Chondrocytes dedifferentiate to a fibroblast‐like phenotype on plastic surfaces. Dedifferentiation is reversible if these cells are then cultured embedded in gels as alginate, agarose or collagen. Chondrocytes cultured in suspension on a non‐adherent surface are also known to form aggregates of differentiated cells. The knowledge of chondrocyte behavior in culture is relevant for tissue engineering purposes. In this report we describe a simple method to culture differentiated or redifferentiated rabbit auricular chondrocytes on plastic surfaces with a stable phenotype. When chondrocyte aggregates formed in suspension are next seeded on plastic surfaces, most of them attach to the plastic as round or polygonal cells, and this morphological differentiation, confirmed by the presence of type II collagen, is stable for long culture periods. We also report that the addition of aggregates to monolayer cultures of dedifferentiated chondrocytes results in their redifferentiation, as is shown by their morphological changes and the synthesis of type II collagen. Therefore, this simple method can be useful for the study of chondrocyte behavior on plastic surfaces and for redifferentiating previously proliferated chondrocytes in tissue engineering techniques. Furthermore, these results demonstrate that, in addition to culture conditions such as cell isolation method or cell‐density, chondrocyte behavior on plastic depends on the presence or absence of aggregates resulting from the dissociation process.  相似文献

Effects of adding different concentrations of melatonin (10?7, 10?9 and 10?11 M) to maturation (Experiment 1; Control, IVM  + 10?7, IVM  + 10?9, IVM  + 10?11) and culture media (Experiment 2; Control, IVC  + 10?7, IVC  + 10?9, IVC  + 10?11) were evaluated on in vitro bovine embryonic development. The optimal concentration of melatonin (10?9 M) from Experiments 1–2 was tested in both maturation and/or culture media of Experiment 3 (Control, IVM  + 10?9, IVC  + 10?9, IVM /IVC  + 10?9). In Experiment 1, maturated oocytes from Control and IVM  + 10?9 treatments showed increased glutathione content, mitochondrial membrane potential and percentage of Grade I blastocysts (40.6% and 43%, respectively). In Experiment 2, an increase in the percentage of Grade I blastocysts was detected in IVC  + 10?7 (43.5%; 56.7%) and IVC  + 10?9 (47.4%; 57.4%). Moreover, a lower number and percentage of apoptotic cells in blastocysts were observed in the IVC  + 10?9 group compared to Control (3.8 ± 0.6; 3.6% versus 6.1 ± 0.6; 5.3%). In Experiment 3, the IVC  + 10?9 treatment increased percentage of Grade I blastocysts with a lower number of apoptotic cells compared to IVM /IVC  + 10?9 group (52.6%; 3.0 ± 0.5 versus 46.0%; 5.4 ± 1.0). The IVC  + 10?9 treatment also had a higher mRNA expression of antioxidant gene (SOD 2) compared to the Control, as well as the heat shock protein (HSPB 1) compared to the IVM  + 10?9. Reactive oxygen species production was greater in the IVM /IVC  + 10?9 treatment group. In conclusion, the 10?9 M concentration of melatonin and the in vitro production phase in which it is used directly affected embryonic development and quality.  相似文献

Objective To evaluate standing, percutaneous, ultrasound-guided, transthoracic liver biopsy in mares, and transabdominal, laparoscopically-guided, liver biopsy under general anaesthesia in foals, as techniques for obtaining tissue for assessment of copper status. The techniques were evaluated with respect to ease of use and effect on the animal.
Procedure Twenty of 24 Thoroughbred mares and 21 of their foals were biopsied. The animals were part of a larger study of the effect of copper supplementation on copper status and the prevalence of developmental orthopaedic disease. Livers were also collected from unrelated horses and sampled to investigate the variability in the distribution of copper in liver tissue.
Result The biopsy technique caused no lasting effect on the mares, but there was an increased risk of viscus penetration associated with taking multiple biopsy cores. The use of ultrasonography to scan the target area for the liver identified four cases that were not appropriate candidates for liver biopsy, because of large intestine being located in the biopsy area. In the foals there were no serious postoperative adverse effects, nor was there any evidence of problems caused by the procedure when the abdomen was examined post-mortem at 5 months of age. In livers collected to investigate the variability of copper concentration, copper appeared to be relatively evenly distributed through the liver.
Conclusion Standing, percutaneous, ultrasound-guided, transthoracic liver biopsy in mares, and transabdominal, laparoscopically-guided, liver biopsy under general anaesthesia in foals are convenient procedures for obtaining liver tissue for assessing copper status in horses. The use of ultrasound to identify liver tissue is recommended, especially in older mares.  相似文献

