Fifteen experiments testing seven sets of genotypes under irrigated non-stress and drought stress situations were conducted from 2004 to 2006 with the objective to study the effect of different severity levels of drought on tolerant and susceptible advanced breeding lines, current varieties, and traditional and improved donors, and to study the effect of selection for high early vigor on yield. With the onset of drought, a yield reduction due to a decrease in biomass was observed in both susceptible and tolerant lines. However, after an initial reduction in yield, tolerant lines were able to sustain a yield loss more effectively than susceptible lines by better maintenance of biomass and higher harvest index. Under intermittent drought stress, genotypes with the ability to maintain higher biomass, reflected in terms of high early vigor recorded 3 wk after transplanting, recorded higher grain yield than genotypes with low early vigor. Lines with high early vigor yielded higher under both irrigated non-stress and intermittent drought stress situations. Under intermittent stress, plants with high vigor, low vigor, or random plants with high or low vigor coming from tolerant × susceptible crosses yielded higher than did the plants with the same vigor coming from susceptible × susceptible crosses. Under both irrigated non-stress and intermittent drought stress situations, early vigor showed significant genetic correlation with grain yield and could be included as an efficient selection criterion to improve yield in the lowlands. 相似文献

A 10-year-old neutered male Airedale Terrier was evaluated for inappetance, weight loss, and lameness. Multiple myeloma was diagnosed based on bone marrow plasmacytosis, multiple lytic bone lesions, and hyperglobulinemia with a clonal gammopathy on serum protein electrophoresis. Splenic plasmacytosis, and retinal lesions consistent with hyperviscosity syndrome also were found. Temporary responses to 2 different chemotherapy protocols (melphalan and prednisone, and cyclophosphamide and prednisone) were seen, with remission of clinical signs and a decrease in the biclonal gammopathy but no resolution of the splenic mass. Eventual return of clinical signs led to euthanasia at 175 days postdiagnosis. Necropsy examination confirmed multiple myeloma involving bone marrow and spleen, and glomerulonephritis. An immunoglobulin-A (IgA) gammopathy was demonstrated by immunoelectrophoresis; biclonality was ascertained by immunofixation electrophoresis. The clonal components consisted of intact Ig with a heavy chain of the a class and a light chain of undetermined class. To our knowledge, this is the first report of undimerized biclonal gammopathy in a dog caused by a single heavy chain class involving IgA. 相似文献

A large-effect QTL for grain yield under drought conditions (qtl12.1) was reported in a rice mapping population derived from Vandana and Way Rarem. Here, we measured the effect of qtl12.1 on grain yield and associated traits in 21 field trials: ten at IRRI in the Philippines and 11 in the target environment
of eastern India. The relative effect of the QTL on grain yield increased with increasing intensity of drought stress, from
having no effect under well-watered conditions to having an additive effect of more than 40% of the trial mean in the most
severe stress treatments. The QTL improved grain yield in nine out of ten direct-seeded upland trials where drought stress
was severe or moderate, but no effect was measured under well-watered aerobic conditions or under transplanted lowland conditions.
These trials confirm that qtl12.1 has a large and consistent effect on grain yield under upland drought stress conditions, in a wide range of environments. 相似文献

The purpose of this paper by the Regulatory Affairs Committee (RAC) of the American Society for Veterinary Clinical Pathology (ASVCP) is to review the current regulatory guidances (eg, guidelines) and published recommendations for best practices in veterinary toxicologic clinical pathology, particularly in the pharmaceutical and biotechnology industries, and to utilize the combined experience of ASVCP RAC to provide updated recommendations. Discussion points include (1) instrumentation, validation, and sample collection, (2) routine laboratory variables, (3) cytologic laboratory variables, (4) data interpretation and reporting (including peer review, reference intervals and statistics), and (5) roles and responsibilities of clinical pathologists and laboratory personnel. Revision and improvement of current practices should be in alignment with evolving regulatory guidance documents, new technology, and expanding understanding and utility of clinical pathology. These recommendations provide a contemporary guide for the refinement of veterinary toxicologic clinical pathology best practices. 相似文献

In preclinical safety assessment, blood volume requirements for various endpoints pose a major challenge. The goal of this working group was to review current practices for clinical pathology (CP) testing in preclinical toxicologic studies, and to discuss advantages and disadvantages of methods for reducing blood volume requirements. An industry‐wide survey was conducted to gather information on CP instrumentation and blood collection practices for hematology, clinical biochemistry, and coagulation evaluation in laboratory animals involved in preclinical studies. Based on the survey results and collective experience of the authors, the working group proposes the following “points to consider” for CP testing: (1) For most commercial analyzers, 0.5 mL and 0.8 mL of whole blood are sufficient for hematology and biochemistry evaluation, respectively. (2) Small analyzers with low volume requirements and low throughput have limited utility in preclinical studies. (3) Sample pooling or dilution is inappropriate for many CP methods. (4) Appropriate collection sites should be determined based on blood volume requirements and technical expertise. (5) Microsampling does not provide sufficient volume given current analyzer and quality assurance requirements. (6) Study design considerations include: the use of older/larger animals (rodents), collection of CP samples before toxicokinetic samples, use of separate subsets of mice for hematology and clinical biochemistry testing, use of a priority list for clinical biochemistry, and when possible, eliminating coagulation testing. 相似文献

An investigation was carried out to assess the efficiency of A2 cytoplasmic-nuclear male-sterility (CMS) system in comparison to the widely used A1 cytoplasm in terms of general combining ability (gca) effects of male-sterile (A-) lines and mean performance, specific combining ability (sca) effects and mid-parent heterosis of hybrids for days to 50% flowering, plant height and grain yield at International Crops
Research Institute for the Semi-Arid Tropics, Patancheru, Andhra Pradesh, India in 2001 and 2002 rainy seasons. The material
for the study consisted of six pairs of iso-nuclear, allo-plasmic (A1 and A2) A-lines and 36 iso-nuclear hybrids produced by crossing these A-lines with three dual restorer (R-) lines. The results revealed
that cytoplasm and its first-order interaction with year, R- and A-lines did not appear to contribute to variation in iso-nuclear
hybrids for plant height and grain yield. Cytoplasm had limited effect on gca effects of A-lines and on sca effects and mid-parent heterosis of iso-nuclear hybrids for days to 50% flowering, plant height and grain yield. The mean
days to 50% flowering, plant height and grain yield of A2 cytoplasm-based hybrids were comparable with those of widely used A1 cytoplasm-based hybrids. The relative frequency of the occurrence of the A1- and A2-based hybrids with significant sca effects and mid-parent heterosis indicated that A2 CMS system is as efficient as A1 with a slight edge over A1 for commercial exploitation. The implications of these results are discussed in relation to opportunities for broadening
