybean hulls led to positive associative effects on diet digestibility, but alfalfa additions led to increased liquid and solid passage rates, suggesting that the benefit was not a result of slower passage of soybean hulls from the rumen. 相似文献

Holstein steers were used in two 5 x 5 Latin square experiments to evaluate the sparing of methionine by alternative sources of methyl groups (betaine and choline). Steers were housed in metabolism crates and limit-fed a soybean hull-based diet high in rumen degradable protein. To increase energy supply, ruminal infusions of volatile fatty acids and abomasal infusions of glucose were provided. An amino acid mixture, limiting in methionine, was infused abomasally to ensure that nonsulfur amino acids did not limit protein synthesis. Treatments for Exp. 1 were abomasal infusion of 1) water, 2) 2 g/d L-methionine, 3) 1.7 g/d L-cysteine, 4) 1.6 g/d betaine, and 5) 1.7 g/d L-cysteine + 1.6 g/d betaine. Treatments for Exp. 2 were abomasal infusion of 1) water, 2) 2 g/d L-methionine, 3) 8 g/d betaine, 4) 16 g/d betaine, and 5) 8 g/d choline. In both experiments, nitrogen retention increased in response to methionine (P < 0.05), demonstrating a deficiency of sulfur amino acids. Responses to cysteine, betaine, and choline were all small and not significant. The lack of response to cysteine indicates that the response to methionine was not due to transsulfuration to cysteine or that cysteine supply did not alter the flux of methionine through transsulfuration. The lack of response to betaine suggests that the steers' needs for methyl groups were met by the dietary conditions or that betaine was relatively inefficient in increasing the remethylation of homocysteine to methionine and, thereby, reducing the synthesis of cysteine from homocysteine. Under our experimental conditions, responses to methionine were likely due to a correction of a deficiency of methionine per se rather than of methyl group donors. 相似文献

Six ruminally cannulated steers (345 +/- 20 kg initial BW) were used in a 6 x 6 Latin square to evaluate effects of diet and antibiotics on ruminal protein metabolism. Two diets and three antibiotic treatments were arranged factorially. One diet contained (DM basis) 72% dry-rolled corn, 12% soybean meal, 10% alfalfa hay, and 4% molasses (SBM), and the other contained 63% dry-rolled corn, 30% wet corn gluten feed, and 5% alfalfa hay (WCGF). Antibiotic treatments included control, virginiamycin (175 mg/d; VM), and monensin/tylosin (250 and 100 mg/d, respectively; MT). Steers were fed at 12-h intervals at a rate of 2.4% of empty BW daily. Each period included 18 d of adaptation and 3 d of ruminal fluid collections. Samples were collected at 0, 2, 4, 6, 8, and 10 h after the morning feeding on d 19 and 20. On d 21, rumens were dosed 2 h after the morning feeding with 350 g of solubilized casein to evaluate in vivo ruminal protease and deaminase activities. Ruminal fluid samples were collected 1, 2, 3, 4, and 6 h after the casein dose. On d 19 and 20, antibiotics had no effect on ruminal pH or concentrations of VFA, lactate, ammonia, ciliated protozoa, alpha-amino nitrogen (AAN), or peptide N, but VM reduced (P < 0.01) the concentration of isovalerate compared to MT and control. After casein dosing (d 21), peptide N concentration was unaffected by antibiotics, but AAN were higher (P < 0.01) for VM than MT and control. Relative to MT and control, VM reduced ruminal isovalerate (P = 0.05) and increased ruminal propionate (P < 0.01) on d 21. Ruminal pH was lower (P < 0.01) in steers fed SBM than in steers fed WCGF, but lactate concentrations were unaffected by diet. Steers fed SBM had higher (P < 0.05) ruminal concentrations of total VFA and propionate. Ammonia concentrations were lower before feeding and higher after feeding for steers fed WCGF (P < 0.01). Steers fed WCGF had higher counts of total ciliated protozoa than steers fed SBM (P < 0.05) due to greater Entodinium sp. (P < 0.05). Steers fed WCGF had higher (P < 0.01) ruminal AAN and peptide N concentrations than those fed SBM on d 19 and 20. After casein dosing, ruminal peptide N concentrations were similar, but AAN were lower (P < 0.01) for WCGF than SBM. Overall, VM appeared to depress ruminal deaminase activity, and MT had minimal effects on ruminal fermentation products. Objective To evaluate standing, percutaneous, ultrasound-guided, transthoracic liver biopsy in mares, and transabdominal, laparoscopically-guided, liver biopsy under general anaesthesia in foals, as techniques for obtaining tissue for assessment of copper status. The techniques were evaluated with respect to ease of use and effect on the animal.
Procedure Twenty of 24 Thoroughbred mares and 21 of their foals were biopsied. The animals were part of a larger study of the effect of copper supplementation on copper status and the prevalence of developmental orthopaedic disease. Livers were also collected from unrelated horses and sampled to investigate the variability in the distribution of copper in liver tissue.
Result The biopsy technique caused no lasting effect on the mares, but there was an increased risk of viscus penetration associated with taking multiple biopsy cores. The use of ultrasonography to scan the target area for the liver identified four cases that were not appropriate candidates for liver biopsy, because of large intestine being located in the biopsy area. In the foals there were no serious postoperative adverse effects, nor was there any evidence of problems caused by the procedure when the abdomen was examined post-mortem at 5 months of age. In livers collected to investigate the variability of copper concentration, copper appeared to be relatively evenly distributed through the liver.
Conclusion Standing, percutaneous, ultrasound-guided, transthoracic liver biopsy in mares, and transabdominal, laparoscopically-guided, liver biopsy under general anaesthesia in foals are convenient procedures for obtaining liver tissue for assessing copper status in horses. Eight ruminally cannulated steers (294 kg, ADG = 1.3 kg/d) were used in a N retention study (8 x 8 latin-square design) to evaluate sulfur-containing (S) amino acid (AA) requirements for growth. Treatments were abomasal infusions of seven levels of L-methionine (0, 3, 6, 9, 12, 15 and 18 g/d) and one level of DL-methionine (6 g/d). All steers were fed a semipurified diet based on ammoniated corn cobs (DMI = 6.56 kg/d) and were abomasally infused with 400 g/d dextrose and 296.4 g/d of crystalline AA that simulated the non-S-AA pattern of casein. Infusion of 3 g/d supplemental L-methionine maximized N retention in steers. Intestinal flows of absorbable S-AA were determined to be 1.89 g/kg DMI. Breakpoint analysis of retained N as a function of total absorbable S-AA yielded a total S-AA requirement of 14.7 g/d. Nitrogen retention for DL-methionine (36.4 g/d) was not different (P greater than .05) from that for 6 g/d L-methionine (38.8 g/d), but because this value was not in the linear response range, the efficacy of DL-methionine in meeting S-AA needs could not be evaluated. Plasma methionine concentrations increased linearly (P less than .05) in response to L-methionine infusion and were greater (P less than .05) for steers infused with 6 g/d DL-methionine (45.3 microM) than for steers receiving 6 g/d L-methionine (30.5 microM). An experiment was conducted to develop a system useful for measuring methionine requirements of growing steers. Seven ruminally cannulated steers (312 kg, gaining .91 kg/d) were fed a diet based on ammoniated corn cobs, corn starch, molasses and urea. Quantities of N and sulfur-containing amino acids disappearing from the small intestine were 96.0 and 14.8 g/d, respectively. Postruminal infusions of Na-caseinate (CAS) resulted in linear (P less than .01) increases in N retention with values increasing from 30.1 g/d with no postruminal CAS infusion to 39.3, 50.8 and 59.2 g/d (averaged across methionine supplementation) when 100, 200 and 300 g/d CAS were infused. Postruminal infusions of a mixture of crystalline L-amino acids (simulating the nonsulfur-containing essential amino acid pattern of casein; SIM) at levels of 100, 200 and 300 g/d also led to linear increases (P less than .01) in N retention with steers retaining 30.9, 38.9 and 50.5 g N/d (averaged across methionine supplementation), respectively. Postruminal infusion of 12 g/d L-methionine across CAS and SIM infusions improved (P less than .01) N retention by 7.6 g/d but infusion of SIM, which is devoid of sulfur amino acids, also increased N retention. Responses to methionine supplementation was greatest when 200 or 300 g/d SIM were abomasally infused. The data are interpreted to demonstrate that, for steers fed a diet containing little true protein, postruminal supplementation with nonsulfur-containing amino acids tended to increase the ability of growing steers to respond to methionine supplementation. 相似文献

