Aequipecten opercularis (Queen scallop) and Mimachlamys (Chlamys) varia (Black scallop) are important natural resources occurring in Atlantic and Mediterranean coasts. To develop an optimal sustainable exploitation plan, it is important to study the genetic structure of the different populations. In this study, we used polymerase chain reaction‐restriction fragment length polymorphisms for the determination of the genetic variation and population structure of these two species in different localities. Ten composite haplotypes were generated for A. opercularis and 15 haplotypes for M. varia. Of these, six and four were unique respectively. The analysis of the distribution of the different haplotypes between the localities showed no clear evidence of subdivision in A. opercularis, while in M. varia the results indicated that the two localities analysed should be managed as separate stocks.  相似文献

Response of western flower thrips, Frankliniella occidentalis (Pergande), to selection for resistance to insecticides commonly used to control this pest in Murcia (south-east Spain) was studied under field and laboratory conditions. In the field, plots within sweet pepper crops in commercial and experimental greenhouses were treated under different selection strategies: insecticide rotation versus formetanate reiteration, formetanate reiteration versus acrinathrin reiteration, and formetanate reiteration versus methiocarb reiteration. Thrips populations were sampled monthly and bioassayed against methiocarb, methamidophos, acrinathrin, endosulfan, deltamethrin and formetanate. In the laboratory, F occidentalis strains were selected against each insecticide for several generations. To evaluate cross-resistance, each selected strain was bioassayed with the other insecticides. Frankliniella occidentalis populations showed a rapid development of acrinathrin resistance, reaching high levels in field and laboratory conditions. Formetanate and methiocarb resistance were also observed, although development was slower and at moderate levels. Cross-resistances between acrinathrin/deltamethrin and acrinathrin/formetanate were detected under field and laboratory conditions. Formetanate/methiocarb cross-resistance was suspected in laboratory selections, but not in field assays. Simultaneous moderate resistance levels to the three specific insecticides against thrips (formetanate, methiocarb and acrinathrin) were shown in laboratory selection strains, indicating a general mechanism of resistance, probably metabolic.  相似文献

Beta-lactoglobulin (beta-Lg) was subjected to high pressures up to 400 MPa and proteolysis with chymotrypsin. The hydrolysates were analyzed by SDS-PAGE and RP-HPLC, and the fragments obtained were identified by ESI-MS/MS. The results obtained showed that beta-Lg was hydrolyzed by chymotrypsin in a "progressive proteolysis" manner at either atmospheric or high pressure. Proteolysis during or after high-pressure treatment showed longer and more hydrophobic peptides than proteolysis at atmospheric pressure. Chymotrypsin showed a behavior similar to that of trypsin, with some differences, probably related to the orientation of the target residues specific for each enzyme. The similarities between proteolytic fragments produced by the two enzymes support that proteolysis enhancement under high pressure depends on the substrate changes rather than the enzyme. High pressure seemed to accelerate the first steps of proteolysis, probably through dimer dissociation, while leaving portions of the molecule more resistant to the enzyme.  相似文献

The effect of tillage on soil properties varies within field due to spatial variability of soils. Mapping changes of soil penetration resistance (PR) would be useful to understand and assess tillage practices to alleviate soil compaction. The objectives were to model the short-term effect of tillage on PR and its spatial structure, and to delineate homogeneous zones based on soil response in a Typic Argiudoll previously managed under no-till. A grid sampling for PR and soil water content (SWC) were performed before and after chiselling. Spatial analysis was performed on the effect of tillage on PR data by 10 cm layers and homogeneous zones were delineated by k-means cluster analysis. The effect of tillage was ?0.33 MPa in 10–20 and 20–30 cm layer. No differences of PR were found at 0–10 cm. Short range (5–7 m) spatial structure on the horizontal plane was observed in all layers. Only 45% of the field showed a marked effect of tillage on PR. Mapping the effect of tillage on PR would be a useful approach for evaluating the global and local response of soil to tillage, as well as for delineating of areas within field for site-specific tillage practices.  相似文献

Context

Amphibians are declining worldwide and land use change to agriculture is recognized as a leading cause. Argentina is undergoing an agriculturalization process with rapid changes in landscape structure.

Objectives

We evaluated anuran response to landscape composition and configuration in two landscapes of east-central Argentina with different degrees of agriculturalization. We identified sensitive species and evaluated landscape influence on communities and individual species at two spatial scales.

Methods

We compared anuran richness, frequency of occurrence, and activity between landscapes using call surveys data from 120 sampling points from 2007 to 2009. We evaluated anuran responses to landscape structure variables estimated within 250 and 500-m radius buffers using canonical correspondence analysis and multimodel inference from a set of candidate models.

Results

Anuran richness was lower in the landscape with greater level of agriculturalization with reduced amount of forest cover and stream length. This pattern was driven by the lower occurrence and calling activity of seven out of the sixteen recorded species. Four species responded positively to the amount of forest cover and stream habitat. Three species responded positively to forest cohesion and negatively to rural housing. Two responded negatively to crop area and diversity of cover classes.

Conclusions

Anurans within agricultural landscapes of east-central Argentina are responding to landscape structure. Responses varied depending on species and study scale. Life-history traits contribute to responses differences. Our study offers a better understanding of landscape effects on anurans and can be used for land management in other areas experiencing a similar agriculturalization process.

