The in vivo and in vitro mechanistic anti-inflammatory actions of cucurbitacin E (CE) (Citrullus lanatus var. citroides) were examined. The results showed that LPS/INF-γ increased NO production in RAW264.7 macrophages, whereas L-NAME and CE curtailed it. CE did not reveal any cytotoxicity on RAW264.7 and WRL-68 cells. CE inhibited both COX enzymes with more selectivity toward COX-2. Intraperitoneal injection of CE significantly suppressed carrageenan-induced rat's paw edema. ORAC and FRAP assays showed that CE is not a potent ROS scavenger. It could be concluded that CE is potentially useful in treating inflammation through the inhibition of COX and RNS but not ROS.  相似文献

A simplex centroid mixture design was used to study the interactions between two chosen solvents, dichloromethane (DCM) and acetone (ACT), as organic-phase components in the formation and physicochemical characterization and cellular uptake of astaxanthin nanodispersions produced using precipitation and condensation processes. Full cubic or quadratic regression models with acceptable determination coefficients were obtained for all of the studied responses. Multiple-response optimization predicted that the organic phase with 38% (w/w) DCM and 62% (w/w) ACT yielded astaxanthin nanodispersions with the minimum particle size (106 nm), polydispersity index (0.191), and total astaxanthin loss (12.7%, w/w) and the maximum cellular uptake (2981 fmol/cell). Astaxanthin cellular uptake from the produced nanodispersions also showed a good correlation with their particle size distributions and astaxanthin trans/cis isomerization ratios. The absence of significant (p > 0.05) differences between the experimental and predicted values of the response variables confirmed the adequacy of the fitted models.  相似文献

 以菠萝(Ananas comosus L. Merrill) 杂交种MD1 无菌苗为材料, 通过培养薄细胞层切片 (Thin cell layer, TCL) 的方法, 研究直接再生胚的固体培养体系。结果证明, 预培养的设计2 (在预培养基PT上黑暗预培养无菌苗1周后, 从预培养苗茎顶端切取TCL, 在MS + 2, 4-D 2 mg·L - 1或1 mg·L - 1胚诱导培养基EI2上黑暗培养1周, 然后转到光照条件下继续诱导胚1周, 再在成熟胚培养基EM上培养2周, 最后转入幼苗培养基PL上培养) 是直接诱导获得体胚的最佳途径; 诱导的体胚最初均来自于薄细胞层上微管束附近的细胞; picloram有利于芽的诱导, 2, 4-D有利于直接再生胚, 但如果球形胚形成后一直培养在较高浓度的2, 4-D培养基中, 将阻碍球形胚和心形胚进一步发育为成熟胚, 反而促进芽的生成。  相似文献

The toxicity of mercury for sequential stages of Macrobrachium rosenbergii (de Man) larvae was studied under static water conditions at a salinity of 12‰ and temperature of 25.5 – 27.0°C. The results revealed that stage 1 larvae had the lowest threshold lethal concentration (TLC) of mercury, 0.041 ppm Hg, while the post-larvae had a TLC of 0.325 ppm Hg. There was an abrupt increase in the TLC of mercury after the larvae had reached stage 5. The lower TLC prior to stage 5 is most probably due to the more frequent moulting and the relatively thinner cuticle of the larvae.  相似文献

Isolation of Neospora caninum from a calf in Malaysia   总被引:2,自引:0,他引:2  
In order to attempt isolate the protozoan parasite Neospora caninum, an N. caninum seropositive pregnant Sahiwal Friesian cross heifer from a large-scale dairy farm in Malaysia was kept for observation until parturition at the Veterinary Research Institute, Ipoh. The heifer gave birth to a female calf that was weak, underweight and unable to rise. Precolostral serum from the calf had an N. caninum indirect fluorescent antibody test titre of 1:3200. It died 12 h after birth and necropsy was performed. Brain homogenate from the calf was inoculated into 10 BALB/c mice that were kept for 3 months after which brain tissue from the mice was inoculated onto 24 h fresh monolayer Vero cell lines. The cell cultures were examined daily until growth of intracellular protozoa was observed. DNA of the organisms from the cell cultures was analyzed by PCR and DNA sequencing. DNA fragments of the expected size were amplified from the isolate using N. caninum-specific primers, and sequence analysis of ITS1 clearly identified the isolate as N. caninum. This is the first successful isolation of N. caninum from a bovine in Malaysia, and the isolate is designated Nc-MalB1.  相似文献

针对香蕉胚性悬浮细胞培养过程中存在的胚性愈伤组织的诱导率低、周期过长、胚性易丢失等问题, 以“苹果蕉”及“威廉斯”香蕉的未成熟雄花为试验材料, 对香蕉胚性愈伤组织的诱导、 悬浮培养的理化条件进行改进。 结果表明: (1)光照条件较暗培养更有利于诱导理想的香蕉胚性愈伤组织; (2)M1培养基中加入酪蛋白和脯氨酸有利于胚性愈伤组织的诱导; (3)用微孔板代替三角瓶悬浮培养, 有利于悬浮细胞分裂增殖, 而且可减少污染的可能。  相似文献