Kenaf (Hibiscus cannabinus L.) is an annual plant which produces a very high fiber yield per hectare and has demonstrated to be a suitable material for pulping. The crop conditions influence fiber production, but there is no information about their effect on pulp quality. The aim of this work was to study the effect of kenaf varieties and crop conditions (watering dose and harvesting time) on sulfate pulp quality. The study has been made on bast fiber sulfate pulps due to their high added value. The quality criteria were fiber dimensions, chemical composition and pulp quality. Kenaf variety or crop conditions have significant effect on fiber dimensions, holocellulose content, kappa number and breaking strength. To obtain high fiber yield and good delignified pulps it is recommended to grow the Salvador variety and to employ advance harvesting. Received: 20 January 1999  相似文献

Diverse actinomycetes produce a family of structurally and biosynthetically related non-ribosomal peptide compounds which belong to the chromodepsipeptide family. These compounds act as bisintercalators into the DNA helix. They give rise to antitumor, antiparasitic, antibacterial and antiviral bioactivities. These compounds show a high degree of conserved modularity (chromophores, number and type of amino acids). This modularity and their high sequence similarities at the genetic level imply a common biosynthetic origin for these pathways. Here, we describe insights about rules governing this modular biosynthesis, taking advantage of the fact that nowadays five of these gene clusters have been made public (thiocoraline, triostin, SW-163 and echinomycin/quinomycin). This modularity has potential application for designing and producing novel genetic engineered derivatives, as well as for developing new chemical synthesis strategies. Annual (Pisum sativum L. and Vicia sativa L.) and perennial (Trifolium repens L. and Lotus corniculatus L.) leguminous species were grown in pots containing samples from the ash layers of two Cambisols under Pinus sylvestris L., which has been affected by high-intensity wildfires 3 and 15 days before the sampling. The gramineous Lolium perenne L. was cultivated as a second plant after Trifolium and Lotus harvesting. Three treatments were compared: soils without fertilization and soils fertilized with two doses of poultry manure (1 and 2 g total N kg-1 dry soil). The aim of the work was to study the capacity of the ash layer to sustain vegetation and the influence of plants and organic manure on the recovery of vegetation cover, ash layer fixation and soil structure formation to avoid erosion. The ash samples were able to sustain vegetation without fertilization. The organic manure increased the yields of all the plants tested, the lower dose being the optimal for the first crop whereas the higher dose was beneficial for the second crop. The annual legumes grew very quickly. The mixture of Trifolium and Lotus seemed very suitable for reclamation of soil degraded by wildfires because Trifolium produced more phytomass than Lotus in the first growing stages whereas the development of Lotus was higher in the later growing stages. Ash layer conditions did not inhibit nodulation, which was, however, stimulated by the organic manure, particularly in the case of Lotus. Lolium after perennial legumes was the best plant combination because it produced the highest phytomass, particularly root phytomass, and thus improved vegetation cover and ash layer fixation. All the plants tested improved the formation of soil aggregates, particularly the combination of perennial legumes and Lolium. However, wet aggregate stability was higher when plants were grown on soils fertilized with poultry manure than when plants were cropped on unmanured soils, which points to the favourable influence of the organic manure on soil aggregation.  相似文献