Soil available water capacity (AWC) is an important factor affecting soil productivity in semiarid and subhumid environments and is mainly determined by the soil textural composition. As the soils of these environments usually present fairly uniform textures across depth, we hypothesized that it would be possible to accurately estimate the whole-profile AWC using surface information. Our objective was to test this hypothesis in the Argentine Semiarid Pampas. Information was collected from 152 sites where AWC was measured in 20?cm layers up to a depth of 140?cm or up to the upper limit of the petrocalcic horizon, when present. In each case, whole profile AWC was estimated using a one-step and a two-step approach, comparing multiple regression and artificial neural networks as modeling techniques. Recently, anti-Leishmania IgG has been detected in urine samples from Leishmania-infected dogs and its concentrations have been correlated with impairment of renal function. The presence and relationship with other anti-Leishmania Ig isotypes in urine have not yet been investigated. The current study analyzed the concentrations of anti-Leishmania IgA and IgG in sera (Ig-S) and urine (Ig-U) samples by ELISA in 64 untreated dogs with clinical leishmaniasis. All 64 serum samples tested were positive for anti-Leishmania IgG. Fifty of them (78.1%) were also positive for anti-Leishmania IgA. The results showed the presence of anti-Leishmania IgA-U in 38% of the 50 dogs that were positive for specific IgA-S. Thirty-eight of the 64 dogs positive for Leishmania-specific IgG-S (59.4%) were also positive for Leishmania-specific IgG in urine (IgG-U). The concentrations of anti-Leishmania IgA-U were significantly correlated with urine protein/creatinine (uP/C) ratio (ρ = 0.542; P < 0.001) and with serum biochemical parameters, such as γ-globulins, urea and creatinine. Goldmann–Witmer coefficient (C value) indicated that detection of specific IgA in urine samples from dogs with leishmaniasis might not only be due to impairment of filtration of the glomerular barrier but also be due to local production of this isotype, which might reflect a local immunological response to the presence of the parasite in the genitourinary tract. Anti-Leishmania IgG-U concentrations were highly correlated with uP/C ratio (ρ = 0.779; P < 0.001) and C value did not support in any case local production of this isotype. IgG isotype might be a more suitable and specific tool to evaluate renal damage due to the lower IgA-U sensitivity and correlation coefficients and evidence of IgA local production. However, dogs found positive for both Ig isotypes in urine presented significantly higher specific IgG-U concentrations and higher uP/C ratios than dogs found positive only for IgG-U, thus suggesting that the first group suffered more severe renal damage. This fact makes it necessary to evaluate the prognosis of dogs showing both anti-Leishmania IgA-U and IgG-U in future studies.  相似文献

A germplasm collection of 2759 pea accessions was screened for resistance against Uromyces pisi (Pers.) Wint. All accessions displayed a compatible interaction (high infection type) both in adult plants under field conditions and in seedlings under growth chamber conditions, but with varying levels of disease reduction. The identified resistance was based on reduction of disease severity with no associated host cell necrosis, which fits the definition of Partial Resistance. No complete resistance or incomplete resistance based on hypersensitivity was observed.  相似文献

The unfolding of beta-lactoglobulin during high-pressure treatment and its refolding after decompression were studied by 1H NMR and 2H/1H exchange at pH 6.8 and 2.5 and at 37 and 25 degrees C. The extent of unfolding increased with the pressure level. The structure of beta-lactoglobulin required higher pressures to unfold at pH 2.5 than at pH 6.8. More flexibility was achieved at 37 degrees C than at 25 degrees C. Results indicated that the structural region formed by strands F, G, and H was more resistant to unfold under acidic and neutral conditions. The exposure of Trp19 at an earlier time, as compared to other protein regions, supports the formation of a swollen structural state at pH 2.5. Refolding was achieved faster when beta-lactoglobulin was subjected to 200 MPa than to 400 MPa, to 37 degrees C than to 25 degrees C, and to acidic than to neutral pH. After treatment at 400 MPa for 20 min at neutral pH, the protein native structure was not recovered. All samples at acidic pH showed that the protein quickly regained its structure. Hydrolysis of beta-lactoglobulin by pepsin and chymotrypsin could be related to pressure-induced changes in the structure of the protein. Compared to the behavior of the protein at atmospheric pressure, no increased proteolysis was found in samples with no increased flexibility (100 MPa, 37 degrees C, pH 2.5). Slightly flexible structures were associated with significantly increased proteolysis (100 MPa, 37 degrees C, pH 6.8; 200 MPa, 37 degrees C, pH 2.5). Highly flexible structures were associated with very fast proteolysis (>or=200 MPa, 37 degrees C, pH 6.8; >or=300 MPa, 37 degrees C, pH 2.5). Susceptibility to spinosad of western flower thrips (WFT), Frankliniella occidentalis (Pergande), from south-eastern Spain was determined. LC(50) values of the field populations without previous exposure to spinosad collected in Murcia in 2001 and 2002 ranged from 0.005 to 0.077 mg L(-1). The populations collected in Almeria in 2003 in greenhouses were resistant to spinosad (LC(50) > 54 mg L(-1)) compared with the authors' highly susceptible laboratory strain. The highly sensitive laboratory strain leads to very high resistance ratios for the field populations (>13 500), but these ratios do not necessarily mean resistance problems and control failures (spinosad field rate 90-120 mg L(-1)). The populations collected in Murcia from some greenhouses in 2004 were also resistant to spinosad (RF > 3682). Spinosad overuse, with more than ten applications per crop, produced these resistant populations in some greenhouses. Spinosad showed no cross-resistance to acrinathrin, formetanate or methiocarb in laboratory strains selected for resistance towards each insecticide. Correlation analysis indicated no cross-resistance among spinosad and the other three insecticides in 13 field populations and in nine laboratory strains. The synergists piperonyl butoxide (PBO), S,S,S-tributyl phosphorotrithioate (DEF) and diethyl maleate (DEM) did not enhance the toxicity of spinosad to the resistant strains, indicating that metabolic-mediated detoxification was not responsible for the spinosad resistance. These findings suggest that rotation with spinosad may be an effective resistance management strategy.  相似文献