The objective of this experiment was to determine whether alkaline hydrogen peroxide-treated oat hulls (termed oat fiber; OF) are nutritionally efficacious as a source of dietary fiber in meat-based dog foods. Thirty female English Pointers were assigned in a completely randomized design to isonitrogenous diets. Treatments were 1) control diet, 2) 7.5% added beet pulp (BP), and 3) 2.5, 4) 5.0, and 5) 7.5% added OF. Inclusion of 7.5% BP increased (P less than .05) DM intake and decreased (P less than .05) digestibility of DM and OM compared with the control. Dry matter intake increased (P less than .05) with increasing level of OF and digestibility of DM, OM, and total dietary fiber (TDF) decreased (P less than .05). Digestibility of DM, OM, and TDF were higher for dogs fed the 7.5% BP than for those fed the 7.5% OF treatment. Digestible energy, expressed as a percentage of GE, was greater for the control treatment than for the 7.5% BP treatment. A linear decrease in DE (percentage of GE) was noted as the concentration of OF increased, and the DE value (percentage of GE) for the 7.5% BP treatment was greater (P less than .05) than that for the 7.5% OF treatment. A linear decrease (P less than .05) was noted in ME, expressed as a percentage of GE, as the level of OF increased. The value of soybean meal (SBM), corn gluten meal (CGM), blood meal (BM) and fish meal (FM) in supplying N and amino acids (AA) escaping ruminal microbial degradation and disappearing from the small intestine (SI) was studied in steers using a regression approach. Replacement of corn starch in diets with protein sources resulted in decreases (P less than .05) in efficiency of microbial protein synthesis. Ruminal ammonia-N (NH3-N) had the greatest increase (P less than .05) when SBM was fed; BM supplementation resulted in only nonsignificant increases in ruminal NH3-N (P greater than .05). Soybean meal had the lowest proportion of N escaping ruminal degradation (.21). Corn gluten meal-N (.86) and BM-N (.92) escaped ruminal degradation to the greatest extent, and FM-N was intermediate (.68). Protein sources followed similar trends in providing absorbable nonbacterial N to the SI. Thirteen (+/- 6.2) percent of SBM-N was absorbed from the SI; 69 (+/- 6.2), 68 (+/- 9.1) and 50 (+/- 10.1)% of CGM-N, BM-N and FM-N, respectively, were absorbed from the SI. Values for ruminal escape and SI availability for individual and total AA are presented. Of the essential AA (EAA), threonine, valine and isoleucine were more resistant to ruminal degradation; methionine, cysteine, histidine and arginine were more extensively degraded than the total AA supply. Of the EAA escaping ruminal degradation, cysteine, histidine and threonine tended to be less digestible, whereas arginine was more digestible in the SI than the total AA supply. 相似文献
